The diversity of heterotrophic flagellates was examined at marine sediments around Cape Tribulation, Australia. The species described belong to the Alveolates, Apusomonadiae, Cercomonadida, Choanoflagellida, Cryptomonadida, Diplomonadida, Euglenozoa incertae sedis, Kathablepharidae, Kinetoplastida, Pedinellids, Stephanopogonidae, Stramenopiles, Stramenopiles incertae sedis, Thaumatomonadidae and Protista incertae sedis. Among the 51 species from 38 genera encountered in this study is one new taxon: Glissandra similis n. sp., and two new names are introduced: Goniomonas abrupta (Skvortzov 1924) nomen nodum and Cercomonas skvortzovi (Skvortzov 1977) nomen nodum. There was little evidence for endemism because most flagellates including one new taxon described here have been reported.

Floating seaweed rafts are a surface-pelagic habitat that serve as substrates for benthic flora and fauna. Since 2008, Sargassum horneri clumps have periodically invaded the Korea Strait. In this study, the polymerase chain reaction-free small-organelles enriched metagenomics method was adopted to identify the species of epibiotic eukaryotes present in floating S. horneri fronds. A total of 185 species were identified, of which about 63% were previously undetected or unreported in Korean waters. The rafts harbored a diverse assemblage of eukaryotic species, including 39 Alveolata, 4 Archaeplastida, 95 Opisthokonts, 4 Rhizaria, and 43 Stramenopiles. Of these 185 taxa, 48 species were found at both Sargassum rafts collection stations and included 24 Stramenopiles, 17 Alveolata, and 7 Opisthokonts. Among these, the highest proportion (50%) of species was photo-autotrophic in basic trophic modes, while the proportion of phagotrophic, osmo- or saprotrophic, and parasitic modes were 43.8%, 4.2%, and 2.1%, respectively. This study demonstrates the contribution of floating Sargassum rafts as dispersal vectors that facilitate the spread of alien species.
본 연구는 메타게놈 분석법을 기초로 낙동강 하류 담수 환경의 물금과 기수 환경의 을숙도대교 정점에서 채수된 환경 시료내의 진핵 플랑크톤 종다양성 및 군집 구조를 비교 분석하고자 하였다. 수환경 시료에서 추출된 DNA에 대한 environmental Polymerase Chain Reaction(PCR)을 수행하여 18S rDNA 클론라이브러리를 구축하였고, colony PCR, PCR-Restriction Fragment Length Polymorphism(RFLP), 염기서열 결정 및 유사도 분석을 통하여 종다양성을 분석하였다. 물금 및 을숙도대교 정점에서 338개의 클론들을 분석하였고(170 clones, 물금; 168 clones, 을숙도대교), 그 결과 총 74개의 phylotype을 발굴하였다(49개, 물금; 25개, 을숙도대교). 발굴된 phylotype에 대한 계통 분석 결과, Stramenopiles, Cryptophyta, Viridiplantae, Alveolata, Rhizaria, Metazoa 및 Fungi 등의 분류군에 속하는 다양한 생물종이 발굴되었으며, 국내 미기록종 및 신종 후보 가능 생물종과 속(genus)이상의 새로운 분류학적 처리가 필요한 생물종의 존재를 확인하였다. 특히 Stramenopiles의 Pirsonia 및 Alveolata의 Perkinsea에 속하는 phylotypes 등 국내 미기록 생물종을 포함한 숨은 종다양성(cryptic species diversity)의 발굴은 분자모니터링 기법이 낙동강 하구역 수생태계 변화 모니터링을 위한 새로운 유용한 생물학적 정보를 제공할 수 있음을 제시하고 있다.
Free-living heterotrophic stramenopile flagellates, which lack chloroplasts, were encountered in deep-sea sediments of Gippsland Basin (Australia) and classified into 10 species (8 genera, 5 families, 3 orders). Their descriptions were based on living specimens by light microscopy. Those species rarely found in this study were Bicosoeca gracilipes, Caecitellus parvulus, Cafeteria minuta, Cafeteria roenbergensis, Pseudobodo tremulans, Spumella sp., Paraphysomonas sp., Actinomonas mirabilis, Ciliophrys infusionum and Developayella elegans. Their morphological characters and geographic distribution are presented.

Biodiversity and the community composition of micro-eukaryotic organisms were investigated in the Upo wetland in Korea using molecular analysis. Molecular identification was performed using cytochrome oxidase I (COI) and small subunit ribosomal DNA (SSU rDNA). The genomic DNA was isolated directly from soil samples. The COI and SSU rDNA regions were amplified using universal primers and then sequenced after cloning. In a similarity search of the obtained sequences with BLAST in the Genbank database, the closely related sequences from NCBI were used to identify the amplified sequences. A total of six eukaryotic groups (Annelida, Arthropoda, Rotifera, Chlorophyta, Bacillariophyta, and Stramenopiles) with COI and six groups (Annelida, Arthropoda, Rotifera, Alveolata, Fungi, and Apicomplexa) with SSU rDNA genes were determined in the Upo wetland. Among 38 taxa in 20 genera, which are closely related to the amplified sequences, 10 genera (50%) were newly reported in Korea and five genera (25%) were shown to be distributed in the Upo wetland. This approach is applicable to the development of an efficient method for monitoring biodiversity without traditional taxonomic processes and is expected to produce more accurate results in depositing molecular barcode data in the near future.
The infection of marine diatom Coscinodiscus wailesii by a parasitic protist from the Yongho Bay of Busan, Korea was observed during the diatom bloom events in 2017 through 2018. The morphological and molecular features suggested that the parasitic nanoflagellate Pirsonia diadema was responsible for the infection. During the study period, the parasite prevalence ranged from 0.3% to 3.3%, and infected C. wailesii cells were observed only at surface seawater temperatures ranging between 10.9 and 19.9℃, although the host population appeared at temperatures above 25℃. The parasite and host system was successfully established as cultures. Using the cultures, we determined the morphological features over the infection cycle, parasite generation time, parasite prevalence as a function of inoculum size, and zoospore infectivity and survival time. The diatom C. wailesii was readily infected by the parasite P. diadema, with a parasite prevalence reaching up to 100% and a zoospore to host inoculum ratio above 20:1. The survival and infectivity of the parasite zoospores decreased with age. While the zoospores could survive up to 88 hours, they quickly lost their ability to infect after 48 hours. These results could lead to a better understanding of the biology and ecology of the parasitoid infecting the giant-sized diatoms in coastal waters.
Blastocystis is a genus of unicellular heterokont parasites belonging to a group of organisms known as Stramenopiles, which includes algae, diatoms, and water molds. Blastocystis includes several species that habitat in the gastrointestinal tracts of organisms as diverse as humans, farm animals, birds, rodents, reptiles, amphibians, fish, and cockroaches. It is important to public health and distributed globally, but its prevalence in dogs in Korea has not been reported to date. Here, we collected 787 canine fecal samples and assessed Blastocystis infection by age, sex, region, season, and diarrhea symptoms. We determined Blastocystis subtypes using phylogenetic analyses based on 18S rRNA gene sequences. We identified, 10 Blastocystis positive samples (1.3%). A higher proportion of infected dogs was asymptomatic; however, infection rates did not significantly differ according to region, age, sex, and season. Phylogenetic analysis showed that the Blastocystis sp. identified belonged to 4 subtypes (STs), ST1, ST5, ST10, and ST14, thus revealed the genetic diversity of Blastocystis sp. in dogs Korean. This is first report on the presence of Blastocystis sp. in dogs Korean. This study revealed a lower infection rate than expected and differed from previous studies in STs. Further studies are warranted to observe the national infection status of Blastocystis in dogs and the genetic characteristics of this genus.
해양 같은 지리적 장벽에도 불구하고, 많은 담수조류는 세계의 다른 대륙에 서식하고 있다. 단세포 담수조류, Mallomonas caudata는 북반구의 미국, 유럽, 및 아시아에서 흔하며, 기후변화 감지나 수화현상 감시 같은 인간생활에 밀접하게 관련되어 있다. 본 종의 계통과 국내출현을 설명하기 위해 여섯 군데의 저수지에서 채집된 균주로부터 엽록체 rbcL과 핵 SSU유전자를 염기서열 분석하였다. 또한, 한국산 균주의 전자현미경적 구조를 조사하였다. 한국과 미국산 종의 SSU염기서열은 0.06%가, rbcL은 0.45%의 차이로 거의 동일하였다. 두 유전자를 이용한 계통수에서 본 종은 속의 다른 종들과 분명히 분리되지만, 단계통군은 아니었다. 근형질은 기저체의 다층판에 부착된 가로무늬의 microfibril들로 구성되어 있었으며, microfibril들의 끝은 핵의 표면 위에 배열되어 있었다. 이 근형질은 Synurophyceae에서 전형적으로 보이는 기저체-핵 연결자이다. Mallomonas가 SSU와 rbcL자료에 의해 지지되지 않는 결과는 분류군 추가와 함께 좀더 연구되어야 할 것으로 사료된다.
