The Treatment Effect of Ulcerative Colitis of Supercritical Heat-Treated Radish Extracts

  • With the recent rapid improvement in the standards of life and westernization of dietary lifestyles, the consumption of high-calorie diets such as high-fat and high-protein red meat and instant foods has increased, while less vegetables containing dietary fiber are consumed. In addition to that, stress, erroneous dietary behaviors, and contaminated environments are linked to the risk of developing ulcerative colitis, which is on the rise. Another cause of ulcerative colitis is that involve laxative abuse, including repeated, frequent use of laxatives, and include such conditions as deteriorated bowel function, irritable bowel syndrome, diarrhea, intestinal inflammation, etc. The present study aimed to investigate the comparative evaluation of pharmacological efficacy between sulfasalazine alone and combination with herbal medicine on dextran sodium sulfate (DSS)-induced UC in mice. Balb/c mice received 5% DSS in drinking water for 7 days to induce colitis. Animals were divided into five groups (n = 9): group I-normal group, group II-DSS control group, group III-DSS + sulfasalazine (30 mg/kg), group IV-DSS + sulfasalazine (60 mg/kg), group V-DSS + sulfasalazine (30 mg/kg) + Radish Extract mixture (30 mg /kg) (SRE). DSS-treated mice developed symptoms similar to those of human UC, such as severe bloody diarrhea and weight loss. SRE supplementation, as well as sulfasalazine, suppressed colonic length and mucosal inflammatory infiltration. In addition, SRE treatment significantly reduced the expression of pro-inflammatory signaling molecules through suppression both mitogen-activated protein kinases (MAPK) and nuclear factor-kappa B (NF-κB) signaling pathways, and prevented the apoptosis of colon. Moreover, SRE administration significantly led to the up-regulation of antioxidant enzyme including SOD and Catalase. This is the first report that Radish extract mixture combined with sulfasalazine protects against experimental UC via the inhibition of both inflammation and apoptosis, very similar to the standard-of-care sulfasalazine.

Effects of Ethanol Addition on the Efficiency of Subcritical Water Extraction of Proteins and Amino Acids from Porcine Placenta

  • In a previous study, hydrolysates of porcine placenta were obtained and the extraction efficiency for proteins and amino acids was compared between sub- and super-critical water extraction systems; optimum efficiency was found to be achieved using subcritical water ($170^{\circ}C$, 10 bar). In this study, the effects of adding ethanol to the subcritical water system were investigated. The lowest-molecular-weight extraction product detected weighed 434 Da, and the efficiency of extraction for low-molecular-weight products was increased when either the concentration of ethanol was decreased, or the extraction time was lengthened from 10 min to 30 min. The highest concentration of free amino acids (approximately 8 mM) was observed following 30 min extraction using pure distilled water. The concentration of free amino acids was significantly lower when ethanol was added or a shorter extraction time was used (p<0.05). Color change of the solution following extraction was measured. There were no significant differences in color between lysates produced with different extraction times when using distilled water (p>0.05); however, using different extraction times produced significant differences in color when using 20% or 50% ethanol solution for subcritical extraction (p<0.05). The range of pH for the hydrolysate solutions was 6.4-7.5. In conclusion, the investigated extraction system was successful in the extraction of $\leq$ 500 Da hydrolysates from porcine placenta, but addition of ethanol did not yield higher production of low-molecular-weight hydrolysates than that achieved by DW alone.

Properties of non-cement mortars with small addition of alkali activator using fly ash and fused waste slag (석탄회 및 용융폐기물 슬래그에 소량의 알칼리 활성화제를 첨가한 무시멘트 모르타르의 특성)

  • Recently the world wide efforts reduce occurrence of $CO_2$; global warming main reason. The aim of this study is to improve recycling rate of the fly ash (FA) and fused waste slag (FWS) from the power plant and to carbonate under supercritical condition ($40^{\circ}C$, $80kgf/cm^2$ pressure, 60 min) for $CO_2$ fixation. Specimens of mortar with various mixing ratios of FA, FWS (from 100:0 to 20:80 in 5 steps of 20 % reduction each time), distilled water and 3 M NaOH alkali activators were prepared. As a result, the proportion of weight change ratio increases with CaO content, to 12 % after carbonation under the supercritical condition. There is difference of compressive strength between the carbonated and the alkali activator mortar specimens. The stabilization of $CO_2$ fixation through carbonation which could confirm the applicability of the eco-friendly materials without loss of compressive strength.

Establishment of the Extraction Process by Evaluation of Proanthocyanidin Contents and Antioxidative Activities of Pine (Pinus densiflora) Needle Extracts. (적송잎 추출물의 proanthocyanidin 함량 분석 및 항산화 활성 검증을 통한 추출공정 확립)

  • We evaluated proanthocyanidin contents out of total polyphenolic compounds in two pine needle extracts prepared by combined extraction methods and compared their antioxidative activities. Amounts of total polyphenolic compounds of sub-supercritical hotwater precipitate (SSHP) and sub-supercritical hotwater supermatant (SSHS) were 225.4 and 116.6 mg/g, respectively. Amounts of proanthocyanidin of SSHP and SSHS were 9.9 and 3.6 mg/g, respectively. Thus, the ratio of proanthocyanidin contents over total polyphenolic compounds of SSHP and SSHS were 4.3 and 3.9%, respectively. Antioxidative activities ($IC_{50}$) of SSHP and SSHS measured by DPPH radical scavenging were 41.5 and $50.7\;{\mu}g/ml$, respectively. These results indicates that two combined extracts, SSHP and SSHS showed higher antioxidative activities than extract prepared by hot water or sub-supercritical. In conclusion, extraction procedures for SSHP and SSHS could be useful methods for preparing the effective components of functional food products.

Preparation of Microparticulate Itraconazole/Hydroxypropyl-$\beta$-cyclodextrin Inclusion Complexes Using a Supercritical Anti-Solvent [SAS] Process (초임계 역용매 공정을 이용한 Itraconazole/Hydroxypropyl-$\beta$-cyclodextrin 포접복합체 미세입자 제조)

  • Microparticles of an inclusion complex between itraconazole and 2-hydroxypropyl-${\beta}$-cyclodextrin (HP-${\beta}$-CD) were prepared using an environmentally-benign supercritical anti-solvent (SAS) process. In order to evaluate the degree of complexation, the thermal behavior of solid micro particulate complexes was investigated using differential scanning calorimetry. The experimental results obatined for the solubility and dissolution rate of the microparticulate inclusion complexes in a buffer solution of pH 1.2 showed that the complexation of itraconazole with HP-${\beta}$-CD results in a significant increase in the solubility and dissolution rate of itraconazole. For the microparticulate itraconazole/Hp-${\beta}$-CD inclusion complexes prepared by the SAS process, about 80% of itraconazole was found to dissolve in the buffer solution. Our experimental results confirmed that the SAS process is a promising method for the preparation of microparticles of water-insoluble drug/cyclodextrin inclusion complexes.

Permeability and Strength of Cements Exposed to Supercritical CO2 for Varying Periods (초임계 CO2 - 시멘트 반응 전후의 투수율 및 강도 변화)

  • Chemical reaction tests were performed to assess the properties of hardened specimens of cement pastes (KS-1 Portland and Class G) exposed to supercritical CO2 for 1, 10, and 100 days. After exposure, the samples' measured permeability and strength were compared with values measured for pristine samples. The pristine cements had permeabilities of 0.009~0.025 mD, which increased by one order of magnitude after 100 days of exposure (to 0.11~0.29 mD). The enhancement of permeability is attributed to the stress release experienced by the samples after removal from the pressure vessel after exposure. Despite its enhancement, the measured permeability mostly remained lower than the API (American Petroleum Institute) recommended maximum value of 0.2 mD. The degradation of the cement samples due to exposure to supercritical CO2 led to a layer of altered material advancing inwards from the sample edges. The Vickers hardness in the altered zone was much higher than that in the unaltered zone, possibly owing to the increase in density and the decrease in porosity due to the carbonation that occurred in the altered zone. Hardness close to the edge within the altered zone was found to have decreased significantly, which is attributed to the conversion of C-S-H into less-strong amorphous silica.

Physicochemical properties of supercritical carbon dioxide defatted mealworm (Tenebrio molitor) powder and protein isolate (초임계이산화탄소 탈지 밀웜(Tenebrio molitor) 분말 및 분리단백의 이화학적 품질 특성)

  • Supercritical carbon dioxide (SCO2) extraction was applied for the defatting of mealworm to prepare defatted powder (DP) and protein isolate (PI) and compare the process to press and hexane extraction, with respect to DP and PI physicochemical properties. SCO2 DP was obtained by extracting 34.40% oil at 41.37 MPa, 40℃ for 180 min, and the product contained 71.66% crude protein, which is similar to that of hexane DP and higher than that of press DP. In using alkali protein extraction to prepare PI from DP, SCO2 was as effective as hexane and better than press. SCO2 produced brighter DP and PI than press, but not as much as hexane. Protein solubility was similar in all DP, with minimum values at pH 5. The highest water adsorption capacity was noticeable for SCO2 PI, and SCO2 DP showed an oil adsorption capacity comparable to that of hexane DP. SCO2 DP and PI had better foaming capacity than press DP and PI and showed superior emulsion activity compared to others.

  • The importance of nanopore structures of carbons is shown in terms of interaction potential for various molecules including supercritical gases such as $H_2$ and $CH_4$. The key factors for adsorption of supercritical $H_2$ and $CH_4$ are shown for single wall carbon nanohorn, single wall carbon nanotube, and double wall carbon nanotube. The cluster formation of molecules is a key process for water adsorption on hydrophobic carbon nanopores. The X-ray absorption spectroscopic examination elucidates an explicit dehydration structure of ions confined in carbon nanopores.

Comparison of Gingerols in Various Fractions and the Antioxidant Effects of Supercritical Fluid Extracts from Ginger (생강 분획의 gingerols 분석 및 초임계 추출물의 항산화 효과)

  • Ginger (Zingiber officinale Rosc.) is a well known edible plant that is traditionally used to treat various illnesses related to inflammation and oxidative stress. Steam distillated ginger oil or water extract are mainly used for related products; however, it is unclear whether these fractions contain most of the bioactive compounds or the highest efficacy. This investigated the antioxidant effects of extracts prepared by supercritical fluid extraction (SFE). 6-Gingerol was the most abundant component in hexane fraction of ethanol extract from ginger. The antioxidative properties of SFE oil and Marc ethanol fractions were demonstrated using the 2,20-diphenylpicrylhydrazyl (DPPH) and 2,2'-azinobis 3-ethyl benzothiazoline-6-sulfonic acid (ABTS) radicals scavenging assays. A clear correlation was observed between total polyphenol contents and $RC_{50}$ values in the Marc ethanol fraction. These results indicate that not only SFE oil but the marc after SFE could be good sources for the food industry.

  • As global warming became a worldwide issue, a significant effort has been made on the development of technology related to $CO_2$ capture and storage. Geologic sequestration of $CO_2$ is one of those technologies for safe disposal of $CO_2$. Geologic sequestration stores $CO_2$ in the form of supercritical fluid into the underground site surrounded by solid rock, and concrete is used for prevention of $CO_2$ leakage into the atmosphere. In such case, concrete may experience severe damage by attack of supercritical $CO_2$, and especially in contact with underground water, very aggressive form of carbonation can occur. In this work, to prevent such deterioration in concrete, calcium phosphates were added to the portland cement to produce hydroxyapatite, one of the most stable mineral in the world. Temperature rise, viscosity, set and stiffening, and strength development of cement paste incorporating three different types of calcium phosphates were investigated. According to the results, it was found that the addition of calcium phosphate increased apparent viscosity, but decreased maximum temperature rise and 28 day compressive strength. It was found that monocalcium phosphate was found to be inappropriate for portland cement based material. Applicability of dicalcium and tricalcium phosphates for portland cement needs to be evaluated with further investigation, including the long term compressive strength development.
