In order to product the furofuranoid lignans, (+)-eudesmin which is one of the secondary products from Magnolia sieboldii. through cell suspension cultures; various culture media, initial sucrose concentration, elicitations, shaking speeds, and inoculum sizes. Among the culture media tested, MS medium had a pronounced effect on suspension cell growth and (+)-eudesmin contents. The maximum dry cell weight (DCW) of 3.71 g per flask was obtained at inoculum size of 0.5 g and in MS medium supplemented with $3\%$ sucrose plus 0.5 mg/L 2,4-D after 8 weeks. (+)-Eudesmin biosynthesis was stimulated with high initial sucrose concentration ,and the maximum (+)-eudesmin production of $3.2{\mu}g/g$ DCW was achieved at 200mg/L chitosan and $5\%$ initial medium sucrose. The optimal shaking speeds for dry biomass accumulation and (+)-eudesmin contents was 130 rpm. This work is considered to be helpful for large-scale bioprocessing of Magnolia sieboldii suspension cell cultures in bioreactor.
Calli and suspension cultures were obtained following inoculation of the explant from leaves of Ginkgo biloba L on the supplemented MS basal medium. The obtained calli and suspension cultured cells were able to produce detectable amounts of ginkgolides which are known as natural specific PAF antagonists. The production of ginkgolides in the calli and suspension cultured celles were identified using GC/MS, GC and HPLC with authentic ocmpounds. Since the production of ginkgolides A and B the calli and suspension cultured cells had been confirmed, effects of types and concentration of plant growth regulators, media and illumination on the induction and growth of the callus were studied. The concentrations of growth regulators for optimal callus induction were 1.0 to 2.0 mg/L for NAA and o.1 mg/L for kinetin. The growth of the Callus seemed to be more simnultaed with the combination of NAA and kinetin than NAA and BA with illumination at all concentration ranges of 1.0 to 4.0 mg/l for NAA and o.1 to 1.0 mg/L for kinetin or BA studied. Amogn 8 different media used, the induction rate of callus on Anderson, Eriksson, and Shenk and Hildebrant at 4 weeks after the innoculation was almost the same as that of MS. However, callus was rarely induced on Heller or White medium. Suspension cultures were easily initiated with 3 g of callus (fresh weight) derived from ginkgo leaves on supplemented MS medium. A typical growth curve of suspension cultured cells could be obtained by measuring the fresh weight of the suspension cultured cells at every 3 days. To improve the growth of suspension cultured cells of ginkgo, effects of concentrations of NAA, sucrose, phosphate ions and molar ratio of $NH_{4}^+\;to\;NO_{3}^-$ ions in the culture medium were studied. The maximum growth of the cells was achieved when the culture medium contained 1.0 mg/L of NAA, 30 g/L sucrose, 1.75 mM phosphate ions and 1:5 molar ratio of $NH_{4}\;to\;NO_{3}^-$ ions.
A metabolic study has been conducted to investigate the conversion of pentachlorophenol(PCP) to water soluble metabolites in soybean and rice cell suspension cultures as well as in intact rice plants. PCP in plant cells was found to be exclusively transformed into water soluble metabolites. The relative rate of the metabolic conversion of PCP in decreasing order was soybean cultures > rice cultures > rice plants. Also observed was that, the older the cultures grown, the lower the conversion rate was. Primary water soluble metabolites isolated from both the 5 day old soybean and 8 day old rice cells were specifically hydrolyzed only by ${\beta}$-glucosidic linkage specific glucosidase, suggesting that the metabolites are ${\beta}$-glucose conjugates. The amount of glucose conjugates was increased with increasing time of incubation of PCP up to 24 hr in both soybean and rice cultures; Thereafter, it was decreased progressively. Most of the glucose conjugates were further metabolized to more polar conjugates in cells, but a portion of them was excreted into the culture medium.
The accumulation of benzophenanthridine alkaloids, sanguinarine, chelerythrine, chelirubine and macarpine occurred in suspension cultures of Eschscholtzia californica. To increase alkaloid production, feeding experiments with the biosynthetic precursors, tyrosine, tyramine, L-dopa, dopamine with and without elicitation were studied. In feeding experiments with various precursors, the total alkaloid production was slightly increased. The precursor feeding with elicitation, however, increased total alkaloid production several times.

Biotransformation of exogenous (-)-(4R)-isopiperitenone in cell suspension cultures of Mentha piperita resulted in oxidized products, with (-)-7-hydroxyisopiperitenone being the major compound. The mass of products obtained $unde^{18}O_2$, atmosphere was two units higher than that under normal atmosphere. The biotransformation was inhibited by several cytochrome P450-specific inhibitors as well as by carbon monoxide. Carbon monooxide inhibition was substantially overcome by irradiation of cells with blue light including light at 450nm wavelength. These results suggested that a cytochrome P450-type monooxygenase was involved in the biotransformation.
Elicited cells with methyl jasmonate continued to produce benzophenanthridine alkaloids throughout medium changes in suspension cultures of Eschscholtzia californica. Large increases in alkaloid production were observed by re-elitations with medium changes. The total alkaloid production increased during the successive elicitation steps reaching a maximum level on the 4th elicitation. The highest total alkaloid produced was 250 mg/I, which was 20fold higher than that of the single elicitation and 4-fold higher than that of the normal culture without elicitation. The large increases in alkaloid production in successive re-elicitations with medium changes are believed to be caused by the accumulation of the signal transduction compound, jasmonate.
Large and rapid increases in benzophenanthridine alkaloid production occured in suspension cultures of Eschscholtzia californica cells treated with elicitors. Response to different biotic elicitors showed that elicitors prepared from yeast extract, Collectotrichum lindemuthianum and Verticillium dahliae induced alkaloid formation. Highest alkaloid accumulation was obtained with $60\;\mu\textrm{g}$ of yeast extract elicitor per gram of fresh cell weight. In time course performance after elicitor addition, more than 40 hours were required to obtain saturated alkaloid accumulation. Compounded silicone fluid, an ideal accumulation phase for two-phase culture of E. californica, accumulated a large amount of alkaloids produced in a specific manner. Elicitation in two-phase culture clearly increased net alkaloid production as well as their concentrations in the accumulation phase.
The productivity of natural flavors in alfalfa suspension cultures is low partially due to their low solubility. Thus, by formation inclusion complex of cyclodextrin(CD) with flavor compounds enhancement of the productivity of flavor was attempted in this research. we observed that CD formed inclusion complex with flavor compounds by GC/MS analysis. Also, there was indication that CD played a role as an elicitor in alfalfa suspension cultures.
Extracts of Escherichia coli as a elicit were treated to suspension cultures of Streptanthus tostus in order to observe the effect on the pholem development. By the elicit treatment, cell wall, sieve endoplasmic reticulum (SER) and p-protein were normally synthesized, but the structure of amyloplast was changed from a round form to irregular and swollen unhalthy form with a tiny starch granular. Oil drops were new synthesized and accumulated in a large oleoplast and proteins were also accumulated in a single membrane. The concentration of sucrose in the phloem, which was induced during the elicit treatment, was higher than normally developed phloem cells. These results suggest that phloem cells might be changed in the normal cycles of metabolism of lipids, carbohydrates and proteins to overcome during the eilicit stress.
In suspension cultures of Streptanthus. the uptake rate of sugar was increased during the ceil starvation of sugar in the medium. The maximal uptake rate obtained with 3 days of cell starvation. Sugar transport system induced by the sugar starvation was completely inhibited by 10 $\mu$M cycloheximide. Plant cells are known to possess only one sugar transport system, but the uptake rate of glucose obtained a saturated kinetic while the one of sucrose had two different kinetics after the sugar starvation. Induced sugar transport systems had different kinetics compared to plant cell. These results showed that higher plants have adaptable ability to induce new sugar transport systems when the environment changed unsuitable.
