Characteristics and antimicrobial resistance patterns of Pasteurella multocida isolated from swine in Gyeongbuk province (경북지역 돼지 유래 Pasteurella multocida 의 특성 및 항생제 내성양상)

  • This study was conducted to investigate the species-specific gene detection, capsular serogroup and antimicrobial resistance pattern of Pasteurella multocida isolated from pneumonic lung lesion of swine in Gyeongbuk province. P. multocida isolates were typed for capsular serogroups by polymerase chain reaction. Of the 32 strains, 28 (87.5%) were typed serotype A, 3 (9.3%) were typed serotype D, and 1 strain was unknown (3.1%), respectively. In antimicrobial agents resistance test, almost of strains were susceptible to amoxicillin (100%), enrofloxacin (96.9%), ampicillin (93.8%), florfenicol (90.6%), chloramphenicol (90.6%) and were resistant to streptomycin (71.9%), spectinomycin (56.3%). All strains were resistant to clindamycin, erythromycin and lincomycin.

  • Biochemical and antimicrobial susceptibility tests were conducted on 40 strains of Pseudomonas aeruginosa originated from diseased chicken submitted for diagnosis to this Institute during 1978-80. An extensive study of the biochemical properties revealed that the tested strains can be identified with Pseudomonas aeruginosa. Antibiogram showed that all the strains were susceptible to gentamicin, colistin and amikacin but resist to nitrofurantoin, trimethoprim+sulfamethoxazole, ampicillin, methicillin and kanamycin, and had varing degreed of resistance to other antimicrobials including carbenicillin, sulfomamides, neomycin, streptomycin, tetracycline and chloramphenicol. Three of the most frequent resitance patterns observed were FM SXT AM ME KM CM TC SM NE SSS Pattern, FM SXT AM ME KM CM TC SM NE SSS CB Pattern and FM SXT AM EKM CM TC SM Pattern, and these resistance patterns contained 72.5% of the tested strains.

  • One hundred and sixty one Escherichia coli strains isolated from 24 swine (11 swine fed with feedstuffs containing 7.5mg/kg of tetracycline and 13 swine not received antibiotic) were studied for the drug resistance and distribution of R factors. About 42 per cent of E. coli strains isolated from pigs of a herd fed with tetracycline (TC)-containing feeds were resistant to TC, streptomycin(SM), sulfisomidine(SA), ampicillin (AP) and kanamycin (KM), alone or in combination thereof, but none of the swine not receiving antibiotic containing feedstuffs excreted E. coli resistant to these drugs, Among resistant strains, 18.2% were found to be singly resistant to TC, whereas 81.8% were resistant to two or more antibiotics. The most common pattern was the triple resistant to TC, SM and SA(30.3%), and follolwed by double ones to TC and SM(24.2%). About one half of resistant strains carried R factors which were tranferable to the recipients by conjugation. In spite of feeding with feedstuffs containing only TC, high incidences of multiple resistance and R factors were observed in the E. coli isolated from these swine.

Virulence and antimicrobial resistance genes of pathogenic Escherichia coli from piglets showing diarrhea before and after ban on antibiotic growth promoters in feed (사료 첨가 항생제 금지 전후 돼지 설사증 유래 대장균의 병원성 인자 및 항생제 내성 유전자)

  • This study examined the prevalence of adherence factors, toxin genes, antimicrobial resistance phenotypes, and resistance genes in Escherichia coli (E. coli) isolated from piglets with diarrhea before and after the ban on antibiotic growth promoters (AGPs) in Korea from 2007 to 2018. In this period, pathogenic 474 E. coli isolates were obtained from diarrheic piglets. The virulence factors and antimicrobial resistance genes were assayed using a polymerase chain reaction, and the susceptibility to antibiotics was tested according to the Clinical and Laboratory Standards Institute guidelines. After the ban on AGPs, the frequency of F4 (12.5% to 32.7%) increased significantly, and LT (31.9% to 20.3%) and EAST-I (46.5% to 35.2%) decreased significantly. In addition, the resistance to streptomycin (45.8% to 67.9%), cephalothin (34.0% to 59.4%), and cefazlin (10.4% to 28.8%) increased significantly. Colistin resistance plasmid-mediated genes, mcr-1 and mcr-3, were detected after the ban on AGPs. The results of this study can provide useful data for analyzing the impact of the ban on AGPs on the virulence profiles and antimicrobial resistance of E. coli isolated from piglets with diarrhea in Korea.

Biochemical Characteristics and Antibiotic Resistant Patterns of Klebsiella pneumoniae (Klebsiella pneumoniae 분리균의 생화학적 특성과 항생물질 내성유형 연구)

  • One hundred and thirty-six strains of Klebsiella pneumoniae were isolated from clinical specimen taken from pediatric patiants at 6 different hospitals and identified, characterized and investigated on the patterns of antibiotic resistance. The 136 strains showed the positive reactions in the 17 tests; Voges-Proskauer, ONPG, cirate utilization, KCN degradation, productions of lysine decarboxylase, acid and gas from glucose, utilizations of malonate, manitol, rhamnose, salicin, sucrose, raffinose, arabinose, lactose, sucrose, inositol and raffinose, but the strains showed the negative reactions in the 8 tests; production of $H_2S$, indole, arginine dehydrolase, ornithine decaraoxylase, phenylalanine deaminase, motility, methly red and gelatin liquefaction. 41 did not utilize dulcitol, and 32 did not utilize adonitol. The 36 out of them produced klebecin. The 136 strains were resistant to ampicillin, 2 to gentamicin, 14 to cephalothin, 16 to chloramphenicol, 8 to kanamycin, 13 to streptomycin, and 17 to tetracycline. Twenty strains were resistant to 2 kinds of antibiotics 5 strains to 3, 4 to 4 and 1 to 6 and 7.

Study on Development of Enzyme-Linked Immunosorbent Assay for the Screening of Chloramphenicol Residues (잔류 Chloramphenicol 검사용 효소 면역측정법의 개발에 관한 연구)

  • The monoclonal antibody to chloramphenicol(CAP) was produced to develop an enzyme-linked immunosorbent assay(ELISA) for residual CAP. An immunogen(CAP-BSA) was prepared by immunogen, antibody titer was measured by indirect ELISA. Spleen cells form the immunized mouse were fused with SP2/OAg14 myeloma cells. Among hybridomas selected in HAT media, 6 clones shown high antibody titer to CAP were subjected to cloning by limit dilution, and all of the monoclonal antibodies(MCA1, 2, 3, 4, 5, 7 and 9) produced by each clone were identified as IgG1 by ELISA isotyping analysis. Competitive ELISA condition was established by using the purified monoclonal antibody MCA1 as primary antibody and CAP-HSA conjugate as coating antigen. Standard curve of CAP(n=28) showed that the lowest detection limit of CAP is 20ng/ml level. The cross-reactivities of the 6 monoclonal antibodies showed that CAP sodium succinate. CAP base, P-nitrophenol, and p-nitrobenzyl alcohol were 89∼178, 0.050∼2.237, 0.056∼0.794 and 0.013∼7.939%, respectively. No cross-reactivities were observed with phenylalanine, tyrosine, glutamine, thiamphenicol, neomycin, streptomycin, gentamicin, sulfamethazine, sulfathiazole, chlortetracycline and p-aminobenzoic acid.

  • A total of 315 microorgainisms were isolated from 256 pneumonic lung samples of pig in abattoirs of northern Chungnam area during February to November 1994, and Identified 97 strains as Pusteurella multocida, 89 strains as Staphylococcus spp, 54 strains as Streptococcus spp, 22 strains as Mycoplasma spp, 21 strains as Escherichia coli, 18 strains as Haemophilus parsuis, 11 strains as Corymebacteroi, pyogenes, and 3 strains as Acrinobacillus spp by Gram's and Dienes stain, and biological properties test Involved API system. After that, they were examined anti biotic susceptibility for ampicillin(AM), cephalothin(CF), chloramphenicol(CP), erythromycin(EM), kanamycin(KM), gentamicin(GM), neomycin(NM), penicillin(PC), streptomycin(SM), tetracycline(TE), tiamulin(TIA), tylosin(TYL), methicillin(DP), colistin(CL) and trimet hoprim(SXT). In antibiotics susceptibility test, 293 isolates except Mycoplasma spp 22 strain were highly susceptible to DF(79.2%) and AM(76.2%), but resistant to PC(14.0%), NM(19.5%) and KM (23.2%) The multiple drug resistant patterns were noted in most isolates, whereas only 7 isolates resistant to single drugs.

  • Salmonella Typhimurium is a virulent pathogen for human and animal. We studied serotypes, biochemical characteristics, and antimicrobial resistance of S. Typhimurium isolated from diseased pigs in Gyeongbuk province over 1998 to 2008. One hundred sixteen isolates were identified as S. Typhimurium by biochemical characteristics and serotypes from 90 farms. The biochemical characteristics of S. Typhimurium isolates was production of $H_2S$, indole-negative, fermentation of mannitol, dulcitol, sorbitol, inositol, rhamnose, and maltose, and ornithine decarboxylase. At antimicrobial susceptibility test, the majority of isolates were highly susceptible to amoxicillin/clavulanic acid, cefepime, ciprofloxacin, while were highly resistant streptomycin, cephalothin, enrofloxacin, nalidixic acid, apramycin, chloramphenicol, tetracycline. The isolates were divided into 65 resistant patterns and 47 of the isolates were shown as a DT104 ASSSuT resistant phenotype.

  • Antimicrobial susceptibility, plasmid profiles and distribution of toxA gene were investigated in Pasteurella multocida isolated from pneumonic lung lesions of swine. The bacteria were highly susceptible to norfloxacin, cabenicillin, enrofloxacin and chloramphenicol, but resistant to colistin, sulfamethoxawle/trimethoprime, bacitracin, streptomycin. Sixty percentage of the isolates was resistant more than 2 drugs used in this experiment and 21 strains (23.6%) were resistant more than 5 drugs. This phenomenon meant that they had highly multi-drugs resistance. In the analysis of plasmid profiles, nineteen strains (47.5%) of 40 P multocida isolates harbored plasmids, ranging from 53.3kb to 2.49kb in size and the plasmid profiles could be classified into 5 groups. However, there was no relationship between the size and the profile of plasmid and the resistance pattern of antimicrobial agents. Thirty strains of 39 P multocida isolates (77%) investigated by PCR harbored toxA gene. This result suggested involvement of the ToxA protein expressed from the gene in pneumonic pasteurellosis of swine.

Epidemiological characteristics of Salmonella spp. isolated from different stages of commercial swine farms

  • Epidemiological characteristics of a total of 48 swine herd with diarrhea or a history of diarrhea in Gyeongsang-do between 1999 and 2000 were performed to evaluate the prevalence of Salmonella spp., their serotypes and antibiotic resistance patterns with respect to the different stages of swine production system. A total of 139 Salmonella spp. (21%) were isolated from 662 fecal samples and the overall herd prevalence of Salmonella spp. ranged from 12.5% to 88%. The average prevalence of Salmonella spp. from swine stages of suckling/nursery, grow/finisher and sow stage were 25.7%, 19.2% and 18.4%, respectively. Ten serotypes of Salmonella spp. were identified with a predominance of S. Typhimurium, S. Derby and S. Agona. Twenty-five isolates (18%) were found to be untypable. One hundred and two Salmonella isolates (73.4%) resistant to more than 1 antibiotic were characterized by 24 diverse resistance patterns, and their frequency of antibiotic resistance was highest in grow/finisher stage (83.3%). Resistance to tetracycline (TE; 67.6%), sulfamethoxazole (SU; 46.8%) and streptomycin (ST; 28%) was most common and the most common resistance patterns were TE SU (31.4%), TE (21.6%) and TE SU ST (20.6%) in order.
