Saccharification and Ethanol Fermentation from Uncooked Starch using Aspergillus niger Koji (생전분의 당화와 주정발효)

  • The energy for cooking starch prior to saccharification could be saved by fermenting raw starch into ethanol using Aspergillus niger koji of wheat bran. Optimum cultivation time to produce glucoamylase was 4 days in wheat bran medium. The rate of saccharification from uncooked corn starch were optimum at pH 3.3 and 40-$50^{\circ}C$. Corn and sweet potato starch were saccharified more efficiently by wheat bran koji than other tested starch sources. 5 days of fermentation were required for optimum yield of ethanol using a mixture of AspergiUus niger koji and dried yeast. Final ethanol yields from raw corn, sweet potato, and rice starch with agitation at the rate of 100 rpm were about 95% at $30^{\circ}C$.

Effect of Planting Dates on Growth and Yield of Late-planted Sweet Corn (Zea mays L.) to Sell Fresh Ears in the Autumn (가을 출하용 단옥수수 극만파재배시 파종기가 단옥수수의 생육과 수량에 미치는 영향)

  • Fresh edible sweet corns demand relatively short period to harvest fresh ears, which can allow farmers to make a choice sweet corns for various cropping systems. For this reason, we were to find the optimum planting date of late-planted sweet corns to sell fresh ears in the autumn linked to cropping system with winter crops, investigating yield and properties of marketable fresh ears and growth traits of sweet corns (cv. 'Godangok' and cv. 'Guseulok') depending on planting dates such as 10 July, 20 July, and 30 July in Suwon 2012 and 2013, respectively. The 20 July-planted sweet corns showed the most fresh ear yield. However, the 10 July-planted and the 30 July-planted had 32% less yield caused by consecutive rainfall from 10 July through 20 July, and 15% less yield due to low air temperature during ripening than the 20 July-planted, respectively. The 10 and 20 July-planted sweet corns had average 140g of a fresh ear weight and 15% heavier ear than the 30 July-planted. For the July-planted sweet corns, silking days after planting ($r=-0.80^{**}$), and harvesting days after silking ($r=-0.97^{**}$) and planting ($r=-0.91^{**}$) were highly negatively correlated with daily mean air temperature during the period, resulting in it takes 1,100 growing degree days (GDD) to harvest fresh ears from the July-planted sweet corns. The fresh ears of the 20 July-planted sweet corns are able to be harvested by early October. Therefore it will be a good choice for the cropping system based on winter vegetable cash crops such as temperate garlic and onion with medium or late maturity. Among three planting dates 20 July-planted sweet corns had the best field performance in every year considering fresh ear yield, ear size, and stability to grow.

Effect of Content of Crop Component on the Bioethanol Production (작물의 성분 함량이 바이오에탄올 생산에 미치는 영향)

  • The contents of starch, moisture, crude protein, crude fat, crude fiber, and ash of different varieties of various crops such as brown rice, barley, corn, sweet potato were analyzed. The average starch contents of brown rice, barley, corn, and sweet potato were $70.1{\pm}0.4\;{\sim}\;72.2{\pm}2.1$, $68.7{\pm}0.2\;{\sim}\;71.4{\pm}1.2$, $67.6{\pm}0.8\;{\sim}\;69.4{\pm}1.8$, and $21.7{\pm}0.9\;{\sim}\;28.3{\pm}0.5%$, respectively. The ground powder of each starchy substrate was suspended in distilled water, and then liquefied, saccharified, and fermented by dried active yeast at 32 for 4 days. By statistical analysis, the effectiveness of the contents of the different components such as starch, moisture, crude protein, crude fat, crude fiber, and ash of the crops on the ethanol production were examined. The results showed that the starch content positively affected the ethanol production in all the tested cereals and sweet potato. In brown rice, ash content affected negatively the ethanol production. In barley, protein content affected negatively the ethanol production, while fiber content affected positively the ethanol production. The sweet potato containing higher content of moisture produced less amount of ethanol.

Genetic Improvement for Yield and Yield Related Traits by Introgressive Hybridization in Sweet Corn

  • Proper choice of source populations contributes to the ultimate success of selection for genetic improvement. The source population should possess the most desirable alleles at as many loci as possible for intra population improvement. Such desirable alleles can be intensified by introgression of exotic germ plasm into locally adapted ones through hybridization followed by selection. The objectives of this study were to determine the mean performance, genetic variability $({\sigma}^2G)$ and heritability of fresh ear yield and other important traits within two sweet corn source populations, $BC1-10{\times}Syn-II$ and BC2-10. One hundred selfed progenies from each of the two source populations were evaluated in a $10\times10$ lattice design, at the Institute of Bioscience (IBS) Farm, University of Putra Malaysia (UPM) following the recommended cultural practices. Significant differences among selfed progenies within $BC1-10{\times}Syn-II$ were observed for all traits, while differences among selfed progenies within BC2-10 were noted for fresh ear yield, ear length, ear diameter, number of kernels per row, ear height, days to tasseling and days to silking. Progenies developed from $BC1-10{\times}Syn-II$ population had higher estimates of ${\sigma}^2G$ than did progenies from BC2-10 population for number of kernel rows per ear, total soluble solids, plant height, days to tasseling and days to silking, showing that selection to improve these traits would be more effective in selfed progenies of $BC1-10{\times}Syn-II$ than that in BC2-10. On the other hand, progenies developed from BC2-10 population had higher estimates of ${\sigma}^2G$ for ear length, ear diameter and ear height, indicating that progenies from this population would have better genetic gain than $BC1-10{\times}Syn-II$. Comparable estimates of genetic variance were found for fresh ear yield, and number of kernels per row, indicating that genetic improvement of the two source populations is expected to produce similar genetic gains for these two traits. Therefore, selfed progenies developed from both source populations could be used to improve the two populations for various traits and thereby develop superior genotypes for immediate use in the production system.

Optimum Plant Populations of a Super Sweet Corn Hybrid at Different Planting Dates (초당옥수수의 파종기별 적정 재식밀도)

  • A super sweet corn hybrid, "Cambella 90", was grown at 5 plant populations (35,000, 45,000, 55,000, 65,000, 75,000 plants/ha) on 1 April, 1 May, and 1 June in 2004 under black polyethylene (P. E.) film mulch to find out the optimum plant populations at different planting dates. Emergence rate and percent stand increased as planting dates delayed. Culm length and ear height were highest at the planting on 1 May and increased with increased plant population. Soluble solids content was highest at the planting on 1 May and decreased as plant population increased. Ear length, seed set ear length, ear diameter, and ear weight decreased as plant population increased. Seed set ear length could not substitute for ear weight to evaluate marketable ears. The optimum plant population of "Cambella 90" was $65,000{\sim}75,000$ plants/ha at the planting on 1 April when emergence rate was low due to low temperature, $55,000{\sim}65,000$ plants/ha at the planting on 1 May, and 65,000 plants/ ha after the planting on 1 June.

Detection of Irradiated Starches from Korea and China by Viscometric Method (국산 및 중국산 전분의 감마선 조사 여부 검지를 위한 점도 측정)

  • A study was carried out to establish the detection method for irradiated cereals. Starchs were ground and irradiated at 2.5, 5.0, 7.5, 10 and 15 kGy using a Co-60 irradiator. The viscosities decreased in all samples by increasing irradiation dose. The viscosity of the potato, corn and sweet potato from Korea dropped from 35.00$\pm$ 0.34, 11.86$\pm$0.86 and 14.02$\pm$0.62 in the control to 11.38$\pm$0.92, 2.98$\pm$0.80 and 2.74$\pm$0.80, respectively, in the samples irradiated at 15 kGy. These trends were similar to samples from China. Regression expression and coefficients of viscosity of potato, corn and sweet potato starch from Korea were 0.97 (y=-4.6197x+38.284), 0.88 (y=-1.5146x+12.023) and 0.88 (y=-1.5146x+12.023), respectively, those of viscosity of starches from China were 0.69 (y=-28.849x+152.25), 0.81 (y=-3.38x+20.56) and 0.88 (y=-2.668x+18.505) at 50 rpm. All samples resulted in a decrease in specific parameter by increasing rpm after irradiation. Parameter values showed dose-dependent relationship between unirradiated and irradiated samples and indicated that all values of unirradiated samples were higher than the those of irradiated ones. These results suggest that the detection of irradiated potato, corn and sweet potato starch at various irradiation doses is possible by the viscometric method.

Evaluation of Regional Adaptability in Introduced Super Sweet Corn Hybrids and Heritability of Agronomic Traits (도입 초당옥수수 교잡종의 지역 적응성 및 농업 형질의 유전력 평가)

  • This study evaluated newly introduced, commercial super sweet corn hybrids (Zea mays L.) for their potential as breeding materials. Agronomic traits were measured and variance components were obtained using a linear mixed model to estimate the heritability. The trials were carried out in 2018 at two locations (Haenam and Oksan in South Korea). All traits had low heritability, except for mid tasseling and silking days. These traits with low heritability mostly had low genetic variance component estimate. In case of ear height ratio, significant genotype by location appeared to be responsible for low genetic variance, which in turn led to low heritability. Low heritability estimates from the trials with commercial hybrids were perhaps because those hybrids were highly improved for commercial success. Hence, this does not necessarily point to them having poor potential as breeding materials. To overcome low heritability, significant genotype by environment interaction, and achieve high selection efficiency, intermating among hybrids is recommended to create new recombinants before inbred line development.

Optimization of Conditions for the Production of Algin-like Polysaccharide by Polyglucuronic Acid C5-Epimerase (Polyglucuronic Acid C5-Epimerase에 의한 Algin 유사 다당류 생산 조건의 최적화)

  • We could produce algin-like biomaterial of polyiduronan using polyglucuronic acid C5-epimerase with polyglucuronic acid prepared by specific oxidation of primary alcohol groups of four kinds of polysaccharides(corn starch, rice starch, sweet potato starch, and cellulose). The enzyme activity was determined by the modified Dische carbazole methodology with the isolated crude enzyme from the supernatant centrifuged at $100,000{\times}g$ for 1 hr after grinding fresh bovine liver. And then, the optimal substrate, pH, and temperature for the enzyme reaction of polyglucuronic acid C5-epimerase were determined as the oxidized sweet potato starch, 7.0, and $30^{\circ}C$, respectively. Conclusively, it could be possible to epimerize polyglucuronic acid in the oxidized sweet potato starch to polyiduronic acid. Therefore, we could obtain algin-like polysaccharide using the oxidized sweet potato starch and polyglucuronic acid C5-epimerase isolated from bovine liver.
