Preterm infants are vulnerable to the oxidative stress due to the production of large amounts of free radicals, antioxidant system insufficiency, and immature oligodendroglial cells. Reactive oxygen species (ROS) play a pivotal role in the development of periventricular leukomalacia. The three most common ROS are superoxide ($O2^{\cdot-}$), hydroxyl radical ($OH^{\cdot}$), and hydrogen peroxide ($H_2O_2$). Under normal physiological conditions, a balance is maintained between the production of ROS and the capacity of the antioxidant enzyme system. However, if this balance breaks down, ROS can exert toxic effects. Superoxide dismutase, glutathione peroxidase, and catalase are considered the classical antioxidant enzymes. A recently discovered antioxidant enzyme family, peroxiredoxin (Prdx), is also an important scavenger of free radicals. Prdx1 expression is induced at birth, whereas Prdx2 is constitutively expressed, and Prdx6 expression is consistent with the classical antioxidant enzymes. Several antioxidant substances have been studied as potential therapeutic agents; however, further preclinical and clinical studies are required before allowing clinical application.
In this study, antioxidant and free radical scavenging activities of the natural antioxidative compound, pyrogallol-phloroglucinol-6,6'-bieckol (PPB) isolated from brown algae, Ecklonia cava was assessed in vitro by measuring the radical scavenging activities (DPPH, alkyl, hydroxyl, and superoxide) using electron spin resonance (ESR) spectrometry, intracellular reactive oxygen species (ROS) scavenging activity, and DNA damage assay. According to the results of these experiments, the scavenging activity PPB against difference radicals was in the following order: DPPH, alkyl, hydroxyl, and superoxide radicals ($IC_{50}$; 0.90, 2.54, 62.93 and $109.05{\mu}M$). The antioxidant activities of PPB were higher than that of the commercial antioxidant, ascorbic acid. Furthermore, PPB effectively inhibited DNA damage induced by $H_2O_2$. These results suggest that the natural antioxidative compound, PPB, can be used by the natural food industry.
Salsolinol, endogenous neurotoxin, is known to be involved in the pathogenesis of Parkinson's disease (PD). In the present study, we have investigated the oxidative damage of DNA induced by the reaction of salsolinol with Cu,Zn-SOD. When plasmid DNA incubated with salsolinol and Cu,Zn-SOD, DNA cleavage was proportional to the concentrations of salsolinol and Cu,Zn-SOD. The salsolinol/Cu,Zn-SOD system-mediated DNA cleavage was significantly inhibited by radical scavengers such as mannitol, ethanol and thiourea. These results indicated that free radicals might participate in DNA cleavage by the salsolinol/Cu,Zn-SOD system. Spectrophotometric study using a thiobarbituric acid showed that hydroxyl radical formation was proportional to the concentration of salsolinol and was inhibited by radical scavengers. These results indicated that hydroxyl radical generated in the reaction of salsolinol with Cu,Zn-SOD was implicated in the DNA cleavage. Catalase and copper chelators inhibited DNA cleavage and the production of hydroxyl radicals. These results suggest that DNA cleavage is mediated in the reaction of salsolinol with Cu,Zn-SOD via the generation of hydroxyl radical by a combination of the oxidation reaction of salsolinol and Fenton-like reaction of free copper ions released from oxidatively damaged SOD.
As the biodegradation of wood constituents has been understood as a multi-basidiomycetes and enzymatic processes, this review will focus on the roles of low molecular compounds and radicals working in harmony with fungal enzymes. Wood rotting basidiomycete fungi penetrate wood, and lead to more easily metabolize carbohydrates of the wood complex. The white-rot fungi, having versatile enzymes, are able to attack directly the "lignin barrier". They also use a multi-enzyme system including so-called "feedback" type enzymes allowing for simultaneous degradation of lignin and carbohydrates. The multi-enzymes including laccase support the proposed route by explaining how the high molecular weight enzymes can function in the wood complex. These enzymes may function separately or cooperate each other. In addition, veratryl alcohol oxidase, cellobiose dehydrogenase, arylalcohol dehydrogenase, and particularly low molecular mediators and radicals have an important role in wood biodegradation. However, the possibility of other mechanism as well as other enzymes, as operating as feedback systems in the process of wood degradation, could not be excluded.
Objectives : The aerial parts of Rumex japonicus Houtt. (RF) is used by traditional clinics to treat parasite infection in East asia. This study aims a verification of anti-oxidative and anti-inflammatory effects of RF methanol extract. Methods : Anti-oxidative effects of RF were measured by scavenging activities of DPPH, superoxide, nitric oxide (NO) and peroxynitrite radicals. And also scavenging activities of anti-oxidation in lipopolysaccharide (LPS)-treated RAW 264.7 cells were measured. The inhibitory effects against the production of inflammatory mediators including NO, prostaglandin $E_2$ ($PGE_2$), tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$), interleukin-$1{\beta}$ (IL-$1{\beta}$), IL-6, inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2), and the translocation of nuclear factor (NF)-${\kappa}B$ in LPS-stimulated RAW 264.7 cells by RF were tested. Results : RF scavenged DPPH, superoxide, NO and peroxynitrite radicals, and RF (at $200{\mu}g/m{\ell}$) reduced the inflammatory mediators definitely. Conclusions : These results indicate that RF may be a potential drug source for oxidative stress related inflammatory diseases.
Objectives : This study was designed to investigate the effects of anti-oxidation of fractionated Pulsatilla koreana $N_{AKAI}$ (PK) extracts. And we examined to determine that a certain fractionated extract has the best anti-oxidative effects between the fractionated PK extracts. Methods : Anti-oxidative effects of fractionated PK extracts was measured by scavenging activities of DPPH, superoxide, nitric oxide (NO) and peroxynitrite radicals. And also scavenging activities of anti-oxidation in lipopolysaccharide (LPS)-treated RAW 264.7 cell was measured. After these examination, we determined a fraction that has best anti-oxidative effects. Results : Fractionated PK extracts inhibited radicals effectively. Also in RAW 264.7 cell, intracellular oxidation has inhibited by PK extracts. In these tests, ethyl acetate (EA) fraction has the best anti-oxidative effects among PK extracts. Conclusions : This results demonstrate that PK extracts exhibit anti-oxidative effects. And EA fraction has the best inhibition effects among the six fractions of PK.
This study was designed to investigate the effects of silkworm powder (SWP) on oxygen radicals and their scavenger enzymes in serum of rats. Sprague Dawley (SD) male rats (160${\pm}$10g) were fed experimental diets (SWP-200and SWP-400 groups)added 200 and 400mg/kg BW/day for 6 weeks. Triglyceride (TG)levels were significantly inhibited (10% and 25%) in SWP-200 and SWP-400 groups, but there were no singificant differences in total, LDL-and HDL-cholesterol levels in both SWP-200 and SWP-400 groups. Hydroxyl radical ($.$OH) formations resulted in a marked decreases (about 20%) in both SWP-200 and SWP-400 groups compared with control group. Superoxide radical (O2.-)and hydrogen peroxide formations resulted in a significant decreases in SWP-400 group compared with control group. Lipid peroxide (LPO)and oxidized protein(>C=O group) productions resulted in a significant decreases (SOD) and catalase (CAT) activiteies were remarkably increased (10∼25% and 40∼50%)in SWP-200 and SWP-400groups. Glutathione peroxidase(GSHPX) activities were significantly increased (about 10%) in SWP-400 groups compared with control group. These results suggest that anti-aging effect of silkworm powder (SWP) may play a pivotal role in attenuation a various chronic degenerative diseases age-related changes.
본 연구의 목적은 인지기능을 가진 약효제를 발굴하는 것이다. 본 연구에서는 토끼의 간과 신경세포로부터 각각 유래한 angiotension converting enzyme과 acetylcholinesterase에 대한 강황 열수추출물(CLWE)의 억제효과 뿐만 아니라 항산화 효과가 조사되었다. 이리하여 먼저 산화적 스트레스와 연관이 있는 환원력과 DPPH radical, superoxide anion, hydroxyl radical, lipid peroxidation에 대한 CLEW의 소거능 및 DNA 산화에 대한 보호효과를 평가하였다. CLEW는 환원력 뿐만 아니라 본 연구에서 시험된 활성산소종 중에서 특히 superoxide anion의 소거능이 가장 우수한 것으로 나타났다. 더욱이 CLEW는 0.25% 이상의 농도에서 angiotensin converting enzyme 활성을 억제효과를 발휘하는 것으로 나타났다. 신경세포에서 CLEW는 또한 산화적 스트레스와 nitric oxide 유발을 억제하였다. 그러므로 이상의 결과는 CLEW의 항산화 효과와 신경세포 보호효과를 입증하여, 이것이 사람의 신경건강을 위한 천연생물소재로 잠재적인 가능성을 가지고 있을 것으로 시사한다.
Phellinus pini (CY001) 추출물에서의 페놀류성분의 농도는 P. pini (CY001)분획 g 당 GAEs mg으로 나타내었고, P. pini의 EtOAc 분획 (436.5 mg GAEs/g)이 다른 분획에 비해 가장 높은 함량을 나타내었다. 환원력과 같은 항산화 활성을 screening 하기 위해 몇 가지 생화학적 시험이 수행되었고, 그것은 2, 2-diphenyl-1-picrylhydrazyl(DPPH) 라디칼 소거작용, NBT/XO superoxide system, DCF/AAPH peroxyl radicals 저해능 등이다. 여섯 개의 버섯추출물 분획 중에서 EtOAc 분획이 DPPH, superoxide 라디칼, peroxyl 라디칼 소거작용이 뛰어났고, $IC_{50}$ values는 각각 $11.49\;{\mu}g/ml$, $8.32\;{\mu}g/ml$, and $1.91\;{\mu}g/ml$이었다. P. pini (CY001)의 EtOAc 분획은 중요한 효소적 지질과산화 저해와 cytotoxicity 없이 RAW 264.7 macrophages의 LPS 유도 NO 생성을 효과적으로 감소시켰다. 또한 EtOAc 분획이 HepG2 cell의 타크린 유도 cytotoxicity에서 간 보호 작용을 나타내었다. 이러한 결과를 바탕으로 P. pini (CY001)가 라디칼 소거작용을 하는 성분을 함유한 천연 항산화제로써의 잠재력을 갖고 있는 것으로 사료된다.
