In the present study, the volatile oil, chloroform extract and methanol extract of the woods of the plants Cedrus deodara and Pinus roxburghii were screened for their antibacterial and antifungal activities against Escherichia coli, Pseudomonas aeruginosa, Staphylococcus aureus, Bacillus subtilis, Streptococcus pyogenes, Candida albicans, Aspergillus niger, and Aspergillus clavatus using the Agar diffusion method. The susceptibilities of the microorganisms to the extracts were compared with each other and with a selected standard antibiotic. It was observed that the volatile oil and chloroform extracts showed the significant antibacterial activities while the least antibacterial activity was recorded with the methanolic extracts. The higher Minimum Inhibitory Concentration value of the extracts and oils against fungus suggested that the plants may possess less antifungal activity. Phytochemical analysis and thin-layer chromatography profiling revealed the presence of flavonoids and terpenoids in the oil and chloroform extracts, which could explain the antimicrobial activity. The findings suggest that the Cedrus deodara and Pinus roxburghii have antimicrobial properties and they can be used in the treatment of infectious diseases. However, further work is required in order to isolate the active constituents of the plants responsible for the antibacterial activity.
1. Objectives : We analyzed the case reports published by the journal of Sasang Constitutional Medicine, to study Sasang Constitution diagnosis and characteristic of clinical practice. 2. Methods : It has been examined 138 case reports of Sasang Constitution society published from 1994 to 2008. We analyzed. patient's number, publication year, distribution of Sasang Constitution, title's characteristics and bases of constitution diagnosis. 3. Results and conclusions : 1) In the analysis of patient's number, 1 case report was almost part of the case reports, and the case reports ware many published after 2001. 2) In the analysis distribution of Sasang Constitution, Soyangin was the most Sasang Constitution, and Tae-umin was the next one and Soeumin, Taeyangin followed them. 3) In the analysis of the journal's title, Sasang Constitution, disease and symptom of Sasang Constitution were more important than prescription recently. 4) In the analysis of the journal's purpose, unique disease and response of treatment were almost part of the case reports. 5) In the analysis of diagnosis bases, subjective bases were more important than objective bases and Sasang Constitutional drug response. therefore we should do researches in objective bases and drug response in the future.
A total of 315 microorgainisms were isolated from 256 pneumonic lung samples of pig in abattoirs of northern Chungnam area during February to November 1994, and Identified 97 strains as Pusteurella multocida, 89 strains as Staphylococcus spp, 54 strains as Streptococcus spp, 22 strains as Mycoplasma spp, 21 strains as Escherichia coli, 18 strains as Haemophilus parsuis, 11 strains as Corymebacteroi, pyogenes, and 3 strains as Acrinobacillus spp by Gram's and Dienes stain, and biological properties test Involved API system. After that, they were examined anti biotic susceptibility for ampicillin(AM), cephalothin(CF), chloramphenicol(CP), erythromycin(EM), kanamycin(KM), gentamicin(GM), neomycin(NM), penicillin(PC), streptomycin(SM), tetracycline(TE), tiamulin(TIA), tylosin(TYL), methicillin(DP), colistin(CL) and trimet hoprim(SXT). In antibiotics susceptibility test, 293 isolates except Mycoplasma spp 22 strain were highly susceptible to DF(79.2%) and AM(76.2%), but resistant to PC(14.0%), NM(19.5%) and KM (23.2%) The multiple drug resistant patterns were noted in most isolates, whereas only 7 isolates resistant to single drugs.
The Korea Centers for Diseases Control and Prevention interpreted that recent outbreaks of scarlet fever in Korea since 2011 was resulted from the expansion of scarlet fever notification criteria. To suggest a relevant hypothesis regarding this emerging outbreak, a time series analysis(TSA) of scarlet fever incidence between 2002 and 2016 was conducted. The raw data was the nationwide insurance claims database administered by the Korean National Health Insurance Service. The inclusion criteria were children aged ≤14 years residing in Jeju Province, Korea who received any form of healthcare for scarlet fever from 2002 to 2016. The season was defined as winter (December, January, February; Q1), spring (March, April, May; Q2), summer (June, July, August; Q3), and autumn (September, October, November; Q4). There were seasonal variations with showing peak season on Q1 and Q3. And three phases as 2002 Q2~2005 Q2, 2005 Q2~2009 Q4, and 2010 Q1~2016 Q4 were found between 2002 and 2016. The results from TSA suggested that the recent outbreak of scarlet fever among children in Jeju Province might be a phenomenon from 'unknown birth-related environmental factors' changed after 2010.
This paper presents the characteristics of gentamicin-loaded into cetyl trimethyl ammonium intercalated montmorillonite (GtM/CTMA/Mt) as a hybrid composite for a slow-released antibacterial delivery systems. The work describes the successful immobilization of gentamicin into the interlayers of surfactant-modified montmorillonite. Physicochemical characterization of the material is carried out by means of X-ray diffraction, scanning electron microscopy, transmission electron microscopy, and Fourier transform infrared spectroscopy. The kinetics of the gentamicin release is investigated by in vitro study and analyzed based on UV-Vis spectrometry. In addition, antibacterial study is performed towards Klebsiella pneumoniae Staphylococcus aureus, Escherichia coli, and Streptococcus pyogenes. The results show that the gentamicin loading into CTMA/Mt increases the effectiveness of the antibacterial activity, as shown by the higher inhibition zone for all tested bacteria, compared to gentamicin as a positive control. The kinetics study suggests that the gentamicin release obeys the modified Korsmeyer-Peppas model. The physicochemical study and activity test demonstrate the feasibility of the GtM/CTMA/Mt for practical applications.
The virus-induced genome editing (VIGE) system aims to induce targeted mutations in seeds without requiring any tissue culture. Here, we show that tobacco rattle virus (TRV) harboring guide RNA (gRNA) edits germ cells in a wild tobacco, Nicotiana attenuata, that expresses Streptococcus pyogenes Cas9 (SpCas9). We first generated N. attenuata transgenic plants expressing SpCas9 under the control of 35S promoter and infected rosette leaves with TRV carrying gRNA. Gene-edited seeds were not found in the progeny of the infected N. attenuata. Next, the N. attenuata ribosomal protein S5 A (RPS5A) promoter fused to SpCas9 was employed to induce the heritable gene editing with TRV. The RPS5A promoter-driven SpCas9 successfully produced monoallelic mutations at three target genes in N. attenuata seeds with TRV-delivered guide RNA. These monoallelic mutations were found in 2%-6% seeds among M1 progenies. This editing method provides an alternative way to increase the heritable editing efficacy of VIGE.

This study was performed in order to investigate changes in platycosides, as well as antimicrobial activities of bronchus diseases-inducing bacteria (Corynebacterium diphtheriae, Klebsiella pneumoniae, Staphylococcus aureus, and Streptococcus pyogenes) of Platycodon grandiflorum root (PGR) fermented by lactic acid bacteria (Leuconostoc mesenteroides N12-4, Leuc. mesenteroides N58-5, Lactobacillus plantarum N76-10, L. plantarum N56-12, Lactobacillus brevis N70-9, and L. brevis E3-8). Growth of L. plantarum on PGR was most active during lactic acid fermentation using different strains. Total platycoside, platycoside E, platycodin A, polygalacin $D_2$, polygalacin D, and diapioplatyco-side E contents of PGR fermented for 96 h at $37^{\circ}C$ by Leuc. mesenteroides and L. plantarum increased, whereas contents of platycodin D and platycodin $D_3$ were reduced. The antimicrobial activity on PGR fermented by L. plantarum N56-12 exhibited strong microbial proliferation for all four kinds of bronchus disease-inducing bacteria and was higher than that of non-fermented PGR extract. MIC of fermented PGR extract by L. plantarum N56-12 on C. diphtheriae, K. pneumoniae, S. aureus, and S. pyogenes were 45, 10, 50, and 25 mg/mL, respectively. Thus, this result shows that the antimicrobial activities of bronchus disease-inducing bacteria and platycoside content of PGR by L. plantarum N56-12 were higher than that of non-fermented PGR extract.
Essential oil of Thuja occidental leaves was extracted by steam distillation method, and chemical composition of essential oil was analyzed by GC-MS. The essential oil was tested to antimicrorbiaI activity against 28 test microorganisms and antioxident assay through DPPH free radical scaveging test and insecticidal activity by mosquito insecticide assay. The major constituents in the essential oil of T. occidental were monoterpene as d-isothujone, ${\alpha}-thujone$, camphor, L-fenchone, and hornyl acetate. The essential oil have broad antimicrobiaI activity, which showed strong antimicrobiaI activity for Streptococcus pyogenes as bacteria, Alternaria alternata, Aspergillus niger, Aspergillus oryzae, Saccharomyces cerevisiae, and Streptococcus mutans as fungi. The essential oil also showed DPPH free radical scavenging ability at concentration of $36\;{\mu}{\ell}/m{\ell}$. In spraying and fumigation tests, the essential oil had strong insecticidal activity against mosquito.
There are conflicting views for the polymerization process of human UDP-glucose dehydrogenase (UGDH) and no clear evidence has been reported yet. Based on crystal coordinates for Streptococcus pyogenes UGDH, we made double mutant A222Q/S233G. The double mutagenesis had no effects on expression, stability, and secondary structure. Interestingly, A222Q/S233G was a dimeric form and showed an UGDH activity, although it showed increased $K_m$ values for substrates. These results suggest that Ala222 and Ser233 play an important role in maintaining the hexameric structure and the reduced binding affinities for substrates are attributable to its altered subunit communication although quaternary structure may not be critical for catalysis.
Fifty three fungi isolated from soils of different microhabitats of eastern Himalayan range (3,400-3,600 msl) were screened for mycosynthesis of silver nanaoparticles (AgNPs) and their efficacy as antimicrobials were assessed in combination with commonly used antibiotics. Three isolates $Aspergillus$$terreus$ SP5, $Paecilomyces$$lilacinus$ SF1 and $Fusarium$ sp. MP5 identified based on morphological and 18S rRNA gene sequences were found to synthesize AgNPs. These nanoparticles were characterized by visual observation followed by UV-visible spectrophotometric analysis. The AgNPs synthesized by $Aspergillus$$terreus$ SP5, $Paecilomyces$$lilacinus$ SF1 and $Fusarium$ sp. MP5 showed absorbance maxima at 412, 419, and 421 nm respectively in the visible region. Transmission electron microscopy micrograph showed formation of spherical AgNPs of 5-50 nm size. The antimicrobial activity of the mycosynthesized nanoparticles were investigated alone and in combination with commonly used antibiotics for analysis of growth inhibition zone against test organisms, namely, $Staphylococcus$$aureus$ MTCC96, $Streptococcus$$pyogenes$ MTCC1925, $Salmonella$$enterica$ MTCC735 and $Enterococcus$$faecalis$ MTCC2729. The mycosynthesized nanoparticles showed potent antibacterial activity and interestingly their syngergistic effect with erythromycin, methicillin, chloramphenicol and ciprofloxacin was significantly higher as compared to inhibitions by AgNPs alone. The present study indicates that silver nanoparticles synthesized using soil borne indigenous fungus of high altitudes show considerable antimicrobial activity, deserving further investigation for potential applications.
