In Vivo $^1H$ MR Spectroscopic Study on Levodopa-Treated Parkison's Disease

  • Authors evaluated alterations of observable metabolite ratios between the cerebral lesion and the contralateral region related to the clinical symptomatic side in levodopa-treated Parkinson's disease (PD) and investigated correlation between age in patients with PD and metabolite ratios of the lesion. Patients with levodopa-treated PD (n = 54) and age-matched normal controls (n = 15) underwent magnetic resonance spectroscopy (MRS) examinations using a stimulated echo acquisition mode (STEAM) pulse sequence that provided 2$\times$2$\times$2 cm3 volume of interest in the selected regions of substantia nigra (SN) and putamed-globus pallidus (PG). To evaluate dependence of metabolite ratios on age, we divided into two groups (i.e., younger and older age). We quantitatively measured N-acetylaspartate (NAA), creatine (Cr), choline-containing compounds (Cho), inositols (Ins), and the sum of glutamate (Glx) and GABA levels and obtained proton metabolite ratios relative to Cr using a Marquart algorithm. Compared with the contralateral region, a significant neuronal laterality of the NAA/Cr ratio in the lesion of SN related to the clinical symptomatic side was established (P = 0.01), but was not established in the lesion of PG (P = 0.24). Also, Cho/Cr ratio tended toward significance in the lesion of SN (P = 0.07) and was statistically significant in the lesion of PG(P = 0.01). Compared with that in the younger age group, NAA/Cr ratio in the older age was decreased in the lesion of SN (P = 0.02), while NAA/Cr ratio was not statistically significant in the lesion of PG ( P = 0.21). Significant metabolic alterations of NAA/Cr and Cho/Cr ratios might be closely related with functional changes of neuropathological process in SN and PG of levodopa treated PD and could be a valuable finding for evaluation of the PD. A trend of NAA/Cr reduction, being statistically significant in older patients, could be indicative of more pronounced neuronal damage in the SN of the progressive PD.

Nutritional Evaluation of Rice with Different Processing Treatments on in vitro Rumen Fermentation Characteristics and in situ Degradation (재고미의 가공처리에 따른 in vitro, in situ 소화율 및 발효성상 평가)

  • This study was conducted to evaluate the effect of different processing of rice on rumen fermentation in in vitro and in situ experiments. Different processing treatments (extruding, roasting, and steaming) were used in this study and all treatments were ground through a cyclone mill (Foss, Hillerød, Denmark) fitted with a 1 mm screen. Non-treated rice was considered to a control substrate. Then, all treatments were used in in vitro and in situ experiments. Total gas production and dry matter digestibility in control were lower than any other treatment at all incubation times (P<0.01). The lowest ammonia nitrogen ($NH_3-N$) concentration was observed in control among treatments at 6, 12, and 24 h incubation (P<0.01). Extruding had a highest total volatile fatty acids (VFA) concentration at 6, 12 h incubation (P<0.01) and Steaming exhibited a highest total VFA at 24 h (P<0.01). The lowest total VFA concentration was observed in control at 6, 12, and 24 h (P<0.01). In an in situ, The highest value of soluble fraction, degradation rates, effective degradability was observed in extruding (P<0.01). It was considered that feed processing increased dry matter digestibility, total VFA concentration, and decreased pH as well as $NH_3-N$ concentration indicating that processing may increase nutrient degradation of rice in the rumen.

Changes in Hydrogen Donating Activities of the Extract from Hololeion maximowiczii Root by Drying Methods (건조방법에 따른 께묵뿌리 추출물의 수소공여능 조사)

  • Hydrogen donating activities(HDA) of solvent extracts of Hololeion maximowiczii roots which were treated with freeze drying, hot air drying and hot air drying after steam blanching were investigated by the method of ${\alpha},{\alpha}'$-diphenyl-${\beta}$-picrylhydrazyl(DPPH). Hydrogen donating activity of hot air drying after steam blanching group showed the highest activity among three drying methods. Among the extracts of freeze dried root extracted with various solvents, color intensity at 420 nm of aqueous extract was highest, but hydrogen donating activity was shown in order of 50%, ethyl alcohol, methyl alcohol, water extracts. By heating the powder of freeze dried root at $100^{\circ}C$, color intensity of its water extract was decreased until 20 min, thereafter it was increased slowly but hydrogen donating activity was rapidly increased until 30 min. Hydrogen donating activity at the concentration of 6.05 mg/ml of 50% alcohol extract was similar to that of ${\alpha}-tocopherol$ at the concentration of $2{\times}10^{-4}M$. Comparison of hydrogen donating activity of the inner and outer solution after dialysis of water extract showed that the outer solution had a stronger hydrogen donating activity than those of the inner one.

Quality Characteristics of Alcohol fermentation Broth and By-Product of Brown Rice Varieties (현미 품종별 알코올발효액 및 부산물의 품질특성)

  • This study was investigated quality characteristics of alcohol fermentation broths and by-produce produced from 4 type of brown rice varieties. The result showed that Daeanbyeo(A), Baekjinju(B) and Sulkyeong(C) produced the high alcohol content(9.5, 9.7 and 9.8%) by non-steamed alcohol fermentation period whereas the Goami(D) gave poor content(3.5%). In addition, the effect of total free sugar content were examined for non-steamed alcohol fermentation period. Among these 4 type, 244.2 mg% showed the highest contents in the Goami(D). The steamed alcohol fermentation showed higher sugar content in overall with slight difference between the various brown rice. The total dietary fiber content variation according to the alcohol fermentation methods in non-steamed and steamed periods came up with the highest content in the Sulkyeong(C) fellowed by Baekjinju(B), Daeanbyeo(A), and Goami(D). The total free amino acid content by alcohol fermentation methods timed out to show the highest content in non-steamed Goami by-product followed by Goami powder steamed Goami by-product Although, the above result revealed the lowest alcohol fermentation efficiency in non-steamed Goami, but it had the highest by-product utilizing capacity. Therefore, Goami(D) was the lowest alcohol yield in alcohol fermentation treated with non-steam among groups, but Goami by-products of the Goami had the various nutritional constitutes including starch, dietary fibers and free amino acids, which can be suggested to be used as various functional food ingredient.

Effects of Various Pretreatment Methods on Physicochemical and Nutritional Properties of Carrot (다양한 전처리 방법에 따른 당근의 이화학 및 영양학적 특성 분석)

  • Pre-thermal treatment is important to minimize quality changes during main cooking or storage. In this study, to optimize pre-thermal treatment of carrots, three types of pre-thermal treatments were applied to carrots and quality changes such as physicochemical, nutritional, or sensory properties were observed. Washed and sliced carrots were thermally treated by hot-water immersion ($100^{\circ}C$, 1~10 min), steaming ($100^{\circ}C$, 1~10 min), and stir-frying with oil (10~80 sec). Carrot tissue was maintained until 2 min hot-water immersion or steaming, and they were damaged by just 30 sec of stir-frying. Color and hardness were significantly affected by treatment time and temperature. Color was completely changed after 5 min and 7 min by hot-water and steam treatments, respectively. Hardness decreased to 44% compared with fresh carrot (4,500 g) after 1 min, 3 min, and 20 sec of hot-water, steam, and stir-frying, respectively. For nutritional changes, ascorbic acid, organic acid, and peroxide activity were reduced by all treatments compared with fresh carrot. Especially, succinic acid was dramatically reduced by hot-water treatment. Otherwise, free sugar contents were increased with greater treatment time in all samples. In this study, pre-thermal treatment of carrot was optimal at 2 min steaming treatment.

Utilization of Steam-treated Oil Palm Fronds in Growing Saanen Goats: II. Supplementation with Energy and Urea

  • The objective of this study was to evaluate the effect of protein and energy on goats fed oil palm fronds (OPF) as roughages. Twenty-four male Saanen goats aged between 7 and 8 months and weighing $23.4{\pm}1.6kg$ were used in a $2{\times}3$ factorial design. Factors were three levels of urea (3%, 4% or 5%) and two levels of energy (low energy (LE) or high energy (HE)). On average, all parameters measured, including dry matter intake (DMI), nutrient digestibility, digestible nutrient intakes, ruminal ammonia-N ($NH_3$-N), ruminal total volatile fatty acid (total VFA) and individual VFA concentrations (mM/L), microbial N supply, P/E ratio and N retention were higher for HE compared to LE diets. Significant (p<0.05) interactions were found between levels of urea and energy for nonstructural carbohydrate (NSC) and energy (DE) digestibilities, ruminal $NH_3$-N and total VFA concentrations. HE diets had higher N absorption and retention than LE diets. Interactions between urea and energy for plasma urea nitrogen (PUN), heat production (HP), and urine and faeces N excretion were significantly lower (p<0.05) for the HE diets than those recorded for the LE diets. The results indicated that supplementation of energy enhanced utilization of urea and resulted in higher animal performance as a consequence of improved ruminal fermentation, microbial yield and N balance. However, the optimal level of urea supplementation remained at 3% in the HE diet.

Elephant Garlic Extracts Inhibit Adipogenesis in 3T3-L1 Adipocytes (코끼리마늘의 3T3-L1 지방세포 분화억제 효과)

  • Elephant garlic (Allium ampeloprasum L.) has been reported to have several pharmacological effects. However, its anti-adipogenic effect and the possible molecular mechanisms have not yet been reported. In this study, we demonstrate that elephant garlic extracts suppress adipogenesis in 3T3-L1 adipocytes. Raw and steamed elephant garlic extracts (REG and SEG, respectively) suppressed the differentiation of adipocytes and cellular lipid accumulation. Of note, the anti-differentiation effect of REG treatment on 3T3-L1 cells resulted in cytotoxicity, whereas SEG-treated cells displayed no such cytotoxicity. Additionally, SEG treatment significantly reduced the adipogenesis-related gene expression of PPAR γ, C/EBPα, adiponectin, Ap2, and LPL. To our knowledge, these results are the first evidence of the anti-adipogenic effects of elephant garlic extracts on 3T3-L1 adipocytes.

Studies on the Artificial Substrates with Rice Straw and the Spawning for Pleurotus florida in Korea (볏짚을 이용(利用)한 사철느타리버섯의 배지(培地) 제조(製造) 및 종균재식(種菌栽植)에 관한 연구(硏究))

  • This study was performed to examine the cultivation of Pleurotus florida that forms pinhead between 5 and $25^{\circ}C$, on rice straw substrates in Korea. The highest yield of P. florida sporophores was obtained from the fermented rice straw substrates added 5% rice bran to the rice straw by dry weight basis at the starting of fermentation. The pile turning of materials for uniform fermenting was made three times when the temperature of the pile reached $60^{\circ}C$ during out door composting. The $60^{\circ}C$ row steam was treated 6 hrs for pasterization of the substrates in the cultivating house. The quantity of spawn affected the mycelial growth and the yield of P. florida. The optimum amount of spawn was 10% to the rice straw by dry weight basis mixed with the substrates. P. florida produced normal sphorophores from September to middle of July of the following year.

Surface Characteristics of Anodized and Hydrothermally-Treated Ti-6Al-7Nb Alloy (양극산화와 열수처리한 Ti-6Al-7Nb 합금의 표면 특성)

  • This study was performed to investigate the surface properties and in vitro biocompatibility of electrochemically oxidized Ti-6Al-7Nb alloy by anodic spark discharge technique. Discs of Ti-6Al-7Nb alloy of 20 mm in diameter and 2 mm in thickness were polished sequentially from #300 to 1000 SiC paper, ultrasonically washed with acetone and distilled water for 5 min, and dried in an oven at $50^{\circ}C$ for 24 hours. Anodizing was performed using a regulated DC power supply. The applied voltages were given at 240, 280, 320, and 360 V and current density of $30mA/cm^2$. Hydrothermal treatment was conducted by high pressure steam at $300^{\circ}C$ for 2 hours using a autoclave. Samples were soaked in the Hanks' solution with pH 7.4 at $36.5^{\circ}C$ during 30 days. The results obtained were summarized as follows; 1. The oxide films were porous with pore size of $1{\sim}5{\mu}m$. The size of micropores increased with increasing the spark forming voltage. 2. The main crystal structure of the anodic oxide film was anatase type as analyzed with thin-film X-ray diffractometery. 3. Needle-like hydroxyapatite (HA) crystals were observed on anodic oxide films after hydrothermal treatment at $300^{\circ}C$ for 2 hours. The precipitation of HA crystals was accelerated with increasing the spark forming voltage. 4. The precipitation of the fine asperity-like HA crystals were observed after being immersed in Hanks' solution at $37^{\circ}C$. The precipitation of HA crystals was accelerated with increasing the spark forming voltage and the time of immersion in Hanks' solution. 5. The Ca/P ratio of the precipitated HA layer was equivalent to that of HA crystal as increasing the spark forming voltage and the time of immersion in Hanks' solution.

Dyeing Properties of Acetate/Functional Polyester Composite Yarn in Different Yarn Twisting Processes (연사방법에 따른 아세테이트/기능성 폴리에스터 복합사의 염색성에 관한 연구)

  • The purpose of this study was to examine the dyeing properties of four kinds of composite yams that were twisted in different processes. The composite yarns consist of acetate and functional polyester in ratio of 70 : 30. The composite yams were dyed at 100$^{circ}C$ and 125$^{circ}C$ using three types of dyes, disperse dyes for acetate fiber, PET fiber and PET-acetate blended fiber, in the three primary colors. The exhaustion($\%$) and K/S value were observed for each case. Also the effects of four different twisting processes to dyeing properties and physical properties were examined. Regardless of twisting methods, the composite yarns that were dyed at 125$^{circ}C$ had higher exhaustion($\%$) than those were dyed at 100$^{circ}C$ with all three types of dyes; However, tendency of the K/S value after reduction cleaning process was measured at 100$^{circ}C$ and the value measured 125$^{circ}C$ had a great difference with disperse dyes for acetate and dyes for PET. The difference of the K/S values of composite yarns, when dyed at 100$^{circ}C$ and 125$^{circ}C$ with disperse dyes for PET-acetate blended fiber, was almost negligible. According to twisting methods K/S values were in the following order: AP1 > AP3 > AP4> AP2. This means that AP1, treated at 220$^{circ}C$, had the highest K/S value and K/S value becomes higher as the yam is higher twisted and becomes lower as lower twisted. On the other hand, the dry shrinkage and wet shrinkage showed low shrinkage rate when the twist was high and steam setting temperature was high.
