Physiological studies on the sudden wilting of JAPONICA/INDICA crossed rice varieties in Korea -II. Artificial induction of sudden wilting in Korea and the effect of nutrio-physiological status (일(日)·인원연교잡(印遠緣交雜) 수도품종(水稻品種)의 급성위조증상(急性萎凋症狀) 발생(發生)에 관(關)한 영양생리학적(營養生理學的) 연구(硏究) -II. 한국(韓國)에서 급성위조증(急性萎凋症)의 인위적(人爲的) 재현(再現)과 영양생리(營養生理)의 영향(影響))

  • The results of the investigation on the effects of plant nutrio-physiological status and reemergence of sudden wilting in rice on the occurrence of sudden wilting were summarized as follows: 1. The occurrence of sudden wilting, degree of damage and occurring time in the investigated variety showed to concur with the results of the previous experiment. 2. Nitrogen and Potassium contents in leaves related an occurrence of sudden wilting at heading stage have a very high negative co-relation (the value is -0.9224 at Yushin and -0.8243 at Milyang 23). 3. Regarding the relation between occurrence of sudden wilting and root activity capacity at the late growth stage, the root activity capacity at productive growth stage showed higher than at reproductive growth stage, which is the same in the two varieties but Milyang 23 is higher than Yushin. It is confirmed that root activity capacity in the two varieties has become very low at reproductive growth stage. 4. The toot activity capacity in the field with potassium showed a small occurrence of sudden wilting and the root activity capacity in the field with no potassium showed a very high co-relation value (the value is 0.8947 at Yushin and 0.7301 at Milyang 23). 5. Regarding the relation between the occurrence of sudden wilting and water evaporation quantity, water evaporation quantity after the heading date has a more direct relation to sudden wilting than that which occurrs prior to the heading date. Water evaporation quantity is higher in the field with no potassium, low leaves and Yushin, than in the field with potassium high leaves and Milyang 23. 6. The starch content in culm of Yushin and Milyang 23 at ripening stage showed high value in proportion to potassium quantity, and the starch content is lower as internode is lower. However, in fourth internode, the starch content is greatly increased in the field with potassium contrasted to the field with no potassium.

Physiological Tolerance of Native Tree Species in Abandoned Coal Mine Spoils (탄광 폐석지내 자생 수종의 생리적 피해 및 내성)

  • This study was conducted to assess the physiological tolerance of native tree species for successful restoration and revegetation of abandoned coal-mine spoils. Study sites were two coal-mine spoils (Sododong and Ssarijae) in Taebaek, Kangwon Province, Korea. Five individuals of Betula costata and of B. schmidtii were analyzed for malondialdehyde (MDA) and hydrogen peroxide ($H_2O$$_2$) content, nitrate reductase (NR) and superoxide dismutase (SOD) activity, and for carbohydrate concentration in the leaves. Trees in the abandoned coal-mine spoils were influenced by deficiencies expressed by MDA and $H_2O$$_2$ content in the leaves of two species being higher at the coal-mine spoils than in the surrounding forest. Low NR activity indirectly represented nitrogen deficiency in the soil of the coal-mine spoils; an unmanageable SOD activity implied that tolerant functions didn't net against a certain stress of the coal-mine spoils. Decreased glucose and increased starch concentration especially showed the inhibition of the carbohydrate metabolism by inadequate factors. Consequently, low nitrogen content in the real-mine soils might increase damage in trees as a result of inhibiting the expression of tolerance mechanisms against stress. Therefore, trees in coal-mine spoils need ample nitrogen to use as a metabolic energy source in order to prevent damage and increase tolerance against stress.

Physiological Responses for Soil Water Stresses in 'Mihong' Peach Tree (복숭아 '미홍'의 토양 수분 스트레스에 따른 생리반응)

  • The present study was conducted to characterize physiological responses of aerial part according to soil water stresses in 'Mihong' peach trees. Discolorations, wilting and falling of leaves were observed in chronological order as response for waterlogging and no irrigation and the phenomena started from basal to end of shoots. Shoot growth in elongation and thickness decreased and fallen leaves were severe in waterlogged trees. Function of water uptake by roots and photosynthesis and leaf respiration decreased by waterlogging. Leaf chlorophyll contents decreased in both treatments. In waterlogging treatment, decrease of chlorophyll was observed in normal leaves with waterlogging using light microscopy. Starch content was lower in both treatment and carbohydrate content was lower in root with waterlogging. These results demonstrated that waterlogging weakened the function of soil water uptake and movement and decreased photosynthesis and fallen leaves. Finally the peach trees would wither or suffer low temperature damage through the shortage of reserve accumulations. We suggested that waterlogging damage in peach trees could be reduced to take notice of irrigation and install drainage facility to improve soil condition.

Effects of Elevated CO2 Concentration on Leaf Phenology of Quercus acutissima (이산화탄소 농도 증가가 상수리나무 잎의 계절현상에 미치는 영향)

  • Effects of elevated $CO_2$ on leaf phenology of Quercus acutissima were examined using open-top chambers, which had ambient and elevated $CO_2$ concentrations (ambient ${\times}1.4$, ambient ${\times}1.8$). To analyze the effect of chamber, non-treatment block was established near outside of the chambers. In 2013, budburst, leaf unfolding, coloring, and shedding were surveyed, and spring phenology was surveyed in 2014. Thermal sum (base temperature $+5^{\circ}C$) of each phenological event occurred was recorded. In addition, bud samples were collected and analyzed for carbohydrate contents in March 2014. Elevated $CO_2$ concentration advanced budburst and leaf unfolding, and delayed shedding in 2013. However, in 2014, the temperature of the spring season was high, and there was no significant effect of elevated $CO_2$ concentration on spring phenology. Carbohydrates content, such as starch, total non-structural carbohydrate and total soluble sugar, were significantly increased in response to elevated $CO_2$ concentration. It has been proposed that elevated $CO_2$ concentration could extend the growing season of temperate species with increased possibility of frost damage due to early bud opening and leaf unfolding. However, our analysis showed that the increased carbohydrate concentration in bud under elevated $CO_2$ would reduce the possibility of early spring frost damage by acting as cryoprotectant.

Ammonia Gas Removal by Bacillus subtilis IB101 and Dctimization of Culture Media (Bacillus subtilis IB101을 이용한 암모니아 가스 제거 및 생산배지 최적화)

  • Ammonia gas is one of the major pollutants which cause environmental pollution and damage to the human and the livestock. The objective of this study was to investigate the important parameters for the development of efficient removal of ammonia gas by Bacillius subtilis IB101 and to optimize the medium composition for the mass production of B. subtilis IB101. The ammonia gas removal efficiency was evaluated at different growth phases and by changing culture conditions (temperature, pH). The effect of $(NH_4)_2SO_4$ concentration in preculture medium was examined. Medium optimization for the mass production of B. subtilis IB101 was performed by using Plackett-Burman design and one factor at a time method. The removal of ammonia gas was more efficient at exponential phase by 20% than at stationary phase. The ammonia gas removal was the highest at pH 4 and 30 $^{\circ}C$. There was not any significant influence of concentration of $(NH_4)_2SO_4$ on the removal of ammonia gas. The components of optimized medium for the production of viable Bacillus subtilis IB101 was yeast extract 10 g/l, soluble starch 2.5 g/l, $MgSO_4$ 6 g/l, $CaCl_2$ 1.55 g/l, $(NH_4)_2SO_4$ 5 g/l, $KH_2PO_4$ 0.75 g/l, soy bean meal 8 g/l.

Effect of Virus Free Stocks of Sweetpotato Cultivated at Different Regions (지역을 달리하여 재배한 고구마 바이러스 무병묘 효과)

  • To analysis of virus free sweetpotato effect, 5 virus free sweetpotato and virus normal sweetpotato varieties were planted in 5 different regions at 2010 year. The average yields of virus free sweetpotato are showed different results according to regions. Sinjami which cultivated at Iksan were increased maximum 68% compare to normal. However, Sinjami which cultivated in Hamyang were decreased yield 11% compare to normal. Analysis of tuber formation ratio of Sinjami, Yenhwangmi, Singeonmi which cultivated in Nonsan were decreased tuber number compare to normal. However, 3 varieties were all increased on Average storage root weight and yield of marketable storage root. In the results of analysis of marketable storage root according to cultivated regions and varieties, all varieties except Sinjami which cultivated in Hamyang were increased yield. Also, quality of virus free sweetpotato were enhanced 7 to 9 compare to virus infected sweet potato which showing average 3. Contents of starch between virus free and virus infected sweetpotato were not affected by virus. Virus free sweetpotato were more increased starch products according to increased total production yield. Also, $Brix^{\circ}$(%) was not showing difference between virus free and virus infected sweetpotatoes. In this experiment, Virus free sweetpotato are enhanced production yields and quality. Therefore, we suggested that virus free sweetpotato is one of the methods to reduce damage by sweetpotato virus.

Physicochemical Studies on the Hard and Soft Wheats Flours (경질(硬質) 및 연질(軟質) 밀가루의 이화학적(理化學的) 성질(性質) 연구(硏究))

  • The physicochemical properties of wheat flours were investigated for hard wheat (Bara and Kameriya varieties), semi-hard wheat (Snisen variety) and soft wheat (Ume variety). There were no sigificant differences in the proximate chemical compositions of the tested wheat fluors; however, the protein contents of them were 12.18 to 8.40 % for the hard wheat flours and 6.81 % for soft wheat flour, and gluten contents were 11.77 to 8.38 % for the hard type flours and 5.53 % for soft flour. The soft wheat flour had higher whiteness, whereas the hard wheat flours showed higher starch damage values and higher flour-water absorption than the soft wheat flour. In farinograph data, the hard wheat flours had better development time, stability and valorimeter value of doughs. There were significant differences in the extensigraph data among the tested flours, i,e, resistance to extention and the area with planimeter of doughs increased with the time and their extensibility decreased. The Bara and Kameriya wheat flours had lower maximum visicosity of amylograph than Suisen and Ume wheat flours.

Corn Growth and Development influenced by Potential CO2 Leakage from Carbon Capture and Storage (CCS) Site (지중저장 이산화탄소의 잠재적 누출 모사에 따른 옥수수 초기 반응 및 생장 연구)

  • Carbon capture and storage (CCS) technology has been suggested as an ultimate strategy for mitigating climate change. However, potential leakage of $CO_2$ from the CCS facilities could lead to serious damage to environment. Plants can be a bio-indicator for $CO_2$ leakage as a cost-effective way, although plants' responses vary with plant species. In this study, a greenhouse experiment was conducted to investigate the relation between the $CO_2$ tolerance of corn species and the initial physiological responses to the elevated soil $CO_2$ concentration. Treatment groups included CI (99.99% $CO_2$ gas injection) and BI (no gas injection). Mean soil $CO_2$ concentration for the CI treatment was 19.5~39.4%, and mean $O_2$ concentration was 6.6~18.4%. The soil gas concentrations in the BI treatment were at the ambient levels. In the CI treatment, chlorophyll content was not decreased until the $13^{th}$ day of the $CO_2$ injection. On the $15^{th}$ day, leaf starch content and stomatal conductance were increased by 89% and 25% in the CI treatment compared to the BI treatment, respectively. This might be due to the compensatory reaction of corn to avoid high soil $CO_2$ stress. However, the prolonged $CO_2$ injection decreased chlorophyll content after 13 days. After $CO_2$ injection, plant biomass was reduced by 25% in the CI treatment compared to the BI treatment. Due to the inhibited root growth, leaf phosphorous and potassium contents were decreased by 54% on average in the CI treatment. This study indicates that corn has a high tolerance to soil $CO_2$ concentration of 30% for 2 weeks by its compensatory reactions such as an maintenance of chlorophyll content and an increase in stomatal conductance.

Studies on the Epidemiology and Control of Bacterial Leaf Blight of Rice in Korea (한국에 있어서의 벼흰빛잎마름병의 발생생태와 방제에 관한 연구)

  • The study has been carried out to investigate the occurrence, damage, characteristics of the pathogen, environmental conditions affecting the disease outbreak, varietal resistance, forecasting, and chemical control of bacterial leaf blight of rice in Korea since 1964. Bacterial leaf blight of rice became a major disease in Korea since 1960. A correlation was found between the annual increase of epidemics and increase of cultivation area of susceptible varieties, Jinheung, Keumnampung etc. Areal damage within the country showed that the more was at southern province, Jeonnam, Gyeongnam and western coast, and at flooded rice paddy. Yield reduction directly related with the amount of infection on upper leaves at heading stage. Fifty per cent of reduction resulted when the lesion area was more than 60 per cent. Less than 20 per cent of lesion area, however, was not affected so much on yield loss One hundred and six isolates collected from all over the country were classified as 8 strains by using 4 different bacteriophages in 1973. It was, however, only two in 1965. There were some specificities on varietal distributions among the strains such as that the Jinheung attacked mainly by strain A, B, C and I, those attack Kimmaze were A, B, H and I. Most strains were found from Tongil except D and E, whereas Akibare was only variety that attacked by strain E. Low temperature, high humidity, heavy rainfall and insutficient daylight favored the disease epidemics. Especially, typhoon and flooding at heading stage were critical factors. The earlier transplanting the more disease was resulted, and more nitrogen fertilizer application accerelated the diseased development in general. The resistance to the disease varied by growing stage of the sane plants. All of recommended varieties in Korea were susceptible to the disease except Norm No. 6 and Sirogane which moderately resistant. The pathogen, Xanthomonas oryzae, was detectable from extract of healthy seedlings that were grown in the field with an heavy infection previous year. The more bacteriophage in irigation water resulted the more disease outbreak, and the existence of more than 50 bacteriophages in 1ml. of irrigation water were necessary to initiate the disease out break. The curves representing occurrence of bacteriophages and disease outbreak were similar with 15 days interval. The survey of bacteriophage occurrence can be utilized in forecasting of the disease two weeks ahead of disease outbreak. Three applications of chemicals, Phenazin and Sangkel, in weekly intervals at the early satage of out-break depressed the symptom development, and increased yield by 20per cent. Proper period for the chemical application was just before the number of bacteriophage reaches 50 in 1ml. of irrigation water.

Studies on the Effects of Copper on the Lactate Dehydrogenase and Esterase Isozymes in Various Tissues of Carassius carassius (붕어(Carassius carassius)의 조직내 젖산수소이탈효소와 에스테라아제 아이소자임에 미치는 동의 영향에 관한 연구)

  • In order to elucidate the effects of copper on Corassius carassius, the following were studied: 1) lactate dehydrogenase isozyme patterns by cellulose acetate electrophoresis, 2) LDH activity and copper effect on LDH enzyme system y spectrophotometry, 3) esterase isozyme patterns by agar thin layer electrophoresis, 4) hemoglobin patterns by starch gel electrophoresis, and 5) histological study. 1. There were two bands of LDH isozymes (LDH-3 and LDH-5) in the gill, three bands (LDH-2, LDH-4, and LDH-5) in the liver, and two bands (LDH-3 and LDH-4) in the muscle of the normal fish. The LDH-1 bond was not found in the above three tissues. When the fish were exposed to copper, LDH-3 appeared in the liver, LDH-5 in the muscle, but no new LDH band appeared in the gill. 2. The sepcific activities of the LDH were lowest in the gill and highest in the muscle of the normal fish, and they were gradually decreassed in the gill and highest in the muscle of the normal fish, and they were gradually decreased in the liver and mucle except in the gill from 1-day to 10-day exposure to copper. It indicates that LDH activities in the liver and muscle of the fish were inhibited by copper. 3. Through in vitro experiment, it is clear that the decrease of the LDH activities of the liver and muscle of the fish exposed to copper is mainly caused by the inhibition on the M-LDH in the fish. 4. The numbers of the esterase isozyme bands of the gill, liver, muscle, blood, brain, and kidney of the normal fish were 3, 6, 2, 2, 2, and 2 respectively, and these numbers were the same as those exposed to copper. The relative mobilities of the esterase bands in the gill, liver, blood, and kidney of the exposed group were different from those of the control. 5. There was one hemoglobin band on the anode in the normal fish. It seems that the nobility of hemoglobin band of the fish exposed to copper was slightly faster than that of the normal fish. 6. The normal gill lamellae of the fish consisted of centrally located pillar cells and a number of mucus cells. When the fish were exposed to copper, the epithelial layer was divorced first, disintegrated, and then destroyed completely. 7. The liver of the normal fish had prominent central veins, cords of hepatic cells, and sinusoids. When the fish were exposed to copper, numerous droplets of fat appeared in the cells around the central vein of the liver. It is assumed that the fatty droplets were accumulated by the lesion due to fatty metamorphosis of the liver caused by copper. 8. There was no histological difference between the muscle of the normal fish and that of the fish exposed to copper. 9. In the normal fish, the tubules of the kidney were surrounded by hemopoetic tissues. However, the kidney tissue of the fish exposed to copper received some damage on the proximal tubules. Since the tubule cells were reduced in height, the lumens of the tubules were enlarged. Consequently many proximal tubules exhibited some pink-stained granular casts and various stages of degeneration.
