Establishment of Artificial Screening Methods and Evaluation of Barley Germplasms for Resistance to Fusarium Head Blight (보리 붉은곰팡이병 검정법과 저항성 품종 선발)

  • Fusarium head blight (FHB) is a severe disease problem that affects the quality and yield of barley grain. The evaluation of FHB resistance is difficult because environmental conditions greatly influence FHB infection and development. The objectives of this study were to: 1) establish an efficient screening method for selecting resistant barley to FHB, 2) compare FHB severity between the cut-spike method and pot-plant method for development of mass screening, and 3) estimate FHB resistance for barley germplasms. Barley cultivars and lines were evaluated for reaction to FHB in controlled-greenhouse condition. Spikes were spray-inoculated with a suspension $(5.0\times10^5\;macroconidia\;mL^{-1})$ of Fusarium graminearum SCK-O4 strain, and then kept in a greenhouse at $18-25^{\circ}C$ with $80-100\%$ relative humidity. Inoculation were employed at 3 different heading growth stages (heading date, three days after heading, and five days after heading). The inoculation was performed in 2 consecutive days in order to avoid escapes. The inoculated plants were maintained in the greenhouse at 4 different free moisture periods (1, 3, 5, and 7 days). The percentage of FHB severity was scored from 0 to 9 according to the rate of infected kernels per spike, and three spikes were evaluated per replication with 3 replicates. There were significant differences of FHB severity depending on the different free moisture periods, but not by the inoculation at different heading stages. The optimum evaluation point of FHB severity in the greenhouse condition was on the 7th day under free moisture condition after inoculation at the heading date. Infection level in cut-spike method highly correlated with that in pot-plant method. This suggested that cut-spike method is useful in evaluating of FHB resistance in barley. Six cultivars, such as Jinkwang, Buheung, Atahualpha 92, Chevron-b, Gobernadora-d, and MNBrite-c, were selected as resistant varieties to FHB. Correlation coefficient for the FHB severity evaluated by the pot-plant method between two seasons was 0.794, indicating the stability and accuracy of the screening method.

  • Weeds may compete detrimentally with the ginseng for moisture and nutrients but hand weeding is the only practical means of eliminating weeds after crop establishment. To define the effects of 2, 4-D herbicide application on the plant growth and root yield of Korean ginseng (Panax ginseng C. A. Meyer). the herbicide 2, 4-D was applied as a foliar spray with the rates of 0.5. 1.0. 1.5 and 2.0 times of the recommended herbicide dosage 70ml/l0a. The Korean ginseng treated with 2, 4-D in the rate of two times concentration was indistinguishable from nontreated plants in visual rating for foliar symptoms. There were no significant differences of the leaf length and width as well as the stem length and diameter in check plants and those recieving 2, 4-D treatments. The. berry maturing in 3 and 4-years old ginseng was not inhibited with 2, 4-D treatment. The root weight of the 4-years old ginseng plant was not reduced by 2, 4-D application of 2 times dosage. However. when the ginseng seedling was treated with 2, 4-D. detrimental phenomena as stem bending and dicoloration of marginal part of seedling leaf were occured but stem bending was recovered in a few days.

  • This study was conducted to increase the shelf life of grape by edible coating material such as methyl cellulose (MC) with antimicrobial substances, n-capric acid isopropyl ester (ci) and sodium nitrate (sn), added by spraying method. The quality changes of packaged grapes with wrapping PE film on EPS tray were investigated for 16 days at $30{\circ}C$. The shelf-lives of C and MCci based on the weight reduction ratio of 7% were 6 days and 9 days, respectively. The reduction rate of acidity of C was higher value than those of treatments during 18 days of storage at $30{\circ}C$. The vitamin C reduction ratios of C, MCsn and MCci were 64.8, 51.5 and 49.8%, respectively, after 16 days at $30{\circ}C$. The reduction rates of firmness of C, MCsn and MCci after 16 days at $30{\circ}C$ were 44.2, 26.5, and 23,2%, respectively compared to that of initial storage grapes. The additions of ci and sn had much affected the reductions of bacteria and yeast counts especially early stage of storage. The hedonic sensory evaluation scores of MCci and MCsn had higher values than those of MC.

Effects of soluble silicon on development powdery mildew(Sphaerotheca fuliginea) in cucumber plants (규소 시용에 의한 오이 흰가루병 발병억제)

  • Effects of silicon application on development of colonies of Sphaerotheca fuliginea were examined. Cucumber plants were applied with nutrient solutions amended with different concentrations of soluble silicon and selected leaves were inoculated with known concentrations of conidia of the pathogen. Colony number per leaf, colony area per leaf, and germination rate of conidia of S. fuliginea collected from the inoculated leaves were reduced as silicate concentrations in the nutrient solutions increased from 0.05 to 4.10 mM. The increase in resistance of plants to mildew infection was apparently due to silicate accumulation in leaves, and there was no correlation between cation or ionic strength effects and the silicate treatments. Silicate treatment in growth medium remarkably suppressed powdery mildew development on cucumber. Colonies of mildew fungus were visible with over approx. 38.3% of the mature leaf surface, while that of the leaves in high Si plants was 2.3% observed at 51 days after transplanting. No significant differences were observed between 1.7 mM and 3.4mM silicate treatments. Conidial germination rates were significantly reduced by increasing Si amendments. Conidial germination ranged from 14.7 to 20.3% for plants grown in low Si solution(<1.40 mM), and from 9.0 to 12.4% for plants grown in high Si solution(>1.8 mM). Foliar applications of Si with ${\geq}$ 17.0 mM decreased the number of powdery mildew colonies. Persistence of Si foliar sprays effects on cucumber demonstrated that the 17 mM Si spray applied 4 days before inoculation with S. fuliginea reduced mildew colony formation. The relationship was positive and linear.

Removal of calcium chloride in road structure using ultra-fine bubble washing machine (미세기포 세척장비를 이용한 도로시설물 염화칼슘의 제거)

  • This study was conducted to evaluate the optimal operating condition of ultra-fine bubble washing machine for removing calcium chloride from road concrete structure. The diameter of the ultra-fine bubble was measured to 196.6 ± 100.6 nm and the ultra-fine bubble concentration was measured to 1.36 × 108 cell/ml. As a result of the performance on the spray device of the washing machine, it was confirmed that the washing efficiency of 93% and 91% appeared at 100cm and 150cm of injection distance at 100bar injection pressure. By increasing the ultra-fine bubble generation cycles from 2 to 6, the chloride removal rate increased from 1% to 7%. As the ultra-fine bubble generation air flow was lowered from 4 ml/min to 0.5 ml/min, it was confirmed that the washing efficiency increased up to 30%. The washing efficiency of ultra-fine bubble water was 25% higher than normal water.

Safety of the Herbicide Fluazifop-butyl application on the Korean Ginseng(Panax ginseng C.A. Meyer) (고려인삼(高麗人蔘)에 대한 제초제(除草劑) Fluazifob-butyl의 안전성(安全性))

  • Weed control in the Korean ginseng(Panax ginseng C.A. Meyer) garden is more difficult than in other crops because the ginseng is a perennial crop and has to be cultivated under the artificial shading. As hand weeding is the only practical means to control weed after crop establishment, a selective herbicide would greatly simplify the control of weed in ginseng garden. In an exploratory experiment, the hervicide Fluazifop-butyl was found to be selectively safe for the Korean ginseng plants. Various rates of Fluazifop-butyl were sprayed on 2-, 3-, and 4-years old ginseng plants as a foliar spray to detect crop injury and to difine their critical concentration. No apparent injury to the ginseng plant was noticable even the doubled application rate of Fluazifop-butyl the based on recommended dosage. Neither abnormal foliar change nor any inhibition in leaf and stem growth was resulted for 2-, 3-, and 4-years old ginseng plants treated with Fluazifop-butyl tripled the recommended usage. The foliar treatments of Fluazifop-butyl did not influence the photosynthesis ability but inhibited the respiration of the ginseng leaf.

Effect of Nematicide-dipping Methods for the Control of Aphelenchoides fragariae in Strawberry (살선충제 침지처리에 의한 딸기잎선충 방제)

  • Effects of different application of nematicides (fosthiazate 5% G, ethoprophos 5% G, and diazinon 34% EC) for the control of Aphelenchoides fragariae in strawberry were evaluated in a greenhouse experiments. Mother strawberry (Fragaria grandiflora) cv. Yeohong were dipped in solution of nematicides (fosthiazate or ethoprophos at 2.5 g a.i./liter in $20^{\circ}C\;or\;46^{\circ}C$) for 10 min. and planted in a greenhouse for dipping treatment. For the compare, mother strawberry were dipped in hot water for 10 min. without chemicals. For soil treatment, fosthiazate or ethoprophos at 3 kg a.i./ha were mixed into soil. For foliar spray, diazinon at 3.4 g a.i./liter was sprayed at foliage until runoff. At 40, 80, and 100 days after planting, runners were harvested from each treatment and the rate of nematode infestation and the number of nematodes per plant were examined. After 100 days of planting, mother strawberry plants dipped in fosthiazate solution (2.5 g a.i./liter, $20^{\circ}C$) for 10 min. produced more number of healthy runners and reduced % of infected runner as much as 90% and also had fewer nematodes per runner. Fosthiazate was more effective than ethoprophos. Foliar application of diazinon was reduced A. fragariae populations only in early season. Hot water treatment and nematicide soil treatment were less effective.

Quality Improvement of Oriental Melon and Watermelon Using Bioceramics (바이오 세라믹을 이용한 수박ㆍ참외의 품질 향상에 관한 연구)

  • Oriental melon and watermelon plants were cultivated in the soil treated with bioceramics in a greenhouse during summer season from June 1st to August 20th, 1995. Two application methods were employed, one was a mixed treatment of soil and bioceramics, and the other was a spray treatment of bioceramic solution on the stems and leaves. And two types of bioceramics were also stopped by five levels. In order to analyze the bioceramic effect on oriental melon and watermelon, the growth rate of stems, leaves and fruits were measured in the greenhouse. After harvest, the sweetness of fruits was measured and the freshness of fruits based on the storage period was tested by human taste and smell sense. The results are summarized as follows. 1. The growth rates of stems, leaves and fruits of oriental melon and watermelon were the largest in the bioceramic treatment of No. 3. 2. The density of oriental melon and watermelon was the largest in the bioceramic treatment of No. 3 and No. 2 respectively. 3. The Brix number of watermelon was 10.6 in non-bioceramic treatment and 11.5 in the bioceramic treatment of No. 2, and that of oriental melon was 8.6 in non-bioceramic treatment and 12.3 in the bioceramic treatment of No. 2. 4. The storage duration of watermelon treated with bioceramics was about 50 days in the condition of the ambient temperature of 25-3$0^{\circ}C$.

Effects of Soil Moisture and Chemical Application on Low Temperature Stress of Cucumber (Cucumis sativus L.) Seedling (토양수분조건 및 화학물질처리가 오이묘의 저온장해에 미치는 영향)

  • This study was conducted to investigate the effects of chemical application and amout of soil moisture on low temperature stress of cucumber seedling under the greenhouse conditions. When chilling treatments ($2^{\circ}C$) were begun at 07:00AM, survival rates of seedlings of two conditions; -0.3 bar and -5.5 bar were 28.3% and 83.3% respectively. But when chilling treatments were begun at 6:00PM - even the soil moisture condition was -0.3 bar - the survival rate was above the 87%. When reducing the soil moisture from -0.3 bar to -9.0 bar, ABA content in leaf was inc.eased by 6.5fo1d. Spraying of abscisc acid (ABA) before or after the chilling significantly increased the survival rates of seedlings, decreased the amounts of leaking electrolytes and prevented the yield reductions. ABA application on the soil before the chilling appeared to be more effective than the application after the chilling with foliar spray. Spraying of ABA ($10^{-5}M$), urea (0.2%) or $KH_2PO_4$ was effective in counteracting the low temperature, which causes growth deterioration and yield reduction in cucumbers.

Combined Foliar Spray of Boron, Calcium, and Silicon can Influence Quality and Shelf Life of Cherry Tomato in Modified Atmosphere Packaging (붕소, 칼슘, 규소의 복합 엽면시비가 방울토마토의 품질과 MAP 조건에서 저장성에 미치는 영향)

  • This study was analyzed the effects of boron (B), calcium (Ca), silicon (Si) on quality and shelf life of 'Unicorn' cherry tomato at the light red maturity-stage. The storage conditions were modified atmosphere packaging (MAP) by oxygen transmission rate (OTR) packaging film at $5^{\circ}C$, $11^{\circ}C$, and $24^{\circ}C$. Respiration and ethylene production were the lowest in B + Ca + Si -treated tomato fruits. The lowest fresh weight loss and the longest shelf life resulted from the B + Ca + Si treatment. And the firmness was enhanced by B + Ca + Si treatment at harvest time, and it was retained after storage at $5^{\circ}C$, $11^{\circ}C$, and $24^{\circ}C$. Significantly lower soluble solids, lycopene, and color development were found at B + Ca + Si-treated tomato fruits compared with control after storage. Moreover, the highest titratable acidity and vitamin C content were observed in B + Ca + Si-treated tomato fruits after storage. From the above results, it was concluded that B + Ca + Si combined treatment can delay the maturity of cherry tomato after harvesting, and retained the firmness and prolong the shelf life.
