Optimal conditions for the supercritical carbon dioxide $(scCO_{2})$ extraction of glycyrrhizin from licorice (Glycyrrhiza glabra) were investigated, with an emphasis on the types and levels of modifiers. The morphology of the licorice tissue remaining after the $scCO_{2} $ extraction of glycyrrhizin was examined by scanning electron microscopy, coupled with measurements of absolute density. Conventional organic solvent extraction was also carried out for purpose of quantitative comparison. At 50 MPa and $60^{circ}C$ glycyrrhizin could not be extracted with pure $scCO_{2}$, while a considerable amount of glycyrrhizin was extracted when water was added to $scCO_{2}$ as a modifier. The highest recovery was found to be about $97\%$ when $70\%$ aqueous methanol was added to $scCO_{2}$ at a concentration of $15\%$. The optimal pressure and temperature for the supercritical fluid extraction of glycyrrhizin were observed to be 30 M Pa and $60^{circ}C$, respectively. Under these conditions, the percentage recovery of glycyrrhizin attained a maximum value of 102.67\pm$$1.13\%$ within 60 min. Furthermore, in the case of $scCO_{2}$ modified with $70\%$ aqueous methanol, the licorice tissue obtained after extraction was found to be severely degraded by excessive swelling, and the absolute density of the licorice residues was observed to be the highest.
Various factors affecting the supercritical carbon dioxide extraction of daidzein from soybean were studied. Daidzein was not extracted with pure supercritical carbon dioxide. The ethanol was an efficient modifier for supercritical carbon dioxide to extract daidzein. The extraction efficiency increased as the pressure increased up to 300 bar. At $35^{\circ}C$ and 300 bar, 93% of daidzein was extracted with supercritical carbon dioxide modified with 15% of ethanol.
Polyacetylenes were extracted from Korean ginseng using supercritical $CO_2$ Yield of extraction of panaxydol and panaxynol was increased as the pressure of supercritical $CO_2$ increased at the range from 200 to 300 bar. The optimal yields of panaxydol and panaxynol was achieved at 65 and $55^{\circ}C$, respectively. Methanol was applied as a modifier. The highest yield of panaxydol and panaxynol were 0.230 and 0.054 mg/g-dry weight at modifier concentration of 10%(w/w), 300 bar, and $65^{\circ}C$. When these results were compared to that of methanol-extraction, the recoveries of panaxydol and panaxynol by supercritical $CO_2$ extraction in SFE were 37.8 and 55.1%, respectively.
Acer tegmentosum is a tree used to treat various liver diseases in Korea. There have been some concern regarding the safety of Acer tegmentosum due to some toxic chemical compounds in its stems. Supercritical fluid extraction (SFE) was employed to develop a removing method of toxic compounds from Acer tegmentosum. The toxic compounds were effectively extracted with ethanol modified supercritical fluid $CO_2$. The optimum condition of SFE was 100 bar of pressure, $40^{\circ}C$ of extraction temperature, 3 mL/min of $CO_2$ flow rate, 0.2 mL/min of modifier (ethanol) flow rate.
A supercritical fluid extraction was performed for the extraction of phenolics from grape seeds which up to now have been discarded. The optimum condition for extraction process was predicted through response surface methodology using central composit experimental design. The extraction amount of grape seed phenolics was increased by increasing extraction temperature, pressure, and concentration of co-solvent (ethanol). The optimum extraction conditions were 84.83$^{\circ}$C, 51.50MPa and 1.27% ethanol. The yield of phenolics using SFE was higher with 3 folds than ethanol and 4 folds than hexane but less than 80% methanol. In the respects of food poisoning, the approved solvents were restricted to ethanol and hexane. So, SFE for extraction of phenolics could be powerful alternative method for solvent extraction.
To develop mil fat fractions with desirable physico-chemical properties, anhydrous milk fat (AMF) was fractionated by one- and two-stage extractions using supercritical $CO_2$(SC-$CO_2$). Tow-stage extraction of AMF was performed by first producing tow fractions, an extract and a residue at 4$0^{\circ}C$/241bar, which were subsequently used as the feed for an extraction at 6$0^{\circ}C$/241bar and 4$0^{\circ}C$/345bar, and separated into five and four fractions, respectively, based one extraction time. These fractions were quantified and analyzed for fatty acids and physico-chemical properties. SHort-chain (C4~C8) fatty acids in extract fractions from an extract were 200~150% compared with those of the original AF. Long-chain (C14~C18) fatty acids in extract fractions from a residue were 118~141%. The ratio of unsaturated fatty acids in the residue fraction was 131%. Melting point ranged from 22 to 43$^{\circ}C$, iodine value 21.8 to 36.9, and saponification value 255 to 221 in the extract and residue fractions. SC-$CO_2$ fractionation of AMF by two-stage extraction offers the possibility of developing ractions with discrete fatty acid compositions and physico-chemical properties such as melting point, iodine value and saponification value.
The whole berry, pulp, and seed of sea buckthorn fruit were extracted with supercritical $CO_2$ to produce edible oils. The effects of extraction pressure, temperature, and $CO_2$ flow rate on the oil yield and extraction rate were investigated, and the fatty acid composition, tocopherol, and carotenoid contents of the oils were compared. The results showed that the extraction rate was affected by pressure, temperature, and $CO_2$ flow rate and, in general, the yield increased with a rise in any of the 3 variables. Fatty acids in the whole berry and pulp oil were dominated by monounsaturated fatty acids (>64%), followed by saturated fatty acids (about 30%). In contrast, fatty acids in the seed oil consisted mainly of polyunsaturated (>60%) and monounsaturated fatty acids (>24%). The seed oil had a slightly higher content of tocopherols, but a much lower content of carotenoids, compared with the whole berry or pulp oil.
The objective of this study was to investigate the effect of ultrasound on the extraction of oil from canola seeds when supercritical carbon dioxide ($SCCO_2$) was used as an extraction solvent. The ultrasound-assisted $SCCO_2$ extraction were carried out while varying such operating parameters as particle size of crushed canola seed, flow rate of $SCCO_2$, aspect ratio of the extraction vessel, and ultrasound power. The extraction rate decreased with increasing particle size of samples, showing a maximun at a $CO_2$ flow rate of 6.2 L/min. Both the extraction rate and extraction yield increased with a decrease in the aspect ratio of the extraction vessel. For the ultrasoundassisted $SCCO_2$ extraction, the extraction yield was slightly increased when the $CO_2$ flow rate was below 6 mL/min with sample A and B.
Polychlorinated biphenyls (PCBs) known as environmental contaminants in soil were analyzed by the soil pollution standard process test and the static supercritical $CO_2$ extraction mode. It was shown that the percent average recoveries of PCBs by the soil pollution standard process test were ranged in 25-35% and the corresponding standard deviations were above 10%. In contrast, the percent average recoveries of PCBs by the static supercritical $CO_2$ extraction mode were 2-2.5 times higher and standard deviations were within 7%. These results indicate that static supercritical $CO_2$ extraction mode may be a useful alternative to sample pretreatment certified by the soil pollution standard process test. The increasing supercritical $CO_2$ pressure from 1130 psi to 1996 psi at $40^{\circ}C$ enhanced the recovery of all PCB congeners from soil. However, at same Tc and Pc, the equilibrium time (5 versus 60 minutes) had no effect on the recovery of each PCB congener. Finally, similar PCB recoveries were obtained under the same extraction condition, regardless of the molecular weight and structure (coplanar versus non-coplanar) of PCB congeners.
