본 연구는 소 배반포의 내부 세포괴로부터 다능성(pluripotency)을 지닌 배아 줄기 세포(embryonic stem cell) 또는 그 유사 세포를 분리 및 배양함으로써 줄기 세포 관련 분야의 기반 기술을 확립하고자 하였다. 소 체외수정란을 $10{\sim}12$일간 체외배양하여 생산된 부화 배반포를 세포분열이 불활성화된 생쥐 태아 섬유아 세포(mouse embryonic fibroblast, MEF) 위에서 배양하여 콜로니 형성을 유도하였으며, 이들로부터 내부 세포괴 유래의 형태를 지닌 것만을 광학현미경 하에서 물리적으로 분리하여 약 $5{\sim}7$일 간격으로 계대배양을 실시하였다. 이러한 방법을 통하여 배아 줄기 유사 세포의 특성을 40계대 이상 유지하는 2개의 세포주를 확립하였다. 각각의 세포주들은 높은 alkaline phosphatase(AP) 활성을 지니고 있었으며, 형광 면역 염색법과 PCR 기법을 사용하여 Oct-4, Nanog, STAT3, SSEA3 및 SSEA4의 발현을 관찰할 수 있었다. 이러한 결과를 종합하여 볼 때, 본 연구에서는 소 배반포로부터 배아 줄기 세포주를 확립하는 제반 기술이 확립되었다고 판단되며, 향후 관련 분야 연구에 활용될 수 있을 것으로 기대된다.
Side population (SP) cells have stem cell-like properties with a capacity for self-renewal and are resistant to chemotherapy and radiotherapy. Therefore the presence of SP cells in human breast cancer probably has prognostic value. Objective: To investigate the characteristics of SP cells and identify the relationship between the SP cells levels and clinico-pathological parameters of the breast tumor and disease-free survival (DFS) in breast cancer patients. Materials and Methods: A total of 122 eligible breast cancer patients were consecutively recruited from January 1, 2006 to December 31, 2007 at Yunnan Tumor Hospital. All eligible subjects received conventional treatment and were followed up for seven years. Predictors of recurrence and/or metastasis and DFS were analyzed using Cox regression analysis. Human breast cancer cells were also obtained from fresh human breast cancer tissue and cultured by the nucleic acid dye Hoechst33342 with Verapami. Flow cytometry (FCM) was employed to isolate the cells of SP and non-SP types. Results: In this study, SP cells were identified using flow cytometric analysis with Hoechst 33342 dye efflux. Adjusted for age, tumor size, lymph nodal status, histological grade, the Cox model showed a higher risk of recurrence and/or metastasis positively associated with the SP cell level (1.75, 1.02-2.98), as well as with axillary lymph node metastasis (2.99, 1.76-5.09), pathology invasiveness type (1.7, 1.14-2.55), and tumor volume doubling time (TVDT) (1.54, 1.01-2.36). Conclusions: The SP cell level is independently associated with tumor progression and clinical outcome after controlling for other pathological factors. The axillary lymph node status, TVDT and the status of non-invasive or invasive tumor independently predict the prognosis of breast cancer.
Pluripotency of human embryonic stem cell (hESC) is one of the most valuable ability of hESCs for applying cell therapy field, but also showing side effect, for example teratoma formation. When transplant multipotent stem cell, such as mesnchymal stem cell (MSC) which retains similar differentiation ability, they do not form teratoma in vivo, but there exist limitation of cellular source supply. Accordingly, differentiation of hESC into MSC will be promising cellular source with strong points of both hESC and MSC line. In this study, we described the derivation of MSC like cell population from feeder free cultured hESC (hESC-MSC) using direct differentiation system. Cells population, hESC-MSC and bone marrow derived MSC (BM-MSC) retained similar characteristics in vitro, such as morphology, MSC specific marker expression and differentiation capacity. At the point of differentiation of both cell populations, differentiation rate was slower in hESC-MSC than BM-MSC. As these reason, to verify differentially expressed molecular condition of both cell population which bring out different differentiation rate, we compare the molecular condition of hESC-MSC and BM-MSC using 2-D proteomic analysis tool. In the proteomic analysis, we identified 49 differentially expressed proteins in hESC-MSC and BM-MSC, and they involved in different biological process such as positive regulation of molecular function, biological process, cellular metabolic process, nitrogen compound metabolic process, macromolecule metabolic process, metabolic process, molecular function, and positive regulation of molecular function and regulation of ubiquitin protein ligase activity during mitotic cell cycle, cellular response to stress, and RNA localization. As the related function of differentially expressed proteins, we sought to these proteins were key regulators which contribute to their differentiation rate, developmental process and cell proliferation. Our results suggest that the expressions of these proteins between the hESC-MSC and BM-MSC, could give to us further evidence for hESC differentiation into the mesenchymal stem cell is associated with a differentiation factor. As the initial step to understand fundamental difference of hESC-MSC and BM-MSC, we sought to investigate different protein expression profile. And the grafting of hESC differentiation into MSC and their comparative proteomic analysis will be positively contribute to cell therapy without cellular source limitation, also with exact background of their molecular condition.
We investigated the effect of transforming growth factor beta 1 ($TGF-{\beta}1$) on equine hyaluronan synthase 2 (HAS2) gene expression and hyaluronan (HA) synthesis in culture models of articular chondrocytes. Equine chondrocytes were treated with $TGF-{\beta}1$ at different concentrations and times in monolayer cultures. In three-dimensional cultures, chondrocyte-seeded gelatin scaffolds were cultured in chondrogenic media containing 10 ng/mL of $TGF-{\beta}1$. The amounts of HA in conditioned media and in scaffolds were determined by enzyme-linked immunosorbent assays. HAS2 mRNA expression was analyzed by semi-quantitative reverse transcription polymerase chain reaction. The uronic acid content and DNA content of the scaffolds were measured by using colorimetric and Hoechst 33258 assays, respectively. Cell proliferation was evaluated by using the alamarBlue assay. Scanning electron microscopy (SEM), histology, and immunohistochemistry were used for microscopic analysis of the samples. The upregulation of HAS2 mRNA levels by $TGF-{\beta}1$ stimulation was dose and time dependent. $TGF-{\beta}1$ was shown to enhance HA and uronic acid content in the scaffolds. Cell proliferation and DNA content were significantly lower in $TGF-{\beta}1$ treatments. SEM and histological results revealed the formation of a cartilaginous-like extracellular matrix in the $TGF-{\beta}1$-treated scaffolds. Together, our results suggest that $TGF-{\beta}1$ has a stimulatory effect on equine chondrocytes, enhancing HA synthesis and promoting cartilage matrix generation.
만능성 인간 배아줄기세포로부터 확립된 신경줄기세포 또는 신경전구세포는 퇴행성 신경질환 세포치료제로 이용될 수 있는 다양한 종류의 신경세포로 분화 유도될 수 있다. 하지만, 인간 배아줄기세포로부터 신경세포를 생산하기 위한 기술은 아직 많은 장애를 가지고 있다. 인간 배아줄기세포 유래 신경전구세포에서 특징적으로 나타나는 신경관 유사로제트에 대한 이해는 인간 배아줄기세포 신경 분화의 효율을 높이는데 유용한 정보를 제공할 것으로 사료된다. 일반적으로 신경로제트(neural rosette)는 분화 중인 배아체를 부착 배양함으로써 유도하지만, 이 방법은 시간이 걸리고 복잡하다는 단점이 있다. 본 연구에서는 신경로제트가 부착배양을 하지 않고 부유배양으로 형성될 수 있는지 조사하였다. 우선적으로, 배아체 형성 및 신경분화에 인간 배아줄기세포 클럼프(clump) 크기가 영향을 주는지를 조사하였고, 사방 $500\;{\mu}m$ 크기의 인간 배아줄기세포 클럼프가 신경 분화 유도에 가장 효과적임을 확인하였다. 로제트 형성을 유도하기 위해, 사방 $500\;{\mu}m$ 크기의 인간 배아줄기세포 클럼프를 1주일 동안 EB 배양배지에 부유 배양함으로써 균일한 크기의 배아체를 얻은 후, NES 배양 배지에서 부가적으로 $1{\sim}2$주 동안 계속 부유 배양한 결과, $7{\sim}10$일 사이에 신경관 유사 로제트가 형성됨을 확인하였다. 로제트 형성 세포의 신경전구세포로서 특성은 RT-PCR과 면역형광염색법을 이용한 신경전구세포 특이적 마커(vimentivi, nestin, MSI1, MSI2, Sox1, Tuj1) 발현을 통해 확인하였다. 또한, 성장인자를 제외한 NES 배양 배지에서 신경로제트를 $2{\sim}6$주 동안 지속적으로 배양하면 성숙 신경세포로의 말단 분화가 유도됨을 확인하였다. 신경세포 특이적 마커(Tuj1, MAP2, GABA)와 신경아교 특이적 마커($S100{\beta}$, GFAP)는 $2{\sim}3$주 또는 4주 후에 각각 발현이 유도됨을 확인하였고, 희소 돌기아교 특이적 마커(O1과 CNPase)는 $5{\sim}6$주 후에 발현이 증가함을 확인하였다. 본 연구결과는 신경로제트가 부유 배양시스템에서 성공적으로 형성됨을 보여주고 있으며, 이는 인간 배아줄기세포의 신경 분화를 이해하고, 신경전구세포 유도 과정을 단순화하는데 효과적으로 이용될 수 있을 것으로 사료된다.
목 적: 사람의 HAD와 HUC를 심근세포로 분화 유도하고자 하였다. 연구방법: 사람의 HAD와 HUC를 분리하여 5-azacytidine을 24시간 처리하고 여러 가지 BMP와 FGF을 첨가하여 배양하였다. 또한 HUC은 BMP와 FGF와 함께 activin A 또는 TGF-$\beta$1 또는 Wnt inhibitor를 첨가하여 배양한 후 심근세포 특이 유전자의 발현을 조사하였다. 결 과: HAD를 5-azacytidine 처리하고 기본배양액에서 4주 동안 배양하였을 때 TnT 유전자가 새로이 발현하였으며 Cmlc1과 kv4.3의 발현 양이 증가하였다. 5-azacytidine 처리 후에 BMP-4와 함께 FGF-4 (B4/F4) 또는 FGF-8 (B4/F8)을 첨가하여 배양하였을 때는 $\beta$-MHC 유전자 발현이 새로이 유도되었으며, Cmlc1, TnT, TnI 그리고 Kv4.3 유전자 발현 양이 더 많이 증가하였다. HUC은 5-azacytidine 및 BMP와 FGF 처리에 의해 유전자 발현 변화가 없었다. 그러나 BMP와 FGF와 함께 activin A 또는 TGF-$\beta$1을 첨가하여 배양하였을 때, BMP-2와 FGF-8 (B2/F8)을 첨가하여 배양한 세포에서 $\beta$-MHC 발현이 새로이 유도되었으며 $\alpha$-CA, TnT 그리고 Kv4.3 유전자의 발현이 증가하였다. 또한 BMP와 FGF와 함께 Wnt inhibitor를 처리하여 1주 동안 배양하였을 때 Cinlc1 유전자 발현이 새로이 유도되었으며 $\alpah$-CA, TnT, TnI 그리고 Kv4.3의 발현이 증가되었다. 결 론: HAD는 BMP와 FGF 처리에 의해 심근세포 특이 유전자의 발현증가를 유도할 수 있었으며 HUC는 BMP와 FGF와 함께 activin A 또는 TGF-$\beta$1 또는 Wnt inhibitor를 처리함으로써 심근세포 특이 유전자의 발현증가를 유도할 수 있었다. 따라서 HAD와 HUC는 심장질환 치료를 목적으로 하는 세포 치료에 이용될 수 있을 것으로 사료된다.
Bone marrow mesenchymal stem cells (MSCs) have shown potential for cardiac repair following myocardial injury, but this approach is limited by their poor viability after transplantation. To reduce cell loss after transplantation, we introduced the fibroblast growth factor-2 (FGF-2) gene ex vivo before transplantation. The isolated MSCs produced colonies with a fibroblast-like morphology in 2 weeks; over 95% expressed CD71, and 28% expressed the cardiomyocyte-specific transcription factor, Nkx2.5, as well as ${\alpha}$-skeletal actin, Nkx2.5, and GATA4. In hypoxic culture, the FGF-2-transfected MSCs (FGF-2-MSCs) secreted increased levels of FGF-2 and displayed a threefold increase in viability, as well as increased expression of the anti-apoptotic gene, Bcl2, and reduced DNA laddering. They had functional adrenergic receptors, like cardiomyocytes, and exposure to norepinephrine led to phosphorylation of ERK1/2. Viable cells persisted 4 weeks after implantation of $5.0{\times}10^5$ FGF-2-MSCs into infarcted myocardia. Expression of cardiac troponin T (CTn T) and a voltage-gated $Ca^{2+}$ channel (CaV2.1) increased, and new blood vessels formed. These data suggest that genetic modification of MSCs before transplantation could be useful for treating myocardial infarction and end-stage cardiac failure.
The Drosophila lymph gland is the hematopoietic organ in which stem-like progenitors proliferate and give rise to myeloid-type blood cells. Mechanisms involved in Drosophila hematopoiesis are well established and known to be conserved in the vertebrate system. Recent studies in Drosophila lymph gland have provided novel insights into how external and internal stresses integrate into blood progenitor maintenance mechanisms and the control of blood cell fate decision. In this review, I will introduce a developmental overview of the Drosophila hematopoietic system, and recent understandings of how the system uses developmental signals not only for hematopoiesis but also as sensors for stress and environmental changes to elicit necessary blood responses. Most of biomaterials come in direct contact with the body, making standardized methods of evaluation and validation of biocompatibility an important aspect to biomaterial development. However, biomaterial validation guidelines have not been fully established, until now. This study was to compare the in vitro behavior of osteoblasts cultured on nanomaterial $TiO_2$ surfaces to osteoblast behavior on culture plates. Comparisons were also made to cells grown in conditioned media (CM) that creates an environment similar to the in vivo environment. Comparisons were made between the different growth conditions for osteoblast adhesion, proliferation, differentiation, and functionality. We found that the in vivo-like system of growing cells in concentrated CM provided a good validation method for biomaterial development and in vivo implant therapy. The $TiO_2$ materials were biocompatible, showing similar behavior to that observed in vivo. This study provided valuable information that would aid in the creation of guidelines into standardization and evaluation of biocompatibility in $TiO_2$ biomaterials.
