Sensitivity of Corynebacterium renale to Some Antimicrobial Agents (축우(畜牛)에서 분리(分離)한 Corynebacterium renale의 각종(各種) 항균제(抗菌劑)에 대(對)한 감수성(感受性))

  • In order to know the current status of sensitivity of Corynebacterium renale to some antimicrobial agents of current use, the sensitivity of 37 strains isolated from urine and vaginal cavity of healthy Korean native cattle and dairy cattle was studied by agar plate dilution method, and their sensitivity according to serological types was compared. The antimicrobial agents tested were streptomycin (SM), erythromycin (EM), oxytetracycline (OT), penicillin G (PC), gentamycin (GM), kanamycin (KM), ampicillin (AP), leucomycin (LM), chloramphenicol (CM), colistin (CL), sulfadimethoxine (SDM) and sulfadiazine (SDZ). Most strains were highly sensitive to PC, GM, AP, LM and KM, and moderate to CM and OT, but resistant to SM, EM, CL, SDM and SDZ. Serological type II was more sensitive to EM, SDM and SDZ than type I and II, but more resistant to SM.

Antibiotic Resistance of Escherichia coli Isolated from Hens (계유내(鷄由來) Escherichia coli의 항생물질내성(抗生物質耐性)및 R 인자(因子)의 분포(分布))

  • Two hundred and twenty-seven strains of Escherichia coli isolated from 25 hens (12 hens received tetracycline neomycin and sulfadimethoxine, and 13 hens not received antibiotics) were studied for the drug resistance and distribution of R factors. About 74 per cent of E. coli strains isolated from hens of a herd received antibiotics were resistance to tetracycline (TC) streptomycin (SM), chloramphenicol (CM), kanamycin (KM), ampicillin (AP) and sulfisomidine (Su), alone or in combination thereof, but only a hen among a herd not received antibiotics excreted E. coli resistant to TC and SM. Among resistant strains, about 7% were found to be resistant to TC and SM, whereas 93% were resistant to three or more antibiotics. The most common pattern was the quadruple resistant to SM, TC, KM and Su (28.7%), and followed by triple ones to SM, TC and Su (25.3%), and SM, TC and KM (24.7%). About 84% of resistant strains carried R factors which were transferable to the recipient by conjugation.

  • One hundred and fifty seven Escherichia coli strains isolated from 18 cattle (9 dairy cattle received penicillin, streptomycin (SM) or sulfadimethoxine for treatment of diseases and 9 Korean native cattle not received antibiotics) were studied for the drug resistance and distribution of R factors. Of 88 E. coli strains isolated from cattle not received antibiotics, only 1 strain was resistant to SM, but about 46 per cent of 69 E. coli strains isolated from cattle received antibiotics were resistant to SM, tetracycline (TC), ampicillin (AP), kanamycin (KM), chloramphenicol (CM), and sulfisomidine (Su), alone or in combination thereof. Of resistant strains, about 72% were resistant to three or more antibiotics, but 28% were found to singly resistant. The most frequent resistant pattern was triple resistance to AP, KM and Su (37.6%), and quadruple one to SM, TC, CM and Su (12.5%). About 28% of resistant strains carried R factors which were transferable to E. coli ML 1410 $NA^r$ by conjugation.

Purification and Characterization of Purine Nucleoside Phosphorylase (PNP) in Micrococcus luteus

  • Purine nucleoside phosphorylase (PNP) was purified in Micrococcus luteus (M. luteus) using streptomycin sulfate and amomonium sulfate fractionation, three times by a Sephadex G-100 gel filtration and a DEAE-Sephadex A-50 ion exchange chromatography. The enzyme was purified 72 folds with a 11% recovery and showed a single band in a nondenaturing gel electrophoresis. The M. W. of PNP turned out to be 1.35 * 10$^{5}$ delton in G-150 gel filtration chromatography. The stability of the enzyme was increased by treatment with both substrates, MgCI$_{2}$ or CaCI$_{2}$, but not significantly kcal/mol. M. luteus PNP catalyzed the phosphorolysis of inosine, deoxyinosine, guanosine and deoxyguanosine with the Km value of 1.5 * 10$^{-3}$ M, 3.0 * 10$^{-3}$ M, 5.0 * 10$^{-4}$ M, respectively. The enzyme was reacted with adenosine, 1-methylnosine and 1-methylguanosine as substrates, which were shown to be poor substrates for mammalian enzyme.

  • Petroleum energy is the major source of the world energy market, and its massive usage, and the corresponding extreme environmental pollution, imposes a serious threat on the ecological cycles. By screening oil-contaminated soil, we isolated, identified, and characterized a novel strain that represents a considerable diesel-degrading potentiality; the Bacillus aryabhattai DA2 strain is registered in the NCBI with the accession number MG571630, and it possesses an efficient tributyrin-degrading capacity. The optimal condition for diesel degradation by DA2 strain was observed at pH between 7-8 and at the temperature of $30^{\circ}C$. The strain is resistant to salt as well as the antibiotics like ampicillin and streptomycin. These results indicate B. aryabhattai is one of the potential candidates for the remediation of the diesel-contaminated sites.

  • After the induction of estrus by the PGFF2$\alpha$-GTH combination on the 118 repeat breeder cow during of 6-13 days of the estrus cycle, the effects of GTH, antibiotics, antibiotics-GTH combination were compared. The results of this study, the following conclusion were obtained: 1. The rate of estrus was reduced the administration of GTH before the administration of PGF2$\alpha$. 2. The rate of ovulation delayed was 24.6% in the group with the administration GTH, but 34.1% without the administration GTH. Duration of ovulation was shortened by GTH. 3. The impregnation induction by the administration of PGF2$\alpha$, PGF2$\alpha$-antibiotics, PGF2$\alpha$-GTH-antibiotics were 41.7%, 69.5%, 55.6% and 80.8% respectively injection in the uterus was more effective than the administration. Before or after the A.I., the above combination was most effective. 4. In antibiotics sensitivity test in vitro of the intravagina bacteria, 5.0%, 12.7% and 60.0% of bacteria were sensitive to Penicilline, Streptomycin and Gentamicin respectively.

Degradation of MS(Linear Alkylbenzene Sulfonate) by Plasmid (LAS(Linear Alkybenzene Sulfonate)의 Plasmid에 의한 분해)

  • Microorganisms capale of utilizing linear alkylbenzene sulfonates(LAS) as sole carbon source were isolated from industrial effluent by using LAS agar plates. The isolated strains were identified as Salmonella sp(BC-2) and Escherichia sp.(BC-3) from the results of morphological, cultural and biochemical tests. The optimal condition for the growth and biodegradation of LAS was the initial pH 7.0 and LAS concentration 0.1%. The isolated BC-2 and BC-3 strains harbored plasmid and LAS-degrading activity was lost when the plasmids were cured by mitomycin C. The plasmids were transformed into E. coli and transformants have the LAS-degrading activity. Isolated strains were examined for primary biodegradation rate of LAS in the medium by methylene blueactive substance(MBAS) method. Of these isolates, BC-2 and BC-3 strains degradated LAS upto 60% and high resistant to CdCl$_{2}$ and HgCl$_{2}$. Isolated strains were sensitive to chloramphenicol, kanamycin, rifampicin, streptomycin and tetracycline but resistant to ampicillin and lincomycin.] Its minimal inhibitory concentration(MIC) for ampicillin was more than 1500 $\mu $g/ml.

Isolation and Characterizaton of Plasmids from Streptomyces

  • Streptomyces spp. purchased from American Type Culture Collection and Institute for Fermentation in Osaka, and donated from Northem Regional Research Laboratory, and those isolated from soil samples were assayed to isolate many plasmids harboring streptomycetes. Among these qrganisms, 5 small size-plasmid carrying organisms SNUS 8810-597A, 8810-600, 8810-754, 8811-344, and 8811-347 were characterized and their plasmids pSJ597, pSJ600, pSJ754, pSJ344, and pSJ347 were isolated in a large scale. The plasmid harboring organisms were sensitive to neomycin, kanamycin, gentamicin, and thiostreptone, but some of them showed weak or strong resistance against streptomycin, chloramphenicol, ampicillin, and tetracycline. It was confirmed that pSJ597 and pSJ600 do not carry antibiotic biosynthetic genes. pSJ600 showed a pock-forming character.

Purification and Characterization of A Thermotolerable Restriction Endonuclease from Streptomyces violochromogenes D2-5

  • A thermotolerable restriction endonuclease. Svil, found in Streptomyces violochromogenes D2-5 was purified. For the purification, streptomycin sulfate and ammonium sulfate precipitation was used. Ph osphocellulose P-ll, DEAE-Cellulose and Sephacryl-S200 HR colum chromatography were also performed. The purified enzyme was found to be homogeneous and the molecular weight of the enzyme estimated by polyacrylamide gel electrophoresis containing 0.1$%$ SDS was about 32, 000 daltons. The recognition sequence and cleavage site of the enzyme were determined to be $5^1$-$TT\downarrow CGAA$-$3^1$ which is the same sequence as that of Asull. Unlike Asull, however, the Svil shows high thermal stability.

Studies on the Streptococcus suis isolated from slaughtered pigs (도축돈에서 분리된 Strptococcus suis에 대한 연구)

  • Streptococcus suis has been identified as a major cause of contagious disease in pigs Ithas been isolated worldwide from pigs suffering from meningitis, bronchopneumonia, polyserositis, polyarthritis and septicemia. Two hundred and fifty-five lung samples of slaughter pigs with gross lung lesions were collected from Jan. to Dec. 1995 in southern Chonnam area. Isolation of S suis were tested by biochemical, serological methods and antimicrobial susceptibility. The results were summerrized as follows ; 1. S suis was Isolated from 30 of 254 pneumonic lungs, 14 Isolates were isolated only, 12 were isolated with p. multocida and 4 were isolated with unidentified Gram positive cocci. 2. In biochemical characteristics studies, all isolates were not grown in 6.5% NaCl medium, and most isolates utilized L-leucine-2-naphtylamide and trehalose, didn't utilize sorbitol, ribose and L-arabinose. 3. In slide agglutination test, 6(20%) Isolates were classified as serotype 2, 4(13.3%) isolates were as serotype l/2, 16, 2 and 2(6.6%) were as serotype 1, 4, 13. 4. S suis isolates showed marked susceptibility to amoxicillin, oxacillin, cephalothin and cepoferazone and high resistance to kanamycin, streptomycin, tetracycline and erythromycin.
