Antioxidant Property of Aqua-Acupuncture Solution from Circium japonicum (대계 약침액의 항산화 효능)

  • Objectives : Circium japonicum is a pharmacologically active used in traditional Korean medicine. An aqua-acupuncture solution of the Circium japonicum was assessed to determine the mechanism of its antioxidant activity. Materials : Circium japonicum was obtained from a Dongguk Korean Medicine Hospital (Kyung-ju, Kyungbuk). The freeze-dry powder was collected (yield 5.1%) for the aqua-acupuncture solution. Scavenging activity on DPPH free radicals by the Circium japonicum aqua-acupuncture solution (CJAS) was assessed according to the method followed by Gyamfi et al.. and then scavenging activity orl superoxide radicals $(O_2^-{\cdot})$ was assessed by the method described by Gotoh et al. with slight modification. Deoxyribose assay to determine the rate constant for the reactions between either antioxidants and hydroxyl radicals or antioxidants and iron ions. We tested by; (1) Non-site-specific scavenging assay (hydroxyl radicals, OH), (2) Site-specific scavenging assay (chelate iron ions), and (3) Pro-oxidant effect of the CJAS on iron dependent hydroxyl radical generation. Finally, we determined hydroxyl radical-mediated DNA nicking formation. Conclusion : Our study demonstrated that CJAS has antioxidant activities and we investigated the potential effectiveness of CJAS in preventing oxidative stress-mediated disease further.

Effects of Extraction Methods on In Vitro Biological Capacities and Rheological Properties of Polysaccharides from Red Pepper Stems

  • The purposes of this study were to produce polysaccharides from red pepper stems using different extraction methods and evaluate their chemical composition, in vitro biological capacities, and rheological properties. Two polysaccharides were extracted from red pepper stems using an autoclave and alkali treatments, and the extracts were named PAU and PAL, respectively. The contents of total phenolics and flavonoids were significantly higher in PAU than those in PAL. PAU exhibited greater scavenging activities on 2,2-diphenyl-1-picrylhydrazyl radicals, 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt radicals, superoxide radicals, and nitrite compared to PAL, suggesting that PAU served as better antioxidants. Similarly, in vitro inhibitory abilities against carbohydrate hydrolyzing enzymes of PAU were higher than those of PAL. Steady shear rheological analysis demonstrated that PAU showed higher psuedoplastic shear-thinning behavior compared to PAL. Based on the results from dynamic shear rheological properties, it was found that both samples had predominantly viscous behavior rather than elastic behavior.

  • There are now increasing evidences that free radicals and reactive oxygen species are involved in a variety of pathological events. Reactive Oxygen Species (ROS) are produced during normal cellular function. ROS lead to lipid peroxidation, massive protein oxdiation and degradation. Under normal conditions, antioxidant are substnaces that either directly or indirectly protect cell against adverse effect of ROS. several biologically important compound include ${\beta}$-carotene, taruine and flavonoids reported have antioxidant function. The various antioxidant either scavange superoxide and free radicals or stimulate the detoxification mechanisms within cells resulting in increased detoxification of free radicals formation and thus in prevention of many pathophysiologic processes. This study carried out to investigate the antioxidant activity of flavonoids, myricetin with other antioxidants, ${\beta}$-carotene and taurine on B16Fl0. In order to investigate the efficacy of antioxidant activity, we measured cell viability, antioxidant enzyme activity (SOD, GPX, CAT) and intracellular reactive oxygen intermediate (ROI). In this results, we show that these flavonoids with other antioxidant substrates are increased antioxidant activity level.

Effects of Oxygen Free Radicals on Extracellular Glutamate Accumulation in Cultured Cells

  • Exogenously applied oxygen free radical generating agent, pyrogallol, highly elevated extracellular glutamate accumulation and augmented N-methyl-D-aspartate (NMDA)-induced glutamate accumulation in cerebellar granule neuronal cells, but did not in astrocytes. Superoxide dismutase remarkably decreased the pyrogallol-induced glutamate accumulation, but either NMDA or kainate antagonists did not. In addition, pyrogallol did not affect the NMDAinduced intracellular calcium elevation. Pyrogallol partially blocked glutamate uptake into astrocytes. These results suggest that oxygen free radicals elevate extracellular glutamate accumulation by stimulating the release of glutamate as well as blocking the glutamate uptake.

  • This study was purposed to investigated the antioxidant effects of water (VLWS) and aqua-acupuncture solution (VLAS) from root of Vitis labrusca. VLWS and VLAS was were assessed to determine the mechanisms of its antioxidant activities. The VLWS and VLAS exhibitde a concentration-dependent inhibition of DPPH free radicals. This VLWS-mediated antioxidant activity was similar to that L-ascorbic acid (vitamin C). In addition, the VLWS and VLAS showed dose-dependent free radical scavenging activity, including superoxide anions and hydroxyl radicals, using different assay systems. The VLWS and VLAS were also found to be strongly inhibited linoleic acid oxidation in a ferric thiocyanate assay (FTC) system. Finally, the VLWS and VLAS significantly prevented Fenton's reagent-induced DNA nicking.

Screening of Antioxidative Components from Red Ginseng Saponin (홍삼 사포닌의 항산화활성 성분 Screening)

  • Aerobic cells are normally protected from the damage of free radicals by antioxidative on , zymes such as superoxide dismutase (SOD), catalase, glutathione (GSH) peroxidase, GSH S- transferase and GSH reductase which scavenge free radicals as well as nonenzymatic antioxidants such as ceruloplasmin, albumin and nonprotein-SH including GSH. The effects of each component (ginsenoside $Rb_1$, $Rb_2$, Rc, Rd, Re, $Rb_1$, Rf, $Rh_1$ and $Rh_2$) of red ginseng on the antioxidative enzyme activities were investigated in the liver in order to screen antioxidative components of red ginseng. Ginsenoside $Rb_1$ and Rc showed a tendency to increase GSH peroxidase activity, while ginsenoside Rc significantly decreased Cu,Zn-SOD activity. Especially, ginsenoside $Rh_2$ significantly increased catalase activity. These results suggest that ginsenoside $Rh_2$ is an important active component among total saponins of red ginseng.

Effects of Endotoxin and Verapamil on Superoxide Production by Rat Alveolar Macrophage (백서폐포대식세포에서의 Superoxide 생산에 미치는 내독소 및 Verapamil의 영향)

  • Background: Superoxide anion which was produced by macrophage and neutrophil has a defensive role to kill invasive microorganisms and also an injurious role to produce self lung damage. Production of oxygen free radicals including superoxide is a main mechanism of acute lung injury caused by bacterial endotoxin. Endotoxin is known to activate alveolar macrophage to produce increased oxygen free radicals after the stimulation with various biological materials (priming effect). Calcium is a very important intracellular messenger in that cellular process of superoxide production. Method: This experiment was performed to elucidate the effects of endotoxin and calcium on superoxide production by phorbol myristate acetate-stimulated alveolar macrophage and the effect of verapamil on priming effect of endotoxin. Results: 1) Preincubation of macrophages with endotoxin (E. coli 055-B5) primed the cells to respond with increased superoxide production after the stimulation with PMA. Priming with endotoxin ($10^{-1}$ug/ml) produced a maximal enhancement of superoxide production (43%). 2) Verapamil could inhibit the superoxide production by PMA stimulated macrophage regardless of the presence of extracellular calcium. This means that the inhibitory effect of verapamil is caused by a mechanism independent of blocking calcium influx. 3) Verapamil could inhibit the priming effect of endotoxin on alveolar macrophage (from 30% increment to 13% increment) and could inhibit the superoxide production by PMA-stimulated macrophage preincubated with endotoxin. Conclusion: We concluded that verapamil could inhibit the superoxide production by PMA-stimulated rat alveolar macrophage and also inhibit the priming effect of endotoxin on alveolar macrophage. These inhibitory effects of verapamil could be one of the mechanisms of verapamil effects on endotoxin induced lung injury.

ALTERATION OF SUPEROXIDE DISMUTASE-AND CATALASE-ACTIVITY IN HUMAN GINGIVAL TISSUES BY THE PERIODONTAL DISEASE SEVERITY (치주질환 심도에 따른 치은조직내의 Superoxide Dismutase와 Catalase의 활성변화에 관한 연구)

  • It has been believed that antioxidant enzymes such as CuZn- and Mn-superoxide dismutase and catalase protect the tissue from damage resulting from the oxygen derived free radicals($O_2\;^-$, $H_2O_2$ and OH ). The purpose of this study was to investigate the relationship between activity of antioxidant enzymes including CuZn- and Mn- superoxide dismutase and catalase and inflammatory periodontal disease and periodontal parameters. For this study, the patients were classified into normal, gingivitis, adult periodontitis and rapidly progressive periodontitis, and then their papillary bleeding index(PBI) and probing depth were checked. Gingival tissues were surgically obtained from the patients during periodontal surgery, extraction, and clinical crown lengthening procedure. The activity of CuZn- and Mn- superoxide dismutase and catalase in the gingival tissues was measured by using UV-spectrophotometer by the same methods as Crapo et al. And Aebi did, respectively. The results were as follows : 1. CuZn- and Mn- and total-superoxide dismutase activity were significantly low in rapidly progressive periodontitis group in comparison to normal group (P<0.05). 2. In comparison of the antioxidant enzyme activity according to papillary bleeding index group(PBI), Mn-superoxide dismutase activity only was significantly lower in PBI 2, 3, and 4 groups than PBI 0 group(P<0.05). 3. Superoxide dismutase activity failed to show any significant difference according to probing depth. But significant]y high catalase activity was shown in deep pocket group (${\ge}7mm$)(P<0.05). In conclusion, these results suggest that the activity of Mn-superoxide dismutase among the antioxidant enzymes may reflect the inflammatory status of gingival tissue and that the decreased activity of superoxide dismutase may be one of responsibe factors for progression of rapidly progressive periodontitis.

  • The superoxide anion radical$(O_2)$ poses a threat to macromocules and cell organelles of the living cells. This toxicity damage to all groups of proteins results in loss of enzyme function concerned with metabolism and ion transport, and peroxidation of unsaturated fatty acids and cholesterol results in a change of permeability characteristics of the membrane, and oxidative of nucleic acids results in genomic damage and thereby cause mutation, potential carcinogenesis and somatic damage that produce cellular aging Superoxide dismutase(SOD) has received substantial attention as a potential therapeutic agent. It has been investigated as a possible agent for the prevention of ontogenesis, the reduction of cytotoxic effect of anticancer drugs, and protection against damage in ischemic tissue. It is suggest that $O_2$ is concerned with cellular aging, thereafter we need to investigate herb that activated to SOD.
