Production of 10-deacetylbaccatin III in Taxus cuspidata Suspension Cell Culture (주목 현탁세포배양을 이용한 10-deacetylbaccatin III 생산)

  • In this study, enhanced production of 10-deacetylbaccatin III(10-DAB), a precursor of taxol in semisynthesis, was investigated in Taxus cuspidata cell suspension cultures. The effects of initial inoculum size and sugar concentration were examined to prove the relationship between the production of 10-DAB and cell growth. The cell growth was found to be stimulated in Schenk and Hildebrandt(SH) medium. The lower the inoculum size as well as initial sugar concentration, the faster the cell growth rate. When the initial sugar concentration was dept low, the production of 10-DAB into medium was increased. By using perfusion culture, continuous cell growth was possible until the end of culture and more than 34.67 g/L of cell concentration could by obtained. This is about 2.5 times higher level than that of control batch culture.

Production of Gomisin J from Suspension Cultured Cells of Schisandra chinensis Baillon in Airlift-type Bioreactor (생물반응기를 이용한 오미자의 현탁배양세포로부터 Gomisin J의 생산)

  • Suspension culture of Schisandra chinensis for production of gomisin J was perfomed in bioreactor. The inoculum size and initial sucrose concentration had significant effect on the cell growth and gomisin J accumulation. The maximum dry cell weight $(DCW;\;43.5\;g/{\ell})$ and gomisin J content $(0.71\;{\times}\;10^{-3}\;{\mu}g/g\;DCW)$ were obtained at inoculum size of 100 g fresh cell weight (FCW) per liter and MB5 medium containing 6% sucrose after 8 weeks of culture. The effect of oxygen supply on the cell growth and gomisin J accumulation was also investigated in an airlift-type bioreactor. The optimal cell growth and gomisin J content was obtained under 0.5 vvm. The productivity of gomisin J was 0.7 fold in bioreactor culture lower than that obtained in a flask cultivation.

  • To develop new elicitors inducing the high productivity of taxane derivatives, plant growth inhibitors, namely, maleic acid hydrazide, N-phosphomethyl glycine and succinic acid 2.2-dimethyl hydrazide, coconut milk and yeast extract were administrated in the cell suspension culture system of Taxus cuspidata, and the production of baccatin III were analysed. The effects of amino acid related with the biosynthesis of baccatin III were also examined in these culture system. As the results, a remarkable enhancement of baccatin III production was observed in the cultivation with coconut water and with maleic acid hydrazide.

  • The growth kinetics and the production of $\beta$-glucuronidase from transgenic tobacco's suspension culture was investigated in the flask culture and a 2.5 L bubble column reactor. The growth of bubble column reactor was similar to that of flask culture. However, in the bubble column reactor, the production of $\beta$ -glucuronidase reached 2850 U/mg (85-fold higher than that of flask culture). In both case, the production level of $\beta$ -glucuronidase was fluctuated, which was resulted from periodical degradation of the protein. Sucrose is important component in plant culture medium. Twice addition of sucrose in bubble column reactor could not improve cell growth, since other components in a medium were already depleted. However, the addition of sugar decreased cell size, which facilitated the operation of bioreactor. The production of $\beta$ -glucuronidase was continuously increased, however final concentration of $\beta$ -glucuronidase was similar to that without sucrose addition.

Effect of Inoculum Size on Biomass Accumulation and Ginsenoside Production by Large-Scale Cell Suspension Cultures of Panax ginseng

  • Cell growth and ginseng saponin production by large-scale suspension (bioreactor) cultures of Panax ginseng were investigated under various inoculum sizes. Cell growth was low at an inoculum size of 40 g FW/L, and the maximum cell growth was obtained with increasing inoculum size up to 100 g FW/L. The cell density of 333 g FW/L and 12.7 g DW/L was obtained at inoculum size of 100 g FW/L after 30 days of cultivation. Maximum saponin production of $4.40\;\cal{mg/g}$ DW was achieved at 60 g FW/L of inoculum size. Thus, inoculum size 60 g FW/L was suitable for optimum biomass accumulation as well as saponin production during bioreactor cultivation of ginseng suspension cells.

Improvement of Peroxidase Productivity by Optimization of Medium Composition and Cell Inoculum Size in Suspension Cultures of Sweet Potato (Ipomoea batatas) (고구마(Ipomoea batatase)현탁배양에서 배지조성 및 세포접종량의 적정화에 의한 Pemxidase생산성 향상)

  • To improve the productivity of peroxidase (POD) of cell line SP-47 derived from cell suspension cultures of sweet potato (Ipomoea batatas (L) Lam.cv White Star), we optimized culture conditions including the composition and concentration of plant growth regulators and carbon source, and the cell inoculum size. When one g (fr wt) of cells was inoculated into 50 mL TL medium supplemented with l mg/L 2,4-D and 30g/L sucrose in 300 mL Erlenmeyer flask at 25$^{\circ}C$ in the dark (100rpm), the POD activity per g cell dry wt was maximized to be about 6,800 units after 25 days of subculture, which was about 30 times higher than that of intact roots of horseradish plants grown in the greenhouse, but the cell growth was maximum after 15 days of subculture. The protein content per g cell dry wt maintained almost plateau and after 25 days of subculture decreased as culture Proceeded further whereas the POD specific activity (unit/mg protein) was about two times higher after subculture and continuously increased from 12 days to the end of cultures (40 days). The POD isozyme patterns showed almost the same regardless of cell growth stage, but some acidic isozymes were slightly increased after 25 days of subculture. These results indicate that POD activity in suspension cultures of sweet potato is closely associated with cell growth and stresses derived from cell culture renditions and medium depletion. Due to its high POD activity the SPL47cell line seems to be suitable for the mass production of POD.

  • Human garnulocyte-colony stimulating factor(hG-CSF), a hematopoietic growth factor$^{1)}$, was produced and secreted from tobacco cell suspension. hG-CSF produced from tobacco cell suspension culture is biologically active form. The produced amount of hG-CSF is about 100${\mu}g/L$ in 9 days after inoculation.

  • Production of shikonin derivatives through cell suspension culture of Lithospermum erythrorhizon was investigated. Optimal concentrations of IAA and kinetin on the growth of cell suspension were 0.2 and 0.1 ppm respectively. Pigment content was markedly increased when aluminum oxide was added to the production medium and its optimal concentration was 1.5g/70ml medium. The most effective concentration of IAA was 0.5 ppm and the production of pigment did not depend on the kinetin concentration.

Identification of Salidroside from Rhodiola sachalinensis A. Bor. and its Production through Cell Suspension Culture (참돌꽃에서 Salidroside의 동정 및 현탁세포배양을 통한 분리)

  • Salidroside was isolated and purified from R. sachalinensis A. Bor. roots. Purified salidroside was obtained from repeated silicagel column chromatography and preparative HPLC, and identified by $^1H-NMR,\;^{13}C-NMR\;and\;^1H-^1H$ COSY spectra analyzer. Callus induction and cell suspension from R. sachalinensis leaf segments were established on 1/2MS solid medium and in $2B_5$ liquid medium containing 0.5 mg/l NAA and 1 mg/l, BA in the dark condition, respectively. The contents of salidroside for suspension culture were ranging from 0.12% to 0.41% in comparison with 0.17% for natural roots.
