Assessment of cutting time on nutrient values, in vitro fermentation and methane production among three ryegrass cultivars

  • Objective: The 3×3 factorial arrangement was used to investigate if either high water-soluble carbohydrates (WSC) cultivars or suitable time of day that the grass cut could improve nutrient values and in vitro fermentation characteristics. Methods: The 3 cultivars were mowed at 3 diurnal time points and included a benchmark WSC ryegrass cultivar 'Premium', and 2 high WSC cultivars AberAvon and AberMagic, which contained, on average, 157, 173, and 193 g/kg dry matter (DM) of WSC, and 36.0, 36.5, and 34.1 g/kg DM of N during 7th regrowth stage, respectively. The fermentation jars were run at 39℃ with gas production recorded and sampled at 2, 5, 8, 11, 14, 17, 22, 28, 36, and 48 h. The rumen liquid was collected from 3 rumen fistulated cows grazing on ryegrass pasture. Results: High WSC cultivars had significantly greater WSC content, in vitro DM digestibility (IVDMD) and total gas production (TGP), and lower lag time than Premium cultivar. Methane production for AberMagic cultivar containing lower N concentration was marginally lower than that for AberAvon and Premium cultivars. Grass cut at Noon or PM contained greater WSC concentration, IVDMD and TGP, and lower N and neutral detergent fiber (NDF) contents, but CH4 production was also increased, compared to grass cut in AM. Meanwhile, the effects of diurnal cutting time were influenced by cultivars, such as in vitro CH4 production for AberMagic was not affected by cutting time. The IVDMD and gas production per unit of DM incubated were positively related to WSC concentration, WSC/N and WSC/NDF, respectively, and negatively related to N and NDF concentrations. Conclusion: These results imply either grass cut in Noon or PM or high WSC cultivars could improve nutrient values, IVDMD and in vitro TGP, and that AberMagic cultivar has a slightly lower CH4 production compared to AberAvon and Premium. Further study is necessary to determine whether the increase of CH4 production response incurred by shifting from AM cutting to Noon and/or PM cutting could be compensated for by high daily gain from increased WSC concentration and DM digestibility.

Isolation and Characterization of Pepsin-soluble Collagens from Bones, Skins, and Tendons in Duck Feet

  • The objectives of this study were conducted to characterize pepsin-soluble collagen (PSC) extracted from bones (PSC-B), skins (PSC-S), and tendons (PSC-T) of duck feet and to determine their thermal and structural properties, for better practical application of each part of duck feet as a novel source for collagen. PSC was extracted from each part of duck feet by using 0.5 M acetic acid containing 5% (w/w) pepsin. Electrophoretic patterns showed that the ratio between α1 and α2 chains, which are subunit polypeptides forming collagen triple helix, was approximately 1:1 in all PSCs of duck feet. PSC-B had slightly higher molecular weights for α1 and α2 chains than PSC-S and PSC-T. From the results of differential scanning calorimetry (DSC), higher onset (beginning point of melting) and peak temperatures (maximum point of curve) were found at PSC-B compared to PSC-S and PSC-T (p<0.05). Fourier transform infrared spectroscopy (FT-IR) presented that PSC-S and PSC-T had similar intermolecular structures and chemical bonds, whereas PSC-B exhibited slight difference in amide A region. Irregular dense sheet-like films linked by random-coiled filaments were observed similarly. Our findings indicate that PSCs of duck feet might be characterized similarly as a mixture of collagen type I and II and suggest that duck feet could be used for collagen extraction without deboning and/or separation processes.

Relationships between Air Pollution by SO2 and Soluble Sulphur Contents in the Leaves and Bark pH in Urban Forest Trees (도심지역(都心地域)의 아황산(亞黃酸) 가스에 의(依)한 대기오염(大氣汚染)과 수목내(樹木內) 엽(燁)의 수용성(水溶性) 유황(硫黃) 함량(含量) 및 수피산도(樹皮酸度)와의 관계(關係))

  • This study was carried out to investigate relationships between atmospheric $SO_2$ concentration and both soluble sulphur contents in the tree leaves and bark pH to test the possibility of using them as bioindicators for air pollution. Mt. Kwanak, Mt. Nam, Mt. Bukhan (located in Seoul) and Mt. Paldal (located in Suwon, Kyonggi-do) as polluted areas and Pyongchang, Kwangwon-do as an unpolluted area were selected for this study. Soluble sulphur contents in the leaves and hark pH of two tree species (Pines densiflora S. et Z. and Quercus mongolica Fisch.) were analyzed in May, August, and October, 1990 by $BaSO_4$ precipitation method and pH measurement, respectively. In both species, concentration of soluble sulphur in the leaves increased with increasing concentration of atmospheric $SO_2$ (correlation coefficient : 0.52). Soluble sulphur contents in the 2-year-old needles of Pines densiflora (0.170%) and current year leaves of Quercus mongolica (0.081%) in Mt. Nam in the center of Seoul were higher than those in unpolluted Pyongchang area (0.023% and 0.034%, respectively). Bark pH decreased with increasing atmospheric $SO_2$ concentration (correlation coefficient : -0.52). Bark pH of P. densiflora (pH 3.42) and Q. mongolica (pH 3.63) in Mt. Nam were lower than those in Pyongchang area (pH 3.94 and pH 4.93, respectively). Both soluble S content in the leaves and bark pH were recognized as suitable bioindicators for air pollution by $SO_2$. Especially, bark pH showed more sensitive response to air pollution by $SO_2$ than soluble S concentration in the leaves. The lowest concentration of soluble sulphur and the highest bark pH in August were considered to be due to heavy rain during the rainy season. Soluble S content in the leaves and bark pH were not significantly different at 5% level between the two species in polluted areas.

Morphological characteristics and nutrient removal efficiency of granular PAO and DPAO SBRs operating at different temperatures

  • Biological nutrient removal is gaining increasing attention in wastewater treatment plants; however, it is adversely affected by low temperatures. This study examined temperature effects on nutrient removal and morphological stability of the granular and denitrifying phosphorus accumulating organisms (PAO and DPAO, respectively) using sequencing batch reactors (SBRs) at 5, 10, and 20 ℃. Lab-scale SBRs were continuously operated using anaerobic-anoxic and anaerobic-oxic cycles to develop the PAO and DPAO granules for 230 d. Sludge granulation in the two SBRs was observed after approximately 200 d. The average removal efficiency of soluble chemical oxygen demand (SCOD) and PO43--P remained >90% throughout, even when the temperature dropped to 5 ℃. The average removal efficiency of NO3--N remained >80% consistently in DPAO SBR. However, nitrification drastically decreased at 10 ℃. Hence, the removal efficiency of NH4+-N was decreased from 99.1% to 54.5% in PAO SBR. Owing to the increased oxygen penetration depth at low temperatures, the influence on nitrification rates was limited. The granule in DPAO and PAO SBR was observed to be unstable and disintegrated at 10 ℃. In conclusion, morphological characteristics showed that changed conversion rates at low temperatures in aerobic granular sludge altered both nutrient removal efficiencies and granule formation.

Glass Formation and Dissolution Characteristics of K2O-CaO-SiO2-P2O5 (K2O-CaO-SiO2-P2O5계 유리의 형성 및 용출특성)

  • The glass-forming conditions are investigated in the $K_2$O-CaO-$SiO_2$ $-P_2$$O_{5}$ system with 10~65 mole% of$ P_2$$O_{5}$. Thermal properties and dissolution properties were investigated according to variation of the SiO and KO contents in selected compositions. The glass formation condition was determined when $K_2$O/$P_2$$O_{5}$ mole ratio 0~0.24, $SiO_2$/($SiO_2$+$P_2$$O_{5}$ ) mole ratio 0~0.29, and CaO/$P_2$$O_{5}$ mole ratio 0~1.00. In the result of the thermal properties, the glass transition temperature($T_{g}$) and softening temperature($T_{s}$ ) were gradually shifted to the lower temperature range with increase of $K_2$O contents. The maximum value of the chemical durability was shown in $K_2$$PO_2$$O_{5}$ mole ratio 0.17 when the $SiO_2$ contents were changed, and $K_2$O was fixed at 10 mole%. Hence the change of the $K_2$O/$P_2$$O_{5}$ mole ratio was found that a new main factor of the chemical durability.

Fungal Infection Occurrence on New Cultivars of Kiwifruit Correlated with Fruit Soluble Solids Content and Firmness

  • Kiwifruit is prone to damage by frost in the harvest season and the occurrence of postharvest diseases. Accordingly, new cultivars overcoming these disadvantages have been developed. Compared to existing cultivars, studies on diseases in new cultivars are very limited. Thus, goal of this study was investigation of fungal infection occurrences in new cultivars. Kiwifruit was harvested in 2014 and 2015 year, respectively. In both years, cvs. Goldone and Redvita showed more resistance against fungal infection but cv. Gamrok was more sensitive compared with cv. Hayward. Major fungi were identified as Botryosphaeria dothidea, Phomopsis sp., Alternaria alternata and Nigrospora sp. Firmness and soluble solids content were also influenced by the occurrences. This study's findings are important for providing practical information on new cultivars in the kiwifruit industry.

  • As an initial step for evaluating hepatoprotective components against alcohol-induced toxicity, the effect of various fractions from Aloe vera on alcohol metabolism in rats were examined and the results were as follows: Water soluble fraction, after a single oral administration to rats, was found to cause a significant decrease in the serum ethanol concentration as well as enhancement of liver cytosolic ADH activity. On the other hand, the fractions soluble in organic solvent was found to cause an increase in the blood ethanol concentration and inhibit ADH activity. Further fractionation of the water soluble fraction by ultrafiltration system gave four subfractions corresponding to molecular weight and treatment of them in rats demonstrated that subfraction of M.W. > 30,000 exhibited the most potent enhancing activity of ethanol methabolism.

Effect of the Elevated Carbon Dioxide on the Growth and Physiological Responses of Peach 'Mihong' (CO2 상승처리가 복숭아 '미홍'의 수체생육 및 생리반응에 미치는 영향)

  • This study was conducted to investigate the effect of elevated carbon dioxide on the growth and physiological responses of peach 'Mihong' (Prunus persica). We simulated three different carbon dioxide conditions based on climate change scenarios RCP 8.5 in the sunlight phytotron rooms from April 22 to July 6, 2020; 400 µmol·mol-1(present condition), 700 µmol·mol-1 treatment(expecting carbon dioxide concentrations in mid-21st century), 940 µmol·mol-1 treatment (expecting carbon dioxide concentrations in late 21st century). The average of maximum photosynthesis rate at 700 µmol·mol-1(16.06 µmol·CO2·m-2·s-1) was higher than those at 400 µmol·mol-1(14.45 µmol·CO2·m-2·s-1) and 940 µmol·mol-1(15.96 µmol·CO2·m-2·s-1) from May 22 to July 2. However, stomatal conductances at 700 µmol·mol-1 and 940 µmol·mol-1 were lower than those at the control. Also, the carbon dioxide saturation point in all treatments was reduced from 1,200 µmol·mol-1 in the early stage of growth to 600-800 µmol·mol-1 in the late stage of growth. The stomatal densities were decreased as carbon dioxide increased. The shoot lengths were decreased while the carbon dioxide was increased, but the increase of trunk diameter and leaf areas, shoot numbers were not statistically different. The fruit weight at 700 µmol·mol-1(152.5 g) was higher than those at the control(141.8 g) and 940 µmol·mol-1(147.4 g). The soluble solids were higher at 700 µmol·mol-1, 940 µmol·mol-1 compared to the control. These results suggest that a carbon dioxide elevated to 700 µmol·mol-1 in the future may give a positive effect on the yield and fruit quality of peach 'Mihong' while a carbon dioxide elevated above 940 µmol·mol-1 may affect negatively such as early senescence and loss of fruit set.

Evaluation of the Color-change and Stability of Hoecheong (Smalt) Pigments When Exposed to Airborne Environmental Pollutants (회청 안료의 보존 환경에 따른 안정성 평가)

  • Recently, as the climate changes rapidly and the prevalence of airborne fine particulate matter increases, the pattern of pollutants in the atmospheric environment is also changing. Therefore, the importance of studying the stability of pigments used in colored cultural properties is emerging. Hoecheong is an inorganic blue glass pigment called smalt; it is made by using cobalt as a coloring element in potash glass, and was widely used in colored cultural assets, such as murals and paintings. In this study, we collected three other hoecheong pigments to analyze their properties. The percentage of Co and K contained are different according to the manufacturer, and the smalt-3 sample has a lower cobalt content (15.1 wt.%) and higher potassium content (29.6 wt.%). After this analysis, colored specimens were prepared. Prepared specimens were exposed to ultra-violet rays, CO2/NO2, and NaCl, which are known to have the greatest influence on the stability of pigments. We found that factors affecting the color stability were NO2 gas, ultra-violet rays, and water-soluble salts (NaCl). Among them, NO2 has the most severe impact on color change of the pigments. Results of the component analysis showed that the color change depends on the potassium and cobalt content of the hoecheong pigment. Among the specimens, smalt-3 showed the most vulnerability after exposure to NO2 gas and water-soluble salts. Pigment film stability is affected by watersoluble salts, giving rise not only to color change, but also weakening the physical properties of the film. However, there was no significant change in composition and color after exposure to CO2 gas. In conclusion, we found that hoecheong pigments underwent color change and increased instability of the coating film when exposed to any of the atmospheric environmental factors used in this study, except for CO2.

A Novel Method for Preparing of Oxoruthenates Complexes: trans-[RuO3(OH)2]2-, [RuO4]-, (n-Pr4N)+[RuO4]- and [RuO4 and Their Use as Catalytic Oxidants

  • The synthesis and characterization of ${K_3[Ru(C_2O_4)3]{\cdot}4H_2O\;(C_2O_4}^{2-}$ = oxalato anoin) complex are described, and its redox properties (in buffer solution of pH = 12) have been investigated. This complex is used for in situ generation of oxoruthenates complexes which have been characterized by electronic spectroscopy. Reaction of ${K_3[Ru(C_2O_4)3]{\cdot}4H_2O$ with excess ${S_2O_8}^{2-}$ in molar KOH generates trans-${[RuO_3(OH)_2]^{2-}/S_2O_8}^{2-}$ reagent while with excess ${BrO_3}^-$ in molar $Na_2CO_3$ generates ${[RuO_4]^-/BrO_3}^-$ reagent. Avoiding the direct use of [$RuO_4$] the organic-soluble $(n-Pr_4N)^+[RuO_4]^-$, (TPAP) has been isolated by reaction of $K_3[Ru(C_2O_4)3]{\cdot}4H_2O$ with excess ${BrO_3}^-$ in molar carbonate and n-$Pr_4$NOH. In a mixture of $H_2O/CCl_4$ ruthenium tetraoxide can be generated by reaction of $K_3[Ru(C_2O_4)3]{\cdot}4H_2O$ with excess ${IO_4}^-$. The catalytic activities of oxoruthenates that have been made from $K_3[Ru(C_2O_4)3]{\cdot}4H_2O$ towards the oxidation of benzyl alcohol, piperonyl alcohol, benzaldehyde and benzyl amine at room temperature have been studied.
