노화처리된 고추종자의 저장성분 누출과 priming 처리가 종자활력 증진에 미치는 영향 (Leakage of Seed Reserve Nutrient in Artificially Aged Pepper Seeds and Enhancement of Seed Vigor by Priming)

  • 노화처리 일수가 경과하여 활력이 저하된 종자일수록 종자에서 누출되는 단백질, 아미노산, 가용성 당의 누출량이 많았다. 20일간 노화처리된 종자는 침지 4일째 종자 1g당 35.8 mg의 단백질이 누출되어 건전종자에 비해 6.9배 많았다. 아미노산 누출량도 노화처리된 저활력 종자에서 증가하였다. 가용성 당의 누출량도 20일 노화처리된 저활력 종자에서 침지 4일째 종자 1g당 $36.5{\mu}g$이 누출되어 건전종자보다. 2.8배 많았다. 무기성분의 누출량은 저활력 종자에서 많았으며 무기성분의 누출량은 단백질, 아미노산 및 가용성 당보다는 적었다. 누출량에 근거하여 비파괴적으로 종자활력을 추정할 수 있는 가용성 물질은 단백질과 $K^+$였다. 노화처리된 종자는 발아율과 발아세가 저하되었으며, 종자활력이 저하된 노화종자를 priming 처리함으로써 종자활력을 회복시킬 수 있었다. 이러한 현상은 저활력 종자를 저온에서 발아시킨 경우에 더욱 뚜렷하였다.

채소 퇴화종자와 건전종자 침지용액의 전기전도도, 유기및 무기성분의 차이 (Differences of Electrical Conductivity, Organic and Inorganic Constituents in Leakage from Aged and Non-aged Vegetable Seeds)

  • 종자가 수분을 흡수할 때 특히 퇴화종자에서 종자의 내용물질을 밖으로 누출하는데 Brassica속의 종자는 총광물질인 sinapine이 누출되는 것을 이용하여 퇴화종자를 선별하는 기술이 개발되어 있다. 따라서 본 시험에서는 무, 배추, broccoli, 참깨, 양파, 당근종자에서 sinapine이외에 전기전도도, 무기성분(K, Ca, Mg, Na), 전당, 아미노산 등의 성분들이 퇴화종자와 건전종자 사이에 어떠한 차이로 누출되는지를 검토하여 이들 성분을 이용할 때 퇴화종자를 검정할 수 있는 지표로 사용될 수 있는지를 검토하였다. 1. 종자침청호액의 전기전도도는 퇴화종자에서 시간이 경과할수록 높아졌는데 십자화과 종자인 무, 배추, broccoli종자에서 뚜렷한 차이가 있었으나 양파와 당근종자에서는 전기전도도도 낮았고 차이도 거의 없었다. 2. 무기물의 누출에서는 무기성분중 potassium이 가장 많이 누출되었으며 퇴화종자와 건전종자 사이의 차이도 컸지만 기타 Ca, Mg, Na에서는 누출양도 적었고 차이도 적었다. 또 십자화과 종자에서는 누출이 많았고 양파와 당근종자에서는 누출이 적었다. 3. 전당의 누출도 퇴화종자에서 건전종자보다 높았으며 역시 십자화과 종자에서 누출양과 차리가 컸으나 양파, 당근종자에서는 누출양과 차이가 적었다. 4. 아미노산의 누출도 퇴화종자와 건전종자사이에 차이가 컸으나 품종간 차이가 다양하였고 십자화과 종자에서 누출양이 많았으나 양파나 당근종자에서는 누출양이나 차이가 적었다. 5. 전당과 아미노산은 종자침청 후 약 4시간에서부터 건전종자와 퇴화종자에서 누출양의 차이를 보였다.

치과용 국소마취제에 포함된 Epinephrine이 당뇨 환자의 소수술시 혈당에 미치는 영향에 관한 연구 (A STUDY ON THE INFLUENCE OF EPINEPHRINE CONTAINED IN DENTAL LOCAL ANESTHETIC AGENTS ON THE BLOOD SUGAR OF DIABETES DURING MINOR ORAL SURGERY)

  • Recently, the number of diabetic patients have been increased with westernized living way and meal habitation. The regulation of blood sugar concentration is very important for diabetic patients to keep homeostasis and, to prevent acute or chronic complications. Epinephrine combined with lidocaine is used in dental clinic extensively. And epinephrine combined with lidocaine also effects on decreasing the blood leakage volume by constricting micro vessle and arteriole. But, So far there are few researches about the effect of epinephrine contained in dental local anesthetic agent on the blood sugar of diabetes during minor oral surgery. The purpose of this study was intended to investigate whether epinephrine which combined with dental local anesthetics influence body glucose level in diabetes patients by glucose monitoring. The subject of this study were 38 diabetic patients and 38 normal adults, each patient was checked body glucose after a meal 2 hours later with resting state, and injection 1:100000 epinephrine with dental lidocaine 54 ml or lidocaine 54 ml only. And then the body glucose level was checked 5 minutes and 30 minutes after injection. The results were analyzed by two way ANOVA test (p<0.05). The results were as follows: In the experimental group 1, the mean of the blood sugar level was 180.3 mg/dl before an anesthetic injection, 182.8 mg/dl after 5minutes of the anesthetic injection and 182.2 mg/dl after 30minutes of the anesthetic injection. The change of the blood sugar level was not statistically by two-way ANOVA tests (p<0.05). In the experimental group 2, the mean of the blood sugar level was 237 mg/dl before an anesthetic injection, 234.5 mg/dl after 5minutes of the anesthetic injection and 231.8 mg/dl after 30minutes of the anesthetic injection. The change of the blood sugar level was not statistically by two-way ANOVA tests (p<0.05). In the controlled group 1, the mean of the blood sugar level was 117.6 mg/dl before an anesthetic injection, 119.1 mg/dl after 5minutes of the anesthetic injection and 129.3 mg/dl after 30minutes of the anesthetic injection. The change of the blood sugar level was not statistically by two-way ANOVA tests (p<0.05). In the controlled group 2, the mean of the blood sugar level was 104.2 mg/dl before an anesthetic injection, 102 mg/dl after 5minutes of the anesthetic injection and 105.3 mg/dl after 30minutes of the anesthetic injection. The change of the blood sugar level was not statistically by two-way ANOVA tests (p<0.05).

Effect of Seed Priming on Quality Improvement of Maize Seeds in Different Genotypes

  • In Korea, production of super sweet corn has been economically feasible and is substituting for traditional sweet corn due to better flavor in recent years. Major limiting factors for super sweet corn production are low field emergence and low seedling vigor. The optimum water potential (WP) for the priming of normal and aged seeds of dent, sweet (su) and super sweet (sh2) corns was studied to improve low seed quality. Seeds were primed at 0, -0.3, -0.6, -0.9, and -1.2 MPa of polyethylene glycol (PEG) 8000 solution at $15^{\circ}C$ for 2 days. Priming effects differed depending on the type of corn, seed quality, and WP of PEG solution. Although WP of priming solution did not influence the emergence rate of extremely high quality normal dent corn seeds, it reduced time to $50\%$ emergence (T50) and increased plumule weight. In contrast, the emergence rate of aged field corn was improved by seed priming at 0 MPa and plumule weight and $\alpha-amylase$ activity was enhanced. The optimum WP for both normal and aged sweet and super sweet corn seeds was between -0.3 and -0.6 Mpa. At the optimum WP emergence rate, $\alpha-amylase$ activity, and content of DNA and soluble protein increased, while T50 and leakage of total sugars and electrolytes reduced.

Lysophosphatidylethanolamine Treatment Delays Leaf Senescence and Improve Fruit Storability in Melon (Cucumis melo L.)

  • The influence of lysophosphatidylethanolamine (LPE) on anti-senescence of melon leaves and the change in fruit quality during the storage at low temperature were studied. In most of the crops, freshness of leaves is important factor for characteristics of fruits, such as sugar contents, color, and firmness. Melon ($Cucumis$ $melo$ L. cv. Prince) plants were sprayed with LPE at 5 and 3 weeks before commercial harvest. In upper part, LPE treatment showed slight high number of fresh leaf compared to no treatment (None). However, in lower part, LPE resulted in apparent inhibition effect on senescence, showing that lower side of melon plant kept fresh upon LPE application up to about 30%. The SSC of melon treated with LPE was similar to that of fruit from None at harvest. There was no change in soluble solids content (SSC) for all treatment during the storage at $7^{\circ}C$. There were no significant differences in firmness of mesocarp from melons given different treatments at harvest. The firmness of mesocarp from melon treated with LPE was higher than none after 2 weeks storage. The electrolyte leakage means for melon treated with LPE did not differ significantly from the means at initial storage after 2 weeks storage among the treatments. None increased 57% from its initial electrolyte leakage during storage. These results suggest that the application of LPE may have potential to inhibit senescence of leaves and maintain fruit quality during the storage in melon.

Optimum Harvest Time for High Quality Seed Production of Sweet and Super Sweet Corn Hybrids

  • The production of sweet (su) and super sweet corns (sh2) has been economically feasible in Korea in recent years. Major factors limiting super sweet corn production are low germination and low seedling vigor. Since seed quality is closely related to seed maturity, the optimum harvest time for the seed production of sweet and super sweet corns was studied and the quality of seeds with varying maturities was investigated in 2001 and 2002 cropping seasons. The parents of the sweet corn seeds were Hybrid Early Sunglow and 'Golden Cross Bantam 70' and those of super sweet corn were Xtrasweet 82 and 'For­tune'. Seeds were harvested at 21, 28, 35, 42, 49, and 56 days after silking (DAS). As the seeds developed, seed weight of sweet corn increased and the seed moisture content decreased faster than that of super sweet corn. Germination rates of sweet corn seeds harvested 21 and 28 DAS at $25^{\circ}C$ and emergence rates in the cold soil test were significantly lower than those of seeds harvested after 42 DAS in both years. Although the germination rates of super sweet corn seeds with varying maturities showed similar patterns as sweet corn seeds at $25^{\circ}C$, the emergence rate of super sweet corn seeds in cold soil test continuously increased with seed maturity. This suggests that seed quality of super sweet corn should be tested in a cold soil test to estimate field emergence. As the seeds developed, leakage of total sugars and electrolytes from the both sweet and super sweet corn seeds decreased up to 42 or 49 DAS. The $\alpha-amylase$ activities of both sweet and super sweet corn seeds increased with seed maturity from 21 to 35 or 49 DAS depending on genotype and year. The optimum harvest time for the seed production of sweet corn was 42 DAS and 49 DAS for super sweet corn considering emergence rate and plumule dry weight in the cold soil test, leakage of sugars and electrolytes from the seeds, and $\alpha-amylase$ activity.

퇴화처리에 따리 유채, 배추, 무 종자의 당 누출 (Sugar Leakage from Differently-aged Seeds of Rape, Chinese Cabbage and Radish)

  • 비파괴적으로 활력이 낮은 종자를 선별해내는 방법의 기초를 확립하고자 유채, 배추, 무 종자를 상대습도 90%, 온도 45$^{\circ}C$에서 인위적으로 퇴화시켜 종자활력과 종자를 물에 침지 할 때 분비되는 당의 종류, 분비량, 분비시간과의 관계를 구명한 결과를 요약하면 다음과 같다. 1. 모든 작물의 건전종자는 전당을 분비하지 않았지만 죽은 종자는 침지 후 4~8시간부터 전당을 많이 분비하였다. 2. 유채, 배추, 무의 건전종자는 sucrose와 g1ucose를 가장 많이 함유하였고, stachyose, fructose, raffinose도 함유하였다. 인공퇴화과정에서 stachyose, raffinose, sucrose는 줄고, glucose와 fructose는 증가하였다.

'원황' 배의 착과량 조절이 과실생산성과 내한성에 미치는 영향 (Fruit Productivity and Cold Hardiness as Affected by Fruit Setting of 'Wonhwang' Pear Trees)

  • '원황' 배(Pyrus pyrifolia Nakai)나무에 착과수준을 주당 54개(60%착과), 90개[100%착과구(관행)], 126개(140%착과)로 조절 하였을 때 가지의 생장, 저장양분 및 과실생산과 익년에 삽수의 생장에 미치는 영향을 조사하기 위하여 2년간 수행되었다. 주당 착과량이 증가하면 도장지 생장이 감소하였고 1년생 가지의 탄수화물과 다량 무기원소 및 유리당 농도가 감소하는 경향이 나타났다. 관행 대비 140% 착과처리구는 두 해 모두 수량이 약 35톤으로 가장 높았지만 평균과중과 과실 크기 및 과육당도는 다소 낮은 수준이었다. 하지만 140% 착과처리구는 60%와 100% 착과 처리구 보다 년간 천만원 이상의 소득증대가 관찰되었다. $-20^{\circ}C$ 처리에서 삽수의 발아율은 뚜렷한 감소가 나타나지 않았고, $-30^{\circ}C$에서는 140% 착과처리구에서 70% 이하로 감소되었다. 저온에서 삽수의 전해질 누출률은 적과수준에 따라서 크게 차이는 없었다.

Effect of postharvest conditions on the characteristics of mulberries harvested from various mulberry cultivar Morus alba

  • Recently mulberry has been commercialized but it loses its marketability rapidly after harvest. In this study, characteristics of mulberry were examined at different storage temperature after harvest using the typical Korean mulberry cultivars. Postharvest spoilage fungi on the various mulberry cultivars, Cheongilppong, Daeshim, and Gwasang No. 2 was observed 2, 2, and 1 d, respectively, after harvest at 28℃. However, at 4℃, the day was 8, 7, and 4 d, respectively. At 28℃ storage condition, the weight loss behavior of mulberry did not showed significantly different among them. However, at 4℃ storage condition, mulberry Cheongilppong loses its weight rapidly compared to Daeshim and Gwasang No. 2. Sugar content of mulberry stored at 4℃ was nearly constant, but stored at 28℃ decreased with storage time. The acidity of mulberry slightly decreased with storage time and then increased. Juice leakage of frozen mulberry leaked abruptly within 6 h from the fruit body and then flattened at 25℃. The results of our study, the postharvest characteristics of mulberry were affected by mulberry cultivar and need to further study to increase the shelf life.
