Effect of Mouthrinses prepared by Sterilized Water-Generating Device on the Control of Periodontal Disease (살균수 발생장치에 의해 생성된 양치용액이 치주질환에 미치는 효과)

  • The aim of this study was to evaluate the clinical efficacy of mouthrinses prepared by sterilized water-generating $device(Purister^{(R)})$ on the control of gingivitis and incipient periodontitis when it was used as a adjunctives to the mechanical plaque control. 40 healthy patients with gingivitis or incipient periodontitis were divided into two groups. Patients in the experimental group use mouthrinses prepared by sterilized water-generating $device(Purister^{(R)})$ after tooth brushing while patients in the control group do only tooth brushing for plaque control. All patients received scaling and tooth brushing instruction. 1 week after scaling was set as baseline. Probing pocket depth, clinical attachment level, and bleeding on probing were scored at baseline, 4 weeks. Gingival index and plaque index were scored at baseline, 2 weeks, and 4 weeks. The results were as follows: 1. In the experimental group, gingival index, plaque index, probing pocket depth, and clinical attachment level showed statistically significant decrease, but in the control group, significant increase(p<0.05). 2. There was no significant difference between the experimental and the control group in bleeding on probing, but significant decrease in the experimental group and Significant increase in the control group In a time-dependent manner(p <0.05). From these results, it can be concluded that regular use of mouthrinses prepared by sterilized water-generating $device(Purister^{(R)})$ as adjunctives of mechanical plaque control, may be effective to prevent and treat gingivitis and incipient periodontitis.

Survival Studies of Some Enteric Pathogens in Sewage (장내 병원균의 하수중 생존 상태에 관한 연구)

  • In order to study the growth and survival of enteric pathogens causing water-borne infections in sewage, the filter-sterilized and autoclaved sewages of Dae Gu City were inoculated with Salmonella typhimuriuim, Shigella flexneri 2a, Sh. sonnei I, Vibrio eltor and V. parahaemolyticus, as test series and Escherichia coli as control. After varying periods of incubation up to 15 days at $4^{\circ}$, $15^{\circ}$, $25^{\circ}$ and $37^{\circ}C$, viable cells in the inoculated sewages were counted by colony count technique. Distilled water and 0.9% saline were subjected to inoculation of the organisms was observed in the filter-sterilized and autoclaved sewages at $4^{\circ}$ and the sewages became sterile within a few days. At $15^{\circ}$, no growth and rapid inactivation of the organisms in the filter-sterilized sewage and slight or no growth in the autoclaved sewage was noted. Some viable cells were found in the autoclaved sewage after 15 days. A considerable growth was observed in the filter-sterilized and autoclayed sewages, at $25^{\circ}$ and $37^{\circ}$, and large numbers of viable cells were found even after 15days of incubation. In general, the autoclaved sewage supproted the growth more noticeably than the filter-sterilized, except for V.parahaemolyticus which grew well in filter-sterilized sewage. No marked difference was noted between incubations at $25^{\circ}$ and $37^{\circ}$, but V. parahaemolyticus showed a slightly more active growth at $25^{\circ}$ than at $37^{\circ}$. Distilled water inactivated the organisms within a few days, but saline supported the growth at $25^{\circ}$ and $37^{\circ}$. Marked differences were noted in the survival test of sewages pathogens of different origins.

  • Dehiscence of ginseng seeds under the various conditions, growing experiment in the different soil conditions, and growing experiment with extracts obtained by water passed the pot of 4 year ginseng plants were performed. The results obtained are as follows: The survival rate of ginseng plants in the soil of ginseng field sterilized by MIYAZAWA's method was higher than that of the non-sterilized, but lower than that of the control. The survival rate of ginseng plants on the sterilized soil of ginseng field was lower than that on the surrounding soil. The growth of ginseng plants cultivated with the water passed through the pot-planted 4-year-old ginseng plants was strongly inhibited in comparison with the control. Besides biological cause, chemical products excreted from the root of ginseng plants might be the important factors of soil sickness in the successive cultivation of the same plants.

Quality Changes in Apple Juice Containing Pulp upon Sterilization by Hot Water (열수 살균 조건에 따른 과육 함유 사과주스의 품질 변화)

  • We investigated the physicochemical quality characteristics of apple juice upon sterilization using hot water, and under various storage conditions. None of sugar content, acidity, or pH differed significantly among various sterilization conditions but chromaticity was considerably reduced in sterilized juice compared with control material. The chromaticity of non-sterilized juice decreased significantly after sterilization compared with material supplemented with vitamin C (0.1%, w/v). Fungi, yeast, and aerobic bacteria were detected in juice sterilized at $65^{\circ}C$ for 10 or 20 min, but no microorganisms were observed in juice sterilized by other procedures. Vitamin C content affected sterilization temperature to a greater extent than sterilization time. When juice was stored at $4^{\circ}C$ or $37^{\circ}C$ for 3 weeks after application of different sterilization conditions, almost no change in acidity, sugar content, or pH was observed, regardless of sterilization mode or storage period. However, chromaticity decreased with longer storage. Vitamin C levels were reduced by higher sterilization temperatures. However, longer storage periods had the greatest effect on reduction of vitamin C levels, which tended toward lower values regardless of differences in sterilization and storage conditions. In sensory evaluation tests, all of taste, color, and overall preference were highest for juice sterilized at $75^{\circ}C$ for 20 min. The ASC value was low at a storage temperature of $4^{\circ}C$ and at high sterilization temperatures, and a long storage period was associated with a greater ASC value. Thus, the quality of apple juice was excellent when juice was hot water-sterilized, with additional vitamin C, at $75^{\circ}C$ for 20 min, followed by storage at $4^{\circ}C$.

Degradation of Dinobuton in Soil and Solution (Dinobuton의 토양(土壤) 및 용액중(溶液中)에서 분해(分解))

  • This study was carried out to investigate the stability of dinobuton (2-sec-butyl-4,6-dinitrophenyl isopropyl carbonate) in distilled water and buffer solutions and its persistence in soils. When dinobuton was incubated at $30^{\circ}C$ and $60^{\circ}C$ in distilled water, the half-lives of dinobuton was 28 and 6 days, respectively. The decomposition of dinobuton was, therefore, faster at high temperature than at low temperature. The half-life of dinobuton was about 27 days in the acidic solution $(pH\; 4{\sim}6)$, whereas 10 and 4 days in the alkaline solutions of pH 9, and 10, respectively. Thus dinobuton was stable in acidic solution, and unstable in alkaline solution. Dinoseb (2-sec-butyl-4,6-dinitrophenol), which is produced in the degradation process of dinobuton, was produced in small amounts in distilled water and buffer solutions. The half-life of dinobuton in sterilized soil was about 16 days longer than in non-sterilized soil. Dinoseb was also more persistent in sterilized soil than in non-sterilized one.

  • This study was attempted to develop surface washing-system of ginseng. The effect of sterilization, washing and keeping freshness of ginseng through analyzing unit process were examined to establish optimal condition for washing system. Surface washing method of fresh ginseng used two way and full cone spray type. Sterilization was used at $2^{\circ}C$ water with electrolysis water of 50 and 80 ppm. Ginseng was sterilized with electrolysis water during 30 and 60 s, dehydrated during 1 min and dried during 1min at 30 and $50^{\circ}C$. Hardness of surface-washed ginseng showed good result on 1 min spraying time with 80 ppm electrolysis water at $10^{\circ}C$ storage. Ginseng with 80 ppm electrolysis water was sterilized better with $1.05{\times}103$. There are no changes with 0% on appearance quality at 80 ppm electrolysis.

Effect of Plant Growth Regulators on Multiple Shoot Formation and Elongation from Shoot Tip Cultures of Grape Species (포도의 경정배양에 의한 다아체형성 및 신장에 미치는 생장조절제의 영향)

  • Shoot tips of grape were cultured in uitro and tried to identify optimal culture conditions for regeneration, multiple shoot formation from meristemoid tissue and those subsequent elongation of multi-shoots. Healthy growing shoots were taken in early May, rinsed with running tap water, soaked in a neutral detergent and washed with soft brushing, and washed out with tap water, then sterilized with 10g Ca(ClO)$_2$/140 mL distilled water (Wilson's solution) for 5 min. Survival percentage of the cultures which were sterilized as above procedures was highly increased, compared with the other sterilized method. Propagation of multi-shoots from meristemoid showed a good response in 3/4 strength MS medium enriched with 0.1 mg/L NAA and 3.0 mg/L BA. Shoot elongation from multi-shooting clump well occurred in 3/4 strength MS medium supplemented with 80 mg/L adenine sulfate, 0.1 mg/L NAA and 1.0~2.0 mg/L BA.

The Outbreak and Propagule formation of black root rot caused by Calonectria crotalariae in Korea (콩 흑색뿌리썩음병의 발생과 Propagule의 형성)

  • The infection rate of soybean black root rot disease caused by Calonectria crotalariae was about $14\%$. The isolated fungi from the infected soybean roots and stems were Calonectria crotalariae, Fusarium solani, F. roseum, Phomopsis sojae, Pythium aphanidermatum, Rhizoctonia solani and Macrophomina sp. Among them, C. crotalariae was the most virulent pathogen under the laboratory conditions. Mycelial growth and microsclerotial formation were good on PSA containing 1000cc of water, 100g of potato and 20g of sugar. Mycelial growth, sporulation and microsclerotial formation were good on sterilized root. Perithecial formation was better in the dark condition than in the light. Survival of macroconidia was not available between $0\~25\%$ soil water content. Microsclerotia and mycelium in infected plant debris were survived for 4 months at to $8\%\;50\%$ soil water content. The plant height, when inoculated with $1.2\%$ inoculum density, reached approximately half of uninoculated plants. Disease severity was much higher at nonsterilized soil than completely sterilized soil. It was determined that the host range of this pathogen includes soybean, peanut, green bean and red bean.

Effect of Non-sterilized Makgeolli Consumption on Blood Glucose Level and Survival Rate in Streptozotocin-induced Type I Diabetic Mice (Streptozotocin으로 유도된 제1형 당뇨 생쥐에서 생막걸리 투여가 혈당 및 생존률에 미치는 영향)

  • Makgeolli is a health beneficial food for diabetes compared to other alcoholic beverages. We examined the effect of Makgeolli on blood glucose level and survival rate in a streptozotocin (STZ)-induced diabetic mouse model. We force fed 30 male STZ-induced diabetic ICR mice Makgeolli consisting of 6% alcohol (DM-MAK), 6% ethanol (DM-EtOH), or distilled water (DM-DW) for 4 weeks. In the DM-MAK group, food intake and water intake were higher than those of other groups after 4 weeks. Body weight, however, was not different among the experimental groups. We also found no significant difference in blood glucose level among the experimental groups. In normal ICR mice fed Makgeolli for 1 week, the area of the blood glucose curve was higher than those of other groups fed 6% ethanol, 2% glucose, or distilled water. Survival rates of STZ-induced diabetic mice fed Makgeolli, 6% ethanol, or DW for 4 weeks were 100%, 25%, and 62.5%, respectively. In conclusion, Makgeolli had no beneficial effect on blood glucose in a STZ-induced diabetic mouse model, although their survival rate was high. These results show that Makgeolli has an effect on type 1 diabetes through other mechanisms than blood glucose control.

Effect of Reed-Bed using Ulsan-habitated P.australis, T.orientalis, and P.aundinacea L. on Removing Pollutants from Sewage (울산지역에서 자생하는 갈대, 부들, 갈풀을 이용한 Reed-Bed의 생활하수 정화능력 연구)

  • We examined whether several reeds, which are found around Ulsan area, could be used for downflow reed-bed to remove pollutants of sewage. Three kinds of reed, such as Phagnltes auspuis, ha orientdls, and Phduis aundinacea L., were collected from their habitats near the Taehwa River in Ulsan City. In the minimized model system of dowMlow reed-bed, P.auskdls appeared to reduce BOD more than others did but s119h11y Increase total amount of nitrogen(N). When p. auspdis were placed in the sterilized water. total nitrogen was found to be signincantly increased dependent on the number of experimental plant In the sterilized state, but it was rather decreased in the non-sterilized state. With these results, nlicroorganisms attached to p.auspuis roots can be thought to work for removal of pollutants. Therefore, these microorganisms and their habitat, p. auskdis reed bed, together can be used for sewage treatment It was suggested that oxygen Is produced by photosynthesis reaction of P ecustrdis. The increased oxygen may help microorganisms in their habitats to work on the removal of pollutants.
