The Near-Infrared Imaging Spectroscopy to Visualize the Distribution of Sugar Content in the Flesh of a Melon

  • To improve the accuracy of sweetness sensor in automated sorting operations, it is necessary to clarify unevenness of the sugar content distribution within fruits. And it is expected that the technique to evaluate the content distribution in fruits contribute to the development of the near-infrared (NIR) imaging spectroscopy. Sugiyama (1999) had succeeded to visualize the distribution of the sugar content on the surface of a half-cut green fresh melon. However, this method cannot be applied to red flesh melons because it depends on information of the absorption band of chlorophyll (676 nm), which is affected by the color of the fresh. The objective of this study was to develop the universal visualization method depends on the absorption band of sugar, which can be applied to various kinds of melons and other fruits. The relationship between the sugar contents and absorption spectra of both green and red fresh melons were investigated by using a NIR spectrometer to determine the absorption band of sugar. The combination of 2$\^$nd/ derivative absorbances at 902 nm and 874 nm was highly correlated with the sugar contents. The wavelength of 902 nm is attributed to the absorption band of sugar. A cooled charge-coupled device (CCD) imaging camera which has 16 bit (65536 steps) A/D resolution was equipped with rotating band-pass filter wheel and used to capture the spectral absorption images of the flesh of a vertically half-cut red fresh melon. The advantage of the high A/D resolution in this research is that each pixel of the CCD is expected to function as a detector of the NIR spectrometer for quantitative analysis. Images at 846 nm, 874 nm, 902 nm and 930 nm were acquired using this CCD camera. Then the 2$\^$nd/ derivative absorbances at 902 nm and 874 nm at each pixel were calculated using these four images. On the other hand, parts of the same melon were extracted for capturing the images and squeezed for the measurement of sugar content. Then the calibration curve between the combination of 2$\^$nd/ derivative absorbances at 902 nm and 874 nm and sugar content was developed. The calibration method based on NIR spectroscopy techniques was applied to each pixel of the images to convert the 2$\^$nd/ derivative absorbances into the Brix sugar content. Mapping the sugar content value of each pixel with linear color scale, the distribution of the sugar content was visualized. As a result of the visualization, it was quantitatively confirmed that the Brix sugar contents are low at the near of the skin and become higher towards the seeds. This result suggests that the visualization technique by the NIR imaging spectroscopy could become a new useful method fer quality evaluation of melons.

Optimization of Ingredient Mixing Ratio for Preparation of Chinese Radish (Raphanus sativus L.) Jam (무 잼 재료 혼합비율의 최적화)

  • This study was performed to find the optimum ratio of ingredients in the Chinese radish jam. The experiment was designed according to the RSM (response surface methodology), which included 18 experimental points with 4 replicates for three independent variables (sugar $45{\sim}70%$, pectin $0.5{\sim}2.0.%$, citric acid $0.2{\sim}0.5%$). The compositional and functional properties of the prepared products were measured, and these values were applied to the mathematical models. By use of F-test, sweetness, color values (L, a, b), and firmness were expressed by a linear model, while the sensory characteristics (color, smell, taste, texture and overall acceptance) were by a quadratic model. In the numeric optimization, the optimal ingredient amounts were 53.7% sugar, 1.0% pectin, and 0.3% citric acid. And in the graphical optimization, 53.9% sugar, 1.0% pectin, and 0.3% citric acid; these data were equivalent to 0.6985 desirability, indicating that the values were almost equivalent to the numerical optimization points. The above results demonstrate the feasibility of Chinese radish jam, and therefore, the commercialization of a Chinese radish jam marketed as a functional food is deemed possible.

Characteristics of the Fermentation products of Paecilomyces tenuipes Fermented Using Different Microorganisms (발효 미생물에 따른 누에동충하초 발효산물의 특성)

  • In order to develop fermented silkworm "Dongchunghacho" (Paecilomyces tenuipes) with improved absorption and increased effectiveness, we fermented Dongchunghacho using four kinds of microorganisms, viz., lactic acid bacteria, Bacillus subtilis, Natto bacillus, and yeast. A total of 15 samples were fermented using a combination of microbial inoculation culture and conditions to produce fermentation products. The contents of basic components such as sugar, reducing sugar, protein, total polyphenol, and total flavonoid were examined as well as the antioxidant, tyrosinase inhibitory, and thrombolytic activities of the fermented products were analyzed. We observed that reducing sugar and protein contents decreased in most of the fermented products, but the products fermented using yeast exhibited higher sugar content and, thus, higher sweetness. Total polyphenol content and antioxidant activity did not increase in fermented products compared to non-fermented Dongchunghachos, but total flavonoid content and tyrosinase inhibitory and thrombolytic activities increased by fermentation. In particular, total flavonoid content and tyrosinase inhibitory and thrombolytic activities primarily increased in the products fermented using yeast and lactic acid bacteria. However, it was not possible to confirm the increase in these activities in samples fermented by single fermentation using only yeast. Therefore, we propose that it will be possible to develop fermented food from silkworm Dongchunghacho (P. tenuipes) with excellent health benefits through additional study of multiple fermentation conditions using lactic acid bacteria and yeast.

Sugar content and expression of sugar metabolism-related gene in strawberry fruits from various cultivars

  • Strawberry (Fragaria ${\times}$ ananassa) is a globally-cultivated and popular fruit crop, prized for its flavor and nutritional value. Sweetness, a key determinant of fruit quality, depends on the sugar composition and concentration. We selected eight strawberry cultivars based on the fruit soluble solids content to represent high and low sugar content groups. The average soluble solid content was $13.6^{\circ}Brix$ (Okmae, Geumsil, Aram, and Maehyang) and $2.9^{\circ}Brix$ (Missionary, Camino Real, Portola, and Gilgyung53), for the high and low sugar content groups, respectively. Sucrose was the main sugar in the cultivars with high sugar content, whereas fructose was the main component in the low sugar content cultivars. Fruit starch concentration ranged from $3.247{\pm}0.056$ to $3.850{\pm}0.055g/100g$, with a 12% higher concentration in the high sugar content cultivars. Additionally, we identified 41 sugar metabolism-related genes in Fragaria ${\times}$ ananassa and analyzed the relationship between their transcripts and the sugar accumulation in fruit. FaGPT1, FaTMT1, FaHXK1, FaPHS1, FaINVA-3, and FacxINV2-1 were highly expressed in the high sugar content cultivars, while FapGlcT, FaTMT2-1, FaPHS2-1, FaSUSY1-1, and FaSUSY1-2 were highly expressed in the low sugar content cultivars. In general, a greater number of genes encoding sugar transporters or involved in sugar synthesis were highly expressed in the high sugar content cultivars. Contrarily, genes involved in sugar degradation were preferentially transcribed in the low sugar content cultivars. Although gene expression was not perfectly proportional to sugar content or concentration, our analysis of the genes involved in sugar metabolism and accumulation in strawberries provides a framework for further studies and for the subsequent engineering of sugar metabolism to enhance fruit quality.

A study on the correlation between the dental caries and dietary habits and snack intake of pre-school children (미취학 아동의 치아우식증과 식습관 및 간식섭취와의 관계)

  • Objectives : The purpose of this study was to correlation between the index of dental caries in primary teeth and the habits of diet and snack which are expected to influence on the index. The subjects of this study were 165 parents of the children who visited a pediatric dental hospital located in Gyeong-gi do from Jun. 15th to Sep. 26th, 2009, and the self-report questionnaire survey was performed. Methods : A questionnaire used in a previous study was revised and completed for this study and it consisted of 15 items on general characteristic, nine items on dietary habit, and five items on snack intake habit. The collected data was analyzed using SPSS program and findings are as follows Results : 1. The index of dental caries in primary teeth by ages was that children of four years old has 4~5 with 27.5%, children of five years old has 2~3 and 4~5 with 33.3% and children of six years old has 1 and 8~9 with 25.6%, showing a statistically significant(${\chi}^2$=37.72, p<.01). 2. The frequency of brushing teeth was found to have statistically significant negative correlation with the number of primary teeth with dental caries (${\chi}^2$=61.20, p<.001). 3. The frequency of taking snack was found to have statistically significant positive correlation with the number of primary teeth with dental caries (${\chi}^2$=51.42, p<.001). 4. The desirability of dietary habit was found to have statistically significant negative correlation with the number of primary teeth with dental caries (${\chi}^2$=36.51, p<.001). 5. The frequency of taking biscuit was found to have statistically significant positive correlation with the number of primary teeth with dental caries (${\chi}^2$=67.74, p<.001). 6. The frequency of taking bread was found to have statistically significant positive correlation with the number of primary teeth with dental caries (${\chi}^2$=29.63, p<.01). 7. The frequency of taking caramel and candy was found to have statistically significant positive correlation with the number of primary teeth with dental caries (${\chi}^2$=38.85, p<.001). 8. The frequency of taking soft drinks was found to have statistically significant positive correlation with the number of primary teeth with dental caries (${\chi}^2$=52.92, p<.001). 9. The frequency of taking ice-creams was found to have statistically significant positive correlation with the number of primary teeth with dental caries (${\chi}^2$=75.07, p<.001). Conclusions : Those findings show that the children with undesirable dietary habit and higher frequency of taking snack have higher index of dental caries in primary teeth, therefore, it is considered that the regular brushing teeth and establishment of desirable dietary habit are very important for keeping health oral cavity. It is recommended that the continuous concern and repeated learning are needed in house and group of rearing infants so that the children take foods of fruits which have function of self-purification than the food of carbohydrates which has high adhesion and level of sweetness.

The effect of a low-sodium label on acceptability and perceived saltiness intensity of a dipping sauce for fried pork cutlets (저염 표시가 포크커틀릿 소스의 짠맛 인식과 기호도에 미치는 영향)

  • This study was conducted to investigate the effects of a low-sodium label on acceptability and sensory attributes of a dipping sauce for fried pork cutlets. Two sauce samples, CON (normal sodium level) and LOW-Na (50% sodium level), were evaluated. Consumers (n=77) rated expectations and actual perception of overall liking, purchase intent, and intensities of saltiness, sweetness, sourness, fruit flavor, and degree of flavor balance without and with a carrier (fried pork cutlet) in blind and informed settings. In the informed test, CON labeled as Low-Na (PLACEBO) was additionally tested to examine the placebo effect of information. The low-sodium labeling significantly increased the expected liking and purchase intent, but decreased the expected saltiness. However, the label did not influence actual liking or purchase intent. A significant decrease in actual saltiness was observed only in Low-Na, not in PLACEBO, indicating the label is influential only when actual perception matched the expectation.

Quality characteristics of functional Nokdujuk prepared with optimum mixing ratio of mulberry leaf and fruit powder by response surface method (반응표면분석법을 이용한 최적 비율의 뽕잎과 오디 분말 첨가 기능성 녹두죽의 품질특성)

  • This study was performed to develop and evaluate functional prepared with optimum mixing of mulberry leaf and fruit powder using response surface method (RSM). In order to develop the optimized functional Nokdujukr using RSM, mulberry leaf powder (MLP:X1) and mulberry fruit powder (MLF:X2) were set as independent variables, and pH (Y1), sweetness (Y2), viscosity (Y3), L (Y4), a (Y5), b (Y6), color (Y7), flavor (Y8), taste (Y9), overall quality (Y10), TPC (Y11), and DPPH radical scavenging ability ($IC_{50}$)(Y12) were set as dependent variables. The optimum mixing ratio of MLP and MLF was determined to be 3.88 g of MLP and 6 g of MLF. The values of color, flavor, taste, overall quality, TPC, and DPPH radical scavenging ability ($IC_{50}$) of optimized Nokdujuk were 5.20, 5.85, 6.00, 6.22, 330.99 mg TAE/g and 650.10 g/mL, respectively. In conclusion, this study has led to the development of an improved version of Nokdujuk that has antioxidative properties and good sensory evaluation and, will likely serve as a functional meal replacement for the busy modern world.

Antimicrobial and Antioxidant activity of the Fruit of Prunus avium L (체리(Prunus avium L.)의 항균 및 항산화 활성)

  • The fruits of sweet cherry are highly appreciated by the consumer due to their precocity and quality, such as their sweetness, color and sourness. In this study, the hot-water extract and its sequential organic solvent fractions were prepared from domestic Napoleon sweet cherry (Prunus avium L.) to investigate antimicrobial and antioxidant activity. The hot-water extract contained about 40% sugars, and the solvent fraction yields for hexane, ethylacetate (EA), butanol, and water residue were 0.01%, 3.45%, 16.30%, and 80.24%, respectively. Contents of total polyphenol and total flavonoid of the fractions were 1.24~5.24%, and 0~3.76%, respectively. Among the fractions, EA fraction showed the highest total polyphenol and total flavonoid concentrations. Evaluation of antimicrobial activity of the extract and fractions revealed that EA fraction and butanol fraction contained strong antibacterial activity against Listeria monocytogenes, Staphylococcus aureus, and Salmonella typhimurium with minimal inhibitory concentration (MIC) of 0.5~1.0 mg/mL. But the extract and fractions tested were not active to Pseudomonas aeruginosa. In a while, only hexane fraction showed anti-Candida activity with 0.5~1.0 mg/mL of MIC. The fraction showed strong activity against different multi-antibiotics resistant strains such as C. albicans CCARM 14020. Antioxidative activity assay showed that EA fraction has a strong DPPH scavenging activity and a reducing power. The $IC_{50}s$ of vitamin E and EA fraction were 15.5 and $195.5\;{\mu}g/mL$, respectively. Our results suggest that the fruit of P. avium L. has high potentials with anti-Candida and antioxidative activity.

Effects of ${\beta}-amylase$ and Transglucosidase on the Qualities of Red Ginseng Extract (${\beta}-amylase$와 transglucosidase의 처리가 홍삼 extract의 품질에 미치는 영향)

  • In order to evalulate the qualities of red ginseng extract and decrease precipitate fonnation in ginseng drink, red ginseng extract was hydrolized with ${\beta}-amylase$ and transglucosidase. $5.2\%$ isomaltose was produced as isomaltooligosaccharides and glucose content was increased in the enzyme treated ginseng extract. Contents of ginsenoside $R-b_1\;and\;R-b_2$ were decreased, whereas ginsenoside-Rd was increased by the enzyme treatments. The growth of 3 strains of bifidus spp. and 4 strains of lactobacillus spp., beneficial intestinal bacteria, were enhanced by adding of the enzymatically hydrolized ginseng extract. Sweetness and sourness were increased, however, bitterness and astringency were decreased in the hydrolized ginseng extract. The fonnation of precipitates in hydrolized red ginseng extract of $pH\;3.0\~4.5$ were significantly decreased in the storage condition of $40^{\circ}C$ for 1 month compared to that of control.

Characteristics of Alcohol Fermentation in Citrus Hydrolysates through Narirutin Addition (Narirutin첨가에 따른 감귤가수분해물의 알코올 발효 특성)

  • The characteristics of the narirutin-added alcohol fermentation of citrus hydrolysates were investigated to produce functionally improved citrus wine. The pH of narirutin-added citrus wine ranged from 3.38 to 3.51, and the titratable acidity was about 0.55% which show no significant differences attributable to the narirutin quantity. With regard to the color and brown color, with the addition of more narirutin, the a value was decreased while the b value and brown color increased. The sugar content decreased from 24 to 8.0-8.5 $^{\circ}Brix$ after alcohol fermentation, and the alcohol content appeared to be between 14.5 and 14.6% in all the sections. For the free sugar contents, the glucose and fructose contents were relatively high in every section. For the total polyphenol content, it was shown to increase when more narirutin was added. and the 2%-narirutin-added section showed the highest polyphenol content(46.44 mg%). The results of sensory evaluation were that the color, flavor and bitterness increased while the sweetness and overall acceptability were reduced by the additional narirutin. Functionally improved citrus wine can be produced by using narirutin, but more researches regarding quality improvement should be carried out to overcome the bitterness.
