Antioxidative and Antiaging Effects of Jeju Native Plant Extracts(II) (제주 자생 식물 추출물의 항산화 및 항노화에 관한 연구(II))

  • In the previous study, we reported the antioxidative and cellular protective effects of Jeju native plant extracts. In this study, we investigated the anti-oxidative, anti-wrinkle and whitening effects of new 37 plant extracts collected from self-growing plants in Jeju island. Their anti-oxidant activities were measured by the 1,1-diphenyl-2-picrylhydrazyl(DPPH) radical scavenging assay and reactive oxygen species(ROS) scavenging assay in $Fe^{3+}-EDTA/H_2O_2$ system. The cytoprotective properties of 37 plant extracts were assessed in the rose-bengal sensitized photohemolysis of human erythrocytes. The inhibitory effect of 37 plant extracts on tyrosinase were investigated to assess their whitening efficacy. Finally, their anti-elastase activities were measured to predict the anti-wrinkle efficacy in the human skin. The results showed that the extracts of Myrica rubra stem bark and Securinega suffruticosa have the free radical scavenging activity($FSC_{50}:\;5,\;8{\mu}g/mL$, respectively), and the extracts of Quercus acutissima leaf and Securinega suffruticosa stem bark have the prominent ROS scavenging activity($OSC_{50}:\;0.009{\mu}g/mL$). Photohemolysis of erythrocytes in the presence of rose-bengal as a sensitizer was inhibited by the extracts of Securinega suffruticosa stem bark and Salix koreensis stem(${\tau}_{50}$, 895 min, 640 min at 50 ${\mu}g/mL$, respectively. Myrica rubra stem bark extract(77.8% at 200 ${\mu}g/mL$) and Salix koreensis stem extract(76.2% at 200 ${\mu}g/mL$) also have the inhibitory effect on tyrosinase and elastase activities, respectively. These results indicated that the stem park of Myrica rubra, Securinega suffruticosa, and Camellia japonica, the stem of Salix koreensis, and the leaf of Quercus aqutissima and Camellia japonica could have e benefitial effects when they are added as ingredients in cosmetics.

Antibacterial and Antibiofilm Activities of Alnus japonica Stem Extract against Porphyromonas gingivalis (Porphyromonas gingivalis에 대한 오리나무 줄기 추출물의 항균활성 및 생물막 형성 억제 효과)

  • This study investigated the potential of dye plants as natural oral health products. The antibacterial activity of ethanol stem extracts of A. japonica, R. verniciflua Stokes, G. jasminoides, D. morbifera, P. amurense Rupr., and S. japonica against P. gingivalis KCTC 5352, S. mutans KCTC3065, S. downei KCTC3634, S. sanguinis KCTC3284, and S. gordonii KCTC 3286 was confirmed. Among the stem extracts from 6 dye plants grown in Korea, ethanol extract from A. japonica stem (1 mg/disc) showed the highest antibacterial activity against P. gingivalis KCTC5352. The A. japonica stem extracts showed antibacterial activity similar to chlorhexidine, which was used as a positive control. The MIC and MBC of P. gingivalis KCTC5352 were 0.4 mg/ml and 0.6 mg/ml, respectively. The biofilm production rate and cell growth of P. gingivalis KCTC5352 in the cultures treated with 0.2-2.0 mg/ml of A. japonica extract were significantly decreased in a concentration-dependent manner. In addition, the mRNA expression of the superoxide dismutase and fimA associated with fimbriae formation in these cultures was suppressed, also in a concentration-dependent manner. Based on these results, it is concluded that A. japonica stem extracts can be used as an oral health product derived from natural materials, as demonstrated by its antibacterial action against and inhibition of biofilm formation of P. gingivalis KCTC5352.

Changes in Ginsenosides Composition of Ginseng Leaf and Stem after an Ultrasonication Process (초음파 처리에 의한 인삼 잎과 줄기의 진세노사이드 성분 변화)

  • The purpose of this study is to develop a new preparation process of ginseng leaf and stem extracts having high concentrations of ginsenoside Rg2, Rg3, Rg5, Rh1, a special component of red and black ginseng. Chemical transformation from ginseng saponin glycosides to prosapogenin was analyzed by the HPLC. Extracts of ginseng (Panax ginseng) leaf and stem were processed under several treatment conditions including ultrasonication treatments. The content of total saponin reached their heights at 17 hr (UGL-17) of ultrasonication treatment, followed by 16 hr (UGL-16) and 7 hr (UGL-7) of ultrasonication treatment at $100^{\circ}C$. UGL-17 findings show that the ginseng leaf and stem that had been processed with ultrasonication for 17 hours peaked in the level of Rg2, Rg3 and Rh1. In addition, UGL-16 contained ginsenoside Rg5 at high concentrations. It is thought that such results provide basic information in preparing ginseng leaf and stem extracts with functionality enhanced.

Antishigellosis and Cytotoxic Potency of Crude Extracts and Isolated Constituents from Duranta repens

  • The crude ethanol extracts (stem and fruits), their fractions and two triterpenes, $\beta$-Amyrin and 12-Oleanene 3$\beta$, 21$\beta$-diol, isolated as a mixture from the chloroform soluble fraction of an ethanolic extract of Duranta repens stem, were evaluated for antibacterial, antifungal activities by the disc diffusion method and cytotoxicity by brine shrimp lethality bioassay. The structures of the two compounds were confirmed by IR, $^1H$-NMR, $^{13}C$-NMR and LC-MS spectral data. The chloroform soluble fraction of stem and ethanol extract of fruits possess potent antishigellosis activity and also exhibited moderate activity against some pathogenic bacteria and fungi but the isolated compound 1 (mixture of $\beta$-Amyrin and 12-Oleanene 3$\beta$, 21$\beta$-diol) showed mild to moderate inhibitory activity to microbial growth. The minimum inhibitory concentrations (MICs) of the extracts (stem and fruits), their fractions and compound 1 were found to be in the range of 32$\sim$128 ${\mu}g/ml$. The chloroform soluble fractions of stem and ethanol extract of fruit showed significant cytotoxicity with $LC_{50}$ value of 0.94 ${\mu}g/ml$ and 0.49 ${\mu}g/ml$, respectively against brine shrimp larvae.

  • Prickly pear(Opuntia ficus-indica var saboten Makino, Cactaceae) is widely used as folk medicine for burned wound, edema and indigestion. To confirm the hypolipidemic action of the extract from Cactus stem and fruit in dietary hyperlipidemic rats, variations of the lipid- or cholesterol-related components were investigated. As results these changes were significantly prevented by treatments with Cactus extracts. The effects of the extract of fruit was better than those of stem.

Antioxidant Activities of Different Parts of Synurus deltoids Nakai Extracts in Vitro

  • The antioxidant activity of hot water extracts of various parts, the leaf, stem, and root of Synurus deltoides was evaluated by various antioxidant assays, including total phenolic content, 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging, hydroxyl radical (${\cdot}OH$) scavenging, superoxide dismutase (SOD), and xanthine oxidase (XOI) activities. The various antioxidant activities were compared with the standard antioxidants such as L-ascorbic acid, $\alpha$-tocopherol, and butylated hydroxyanisole (BHA). Among the different plant parts, stem has been found to possess the highest activity in all tested model systems, the activity decreased in the order stems>roots>leaves. These results indicate that stem extract could be used as potential source of natural antioxidant.

  • The effect of alcohol and aqueous extracts of the leaves and stem of Pluchea lanceolata on the spontaneous movements of both the whole worm and the nerve muscle preparation of Setaria cervi, and on the survival of microfilariae in vitro was studied. Alcohol and aqueous extracts of the leaves and stem of P. lanceolata caused the inhibition of spontaneous movements of the whole worm and the nerve muscle preparation of S. cervi, characterized by short lasting small increase in tone and amplitude of contractions followed by paralysis. The concentrations required to inhibit the movements of whole worm and nerve muscle preparations for alcohol extract were 200 and $25\;{\mu}g/ml$, and for aqueous extract were 250 and $100\;{\mu}g/ml$, respectively, suggesting a cuticular permeability barrier. Both the extracts (alcohol and aqueous) caused death of microfilariae in vitro, $LC_{50}$ and $LC_{90}$ being 12 and 18 ng/ml for alcohol extract and 25 and 40 ng/ml for aqueous extract, respectively.

  • Ulmus davidiana var. japonica is a deciduous tree used in traditional medicine. In this study, we examined the antioxidant and anti-inflammatory activity of extracts and compounds isolated from U. davidiana var. japonica stem barks for development of cosmetic phyto-materials. Phytochemical examination of the stem bark led to the isolation and characterization of three known compounds, (+)-catechin (1), (+)-catechin-7-O-${\beta}$-D-apiofuranoside (2), and procyanidin B3 ((+)-catechin-($4{\alpha}{\to}8$)-(+)-catechin) (3). Their bioactivities including antioxidant (DPPH ree radical scavenging assay) activity, anti-inflammatory (nitric oxide nhibition assay) were evaluated. Most of the crude extracts and isolates indicated significant antioxidant potential compared with BHT and ${\alpha}$-tocopherol as controls. Futhermore, all compounds showed higher inhibitory activities for NO production in Raw 264.7 cells than the L-NMMA using the positive control. Eespecially, (+)-catechin (1) and (+)-catechin-7-O-${\beta}$-D-apiofuranoside (2) which could inhibit more than 90% of the NO production at a concentration of 100 ${\mu}g/m{\ell}$, implying excellent anti-inflammatory activity.

Isolation of main component and antioxidant activities on the stem and root of Rosa rugosa (해당화 줄기 및 뿌리의 주성분 분리 및 항산화활성)

  • Total phenol content was obtained from the stem extract of 7.8 (g/100g, D.W.) and root extract of 10.5 (g/100g, D.W.) on Rosa rugosa. S-4 and R-2 compounds were isolated from the stem and root of R. rugosa. The structure of S-4 and R-2 compounds was assigned as catechin by $^{1}H,\;^{13}C$ CNMR signal and TOF-MS. The tannin content of stem extract was 752.5 (mg/100g, D.W.) as 6.3 of gallic acid, 61.5 of epicatechin and 684.8 of catechin. The tannin content of root extract was higher about 2 times (1676.0) as 6.8 of gallic acid, 160.3 of epicatechin and 1508.5 of catechin than that of stem. It was shown that catechin was main compound in the stem and root extracts of R. rugosa. Antioxidant activities were stronger low concentration, and were not different of stem and root extracts. As the results, stem and root of R. rugosa were evaluated, as natural antioxidant resource.

Functional Properties of Water Extracts from Different Parts of Acanthopanax sessiliflorus (오가피 부위별 열수 추출액의 기능적 특성)

  • Acanthopanax sessiliflorus (A. sessiliflorus) has been known as a traditional medicine having anti-stress, antioxidative and platelet aggregation inhibitory effects. This study was undertaken to investigate the functional properties of water extracts from four parts of A. sessiliflorus. Root, stem, leaf and fruit extracts from A. sessiliflorus were prepared with hot water ($80^{\circ}C$). The contents of functional substances, eleutheroside B and E, polyphenol, antioxidative activity, nitrite scavenging ability and anti-cancer activity of the extracts were determined. The contents of eleutheroside E in stem, root and fruit extracts were 542.50 ${\mu}$g/g, 343.35 ${\mu}$g/g and 30.78 ${\mu}$g/g, respectively. A large part of eleutheroside B was found in fruit (372.01 ${\mu}$g/g) and root (289.33 ${\mu}$g/g) extracts. Root and stem extracts contained 227.21 mg/100g and 131.22 mg/100g of polyphenols, respectively. Antioxidative activities (electron donating ability) of stem and root extracts were 79.87% and 77.27%, respectively. It appears that the antioxidative activities were related to polyphenol contents of the extracts. Most extracts showed 76-81.5% of nitrite scavenging ability at pH 1.2. It reveals that water extract from parts of A. sessiliflorus can inhibit formation of nitrosoamine in food. Effects of the extracts on the growth of normal and cancer cell lines were investigated. Extracts showed no cytotoxicity to normal dendritic cell line (DC2.4). Especially, the root extract promoted the growth of normal cell line. Root and stem extracts had 20-23% of inhibitory effect against stomach cancer cell line (SNU-719) and liver cancer cell line (Hep3B). These result indicated that the extracts from A. sessiliflorus can be used as functional food materials with antioxidative activity and nitrite scavenging ability to eliminate nitrosoamine in food.
