The objective of this work was to study the effect of mixed culture solid substrate fermentation of C. tropicalis ZD-3 with A. niger ZD-8 on detoxification of cottonseed meal (CSM), and to investigate the effect of fermentation period, proportion of CSM in substrate, sodium carbonate, minerals and heat treatment on the reduction of free gossypol levels during mixed culture solid substrate fermentation of CSM. Experiment 1: Three groups of disinfected CSM substrate were incubated for 48 h after inoculation with either of the fungi C. tropicalis ZD-3, A. niger ZD-8 or mixed culture (C. tropicalis ZD-3 with A. niger ZD-8). One non-inoculated group was used as the control. Levels of initial and final free gossypol (FG), CP and in vitro CP digestibility were assayed. The results indicated that mixed culture fermentation was far more effective than single strain fermentation, which not only had higher detoxification rate, but also had higher CP content and in vitro digestibility. Experiment 2: CSM substrates were treated according to experimental variables including fermentation period, proportion of CSM in substrate, sodium carbonate, minerals and heat treatment, Then, the treated CSM substrates were inoculated with mixed culture (C. tropicalis ZD-3 with A. niger ZD-8) and incubated at $30^{\circ}C$ for 36 h in a 95% relative humidity chamber. After fermentation ended, FG and CP content of fermented CSM substrate was assayed. The results showed that the appropriate fermentation period was 36 h, and the optimal proportion of CSM in substrate was 70%. Addition of sodium carbonate to CSM substrate was beneficial for fermentative detoxification. Heat treatment could facilitate fermentative detoxification, and supplementation with minerals was instrumental in reducing gossypol levels during mixed culture solid substrate fermentation of CSM.
Cathamus tinctorius, Coptis japonica and Asarum sieboldii were tested as substrate for the production of Flammulina velutipes. Among the C. tinctoris , C. japonica and A. sieboldii , C. tinctoris was the best substrate for the production of fruitbody. The effects of addition of C. tinctoris to sawdust substrate resulted in the increased mycelial growth on inoculum culture, 3.1% in ratio of fully culture and shorted one day in culture period. C. tinctoris was decreased 6.1% in ratio of fully culture, 11.0% in ratio of fruitbody productive culm. The addition of C.tinctoris, C.japonica to sawdust substrate increased 134.6%, 114.1% on the yield of the mushroom fruitbody respectively . But A. sieboldii decreased the mycelial growth and pineheading ratio delayed the production of fruitbody.
The microbial adsorption characteristics of two different media for biological treatment were studied using attached diverse microbes onto activated carbon and ceramic. The results in the experiments of the characteristics of physical adhesion on two different media with addition of high and low concentrated substrate in the culture were observed that the efficient of adhesion onto F-400 activated carbon was higher over that of ceramic due to the surface area of media. The irradiation treatment by ultrasonication with 400 W power and 3 min retention time on the media without addition substrate conditions and subsequent mixing throughly the culture showed the highest efficiency of cell detachment on the media. Three different microbes, P. ovalis, A calcoaceticus, and B. subtillis were used for the study of the characteristics of microbial adhesion on the media. p ovalis showed the highest adhesion capability while B. subtillis showed the lowest capability adhesion onto media either addition of substrate in the culture. The mixed bacterial culture showed $10\%$ lower removal efficiency of DOC in the low concentrated substrate culture compared to the single pure culture. Whileas, it did not show significant difference between two cultures at high concentrated substrate. It was also observed same population density of microorganism by counting of microbes adhered to microbial media with an ultrasound treatment.
Irrigation management methods controlled by integrated solar radiation (ISR) or drainage level sensor were evaluated in rockwool or coir bag culture as tomato (Solanum lycopersicum L.) production system. Substrate water content and drainage percentage were more stable in the drainage level sensor method than in the ISR method regardless of substrate type. Total yield and marketable yield were high in the drainage level sensor method, but not between substrates in the same irrigation management method. Sugar content was affected more by the substrate type than irrigation method. The drainage level sensor method was elucidated to be better than the ISR method regardless of substrate type.

In this paper, we have examined the MTBE cometabolic degradation by pure culture, which is isolated gasoline contaminated aquifer. Propane was more effectively utilized as a growth substrate to oxidize MTBE. Specific substrate degradation rate was Increased with increasing initial propane amount. Respiking propane was enhanced and continued MTBE degradation and TBA observation was supported MTBE degradation. The mass balance of MTBE and TBA indicated that MTBE was oxidized to TBA as well as further oxidation of TBA.
From several industrial wastewaters, 14 bacterial strains which degrade benzene, toluene, o-xylene, m-xylene, or p-xylene (BTX) were obtained. These strains were characterized as to their species composition and the substrate range, kinetic parameters and the substrate interactions were investigated. Although BTX components have a similar chemical structure, isolated strains showed different substrate ranges and kinetic parameters. None of the strains could degrade all of BTX components and most of them showed an inhibition (Haldane) kinetics on BTX, BTX mixtures were removed under inhibitory substrate interactions with variation in the intensity of inhibition. For a complete degradation of BTX, a defined mixed culture containing three different types of patyways was constructed and all of the BTX components were simultaneously degraded with the totla removal rate of 225.69 mg/g biomass/h Judging from the results, the obtained mixed culture seems to be useful for the treatment of BTX-contaminated wastewater or groundwater as well as for the removal of BTX from the contaminated air stream.
A kinetic model incorporating cell morphology in cephalosporin C biosynthesis by Cephalosporium amemoniurn was developed. The double-substrate Double-substrate kinetic model was used to describe cell growth. Methionine controlled the rate of growth while glucose ultimately controlled the extent of growth. The changes in specific product formation rate were associated with morphologenesis, especially cell differentiation. To increase the productivity of cephalosporin C, the proposed model equations were applied to a fed-batch culture. The algorithm to optimize the fed-batch culture consists of two steps; cell growth was maximized in the growth phase and then cephalosporin C production was maximized in the production phase. The increase of about 33% in the cephalosporin C titre was obtained by the optimal feeding scheduling in comparison with that of batch culture.
The effects of transformation capacity on cometabolic degradation of trichloroethene (TCE) were evaluated using TCE-degrading actinomycetes pure and mixed culture under various culture conditions. The TCE transformation capacity of the actinomycetes enrichment culture in a batch test with phenol addition was 1.0 mg of TCE/mg of volatile suspended solids (VSS). The resting cell TCE transformation capacity of the actinomycetes pure culture cell was 0.75 mg TCE/mg VSS, which increased to 2.0 mg TCE/mg VSS when phenol was added as an external substrate. When the pure culture had an internal substrate in the form of poly-β-hydroxybutyrate (PHB) at 19% of the cell mass, the resting cell TCE transformation capacity increased from 0.47 to 0.6 mg TCE/mg VSS. The presence of PHB increased transformation capacity by 57%, whereas, the addition of phenol caused more than two fold increase in transformation capacity. The actinomycetes culture showed the highest transformation capacity.
New problems have been recently posed on the abnormal foaming (Scum) in an aeration tank and the sludge flotation in a final sedimentation tank during the activated sludge process. However, the activities of the causing bacteria, Nocardia-amarae in an aeration tank have not been searched out at all. Therefore, in this article the activities of Nocardia-amarae in an aeration tank have been closely examined by means of the changes of (F/M) ratio, SRT and inflowing substrate using continuous type and fed-batch type. Summarized results of experiments are as follows. 1. Regrading continuous culture when synthetic wastewater was used substrate neither the increase in the number of Nocardia-amarae in the aeration tank nor the Occurrence of Scum was observed. 2. In the case of fed-batch culture, Nocardia-amarae in the aeration tank increased due to the partial change in substrate and the effect of SRT was significant. 3. Once the scum was formed and the quantity of added Nocardia-amarae and substrate were not changed, the effect of STR was not significant.
Cultures of Saccharomyces cerevisiae were subjected to the effect of PEF (pulse electric field) and a source of selenium. The culture period after which yeast cells were subjected to PEF treatment was optimized, as was the duration of the exposure. Optimization of the nutrient medium composition in S. cerevisiae cultures resulted in an over 1.8-fold increase in selenium accumulation with relation to cultures on the initial substrate. Optimization of the pH value and of culture duration resulted in selenium accumulation increase by approximately 78%. A significant correlation was found between the accumulation of selenium in yeast cells and its concentration in the culture substrate. The highest accumulation of selenium in the biomass of yeast, approx. $240\;{\mu}g/g$ d.m., was obtained after 15-min exposure to PEF on a 20-h culture. An approx. 50% higher content of selenium in cells was recorded, as compared with the control culture without the application of PEF.
