Anti-oxidant and Anti-aging Activities of Essential Oils of Pinus densiflora Needles and Twigs (소나무 잎과 가지의 정유와 그 성분들의 항산화 및 항노화 작용)

  • To investigate effects of pine essential oil isolated by steam distillation from Pinus densiflora needles (PN) and twigs (PT) on anti-oxidant and anti-aging activities, the experiments were carried out to determine anti-oxidant and antiaging activities on DPPH radical scavenging activity, NBT/xanthine oxidase-superoxide scavenging activity, silica-induced intracellular $H_2O_2$ and hydroxyl radical generation in RAW264.7 cells, hyaluronidase and elastase activities in vitro. Essential oil of PN and PT were contained 0.225 and 0.176 (w/v) %, respectively. PN was contained with 30 kinds of essential oil and its major constituent is $\alpha$-pinene (21.5%). Further PT was contained with 40 kinds of essential oil and its major constituent is $\beta$-pinene (22.4%) in GC/MS assay. Other essential oils of PN were $\beta$-pinene, camphene, myrcene, limonene, terpinolene, $\alpha$-terpineol, bornyl acetate, $\alpha$-copaene, caryophellene and humulene and PT were $\alpha$-pinene, camphene, phellandrene, limonene, terpinolene, $\alpha$-terpineol, bornyl acetate, $\alpha$-copaene, caryophellene and humulene. The essential oil of PT have more active than that of PN in anti-oxidant activity which has significant DPPH radical and superoxide scavenging activity, and significant inhibitory activities on silica-induced intracellular $H_2O_2$ and hydroxyl radical generation, as well as, significantly inhibited elastase and hyaluronidase activities. Further, phellandrene, myrcene and $\alpha$-pinene have DPPH radical and superoxide scavenging activities, $\beta$-pinene, terpinolene, myrcene and phellandrene inhibited silica-induced intracellular $H_2O_2$ and hydroxyl radical generations. And also phellandrene and $\beta$-pinene inhibited hyaluronidase and elastase activities. In conclusion, the essential oils isolated from PN and PT have anti-oxidant and anti-aging activities.

Change of ROS Generation and Antioxidant Enzyme Activity of Flavonol Quercetin in the Presence of Vitamin E, L-Ascorbit acid, Reduced Glutathione on the B16F10 Murine Melanoma Cells (B16F10 세포에서 Quercetin과 Vitamin E, L-Ascorbic acid, 환원형 글루타치온과의 병용 투여에 의한 활성산소종 발생과 항산화 효소의 활성 변화)

  • It has been known that quercetin, a bioflavonoid widely distributed in fruits and vegetables as dietary-derived flavonoid and exert significant multiple biological effects such as antioxidant and anti-inflammatory, anti-tumor effects. In addition, it has been shown to have a chemoprotective role in cancer, though complex effects on signal transduction involved in cell proliferation and angiogenesis. The present study investigated whether quercetin can enhance antioxidant enzyme activity (glutathione peroxidase: GPx, superoxide dismutase: SOD, catalase: CAT) and regulate the reactive oxygen species (ROS) generation in the presence of vitamin E, L-ascorbic acid, reduced glutathione (GSH) on B16F10 murine melanoma cells. After 48h treatment of cells with quercetin in the presence of vitamin E, L-ascorbic acid, GSH, we measured the cytotoxicities by MTT assay. The cells exhibited a dose-dependent inhibition in their proliferation in the presence of vitamin E, L-ascorbic acid, GSH respectively. We also investigated the effects of antioxidant enzyme activity and ROS generation. The antioxidant enzyme activity of quercetin in the presence of vitamin E was stronger than GSH, L-ascorbic acid, the same treatments decreased ROS generation in B16F10 murine melanoma cells. Taken together, these result demonstrate that the antioxidant effect of quercetin can enhanced in the presence of vitamin E and it might plays an important role in anti-oxidative effects.

Antioxidative Characteristics of Extracts from Aromatic Herb Elsholtzia splendens

  • We investigated the antioxidant activity of ethanol extracts obtained from the flowers of Elsholtzia splendens on Chinese hamster ovary CHO-K1 cells. When cells were treated with E. splendens extracts (ESEs), low concentrations (<12.5 ug/mL) of stimulated cell proliferation via radical generation. Relative mRNA expression of Cu/Zn-superoxide dismutase (SOD) and Mn-SOD in cells exposed to ESEs (1-20 ug/mL) was significantly induced in a dose-dependent manner (p<0.05). In the case of catalase, ESEs had opposing effects; that is, a low-level treatment caused a decrease, and a high-level treatment induced elevated levels (p<0.05). The results demonstrated that components of ESEs exhibit potential antioxidant properties. Also, further studies are required to clarify the active components of Elsholtzia splendens extracts responsible for such biological activities.

Role of Calcium and Protein Kinase C in Platelet Activating Factor-induced Activation of Peritoneal Macrophages (Platelet Activating Factor에 의한 대식세포의 활성화에 있어서 칼슘과 Protein Kinase C의 역할)

  • Particulate or soluble stimuli appear to stimulate phagocytic cell's response by the change of $Ca^{2+}$ mobilization and by the activation of protein kinase C. In contrast, it is reported that activation of protein kinase C could attenuate agonist-stimulated elevation of $Ca^{2+}i$ in neutrophils. PAF elicited an increase of $Ca^{2+}i$ in peritoneal macrophages in a dose dependent fashion and $Ca^{2+}$ extrusion was accompanied. PAF-induced elevation of $Ca^{2+}i$ was not affected by TMB-8, verapamil and TTX. TEA stimulated PAF-induced mobilization of $Ca^{2+}i$ and delayed lowering of $Ca^{2+}i$. Five mM EGTA almost completely inhibited PAF-induced mobilization of $Ca^{2+}i$. After the addition of PAF, membrane permeability was markedly increased up to 5 min and then slowly increased. PAF-induced LDH release was slightly decreased by EGTA plus TMB-8. PAF-stimulated superoxide generation was inhibited by EGTA, TMB-8 and verapamil but not affected by TTX and TEA. PAF-induced elevation of $Ca^{2+}i$, increased membrane permeability and superoxide generation were inhibited by IQSP, chlorpromazine and propranolol. PAF-induced LDH release was significantly inhibited by chlorpromazine and minimally decreased by propranolol. After the pretreatment with PMA, the stimulatory effect of PAF on the elevation of $Ca^{2+}i$ and LDH release in macrophages was significantly decreased. These results suggest that PAF may exert the stimulatory action on peritoneal macrophages of mouse by the elevation of $Ca^{2+}i$ and by the activation of protein kinase C. Preactivation of protein kinase C appears to attenuate the stimulatory action of PAF on macrophage response.

The Anti-Oxidative Effects of Oldenlandiae Diffusae Herba Extract on Oxidative Hepatic Injury (간세포(肝細胞)의 산화적 손상에 대한 백화사설초(白花蛇舌草)의 항산화효과)

  • Objective : This study was designed to investigate the anti-oxidative effects of Oldenlandiae Diffusae Herba Water extract (ODHW) on lipid peroxidation by free radicals oxidative hepatic injury. Methods : In order to evaluate anti-oxidative activities of ODHW in the liver cell, cultured normal rat liver cells(Ac2F) were incubated with or without ODHW. After 16 hours to 18 hours of experiment, cells were placed in DMEM medium without serum, and then incubated with 1mM tert-butyl hydro-peroxide(t-BHP) for two hours. Viable cells were detected by MTT assay. The levels of LPO induced by hydroxyl radical derived from H2O2-Fe2+ system in rat liver homogenate were determined by means of TBA. Inhibitory effect of ODHW on superoxide generation was measured by xanthine-xanthine oxidase system. Results : In the linoleic acid autoxidation system, ODHW exhibited antioxidant activity, which inhibited 85% of linoleic acid peroxidation. These effects were similar to those of dl-a-tocopherol. ODHW showed scavenging effects on DPPH radical, inhibited superoxide generation in xanthine-xanthine oxidase system, and also inhibited lipid peroxidation of rat liver tissue with hydroxyl radical derived from $H_2O_2-Fe^{2+}$ system. In addition, ODHW protected the cell death induced by t-BHP and it significantly increased cell viability in a normal rat liver cell(Ac2F)

Dietary Ascorbate Supplementation Reduces Oxidative Tissue Damage and Expression of iNOS in the Kidney of Streptozotocin Induced Diabetic Rats

  • Reactive oxygen species (ROS) have been suggested to be contributory factors in complications of diabetes mellitus. In the present study, we investigated the generation of superoxide, the lipid peroxide level measured as thiobarbituric acid reactive substances, the vasorelaxation of isolated thoracic aorta and the iNOS expression in kidney of streptozotocin induced diabetic rats. Sprague Dawley rats were divided into four groups: control, ascorbate (400 mg/kg rat weight daily in drinking water), diabetic (single dose of 50 mg of STZ/kg i.p.) and diabetic simultaneously fed with ascorbate for 12 wk. Rats in groups were studied at tri-weekly intervals (0 to 12 wk). Diabetic rats were evaluated periodically with changes of plasma glucose levels and body weight. The ascorbate supplimentation attenuated the development of hyperglycemia and weight loss induced by STZ injection in rats. In the present experimental condition, the ascorbate supplimentation had no significant effect on plasma glucose levels and changes in body weight of normal rate. The superoxide generation, formation of thiobarbituric acid reactive substance and iNOS expression in kidney were significantly increased in STZ-treated rats that were decreased by ascorbate supplimentation. The ascorbate supplimentation had no effect on vasorelaxation of isolated thoracic aorta. These results indicate that ascorbate supplimentation may exert an inhibitory effect on STZ-induced oxidative tissue damage through protection of pancreatic islet cells by scavanging reactive oxygen species. The ascorbate supplimentation may possibly attenuate the renal complication of diabetes mellitus.

Protective effect of ginsenosides Rk3 and Rh4 on cisplatin-induced acute kidney injury in vitro and in vivo

  • Background: Nephrotoxicity is the major side effect in cisplatin chemotherapy. Previously, we reported that the ginsenosides Rk3 and Rh4 reduced cisplatin toxicity on porcine renal proximal epithelial tubular cells (LLC-PK1). Here, we aimed to evaluate the protective effect of ginsenosides Rk3 and Rh4 on kidney function and elucidate their antioxidant effect using in vitro and in vivo models of cisplatin-induced acute renal failure. Methods: An enriched mixture of ginsenosides Rk3 and Rh4 (KG-KH; 49.3% and 43.1%, respectively) was purified from sun ginseng (heat processed Panax ginseng). Cytotoxicity was induced by treatment of $20{\mu}M$ cisplatin to LLC-PK1 cells and rat model of acute renal failure was generated by single intraperitoneal injection of 5 mg/kg cisplatin. Protective effects were assessed by determining cell viability, reactive oxygen species generation, blood urea nitrogen, serum creatinine, antioxidant enzyme activity, and histopathological examination. Results: The in vitro assay demonstrated that KG-KH ($50{\mu}g/mL$) significantly increased cell viability (4.6-fold), superoxide dismutase activity (2.8-fold), and glutathione reductase activity (1.5-fold), but reduced reactive oxygen species generation (56%) compared to cisplatin control cells. KG-KH (6 mg/kg, per os) also significantly inhibited renal edema (87% kidney index) and dysfunction (71.4% blood urea nitrogen, 67.4% creatinine) compared to cisplatin control rats. Of note, KG-KH significantly recovered the kidney levels of catalase (1.2-fold) and superoxide dismutase (1.5-fold). Conclusion: Considering the oxidative injury as an early trigger of cisplatin nephrotoxicity, our findings suggest that ginsenosides Rk3 and Rh4 protect the kidney from cisplatin-induced oxidative injury and help to recover renal function by restoring intrinsic antioxidant defenses.

Antioxidant Activities of Phenolic Derivatives from Dipsacus asper Wall. (II)

  • The six phenolic compounds isolated from the root of Dipsacus asper, 3,4-di-O-caffeoylquinic acid (1), methyl 3,4-di-O-caffeoyl quinate (2), 3,5-di-O-caffeoylquinic acid (3), methyl 3,5-di-O-caffeoyl quinate (4), 4-5-di-O-caffeoylquinic acid (5), methyl 4,5-di-O-caffeoyl quinate (6) were continuously evaluated for their antioxidant activity using superoxide radical scavenging and AAPH-mediated (LDL) oxidation assay. The results demonstrated that compounds 1 - 6 had remarkable antioxidant activities with the $IC_50$ values ranging from 12.0 to $2.8{\mu}M$ in superoxide radical scavenging. They also inhibited AAPH-mediated low-density lipoprotein LDL oxidation by the generation of thiobarbituric acid reactive substances (TBARS) with $IC_50$ ranging from 6.7 to $8.7{\mu}M$.

Antioxidant and Protective Effects of Palmul-tang on Ultraviolet B (UVB)-induced Damage in Human Keratinocytes (팔물탕의 항산화 효과와 자외선으로 유도된 각질형성세포 손상에 대한 보호효과)

  • Objective : In this paper, we investigated the anti-oxidative capacities and protective effects of water extract of palmul-tang (PMT) against Ultraviolet B(UVB)-induced oxidative damage in human keratinocytes(HaCaT). Method : To evaluate the anti-oxidative activities of PMT, we measured scavenging activities on 1,1-diphenyl-2-picrylhydrazyl(DPPH) radical, hydroxyl radical, hydrogen peroxide, superoxide anion, lipid peroxidation and reducing power of PMT. To give an oxidative stress to HaCaT cells, UVB was irradiated with $40mJ/cm^2$ to HaCaT cells. To detect the protective effects of PMT against UVB, we measured cell viability, apoptotic bodies and reactive oxygen species(ROS) generation. Results : PMT showed the anti-oxidative activities by scavenging DPPH radical, hydroxyl radical, hydrogen peroxide, superoxide anion, lipid peroxidation. Also PMT showed high reducing values. The UVB-induced oxidative conditions led to the cell apoptosis. However, treatment with PMT reduced oxidative stress conditions, including inhibition of cell apoptosis and expression of ROS. Conclusion : PMT had anti-oxidative activities and exhibited protective effects against UVB on HaCaT cells. PMT would be useful for the development of cosmetics treating UVB-induced skin aging.
