갓김치 Chlorophylls 및 Carotenoids의 항산화 효과 (Antioxidative Effect of Cholorophylls and Carotenoids in Mustard Leaf Kimchi Activity)

  • Antioxidative effects of crude chlorophylls and carotenoids extracts from mustard leaf kimchi on the lipid peroxidation in rat liver homogenate, egg phosphatidyl choline (EPC) liposome and superoxide anion radical were examined. The extracts were found to inhibit the increase of the thiobarbituric acid (TBA) value and show the effect of antioxidative activity on the liposomal phospholipid membrane. The oxidation index of EPC liposome was markedly decreased in the prescence of the extracts. The antioxidative activity of the extracts from mustard leaf kimchi was not related with fermentation period of the kimchi. The extracts from mustard leaf showed the similar antioxidative activity of $\alpha$-tocopherol within in the given level of addition. However, the oxidation index. When the effect of the extracts from mustard leaf kimchi on free radical scavenging was observed by the determination of the superoxide anion radical scavenging activity, it had similar value to that of $\alpha$-tocopherol.

Antioxidant activity of ethanol extracts from the root and bark of Ulmus davidiana

  • Antioxidant activities of Ulmus davidiana root and bark extracts were evaluated by various antioxidant tests, including DPPH radical-scavenging, nitric oxide-scavenging, superoxide anion radical-scavenging, and ABTS radical-scavenging assays, and superoxide dismutase (SOD)-like activity and reducing power analysis, along with the determination of total phenolic and flavonoid contents. Both the extracts showed strong antioxidant activities by these testing methods. Ulmus davidiana root extract possessed strong reducing power and nitric oxide-scavenging activity, and high scavenging activities against free radicals including the superoxide anion, and the ABTS and DPPH radicals, but a weaker scavenging activity of SOD. In contrast, the Ulmus davidiana bark extract exhibited a strong SOD-like activity, but all the other activities were weak. It was observed that the total phenolic and flavonoid contents of the Ulmus davidiana root extract were higher than those of the Ulmus davidiana bark extract.

부위별 꽈리(Physalis alkekengi var. francheti) 추출물의 항산화효과 (Antioxidative Activities of Different Part Extracts of Physalis alkekengi var. francheti (Winter Cherry))

  • 본 실험에서는 꽈리를 열매, 꽃받침, 잎, 줄기, 뿌리 등 부위별로 구분하여 메탄올로 추출하고 추출 수율, 총페놀 함량, 총플라보노이드 함량, DPPH radical 소거능, 아질산염 소거능, superoxide anion radical 소거능, 환원력, 철 이온에 대한 킬레이트 효과 등을 측정하였다. 추출 수율은 열매가 가장 높았고 그 다음으로 꽃받침, 잎, 줄기, 뿌리 순이었다. 총페놀 및 총플라보노이드 함량은 잎 추출물이 가장 높았고 그 다음으로 꽃받침 추출물이었다. DPPH radical 소거능은 1 mg/mL 및 5 mg/mL의 농도에서 잎 추출물이 가장 높았고 꽃받침 추출물도 높은 활성을 나타내었다. 아질산염 소거능은 모든 부위에서 농도에 따라 유의적인 차이 없이 높게 나타났다. 꽈리 추출물의 superoxide anion radical 소거능은 꽃받침과 잎 추출물이 높은 활성을 나타내었다. 환원력은 잎 추출물에서 가장 높게 나타났고 철 이온에 대한 킬레이트 효과는 꽃받침, 줄기 및 뿌리 추출물이 높은 활성을 나타내었다. 이상의 결과를 종합하여 보면 꽈리의 잎 추출물에서 총 페놀 함량, 총플라보노이드 함량과 항산화 활성이 탁월하게 높게 나타났고 그 다음으로 꽃받침 추출물이 우수한 항산화 활성을 나타내어 향후 이를 이용한 천연 항산화제로 개발 가능성이 시사되었다.

냉이(Capsella bursa-pastoris) 에탄올 추출물의 유리라디칼 소거 및 항산화 활성 (Free Radical Scavenging and Antioxidative Activities by Ethanol Extract from Capsella bursa-pastoris)

  • Screening was performed on edible natural sources to examine superoxide radical scavenging activities by using the method of superoxide dismutase assay. Among 47 kinds of samples, the extract of Capsella bursa-pastoris showed a potent superoxide radical scavenging activity of 11.6 unit / mg solid and was selected for further studies. In order to select optimal extraction solvent system, Cappella bursa-pastoris was extracted with various solvents and the electron donating abilities and inhibitory effects on lipid peroxidation of linoleic acid were measured. Among them, the ethanol extract of Capsella bursa-pastoris possessed the highest level of activities. The ethanol extract of Cappella bursa-pastoris was found to have an inhibitory effect on autoxidation of soybean oil at 5$0^{\circ}C$ from peroxide value, conjugated diene value and refractive index. In the soybean oil containing 0.2% of ethanol extract, induction period was increased 2 times in comparison with control.

Radical Scavenging Activities of Phenolic Compounds Isolated from Mulberry (Morus spp.) Cake

  • A methanol extract of mulberry cake prepared from mulberry fruits (Morus spp.) was shown to have strong scavenging activities against DPPH, superoxide and hydroxyl radicals. Eleven phenolic compounds were isolated from the mulberry cake by a combination of Diaion HP-20, silica gel (or polyamide), Sephadex LH-20 column chromatographies, preparative HPLC and TLC. Their chemical structures were characterized as procatechuic acid (PCA), caffeic acid (CA), cyanidin 3-O-$\beta$-D-glucopyranoside (CyG) and cyanidin $3-O-\beta­D-rutinoside$ (CyR), rutin (RT), isoquercitrin (IQT), astragalin (AG), quercetin (QT), morin (MR), di-hydroquercetin (DHQ), and 4-prenylmoracin (PM) by spectral analysis and the published data. Most of the phenolic constituents were effective scavengers of DPPH, superoxide and hydroxyl radicals, and especially caffeic acid and 4-prenylmoracin showed potent superoxide and hydroxyl radical scavenging activity, in which their activities were higher than that of the well-known antioxidant, BHT (p< 0.05). Dehydroquercetin and quercetin also exhibited strong superoxide and hydroxyl radical scavenging activities. These results suggest that mulberry cake containing antioxidant phenolic compounds may be useful as natural antioxidants in functional foods and cosmetics.

홍화자 분획물의 사염화탄소 유발 간손상 흰쥐에서 지질과산화와 oxygen free radical 제거 효소 활성도에 미치는 영향 (Effect of Subfractions of Carthamus tinctorius L. Semen on the Lipid Peroxidation and Oxygen Free Radical Scavenging Enzyme Activities in CCl_4$-induced Hepatotoxic Rats)

  • Previous studies have shown that methanol extract and its butanol fraction of Carthamus tinctorius L. Semen have the hepatoprotective effect on the CCl4-induced hepatotoxicity. The hepatoprotective effect of the subfractions of butanol fraction has been evaluated by analyzing oxygen free radical scavenging enzyme activities and histopathological examinations. In BS-5 subfraction treated group, the activity of superoxide dismutase has been significantly increased as compared with that of CCl4, treated rats. Antioxidant activity has been evaluated by the examination of the scavenging activity against 1,1-diphenyl-2-picrylhydrazyl radical. BS-5 subfraction has shown strong antioxidant activities. The histopathological examination showed that the treatment of BS-5 subfraction has relieved the ballooning degeneration of hepatocytes which had been generated by CCl4. It appears that the protective effect of BS-5 subfraction would be mediated of the attenuation of lipid peroxidation by acting as a free radical scavenger, which were based on the increase of superoxide dismutase activity.

생강나무 추출물의 항산화 활성과 미백효과 (Antioxidant Activities and Whitening Effect from Lindera obtusiloba BL. Extract)

  • In this study we investigated antioxidant activity of against several free radicals and skin whitening effect of 70% ethanol extract (leaf extracts and branch/stem mixed) of Lindera obtusiloba BL. Antioxidant activity was assessed by DPPH, superoxide radical and hydroxyl radical assays. The Lindera obtusiloba BL. extract had antioxidant activity dose dependently with an ${IC}_{50}$ value of 243.14 and 181.10 ${\mu}g$/ml for DPPH, 165.77 and >1500 ${\mu}g$/ml for non-enzymatic system of superoxide radical assay, 35.47 and >100 ${\mu}g$/ml for enzymatic system of superoxide radical assay, 1.21 mg/ml for hydroxyl radical assay. In addition we tested tyrosinase inhibition activity and melanin contents on B16 melanoma F10. B16 melanoma cell was treated by such sample as 1, 5, 10 and 50 ${\mu}g$/ml for 72 hr and tyrosinase inhibition was tested. Melanogenesis was inhibited to 22% at the dose of 50 ${\mu}g$/ml and tyrosinase was inhibited to 45.2% at the same dose. In conclusion Lindera obtusiloba BL had potent antioxidant activity and inhibitory activity of tyrosinase and melanin formation. It could be developed as the health functional food and functional cosmetic resources.

오계란 단백질 가수 분해물 제조 및 한외여과 분획물의 in vitro 항산화 활성 특성 (In vitro Antioxidant Activity of Ogae (Korean Native Black Fowl) Egg White Protein Hydrolysates Fractionated by Ultrafiltration)

  • 식물 및 동물성 단백질 유래 펩타이드 형태의 단백질 가수 분해물들은 항산화, 고혈압 완화, 면역조절, 진통완화 및 항균작용 등 생리활성이 있는 것으로 알려져 왔다. 본 연구는 연산오계란 단백질 가수 분해물을 Ultrafiltration를 이용하여 HDS(분획되지 않은 가수 분해물), 1 kDa, 5 kDa, 10 kDa, 50 kDa로 분획된 기능성 펩타이드의 DPPH radical scavenging activity, superoxide anion radical scavenging activity, hydroxyl radical scavenging activity 및 $Fe^{2+}$ chelation ability을 평가하였다. 그 결과 DPPH radical scavenging activity 최대값은 1 kDa(70.83 %), hydroxyl radical scavenging activity 최대값은 5 kDa (47.01 %), superoxide anion radical scavenging activity 최대값은 5 kDa(40.57 %), $Fe^{2+}$ chelation ability 최대값은 5 kDa(29.87 %)로 나타났다. Ultrafiltration를 이용하여 fractionation된 단백질 가수 분해물의 항산화 저해 능력 $IC_{50}$ 평가하였다. 그 결과 HDS의 최대값은 superoxide anion radical scavenging activity($IC_{50}$, 5.42 mg/ml)이고, 1 kDa의 최대값은 $Fe^{2+}$ chelation ability($IC_{50}$, 1.67 mg/ml)이고, 5 kDa의 최대값은 $Fe^{2+}$ chelation ability($IC_{50}$, 2.09 mg/ml)이고, 10 kDa의 최대값은 $Fe^{2+}$ chelation ability($IC_{50}$, 2.61 mg/ml)이고, 50 kDa의 최대값은 $Fe^{2+}$ chelation ability($IC_{50}$, 4.53 mg/ml)이다. 그러므로 본 연구 결과를 바탕으로 5 kDa를 이용하여 오계란 단백질에서 분획한 펩타이드는 항산화 기능성 식품소재로서 활용할 가치가 높을 것으로 기대한다.

  • The superoxide anion radical$(O_2)$ poses a threat to macromocules and cell organelles of the living cells. This toxicity damage to all groups of proteins results in loss of enzyme function concerned with metabolism and ion transport, and peroxidation of unsaturated fatty acids and cholesterol results in a change of permeability characteristics of the membrane, and oxidative of nucleic acids results in genomic damage and thereby cause mutation, potential carcinogenesis and somatic damage that produce cellular aging Superoxide dismutase(SOD) has received substantial attention as a potential therapeutic agent. It has been investigated as a possible agent for the prevention of ontogenesis, the reduction of cytotoxic effect of anticancer drugs, and protection against damage in ischemic tissue. It is suggest that $O_2$ is concerned with cellular aging, thereafter we need to investigate herb that activated to SOD.

조간탕가감방(調肝湯加減方)의 항산화활성(抗酸化活性) 및 항염증작용(抗炎症作用)에 대한 실험적(實驗的) 연구(硏究) (The Experimental Study on Anti-oxidant and Anti-inflammatory Effect of Jogantanggagambang(JGTG))

  • Purpose: This study was performed to evaluate anti-inflammatory effects of Jogantanggagambang(JGTG). Methods: In the study of anti-oxidant activities, JGTG was investigated by DPPH radical scavenger activity, superoxide dismutase activity and superoxide anion radical scavenger activity. In the study of anti-inflammatory effects, JGTG was investigated using cultured cells and murine models. As for the parameters of inflammation, levels of several inflammatory cytokines and chemical mediators which are known to be related to inflammation were measured in mouse lung fibroblast cells(mLFCs) and RAW264.7 cells. Results: 1. JGTG showed a safety in cytotoxicity and toxicity of liver. 2. JGTG effected scavenging activity on DPPH free radical, superoxide dismutase and superoxide anion radical. 3. JGTG in RAW 264.7 cell decreased IL-$1{\beta}$ mRNA expression, IL-6 mRNA expression, TNF-${\alpha}$ mRNA expression at 50, $100{\mu}g/m{\ell}$ and also decreased NOS-II mRNA expression at $100{\mu}g/m{\ell}$, and decreased COX-2 mRNA expression at 10, 50, $100{\mu}g/m{\ell}$. 4. JGTG in RAW 264.7 cell decreased significantly IL-$1{\beta}$, IL-6 and TNF-${\alpha}$ at 50, $100{\mu}g/m{\ell}$. 5. JGTG inhibited significantly IL-$1{\beta}$, IL-6 and TNF-${\alpha}$ production in serum of acute inflammation-induced mice. 6. JGTG decreased significantly IL-$1{\beta}$ mRNA production in spleen tissue. Conclusion: These results suggest that JGTG can be used for treating diverse female diseases caused by inflammation
