Tramadol is an opioid analog used to treat chronic and acute pain. Intradermal injections of tramadol at hundreds of millimoles have been shown to produce a local anesthetic effect. We used the whole-cell patch-clamp technique in this study to investigate whether tramadol blocks the sodium current in HEK293 cells, which stably express the pain threshold sodium channel Nav1.7 or the cardiac sodium channel Nav1.5. The half-maximal inhibitory concentration of tramadol was 0.73 mM for Nav1.7 and 0.43 mM for Nav1.5 at a holding potential of -100 mV. The blocking effects of tramadol were completely reversible. Tramadol shifted the steady-state inactivation curves of Nav1.7 and Nav1.5 toward hyperpolarization. Tramadol also slowed the recovery rate from the inactivation of Nav1.7 and Nav1.5 and induced stronger use-dependent inhibition. Because the mean plasma concentration of tramadol upon oral administration is lower than its mean blocking concentration of sodium channels in this study, it is unlikely that tramadol in plasma will have an analgesic effect by blocking Nav1.7 or show cardiotoxicity by blocking Nav1.5. However, tramadol could act as a local anesthetic when used at a concentration of several hundred millimoles by intradermal injection and as an antiarrhythmic when injected intravenously at a similar dose, as does lidocaine.

In this two-part paper, we consider dynamic resource allocation in orthogonal frequency division multiple access(OFDMA). To reduce the reverse link overhead for channel quality information(CQI) feedback, a set of sub-carriers are tied up to a sub-channel to be used as the unit of CQI feedback, user-multiplexing and the corresponding power/rate allocation. Specifically, we focus on two sub-channel structures, either aggregated or distributed, where the SNR distribution over a sub-channel is modeled as Ricean in general, and the channel quality of a sub-channel is summarized as the mean and variance of channel gain envelop divided by noise standard deviation. Then, we develop a generalized two step channel/resource allocation algorithm, which uses the two statistical measurements, and analyze the spectral efficiency of the OFDMA system in terms of average frequency utilization. An extension to proportional fair algorithm will also be addressed. As confirmed by numerical results, the aggregated structure is preferred especially when intending aggressive link adaptation.

This paper solves the problem of finding a suitable sub-channel and power joint allocation method for multiple users in 802.16e OFDMA/TDD cellular systems. The joint allocation is thatfirstly the sub-channel is allocated to the users and then suitable power is allocated. We propose a FASA (Fairness insured Aggressive Sub-channel Allocation) algorithm which is a dynamic channel allocation algorithm considering all users' channel state information conditionally to maximize fairness and throughput. The improved CHC algorithm, which is dynamic power allocation algorithm, is also proposed in this paper The Improved CHC algorithm collects the extra of the downlink transmit power and then re-allocates it to other users. Simulation results show that the proposed improved CHC algorithm additionally increases the fairness and sector throughput.

This paper reports the optimized mixed-signal performance of a high-voltage (HV) laterally double-diffused metaloxide-semiconductor (LDMOS) field-effect transistor (FET) with a dual gate oxide (DGOX). The fabricated device is based on the split-gate FET concept. In addition, the gate oxide on the source-side channel is thicker than that on the drain-side channel. The experiment results showed that the electrical characteristics are strongly dependent on the source-side channel length with a thick gate oxide. The digital and analog performances according to the source-side channel length of the DGOX LDMOS device were examined for circuit applications. The HV DGOX device with various source-side channel lengths showed reduced by maximum 37% on-resistance (RON) and 50% drain conductance (gds). Therefore, the optimized mixed-signal performance of the HV DGOX device can be obtained when the source-side channel length with a thick gate oxide is shorter than half of the channel length.

We fabricated the newly proposed nano structure memory with SOI edge channel and a nano dot. The width of the edge channel of this device, which uses the side wall as a channel and has a nano dot on this channel region, was determined by the thickness of the recessed top-silicon layer of SOI wafer. The size of side-wall nano dot was determined by the RIE etch and E-Beam lithography. The I$_{d}$-V$_{d}$, I$_{d}$-V$_{g}$ characteristics of the devices without nano dots and memory characteristics of the devices with nano dots were obtained, where the voltage scan was done between -20 V and 14 V and the threshold voltage shift was about 1 V.t 1 V.

Early afterdepolarization (EAD), a significant cause of fatal ventricular arrhythmias including Torsade de Pointes (TdP) in long QT syndromes, is a depolarizing afterpotential at the plateau or repolarization phase in action potential (AP) profile early before completing one pace. AP duration prolongation is related to EAD but is not necessarily accounted for EAD. Several computational studies suggested EAD can form from an abnormality in the late plateau and/or repolarization phase of AP shape. In this sense, we hypothesized the slope during repolarization has the characteristics to predict TdP risk, mainly focusing on the maximum slope during repolarization (dVm/dtmax_repol). This study aimed to predict the sensitivity of dVm/dtmax_repol to ion channel conductances as a TdP risk metric through a population simulation considering multiple effects of simultaneous reduction in six ion channel conductances of gNaL, gKr, gKs, gto, gK1, and gCaL. Additionally, we verified the availability of dVm/dtmax_repol for TdP risk prediction through the correlation analysis with qNet, the representative TdP metric. We performed the population simulations based on the methodology of Gemmel et al. using the human ventricular myocyte model of Dutta et al. Among the sixion channel conductances, dVm/dtmax_repol and qNet responded most sensitively to the change in gKr, followed by gNaL. Furthermore, dVm/dtmax_repol showed a statistically significant high negative correlation with qNet. The dVm/dtmax_repol values were significantly different according to three TdP risk levels of high, intermediate, and low by qNet (p<0.001). In conclusion, we suggested dVm/dtmax_repol as a new biomarker metric for TdP risk assessment.

The effect of the sub-optimal beam-forming and the fading channel correlation on the closed loop transmit antenna diversity(CTD) system is investigated in frequency flat Rayleigh fading channels. The fast channel fading prevents the perfect channel estimation at a mobile station, hence the imperfect weight is applied to the antenna branch of transmitter. The weight causes sub-optimalbeam-forming and aggravates the performance of CTD system. The fading correlation or a wireless channel also is one of the factors decreasing the diversity gain. A bit error rate expression for the CTD system is analytically derived as a function of the channel estimation error, the channel correlation coefficient the feedback delay, and fading index. It is shown that the channel estimation error gives more severe effect to the system performance than the channel correlation.

Sophora flavescens Ait. (Fabaceae) is a medicinal plant widely founded in Northeast Asia, and its dried root (Kushen) has been used as a traditional Chinese herbal medicine. The therapeutic effects of Kushen in micturition disorder was not investigated comprehensively yet. In the present study, we examined and compared the efficacy of three batches of Kushen extract using different ethanol content through an in vitro cell-based assay. Among them, we chose the batch with the highest efficacy and augmented the volume of extract for industrial purpose. The bulk extract was examined in its efficacy in the in vitro cell-based assay, and the therapeutic effects through an in vivo behavioral assay of OAB rats. The main components of the extracts were analyzed by liquid chromatography. Cytotoxicity of the extracts was investigated by MTT assay. The overall efficacy of the extract was as much as, or more than, kurarinone, a potent BKCa channel activator. Thus, the extract was a potent relaxant of urinary smooth muscle by upregulating the activity of BKCa channel. The Kushen extract could be explored as an alternative medicine against overactive bladder patients indicating severe dysfunction of BKCa channel.

ECMA-392 is the first CR (cognitive radio) specification for personal/portable devices in TV white space. In ECMA-392, in addition to the long preamble of 2 OFDM symbols, 4 pilot sub-carriers per OFDM symbol are repeatedly transmitted with the period of 13 OFDM symbols. In this paper, we consider channel estimation schemes using both long preamble and pilot sub-carriers: we propose channel estimation schemes in which a first order recursive filter is used, and present a method for reducing channel estimation delay. Computer simulation results indicate that the proposed schemes perform well over slow Rayleigh fading channels.
We propose a sub-band spreading technique for adaptive modulation (AM) in orthogonal frequency division multiplexing (OFDM) systems in order to reduce signaling overheads and to average frequency selective fading channels causing different signal-to-noise ratio (SNR) values for subcarriers in each subband. The conventional sub-band based AM schemes can also reduce signaling overheads and complexity for allocating a resource per sub-band at a time. However, they may suffer from the channel variation in a sub-band when the sub-band size is larger than the channel coherence bandwidth (BW). The sub-band spreading at the transmitter enables the received symbols in each sub-band to have an identical reliability even in a frequency selective fading channel. We rigorously analyze the averaged SNR value at the receiver of the sub-band spreading system and the analyzed average SNR in a sub-band is used for an adaptation criterion. The proposed AM scheme outperforms the conventional sub-band based OFDM scheme without spreading, and it can yield better throughput performance than the conventional subcarrier based AM schemes when we consider the signaling overheads.
