Physicochemical Properties of Starch Granules from Endosperm Mutants in Rice (배유돌연변이체 쌀전분의 이화학적 특성 비교)

  • Starches from the eight varieties of rice were analyzed using scanning electron microscope(SEM), differential scanning calorimetry(DSC) and X-ray diffractometry, and tested on the starch-granule susceptibility to 15% $H_2SO_4$ and glucoamylase. The shape of starch granules from normal rice varieties and low-amylose mutants were polygonal while shrunken and floury mutants were globular. According to DSC, starches from Nampung CB243 showed higher onset temperature$(T_o)$, completion temperature$(T_c)$ and Punchilmi, Nampung EM90 showed higher enthalpy$({\triangle}H)$ of gelatinization than others. shr showed the highest hydrolysis rate to 15% $H_2SO_4$ while Nampung CB243 showed the lowest one. Eight varieties of rice starch granules showed A-type pattern on X-ray diffractograms. The lower amylose content rice varieties showed the higher hydrolysis rate treated with glucoamylase.

  • Exo-maltotetraohydrolase produced by Pseudomonas stutzeri IAM 12097 was characterized with respect to substrate specificity, the reaction products and hydolysis rate on various carbohydrates. Maltopentaose, maltoheptaose, soluble starch, amylose, amylopectin, oyster glycogen and gelatinized starch of corn, potato, glutinous rice, green banana and arrow root were hydolyzed by this enzyme, but ${\alpha},{\beta},{\gamma}-cyclodextin$, sucrose, raffinose, lactose, pullulan, maltose, maltotriose and maltotetraose were not hydrolyzed. Among oligosaccharides, maltohexaose was favorably hydrolyzed by this enzyme and the main reaction product of oligosaccharides and polysaccharides was maltotetraose. Addition of pullulanase to this enzyme increased the hydolysis rate on gelatinized starches. tut it did not on raw starches. Among various starches, corn starch was favorably hydrolyzed by this enzyme, whereas it acted on potato starch, arrow root starch and high amylose corn starch weakly.

  • The in-vitro starch digestibility of white and brown rice flours was continuously characterized from a rheological point of view. Specifically, the in-vitro viscosities of the rice digesta samples were monitored under simulated oral, gastric, and intestinal conditions. A trend of decreasing viscosities in all the digesta samples was observed during the in-vitro digestion. After cooking, the brown rice sample exhibited lower viscosity than that of the white rice flour due to the presence of more non-starch components. A similar tendency was observed during the simulated oral and gastric digestions. However, the viscosity crossover between the white and brown rice samples was observed during intestinal digestion. In addition, the amount of glucose released from the brown rice flour was significantly lower than that from the white rice flour. Thus, the slower rate of starch hydrolysis in the brown rice flour could be related to its in-vitro rheological behaviors.

  • Characteristics on lintnerization of dry type (Suwon 147) and moist type (Hwangmi) sweet potato starches were investigated. Chain distribution of lintnerized starches was also studied by debranching with pullulanase. Hydrolytic patterns of two starches showed two distinct stages and hydrolysis extents of Suwon 147 starch were lower than those of Hwangmi starch. The relative crystallinities of Suwon 147 starches were higher than those of Hwangmi starch. The elution profiles of lintnerized starches were composed of two peaks about degree of polymerization (DP) 25 and DP 15. The elution profiles of debranched samples showed only one peak about DP 15 and peak DP of Suwon 147 lintnerized starch was higher than that of Hwangmi.

  • The aim of this study was to produce resistant starch preparations from acid-modified com starches prepared at various hydrolysis levels (0.5-4.0 hr). Effect of autoclaving cycles on resistant starch (RS) formation was investigated. Molecular weight distribution, pasting and functional properties of acid-modified com starches were determined. For RS formation native and acid-modified starch samples were gelatinized and autoclaved (1 or 2 cycles). While native and acid-modified starches did not contain any RS, the levels increased to 9.0-13.5% as a result of storage at $95^{\circ}C$ after first autoclaving cycle. Second autoclaving cycle together with storage at $95^{\circ}C$ brought final RS contents of the samples incubated at 4 and $95^{\circ}C$ after the first cycle to comparable level. As acid modification level increased, the amount of high molecular weight fractions decreased, resulting in significant decreases in viscosities (p<0.05). The samples produced in this study had low emulsion stability and capacity values.

  • Lipomyces starkeyi KSM 22 elaborates an enzyme that has both dextranase and amylase activities in a single protein of 100 kDa. Competition studies, using different amounts of dextran and starch as substrates, gave a competition plot consistent with the hypothesis that the hydrolysis of dextran and starch occurs at two independent active sites, each specific for starch and dextran, respectively.

  • The properties of mung bean were investigated depending on hydrolysis condition. The results showed that enzyme treatments (${\alpha}$-amylase and protease each at 0.1% (w/w)) by varying hydrolysis temperature showed better properties than non-pretense treatment (control group). The treatment with 0.08% ${\alpha}$-amylase was best for optimum hydrolysis of mung bean starch The treatment using a mixture of 0.08% (w/w) ${\alpha}$-amylase and 0.12% (w/w) protease was best for optimum hydrolysis of meg bean protein. The effects of Hydrolysis time of mung bean showed that the optimum time was 60 and 90 min and there fore the optimum time was set at 60 min. These result showed that the best hydrolysis conditions of mung bean were the treatment at $60^{\circ}C$ for 60 min using a mixture of 0.08%(w/w) ${\alpha}$-amylase and 0.12% (w/w) pretense, with the sugar level shown at $5.8^{\circ}Brix$, reducing sugar at 2,022.13 mg% and crude protein at 7,666.17 mg%.

Measurement for Determining the Biodegradation of Starch-Filled Polyethylene Film by $\alpha$-Amylase (전분 충전 폴리에틸렌 필름의 아밀레이스 반응에 의한 생분해도 측정)

  • Optimal reaction condition for the starch hydrolysis by ${\alpha}$-amylase was determined and the sugar produced under the optimal condition was measured for estimating the biodegradation of strach-filled polyethylene film. Optimal ranges of temperature and pH were 70~$80^{\circ}C$ and 6.3~7.3, respectively. The 100 units of ${\alpha}$-amylase per mg starch were enough for the enzyme reaction. Reaction with polyethylene film containing 5%, 10%, 15% and 20% starch in the above condition showed that the sugar produced was proportional to the starch content in film. This relationship provides a calibration curve for determining the starch content of search-filled polyethylene film. The average amount of hydrolyzed starch was about 40% of total starch in film. The rest of the starch is considered to be still dispersed in the film and not to be attacked by ${\alpha}$-amylase. In this experiment, we could obtain the higher biodegradability through the $\alpha$-amylase reaction in the above optimal condition than the reported one which had been Improved by adding surfactant.

Direct Alcohol Fermentation of Starch by Schwanniomyces castellii (Schwanniomyces castellii에 의한 전분의 직접 알콜발효)

  • Alcohol fermentations were carried out to confirm the capacity of ethanol production from glucose, starch and soluble starch(dextrin) by Schwanniomyces castellii NRRL Y-2477. Schw. castellii NRRL Y-2477 was able to produce the 63.9g/l ethanol using 94% subtrate from 150g/l-glucose medium. The direct alcohol fermentation of starch having the maximum solubility of 20g/1 at $30^{\circ}C$ yielded 9.1g/l ethanol upon complete depletion of starch, whereas 34.5g/1 ethanol was produced by utilizing 82% of 100g/1 soluble starch medium. The fermentation of 150g/1 soluble starch produced 52.1g/l ethanol using about 79% of substrate. Thus, it was found that the limiting step of direct alcohol fermentation of starch by Schwanniomyces castellii NRRL Y-2477 was a hydrolysis of starch.

The Effect of Medium-Cooked Rice on the Production of Korean Traditional Yakju (한국 전통 약주의 제조시 반숙미의 사용효과)

  • This study was carried out to investigate the effect of medium-cooked rice on brewing of yakju which was a traditional rice wine in Korea. The influences of cooking temperature of rice on hydrolysis of rice starch and rice protein were tested, and experimental brewings were done according to the traditional brewing method of Bangmunju in which some medium-cooked rice was used. The results obtained were as follows The hydrolysis of starch and protein in medium-cooked rice at 60~$65^{\circ}C$ was easier than that of full-cooked rice at 80~10$0^{\circ}C$. The amounts of saccharides, total amino acids and extracts In Yakju brewed with the combined use of medium-cooked rice and full-cooked rice were twice as much as those brewed with full-cooked rice only. The results of sensory test of Yakju brewed with the combined use of medium-cooked rice and full-cooked rice were better in taste, color and flavor than those brewed with full-cooked rice only. It was thought that our ancestor's traditional brewing method of Yakju in which medium cooked rice and full-cooked rice were used combinedly was excellent Judging from zymological point of views.
