Germination and Early Growth Characteristics of the Halophyte Suaeda Asparagoides Under Various Environmental Conditions (염생식물 나문재의 여러 가지 환경 조건 하에서의 종자 발아 및 초기생장특성)

  • This research was carried out to investigate the germination and seedling growth characteristics of the halophyte Suaeda asparagoides under various salt concentrations, temperatures, and the presence or absence of light S. asparagoides has a non-endospermic dicotyledonous seed structure that is go stone-shaped with a dark brown color and covered by a star-shaped perianth. Germination within the temperature range $5{\sim}40^{\circ}C$ varied from 8% to 65%. The highest germination occurred at $15^{\circ}C$. In seeds exposed to salt concentrations in the range 0~400 mM, the highest germination occurred at 0 mM, and the germination progressively decreased as the salt concentration increased. When the perianth of the seed was removed, the germination increased significantly. In the presence or absence of light, germination under white light was 5~20% higher than that under darkness, which varied according to salt concentrations in the range 0~300 mM. The plant growth of S. asparagoides, evaluated by plant height, branch number, root length, fresh weight, and dry weight, was higher in the concentration range 50~100 mM, and it decreased in the other concentrations. In conclusion, the seeds of S. asparagoides showed higher germination in fresh water at low temperature of approximately $15^{\circ}C$ under light when the perianth was removed, and the plant growth was higher at salt concentrations of 50~100 mM.

Ultrastructural Study on the Development of the Carotid Body in Human Fetus (인태아(人胎兒) 경동맥체(頸動脈體)의 발육(發育)에 관(關)한 전자현미경적(電子顯微鏡的) 연구(硏究))

  • The morphological development of the carotid body was studied by electron microscope in human fetuses from 40mm to 260mm crown rump length (10-30 weeks of gestational age). At 40mm fetus, the carotid body was composed of cluster of primitive glomus cells, primitive supporting cells, unmyelinated nerve fibers, and blood capillaries. In connective tissue between internal and external carotid arteries adjacent to the superior cervical sympathetic ganglion, two types of glomus cells through all prenatal period were found. Dark cells contained a dense cytoplasm with conspicuous large dense-cored granules, whereas light cells had a less dense cytoplasm with dense-cored granules. The light cells contained dense-cored granules that were smaller and less abundant than those in the dark cells. The primitive supporting cells appeared star-shaped with attenuated cytoplasmic extensions intervening between the adjacent glomus cells. Synaptic contact between the axon terminals and soma of the glomus cells were first observed at 40mm fetus. In 80-100mm fetus, the carotid body contained tightly packed collection of glomus cells and supporting cells which surrounded the abundant thin-walled blood vessels. Intercellular junctions between the glomus cells and adjacent cells were commonly seen. Nerve endings on the glomus cells have the form of small boutons and the other from of large calyces. During the second half of the fetal period, the glomus cells were completely enveloped by supporting cells and nerve terminals. At 260mm, the morphological features of carotid body were similar to those of human adult. The result of this study demonstrates that there are differences between the carotid body and aorticopulmonary bodies, especially with respect to their synaptic complexes, abundant blood capillaries, and two glomus cell types.

MULTI-FREQUENCY RADIO OBSERVATIONS OF MOLECULAR CLOUDS IN THE IMMEDIATE VICINITIES OF HB3 (초신성 잔해 HB3와 인접 분자운의 다파장 전파관측)

  • The structure and environments of the molecular clouds near the SNR $HB3(G132.7\pm1.3)$ are studied. The molecular complex which is located at the southern rim of HB3 was proposed by former investigators as the one interacting with HB3. This complex region of $2^{\circ}\times2^{\circ}\;at\;l=133^{\circ}$ has been observed at $^{12}CO,\;^{13}CO,\;J=1-0\;at\;a\;1'$, resolution with the 14-m radio telescope at Taeduk Radio Astronomy Observatory. We have reached to the following four conclusions. The possibility that these molecular complex and HB3 are interacting with each other cannot be supported with any of our data. The morphologies of the two show no similarities. Neither particular features for the interaction are found in the CO lines. The hypothetical 'Molecular wall' which was expected to exist on the northwestern rim of HB3 as a cause for the noncircular morphology of HB3 is turned out to be nonexistent in CO. The molecular complex which resembles a 'bar' at a low resolution is now resolved into a U-shaped shell. It seems that the U-shape is consist of two independent components. No peculiarities, such as unseen masses or bright stars capable of forming HlI regions, are found within the U-shape region. The total mass included in the complex is estimated to be $M_{total}\;=\;2.9\~8.4\times10^5\;M_\bigodot$, which is in good agreement with previous observations within errors. Considering about 12 clumps distinguishable within the complex, the total mass implies that masses of each of clumps are on the order of $10^4\;M_\bigodot$, which makes these good objects for further studies in relation to star-formation. Especially the clumps associated with W3 are worthy for more high resolution observations for better understanding of astrophysical phenomenon ongoing in them.

Acellular Dermal Matrix as a Core Strut for Projection in Nipple Reconstruction: Approaches for Three Different Methods of Breast Reconstruction

  • Background The objective of this paper was to describe a novel technique for improving the maintenance of nipple projection in primary nipple reconstruction by using acellular dermal matrix as a strut in one of three different configurations, according to the method of prior breast reconstruction. The struts were designed to best fill the different types of dead spaces in nipple reconstruction depending on the breast reconstruction method. Methods A total of 50 primary nipple reconstructions were performed between May 2012 and May 2015. The prior breast reconstruction methods were latissimus dorsi (LD) flap (28 cases), transverse rectus abdominis myocutaneous (TRAM) flap (10 cases), or tissue expander/implant (12 cases). The nipple reconstruction technique involved the use of local flaps, including the C-V flap or star flap. A $1{\times}2-cm$ acellular dermal matrix was placed into the core with O-, I-, and L-shaped struts for prior LD, TRAM, and expander/implant methods, respectively. The projection of the reconstructed nipple was measured at the time of surgery and at 3, 6, and 9 months postoperatively. Results The nine-month average maintenance of nipple projection was $73.0%{\pm}9.67%$ for the LD flap group using an O-strut, $72.0%{\pm}11.53%$ for the TRAM flap group using an I-strut, and $69.0%{\pm}10.82%$ for the tissue expander/implant group using an L-strut. There were no cases of infection, wound dehiscence, or flap necrosis. Conclusions The application of an acellular dermal matrix with a different kind of strut for each of 3 breast reconstruction methods is an effective addition to current techniques for improving the maintenance of long-term projection in primary nipple reconstruction.

Cloning, expression, and activity of type IV antifreeze protein from cultured subtropical olive flounder (Paralichthys olivaceus)

  • Antifreeze proteins (AFPs) lower the freezing point but not the melting point of aqueous solutions by inhibiting the growth of ice crystals via an adsorption-inhibition mechanism. However, the function of type IV AFP (AFP IV) is questionable, as its antifreeze activity is on the verge of detectable limits, its physiological concentration in adult fish blood is too low to function as a biological antifreeze, and its homologues are present even in fish from tropic oceans as well as freshwater. Therefore, we speculated that AFP IV may have gained antifreeze activity not by selective pressure but by chance. To test this hypothesis, we cloned, expressed, and assayed AFP IV from cultured subtropical olive flounder (Paralichthys olivaceus), which do not require antifreeze protein for survival. Among the identified expressed sequence tags of the flounder liver sample, a 5'-deleted complementary DNA (cDNA) sequence similar to the afp4 gene of the longhorn sculpin was identified, and its full-length cDNA and genome structure were examined. The deduced amino acid sequence of flounder AFP IV shared 55, 53, 52, and 49 % identity with those of Pleuragramma antarcticum, Myoxocephalus octodecemspinosus, Myoxocephalus scorpius, and Notothenia coriiceps, respectively. Furthermore, the genomic structure of this gene was conserved with those of other known AFP IVs. Notably, the recombinant AFP IV showed a weak but distinct thermal hysteresis of $0.07{\pm}0.01^{\circ}C$ at the concentration of 0.5 mg/mL, and ice crystals in an AFP IV solution grew star-shaped, which are very similar to those obtained from other polar AFP IVs. Taken together, our results do not support the hypothesis of evolution of AFP IV by selective pressure, suggesting that the antifreeze activity of AFP IV may have been gained by chance.

Development of the Korean 2.75 inch Rocket Propulsion System (한국형 2.75 인치 로켓 추진기관 개발)

  • In this paper, the development of unique model of the 2.75 inch rocket propulsion system is described. Recently developed korean 2.75 inch rocket propulsion system shows the improvement of a flame stability resulted from a change in the configuration of propellant grain, and of an incidental ignition protection function using the EMI(electromagnetic interference) filter on ignition system. Moreover it is shown that a directional flight stability is improved by increasing the number of fins and changing the nozzle configuration. Static firing test and thermal shock test were conducted for the validation before flight, and flight test of 210 rounds of rockets was conducted to verify the trajectory uniformity. In addition, intellectual property issues can be overcome with the unique korean 2.75 inch rocket motor as well as the performance improvement.

Fast Algorithms for Computing the Shortest Path between Two Points inside a Simple Polygon (다각형 내부에 있는 두 점 사이의 최단 경로를 구하는 빠른 알고리즘)

  • In this paper, we consider the shortest path problems in a simple polygon. The shortest path between two points inside a polygon P is a minimum-length path among all paths connecting them which don't pass by the exterior of P. A linear time algorithm for computing the shortest path in a general simple polygon requires triangulating a polygon as preprocessing. The linear time triangulating is known to very complex to understand and implement it. It is also inefficient in cases without very large input size. In this paper, we present the customized shortest path algorithms for specific polygon classes such as star-shaped polygons, edge-visible polygons, and monotone polygons. These algorithms need not triangulating as preprocessing, so they are simple and run very fast in linear time.

Embryonic and Larva Development of Slime Flounder, Microstomus achne (찰가자미(Microstomus achne)의 난발생 및 자치어 형태 발달)

  • The slime flounder Microstomus achne were caught at Geomun Island, Yosu-si, Jeollanamdo from January to March in 2006. The fertilized eggs were observed for morphological development of egg, embryo and larva. Eggs were colorless transparent, separative pelagic, absent of oil globule, and the diameter was 1.64${\pm}$0.03 mm (n=50). The eggs were hatched at 168 hours 40 minutes after fertilization in the range of $9.8\sim13.0^{\circ}C$ (mean $11.4{\pm}1.6^{\circ}C$). Total length of newly hatched larva was 4.05${\pm}$0.18 mm (n=20). The larva had developed membranous fin showing waterdrop-shaped structure, and their mouth and anus were not open. The myotomes were 14~15+33~34=47~49. The egg yolks were 1.64${\pm}$0.12 mm in major axis, and 1.23${\pm}$0.19 mm in minor axis. At 12 days after hatching, the total length was 7.32${\pm}$0.42 mm(n=20). The egg yolk was completely absorbed and transferred to post larval stage. Star-shaped melanophores and branch-shaped xanthophores in the edge of membranous fin were more densed. Chrysanthemum-shaped melanophores in the notochord were densed and formed 4~5 melanophore bands. At 90~93 days after hatching, morphological features of the larva, 19.91${\pm}$1.63 mm TL(n=20), were transferred to juvenile stage showing similar features with those of the adult fish.

Studies on Growth Characteristics and Shallow Green-Roof Systems of Sedum album L. Introduced in Korea (국내에 도입된 Sedum album L.의 생육 특성 및 저토심 옥상 녹화 시스템에 관한 연구)

  • These studies were carried out (1) to investigate the growth characteristics of Sedum album L. in the field, (2) to propose a suitable shallow peen roof system for this plant, and (3) to evaluate plant growth in the proposed system over the long term. The growth characteristics, such as morphological properties, growth habit, shade tolerance, and flowering, were surveyed. In experimental shallow green-roof systems, the effects of drainage type, substrate type, and soil depth on plant growth were investigated. Then drought tolerance was investigated. After planting Sedum album L. in the proposed system survival rate, cover, and resistance to insects, heal and cold were evaluated for about 2 years. The results of these studies are summarized below. 1. In the field, the aboveground part of Sedum album L. did not die back during the winter. Plant height was 4$\sim$7 cm. Roots were distributed to a depth of 5$\sim$7 cm. Sedum album L. is a compact ground-cover plant that spreads vigorously. Shading condition of less than $30\%$ of full sunlight didn't cause any trouble, but shading conditions above $87\%$ made the shape of the shoots and leaves abnormal. The plant bloomed from June to August and had a rather large compound umbel of white, star-shaped flowers. 2. Two systems, a drainage-blend-10 cm soil depth and a reservoir$\cdot$drainage-blend-15 cm soil depth, performed best in terms of cover, fresh weight, and dry weight. The first has an advantage for green roofs because it is lighter than the latter. 3. In drainage-blend-10 m soil depth and modified reservoir · drainage-blend-10 cm soil depth system no plants died for about 4 months after stopping the irrigation. The visual quality of the latter system was above 5 for 4 months and that of the former was under 5 after 2 months. In the field, however, the drought tolerance of Sedum album L. grown in the former would be enough to withstand the dry season. Considering the urban ecosystem and the importance of healthy growth the modified reservoir $\cdot$ drainage-blend-10 cm soil depth system was finally recommended. This system was composed of a 4 cm thick drainage layer and drain outlets placed at a height of 2.5 cm. 4. In the proposed system, the survival rate was $100\%$, and there was no injury induced by insects and heat. The leaf density decreased a little in winter. Cover increased throughout the year. Sedum album L. was planted with a cover of 72$cm^{2}$ on 3 April 2003; on 16 June 2003 and 15 June 2004, cover was $132.66\pm$5.87 $cm^{2}$(1.8 times) and $886.98\pm$63.51 $cm^{2}$(12.3 times), respectively.

Antifreeze Activity of Dimerized Type I Antifreeze Protein Fragments (Type I 결빙방지 단백질 조각 이량체의 결빙방지 활성)

  • Antifreeze proteins (AFPs) bind to ice crystals and inhibit their growth. AFPs are essential for the survival of organisms living in subzero environments. Type I AFP (AFP37) isolated from winter flounder is an ${\alpha}$-helical peptide of 37 residues long. In this study, we attempted to develop short AFP fragments with higher activity and solubility. We designed and synthesized N-terminal 15 and 21 residue-long AFPs, designated AFP15 and 21. Also dimerized AFP15 and 21, designated dAFP15N and dAFP21N, respectively, were generated through disulfide bonds between peptides containing CGG residues added to the N-terminus of AFP15 and AFP21 (designated AFP15N and 21N). Their helical contents and antifreeze activities were assessed using circular dichroism (CD) spectroscopy and a nanoliter osmometer, respectively. The helical content of AFP15 AFP21, AFP15N, AFP21N, dAFP15N and dAFP21N was 47, 48, 23.8, 28, 49.1, and 52%, respectively compared to that of wild type AFP37; the antifreeze activity was 8.4, 9.3, 0.05, 5.6, 12.1, 11.2% respectively, compared to that of wild type AFP37. Contrary to our anticipation, the dimerized peptides showed almost the same antifreeze activity as their monomeric counterparts. These results indicate that the dimerized peptides behave as monomeric peptides due to the high rotational freedom of disulfide bonds connecting two monomeric peptides. The star-shaped ice crystals generated by the peptides also demonstrated weak interaction between ice and peptides.
