Karyotype of an Endangered Freshwater Fish, Microphysogobio koreensis (Pisces: Cyprinidae) from Korea (멸종위기어류 모래주사 Microphysogobio koreensis (Pisces: Cyprinidae)의 핵형분석)

  • The karyotype analysis of an endangered freshwater fish, Microphysogobio koreensis, was performed to obtain basic data for phylogenetic information. To carry out our study, 4 specimens were collected in Seomjingang River and Nakdongang River and its kidney was treated by flame-drying method. The chromosome number of this species demonstrated 50 diploid chromosomes, with two populations of M. koreensis not significantly different. The karyotype revealed 2n=26m+24sm, consisting of 26 metacentric (m) and 24 submetacentric (sm) chromosomes with the total fundamental arm number determined as FN=100. Total arm length and arm ratio of the chromosomes were $1.44{\sim}2.68{\mu}m$ and 1.27~2.27, respectively. The karyotype of M. koreensis was first reported in this study.

  • Lilium Miquelianum Makino is a species which originated in Korea. The Karyotype of the species was examined in materials collected at Mts. Kaya, Kasan, Chejung, and Kaji. The results are as follows: 1) The somatic chromosome number was found to be 2n=24. 2) The karyotype is described as: K=2Am+2Bkm+2Csst+2Dsst+2Ests+2Fst+2Gst+2Hst+2Ist+2Jsst+2Kst+2Lst m: metacentric, sm: submetacentric st: subtelocentric, s: secondary constriction 3) A single subcentric supernumerary B-chromosome was found in some bulbs from Mt. Kasan. 4) The shape of the supernumerary B-chromosome was similar to that of the E chromosome which had separated at its secondary constriction and lost its lower chromosome fragment. 5) From three to eight nucleio of varying sizes were found in the telophase or interphase nucleus of root tip cells. The maximum number of eight nucleoli corresponds to the number of chromosomes that have a secondary constriction.

  • The chromosome of Cipangopaludina chinensis malleata Chunchon area in 1988 was analysed by using aceto-orcein squash techniques of spermatogonial tissues to obtain mitotic and meiotic chromosomes. The chromosome cycle did not differ, in general, from that found in other snails, C. chinensis malleata has 18 diploid chromosomes and they were identified and classified into 2 groups. The mitotic chromosome complement of this species consists of 2 pairs of metacentric and 7 pairs of submetacentric chromosomes, Spermatogonial metaphase chromosomes range in length from 4.10 ${\mu}{\textrm}{m}$ for the largest pair to 2.20 ${\mu}{\textrm}{m}$ for the smallest pair.

  • Geographical distribution of diploid plant with AA genome (2n=16) and allotetraploid with AABB genome (2n=34) of Scilla scilloides Complex from Korea has been studied. The composition and frequencies of B chromosomes ere also investigated. Plants with AABB genome were predominant over AA genome plants. A mixed population of AA and AABB genome plants was found for the first time. Aneuploid plants have not been found. Chromosomes of AA genome were composed of three pairs of metacentric, two pairs of submetacentric, two pairs of subtlocentric and one pair of telocentric chromosomes, whereas BB genome was four pairs of metacentric and five pairs of subtelocentric chromosomes. B chromosomes were classified into two categories, isochromosome (F) and chromosome fragment (f). The frequencies of B chromosomes were 43% in AA genome plants and 44% in AABB genome plants. The number of B chromosome ranged from 1 to 3 and 1 to 7 in AA and AABB genome plants, respectively. B chromosome combinations were F and F+f in AA genome plants and F, F+f and f in AABB genome plants.

  • The karyotypes of Biomphalaria tenagophila collected from Rio de Janeiro, Brazil were studied using the air-drying method. Somatic cells of this species had 2n=36. The 18 chromosome pairs were identified and classified into 3 groups. The diploid cell has 7 pairs of metacentric, 8 pairs of submetacentric, and 3 pairs of subtelocentric chromosomes. Observed chromosomes ranged from 2.4 to $6.4{\mu}m$, and the total length was $122.3{\mu}m$. This is the first report on the chromosome of B. tenagophila.

  • The comparative karyological analysis of the Korean treefrog, Hyla japonica and Hyla suweonensis performed by conventional giemsa-staining and NOR-staining method. The karyotypes of both species were the same (2n=24). However, according to the Karyological analysis, the 6th chromosomes of the two species distinctly different. Eventhough the 6th chromosomes of the two species shown the same % length, construction of the 6th chromosome of H.japonica was subtelocentric chromosome while H. suweonensis was submetacentric chromosome. This phenomena could explain that the morphological differences in the 6th chromosomes might be caused by pericentric inversion. The two species have 1 pair of NOR site in the 6th chromosome.

  • The karyotype of Lilium cernum has been analysed by means of C-banding technique. Most of clones observed were 2n=24 chromosomes which consist of two pairs of submetacentric and ten pairs of subtelocentric chromosomes, among which two pairs of chromosomes(B and E) showed secondary constriction in the short arm. In addition to these chromosomes a small supernumerary telocentric chromosome was seen in the eight clones. Sixtyeight bands were observed in the twentytwo chromosomes of complement and one band in the supernumerary chromosome. A pair of chromosome (L) did not show any band. The bnads on the chromosome. A pair of chromosome (L) did not show any band. The bands on the chromosomes were distributed in the centromere, secondary constriction and intercalary regions of arms. Of the twelve pairs of chromosomes ten pairs showed symmetric banding patterns in each, but two pairs (I and K) showed asymmetric banding patterns.

  • Three species of Korean sticklebacks family Gasterosteidae were reviewed based on the specimens of several populations for the study of their morphological variations and taxonomical positions. All specimens of Gasterosteus aculeatus observed have a complte row of lateral plates ranging from thirty-two to thirty-five in number. The low-plate morph regarded as landlocked type is virtually absent. And no difference was recognized among six populations of G. aculeatus in the meristic chracters, i.e. number of vertebrae, gill rakers and finrays. The Pungitius sinensis and P.kaibarae ssp. shown local variations have scutes ranging from 31 to 35, but not clinal in the morphometric characters. But P.sinensis was statistically different from P.kaibarae ssp. in some characters such as the number of dorsal spine rays and vertebrae, and body depth, although their ranges overlapped. No significant taxonomic difference was detected between white from and black from in dorsal spinous membrane of P.kailbarae ssp. Although these three species of Korean sticklebacks have the same diploid chromosome number (2N=42), G. aculeatus is obviously different from the two species of genus Pungitius in their karyotype ; G. aculeatus consists of six metacentrics, six submetacentrics, and thiry subtelo-acrocentrics, and both P.sinensis and P.kaibarae ssp. have four metacentrics, six submetacentrics, and thirty-four subtelo-acrocentrics.

  • Karyotypes were established in five Pulsatilla species from Korea : P. cernua, P. davurica, P. koreana, P. chinensis and P. tongkangensis. The somatic chromosome numbers of five species were all 2n=2x=16 with the basic number of x=8. The chromosome complement of P. cernua consisted of 5 pairs of metacentric, 1 pair of submetacentric and 2 pairs of subtelocentric. P. davurica, P. koreana and P. chinensis consisted of 5 pairs of metacentric and 3 pairs of subtelocentric. P. tongkangensis consisted of 5 pairs of metacentric, 2 pairs of submetacentric, and 1 pair of subtelocentric. Karyotype formulas of P. davurica, P. koreana, and P. chinensis were the same as K (2n) = 2x = 16 = 10m + 6st, while those of P. cernua was K (2n) = 2x = 16 = 10m + 2sm + 4st and P. tongkangensis was K (2n) = 2x = 16 = 10m + 4sm + 2st, respectively.

Comparison of Morphological and Chromosomal Characteristics and Cross Breeding of the Two Types Korean Mandarin Fish, Siniperca scherzeri (쏘가리와 황쏘가리(Siniperca scherzeri)의 염색체와 외부형태 비교 및 교배 실험)

  • This study was carried out to confirm the taxonomical status by comparison of morphological and chromosomal charateristics and cross breeding in the two types of black and yellow body color of Korean mandarin fish, Siniperca sherzeri from Han River, Korea. Black and yellow types of Korean mandarin fish were similar in having 90-98 lateral line scale pores, 13 dorsal fin rays and XII XIII spines, 9 anal fin rays and 15 pectoral fin rays, but yellow type was differ from blacks type by yellow body color pattern. Diploid chromosome and arm number (fundamental number, NF) of the two types were the same to 2n=48 and NF=52. Karyotypes in the two types are consisted of 2 pairs submetacentric chromosome and 22 pairs of acro and/or telocentric chromosome. The black type females of Siniperca scherzeri were artificially crossed with yellow type males and black type males, respectively. The progenies from each cross breeding produced the normal individuls that possessed with parent types in the body color pattern. These data suggest that two types of Korean mandarin fish were same species, and yellow type was mutant by albino.
