Biological Control of Plant Pathogen by Pmdornonas sp. (Pseudomondas sp.에 의한 채소병원균의 생물학적 억제)

  • For the selection of powerful antagonistic bacterium for biological control of soil borne Eminia carotovora subsp. carotovora causing rot of vegetable, excellent strains (S4, S14, 565) were selected from 1,196 strains of bacteria which were isolated from rhizosphere in vegetable root rot-suppresive soil. Strains were identified to be Pseudomonas species with Api 20NE kit. Antagonistic substance was produced in 523 synthetic broth medium at pH 7~8 and $30^{\circ}C$ during 3 days culture. The substance was stable in the pH range of 6 to 9. When the basal medium was supplemented with mannitol and sorbitol as carbon source and calcium chloride as metal salt, the production of the inhibitory substance was increased. The inhibitory acitivity was increased by the addition of fertilizer in soil. The isolated strains were resistant to the agricultural chemical such as benomyl and fosethyl-Al-folpet, and the antibiotics such as penicillin and lincomycin. We had found that Pseudomonas sp. S14 strain had a single plasmid. After treated with acridin orange for curing, we confirmed the existence of antagonistic gene in the chromosomal DNA.

Isolation and Characterization of Exopolysaccharide-Producing Bacteria from Korean Fermented Vegetables (전통 침채류 유래 다당 생산균의 분리 특성)

  • Four bacteria producing viscous exopolysaccharides (EPSs) were isolated from Korean fermented vegetables (Cucumber kimchi, Young radish kimchi, Green onion kimchi) using a selection medium intended for isolating bacteria with tannin-degrading activity. They were identified phylogenetically by 16S rDNA sequence analysis and found to be very close to Enterobacter cowan ii, Escherichia senegalensis, Enterobacter asburiae, and Enterobacter ludwigii. Strain CK31, the most efficient EPS-producer, produced a heteropolysaccharide with an approximate molecular weight of 420 kDa. The neutral sugar fraction of the EPS was composed of rhamnose, fucose, arabinose, mannose, galactose, and glucose.

Biodegradation and Saccharification of Wood Chips of Pinus strobus and Liriodendron tulipifera by White Rot Fungi

  • Degradation and glucose production from wood chips of white pine (Pinus strobus) and tulip tree (Liriodendron tulipifera) by several white rot fungi were investigated. The highest weight losses from 4 g of wood chips of P. strobus and L. tulipifera by the fungal degradation on yeast extract-malt extract-glucose agar medium were 38% of Irpex lacteus and 93.7% of Trametes versicolor MrP 1 after 90 days, respectively. When 4 g of wood chips of P. strobus and L. tulipifera biodegraded for 30 days were treated with cellulase, glucose was recovered at the highest values of 106 mg/g degraded wood by I. lacteus and 450 mg/g degraded wood by T. versicolor. The weight loss of 10 g of wood chip of L. tulipifera by T. versicolor on the nutrient non-added agar under the nonsterile conditions was 35% during 7 weeks of incubation, and the cumulative amount of glucose produced during this period was 239 mg without cellulase treatment. The activities of ligninolytic enzymes (lignin peroxidase, manganese peroxidase, and laccase) of fungi tested did not show a high correlation with degradation of the wood chips and subsequent glucose formation. These results suggest that the selection of proper wood species and fungal strain and optimization of glucose recovery are all necessary for the fungal pretreatment of woody biomass as a carbon substrate.

Screening of Domestic Silkworm Strains for Efficient Heterologous Protein Expression by Bombyx mori Nuclear Polyhedrosis Virus (BmNPV)

  • Recombinant proteins can be generated quickly and easily in large amounts and at low-cost in silkworm larvae by using Bombyx mori nuclear polyhedrosis virus (BmNPV). We searched for high-permissive silkworm strains that have high production levels of heterologous proteins and are thus suitable for use as biofactories. In this study, we performed the analysis using a BmNPV vector expressing luciferase as a marker, and we confirmed protein expression by evaluating luciferase activity, determined by western blotting and luciferase ELISA, and confirmed transcription expression by semi- and quantitative real time PCR. For the selection of host silkworm strains, we first chose 52 domestic BmNPV sensitive strains and then identified 10 high-permissive and 5 low-permissive strains. In addition, to determine which hybrid of the high-permissive strains would show heterosis, nine strains derived through three-way crossing were tested for luciferase activity by western blotting, and luciferase ELISA. We found a correlation between luciferase activity and luciferase protein expression, but not transcription. There was no noticeable difference in protein expression levels between Jam313 as the high-permissive control strain and the three-way hybrid strains; however, the three-way cross strains showed lower luciferase activity compared with Jam313. In this study, luciferase protein production in the larvae of 52 domestic silkworm strains was elucidated using BmNPV.

Consolidation Behaviour of Dredged Clay Ground Improved by Horizontal Drain Method (수평배수공법에 의해 개량된 준설점토지반의 압밀거동에 관한 연구)

  • In this study, a large consolidation test was carried out to estimate the consolidation behaviour of dredged clay ground improved by horizontal drain using plastic board drain with a vacuum pressure. The test results were analyzed by a numerical simulation using potential consolidation theory applied to a hollow cylinder. The rapid decreases in pore pressure and the drain speed in the plastic board indicate that the consolidation occurred quickly after the vacuum state was applied to the test soil. According to the numerical analysis obtained by applying the linear potential consolidation theory to a clay hollow cylinder with external radial drainage, the pore pressure is affected by the strain and the permeability of the soil rather than by the diffusion types. Therefore, measured surface settlement agreed with the numerical solution at the point where consolidation pressure increasing rate u: -0.5. Also the behaviour of the clay layer settlement in the place where the drain was installed was similar to that shown in Barron's consolidation theory. Finally, the design and construction procedure including the selection of the appropriate arrangement of horizontal drains were discussed based on the results of the laboratory tutsts. It is also shown that the potential consolidation theory make it possible to predict consolidation behaviour in the field using horizontal drains exactly.

  • The better industry develops, the more spaces need but in the limited area. Most building become larger and more complicated if the more spaces need in the constant area. And this leads to do underground work in long period generally six(6) months for 6 basement stories due to the selection of TOP DOWN technique. Working environment in this underground area can be problems and should not be overlooked, because air quality in underground spaces become quickly worse. Recently, department name to control construction safety has been changed to ENVIRONMENT & SAFETY TEAM from SAFETY TEAM. This means that it is very important to control against environmental condition at site so much. Overall construction work as well as underground work should conform to the requirement of working environment, particularly against inhabitants around the construction area. Strut protection, one of earth protection method, in case to 40m long strut may become weaker due to thermal stress or its longitudinally compressive strain and the another one, earth anchor protection may not be applied to the site In case of encroaching on vertical underground borderline because of regulation to prohibit it. It is necessary that TOP DOWN technique should be introduced in order to solve the external and internal problem of the site such as difficulty level of the work, potential danger with excavating depth, and shortening workperiod. It is needed that improving way of working condition should be shown and simplified computer simulation program should be also provided for checking pollution level & ventilation, excluding of lighting problem here. Results measured with conformance to the Regulation for Working Environment Measurement, enforced by Ministry of Labor have been applied to the computer program developed here. Sample air taken at unit workplace which was considered as exposing condition of pollutant at breathing point and within a range of behavior of the workers, Identified exposing group in underground work, using Moded Flow Life Finally, three types of ventilation system, type I with blower & ventilator, type II natural supply with mechanical ventilation system, and type I mechanical ventilation with Drivent Fan Unit System are selected for this study.

IVET-based Identification of Virulence Factors in Vibrio vulnificus MO6-24/O

  • Vibrio vulnificus is an opportunistic pathogen that causes septicemia in humans. To identify the genes associated with its pathogenicity, in vivo expression technology (IVET) was used to select genes specifically expressed in a host, yet not significantly in vitro. Random lacZ-fusions in the genome of V vulnificus strain MO6-24/O were constructed using an IVET vector, pSG3, which is a suicide vector containing promoterless-aph and -lacZ as reporter genes. A total of ${\sim}18,000$ resulting library clones were then intraperitoneally injected into BALB/c mice using a colony forming unit (CFU) of $1.6{\times}10^6$. Two hours after infection, kanamycin was administered at $200{mu}g$ per gram of mouse weight. After two selection cycles, 11 genes were eventually isolated, which were expressed only in the host. Among these genes, VV20781 and VV21007 exhibiting a homology to a hemagglutinin gene and tolC, respectively, were selected based on having the highest frequency. When compared to wild-type cells, mutants with lesions in these genes showed no difference in the rate of growth rate, yet a significant decrease in cytotoxicity and the capability to form a biofilm.

  • 590 strains of Rhizobia were isolated from root nodules of the legumes collected at 223 sites in Korea. According to their host specificities they were classified into R. japonicum(218 strains), R. phaseoli(101 strains), R. trifolii(97 strains), R. meliloti(4 strains), R. leguminosarium(1 strain), Rhizobium species(101 strains), and unidentified species(159 strains). 3 potent strains R-138, R-168, and R-214 of R. japonicum have been selected based on the infectivity to soybean cultivar and effeciency of nitrogen fixation. It was observed that the fast-growing strains of R. japonicum contained 1 to 4 plasmids of M.W. of 35-300 Md. However, plasmids were hardly detected for the slow-growing strains.

A Model Study on Deformability of A Transversely Isotropic Rock (평면이방성 암석의 변형특성 모델연구)

  • In the uniaxial compressive test of a single specimen of transversely isotropic rock, its 5 independent elastic constants can not be defined since maximum 4 independent strain measurements are available theoretically. In order to solve this problem, one equation proposed by Saint Venant in 19C and confirmed by Lekhnitskii through the test experiences has been used for long time. Accordign to authors' experiences, however, this equation turned out to give erroneous elastic constants in some cases. Three new equations are suggested and their compatibilities are discussed in this paper. As the results of the analyses of the models, Lekhnitskii's suggested equation is effective for the specimen with the high dip angle whereas it results in the large erred output for that with dip angle less than $25{\sim}30$. It was found that the effectivenesses of three suggested equations and their compatibilities are subject to the dip angle and not to the amounts of elastic constants. Guide map to the selection of the compatible one of those suggested equations is presented as a result of the study.

Genetic Transformation and Plant Regeneration of Codonopsis lanceolata Using Agrobacterium (Agrobacterium에 의한 더덕의 형질전환과 식물체 재분화)

  • To obtain transformed plants, we cocultured cotyledonary explants of Codonopsis lanceolata with Agrobacterium tumefaciens LBA4404, a disamed strain harboring a binary vector pBI121 carrying the CaMV35S promoter-$\beta$-glucuronidase (GUS) gene fusion used as a reporter gene and NOS promoter-neomycin phosphotransferase gene as a positive selection marker in MS liquid medium with 1mg/L BA. After 48 h of culture, explants were transferred onto MS solid medium with Img/L BA, 250mg/L carbenicillin, and 100mg/L kanamycin sulfate and cultured in the dark. Numerous adventitious buds formed on the cut edges of the explants after 2 weeks of culture. When subjected to GUS histochemical assay buds showed a positive response at a frequency of 15%. Explants formed adventitious shoot at a frequency of 56.7%, after 6 weeks of culture. Upon transfer onto the basal medium, most of the shoots were rooted and subsequently the regenerants were transplanted to potting soil. Southern blot analysis confirmed that the GUS gene was incorporated into the genomic DNA of the GUS-positive regenerants.
