  • A marine bacterium producing superoxide dismutase, strain number B446, was screened with nitrite quantitation method using hydroxy amine and photochemically generated superoxide ion, and the superoxide dismutase was purified through 35-75% ammonium sulfate precipitation, DEAE-Sephadex A-25 ion exchange chromatography, Sephadex G-200 gel filtration chromatography, and High-Q anion exchange chromatography to a yield of 6% and purification fold of 32.3.

Identification and Molecular Characterization of Superoxide Dismutase Genes in Pseudomonas rhodesiae KK1 Capable of Polycyclic Aromatic Hydrocarbon Degradation (PAH를 분해할 수 있는 Pseudomonas rhodesiae KK1의 SOD 유전자의 동정 및 분자학적 특성 분석)

  • Pseudomonas rhodesiae KK1 has been reported to degrade polycyclic aromatic hydrocarbons (PAHs), such as anthracene, naphthalene, and phenanthrene, which are considered major environmental contaminants. Interestingly, antioxidant genes, including superoxide dismutase, are known to be expressed at different levels in response to environmental contaminants. This study was performed to identify the superoxide dismutase gene in strain KK1, which may be indirectly involved with degradation of PAHs, as well as to investigate the expression pattern of the superoxide dismutase gene in cells grown on different PAHs. Two types of superoxide dismutase genes responsible for the antioxidant defense mechanism, Mn-superoxide dismutase (sodA) and Fe-superoxide dismutase (sodB), were identified in P. rhodesiae KK1. The sodA gene in strain KK1 shared 95% similarity, based on 141 amino acids, with the Mn-sod of P. fluorescens Pf-5. The sodB strain, based on 135 amino acids, shared 99% similarity with the Fe-sod of P. fluorescens Pf-5. Southern hybridization using the sod gene fragment as a probe showed that at least two copies of superoxide dismutase genes exist in strain KK1. RT-PCR analysis revealed that the sodA and sodB genes were more strongly expressed in response to naphthalene and phenanthrene than to anthracene. Interestingly, sodA and sodB activities were revealed to be maintained in cells grown on all of the tested substrates, including glucose.

Secretion of the iron containing superoxide dismutase of Streptomyces subrutilus P5 (Streptomyces subrutilus P5가 생산하는 철 함유 superoxide dismutase의 분비)

  • We tried to analyze the growth time for secretion of the iron containing superoxide dismutase by comparing the intra-and extracellular enzyme activity from Streptomyces subrutilus P5 and analyze possible genetic information for this enzyme secretion. The mycelial dry weights and glucose concentrations in culture filtrates were determined during growth. Glucose was consumed rapidly during logarithmic growth phase and almost exhausted at 24 h of cultivation. While the intracellular activity of iron containing superoxide dismutase was first appeared at three hours, the extracellular activity of this enzyme appeared from 7.5 h of cultivation, early logarithmic growth phase. This early presence of the superoxide dismutase might not be the result of cell lysis but active secretion pathway. There was no information for signal peptide responsible for the enzyme secretion in sodF. However, we found a type three secretion box in the promoter region of sodF that has been known for the genes of type III secreted proteins in other bacteria. This is the first report on the possible existence of type III secretion in Streptomyces.

ALTERATION OF SUPEROXIDE DISMUTASE-AND CATALASE-ACTIVITY IN HUMAN GINGIVAL TISSUES BY THE PERIODONTAL DISEASE SEVERITY (치주질환 심도에 따른 치은조직내의 Superoxide Dismutase와 Catalase의 활성변화에 관한 연구)

  • It has been believed that antioxidant enzymes such as CuZn- and Mn-superoxide dismutase and catalase protect the tissue from damage resulting from the oxygen derived free radicals($O_2\;^-$, $H_2O_2$ and OH ). The purpose of this study was to investigate the relationship between activity of antioxidant enzymes including CuZn- and Mn- superoxide dismutase and catalase and inflammatory periodontal disease and periodontal parameters. For this study, the patients were classified into normal, gingivitis, adult periodontitis and rapidly progressive periodontitis, and then their papillary bleeding index(PBI) and probing depth were checked. Gingival tissues were surgically obtained from the patients during periodontal surgery, extraction, and clinical crown lengthening procedure. The activity of CuZn- and Mn- superoxide dismutase and catalase in the gingival tissues was measured by using UV-spectrophotometer by the same methods as Crapo et al. And Aebi did, respectively. The results were as follows : 1. CuZn- and Mn- and total-superoxide dismutase activity were significantly low in rapidly progressive periodontitis group in comparison to normal group (P<0.05). 2. In comparison of the antioxidant enzyme activity according to papillary bleeding index group(PBI), Mn-superoxide dismutase activity only was significantly lower in PBI 2, 3, and 4 groups than PBI 0 group(P<0.05). 3. Superoxide dismutase activity failed to show any significant difference according to probing depth. But significant]y high catalase activity was shown in deep pocket group (${\ge}7mm$)(P<0.05). In conclusion, these results suggest that the activity of Mn-superoxide dismutase among the antioxidant enzymes may reflect the inflammatory status of gingival tissue and that the decreased activity of superoxide dismutase may be one of responsibe factors for progression of rapidly progressive periodontitis.

  • Culexpfpfens pollens에 존재하는 superoxide dismutase의 연구를 위한 적정 분석 조건과 우화 전후. 시간 경과에 따른 superoxide dismutase 활성 경향에 대해서 연구하였다. C. pipiens에 존재하는 superoxide dismutase의 활성은 부화 직후부터 지속적으로 감소하다가. 우화 후 흡혈 자극에 의해서 급격하게 증가한다 흡혈 후 36시간이 경과했을 때. 최대의 논성도를 보인 후 급격히 감소하였다. 기관별 분석에서 흡혈 후 가장 높은 활성 증가는 중장에서 일어나고, 머리 흥부. 지꼴체 및 난소에서도 활성의 증가가 나타났다. 흡혈 전에는 2개의 동위효소(SOD-1, soD-2)가 존재하였으나. 흡혈후 36시간된 성체에서는 3개의 동위효소 밴드가 보이는데, 머리에서는 SOD-1만이 나타나고, 가슴에서는 SOD-2. 지방체와 난소에서는 SOD-1과 SOD-2이 존재하였고. 중장에서는 SOD-3가 나타났다. 그러므로 흡혈 후 나타나는 새로운 동위효소는 중장에서 합성됨을 알 수 있다.

  • To investigate the effect of hyperbaric oxygenation on superoxide dismutase activity, neonatal rats (7-10 days old) and adult rats (approximately 100 days old) were continuously exposed to hyperbaric oxygen environment of 2.4ATA for 8 hours and their superoxide dismutase activity were measured. Neonatal rats, all survived through exposure, showed significant increases in the pulmonary superoxide dismutase activity at immediately and 24 hours after exposure. Adult rats, whose 8 hour survival rates were 14%, did not show any significant increase in the activity of pulmonary superoxide dismutase as compared to the control adult rats. These findings are indicating that increased tolerance to oxygen toxicity in neonatal animals during exposure may be attributed to the increase in activity of superoxide dismutase in neonatal rats.

  • Production of free radicals of superoxide anion in tissues by cadmium, activities of superoxide dismutase and catalase to protect tissue damages caused by the free radicals and ATPase that plays an important role in energy metabolism at cellular level were investigated. Experiments in vivo were conducted with liver, kidney and testicle tissue homogenates of rats adding $0.05{\sim}0.50mM$ cadmium chloride, and in vivo experiments administering single dose of 5 mg of cadmium/kg of body weight in 0.1% cadmium chloride solution intraperitoneally 48 hours prior to evisceration. Production of superoxide radicals in liver and testicle increased with addition of cadmium in vitro, but not in kidney. In vivo experiments, however, superoxide radicals slightly increased in liver and kidney but not in testicle. Superoxide dismutase (Cu, Zn-SOD and Mn-SOD), catalase and ATPase (total, $Mg^{++}-\;&\;Na^+,\;K^+-$) activity decreased in the presence of cadimium in dose dependent manner. Reduction of these enzyme activities varied not only with dosage of cadmium but also with type of tissue and between in vitro and in vivo experiment.

Purification and Characterizatlon of a Cu, Zn-Superoxide Dismutase from Adult Paragonimus westermani (폐흡충 성충 Cu, Sn-Superoxide Dismutase의 정제 및 생화학적 특성)

  • In cytosolic (raction of adult Paragonimus westermani, superoxide dismutase activity was identified (4.3 units/mg of specific activity) using a xanthine-xanthine oxidase system. The enzyme was purified 150 fold in its activity using the ammonium sulfate precipitation, DEAE-Trisacryl M anion-exchange chromatography and Sephadex G-100 molecular sieve chromatography. The enzyme exhibited the enhanced activity at pH 10.0. The enzyme activity totally disappeared in 1.0mM cyanide while it remained 77.8% even in 10 mM azide. These findings indicated that the ensyme was Cu, Zn-SOD type. Molecular mass of the enzyme was estimated to be 34 kDa by gel filtration and 17 kDa on reducing SDS-polyacrylamide gel electrophoresis which indicated a dimer protein.

  • Effects of exposure to a neurotoxicant, fenitrothion on antioxidant enzyme activities in Chironomus riparius Mg. (Diptera, Chironomidae) larvae were evaluated under laboratory conditions. Exposure to this chemical led to an increase of cupper, zinc type superoxide dismutase and manganese type superoxide dismutase activities and to a decrease of glutathion peroxidase activity. An activation of catalase was observed in the larvae exposed to high fenitrothion concentration. The response of superoxide dismutase was rapid and sensitive to low chemical concentrations, but changes in catalase, total peroxidase and glutathion peroxidase were less sensitive. In this study, antioxidant enzyme activities in Chironomus riparius larvae were identified as pertinent biomarkers for environmental monitoring.

  • This study was designed to observe the effect of tohiene pretreatment on leukocyte variation in whole blood and the oxygen free radical generating, scavenging enzyme activities in neutrophil of bacteria infected rats. Toluene was administered 7 times intraperitoneally at levels of 9.45 mM/kg body weight to the rats and then infected with S. aureus $2\times10^7$ cfu/ml. The toluene treated-rats showed the significantly decreased numbers of lymphocyte and monocytes, but the similiar numbers of neutrophils with the control. Furthermore the increased neutrophils in blood of bacteria infected rats were reduced by the toluene pretreatment. Concomitantly the increased activities of xanthine oxidase and superoxide dismutase in neutrophil of bacteria infected rats were also decreased by the toluene pretreatment. On the other hand, injection of benzaldehyde to rats also led to similiar results in the count of leukocytes, xanthine oxidase and superoxide dismutase activities of neutrophil with those of toluene treated rats. These data suggest that toluene and its intermediate metabolite, benzaldehyde influence on the phagocytosis and defence mechanism of neutrophil.
