Effects of Culture Environments on Alkaline Protease Biosynthesis in Streptomyces sp. (Streptomyces속 세균에서 호염기성 단백질 분해효소 생합성에 미치는 배양환경의 영향)

  • The aims of the present study were to evaluate the effects of culture conditions on the biosynthesis of extra-cellular alkaline protease in Streptomyces sp. The formation of aerial mycelia and spores were compared with the protease production in order to know the relations between the alkaline protease and the cell differentiation. As results, it was found that substrate concentration was very critical to regulate the formation of the protease, aerial mycelia, and spores, which were resulted from the changes of culture pH to acid. When the culture pH was adjusted with phosphate buffer from pH 6 to pH 9, the alkaline protease production was increased as the culture pH increased whereas aerial mycelia and spore formation were reversely related to the culture pH. Therefore, it was thought that the culture pH was an important factor to regulate the alkaline protease synthesis.

  • From apple skin, we isolated a bacterial strain which strongly inhibited the growth of apple white rot fungus, Botryosphaeria dothidea. The isolated strain, designated as SS279, was identified to be the genus Bacillus. The antifungal activity of Bacillus sp. SS279 was found in the culture filtrate. The production of antifungal substances occurred during logarithmic phase and was the highest when cultures reached the stationary growth phase. The optimum ranges of temperature and pH for its production were 25-30$\circ$C and 4.5-9.0, respectively. The culture filtrate of Bacillus sp. SS279 exhibited a strong inhibitory effect on the spore germination and germ tube elongation of B. dothidea. Autoclaved culture filtrate of Bacillus sp. SS279 showed only a slight decrease in antifungal activity, indicating that the Bacillus sp. SS279 produce heat-stable antifungal substances. In in vivo bioassay, Bacillus sp. SS279 also showed antagonistic activity against apple white rot caused by B. dothidea.

  • The purpose of this study is to investigate the liquid culture conditions for mass production of Metarhizium anisopliae var. majus ARS 978 which is a potential microbiological pesticide. The temperature and pH range for optimal cultivation were $28^{\circ}C$ and pH 7.0, respectively. For M. anisopliae ARS 978, 1.0 %(v/v) molasses, 1.0 %(w/v) distiller's dry solubles, and 0.3 %(w/v) $CaCO_{3}$ were found to be the proper nutrients, considering cell mass, enzyme activities and spore concentration.

  • The enzyme-producing potentials of industrially important strains of Aspergillus spp. were studied. Irradiation of three original isolates of Aspergillus oryzae to ultra-violet rays resulted in the production of mutants which differed from the parent riboflavin and vitamin $B_{12}$ in culture media. 1. Irradition three strains of Aspergillus oryzae to ultraviolet light produced mutants and two strains of them were selected for soy sauce brewing. 2. The two strains are the physiological mutants of Aspergillus oryzae. Both were found to have superior enzyme activity to their relatives. 3. Aspergillus oryzae UV-induced mutant 172-722 and 569-713 were more powerful than others in the production of riboflavin and vitamin $B_{12}$. The enzyme activity of these strain were high and decreased only slightly even in 20 percent solution of NaCl. 4. Aspergillus oryzae UV-induced mutant 172-722 had more powerful protease producibility in wheat bran media than in modified Czapek's solution. On the contrary, Aspergillus oryzae UV-induced mutant 569-713 had more powerful producibility of saccharogenic and dextrinogenic amylase in modified Czapek's solution than in mold bran. 5. Aspergillus oryzae UV-induced mutant 172-722 formed the spore rapidly and Aspergillus oryzae UV-induced mutant 569-713 did ordinarily. 6. It is found from the results that Aspergillus oryzae UV-induced mutant 172-722 is valuable material for the manufacture of soy sauce because of its high protease activity in 20 percent solution of NaCl. Aspergillus oryzae UV-induced mutant 569-713 is suitable for soy bean mash and for fermented red pepper sauce for its high saccharogenic and dextrinogenic amylase activity in 20 percent solution of sodium chloride.

  • This study analyzed and compared growth characteristics under large-scale fermentation at 35℃ of three microorganisms with the ability to reduce odor-producing substances in livestock. The three microorganisms (Bacillus subtilis FWC1, Bacillus amyloliquefaciens NAAS1, and Pichia farinosa NAAS2) evaluated in this study have been proven effective in reducing odor-inducing substances. Bacillus subtilis FWC1 exhibited the highest viable cell count when using 2% maltodextrin as carbon source, 0.05% soy-peptone as nitrogen source, and 0.3% yeast extract. The optimum media composition for B. amyloliquefaciens NAAS1 was 1.2% modified-starch with 0.8% yeast extract. The spore formation rate in the mass production of the Bacillus strains was over 90%, indicating that optimal growth medium compositions have been identified. In the case of P. farinosa NAAS2, our optimized growth medium [2% (w/v) glucose and 1% (w/v) yeast extract] improved biomass production.

  • Two hundred forty seven isolates obtained from 21 white or cream-colored mushroom strain by single spore isolation or multiple spore germination were compared type of mycelial growth in vitro and yield trial in a preliminary test. As a result of these tests, four isolates were selected and compared the yields of sporophores with those of 505 and 702 which are leading strains in mushroom production. The newly selected isolate No. 705 showed high yield of mushroom with good quality as described below. 1. The isolate No. 705 produced 13% more mushrooms than those from the strain No. 703. Both produced creamy type of mushroom. The isolate No. 705 showed high blanching yield ratio and moderate resistance to Mycogone perniciosa. 2. For the isolate No. 705 obtained by multiple spore germinations, the optimum temperature of mycelial growth was $25^{\circ}C$, also the mycelial growth was better at $15^{\circ}C$ than others, optimum moisture content of the compost was 65% and optimum casing soil pH for mycelial growth was 7.8. 3. The new isolate No. 705 produced more number of sporophores and the ratio between parts of sporophores were intermediate of those from the strains No. 505 and No. 703.

Effect on the Production of Toxin Protein of Bacillus thuringiesis by Culture Conditions (배양 조건이 Bacillus thuringiesis 독소단백질의 생산에 미치는 영향)

  • We was investigated parasporal inclusion proteins change to use industrial medium of new strain Bacillus thuringiensis CAB 565, CAB 566. To confirm medium's oxygen efficient consist of glucose and yeast extract, we was conducted oxygen transfer coefficients (KLa) of medium's concentration and impeller in 20 l-Jar fermentor. When to increase air flow rate and medium concentration, KLa rate is rise. Also it is effective on agitation rate 200 rpm, but KLa rate is decrease when to rise agitation rate. To hold dissolved oxygen rate (upper 50%), Air flow rate is steadily increase on culture to use microsparger. When 16 hour of culture stage, B.t. CAB 565 and B.t. CAB 566 harvested respectively $2.3{\times}10^{10}$, $1.8{\times}10^{10}$ viable cell/ml. When 54 hour, B.t. CAB565, 566 harvested respectively $1.9{\times}10^{10}$, $1.4{\times}10^{10}spore/ml$. To resulting carbon's concentration, It is the most effective that glucose concentration is contained 5% in medium.

  • This experiment was conducted to select prominent microorganisms with a good insecticidal activity among the ten species, which isolated from soil at the near of Chung-buk, Chung-nam, and Gang-won provinces and made protein crystal endotoxin. As a result, GB-413 strain was finally selected, which showed the high insecticidal activity against susceptible diamondback moth (Plutella xylostella), beet army worm (Spodoptera exigua) and tobacco cutworm (Spodoptera litura) as well as resistant diamondback moth strains. By modifying the cultivation process f.g. lowing the glucose concentration at early cultivation stage and adding the carbon after inducing the spores, the percentage of making spore as well as the number of active spore were increased and the time for cultivation and spore forming was reduced without a reduction of insecticidal activity. These results were not only applied successfully for the optimized cultivation process for a fermentation tank containing five tons capacity, but also improved the possibility of mass cultivation for commercial production.

Chemosensitization of Fusarium graminearum to Chemical Fungicides Using Cyclic Lipopeptides Produced by Bacillus amyloliquefaciens Strain JCK-12

  • Fusarium head blight (FHB) caused by infection with Fusarium graminearum leads to enormous losses to crop growers, and may contaminate grains with a number of Fusarium mycotoxins that pose serious risks to human and animal health. Antagonistic bacteria that are used to prevent FHB offer attractive alternatives or supplements to synthetic fungicides for controlling FHB without the negative effects of chemical management. Out of 500 bacterial strains isolated from soil, Bacillus amyloliquefaciens JCK-12 showed strong antifungal activity and was considered a potential source for control strategies to reduce FHB. B. amyloliquefaciens JCK-12 produces several cyclic lipopeptides (CLPs) including iturin A, fengycin, and surfactin. Iturin A inhibits spore germination of F. graminearum. Fengycin or surfactin alone did not display any inhibitory activity against spore germination at concentrations less than 30 ug/ml, but a mixture of iturin A, fengycin, and surfactin showed a remarkable synergistic inhibitory effect on F. graminearum spore germination. The fermentation broth and formulation of B. amyloliquefaciens JCK-12 strain reduced the disease incidence of FHB in wheat. Furthermore, co-application of B. amyloliquefaciens JCK-12 and chemical fungicides resulted in synergistic in vitro antifungal effects and significant disease control efficacy against FHB under greenhouse and field conditions, suggesting that B. amyloliquefaciens JCK-12 has a strong chemosensitizing effect. The synergistic antifungal effect of B. amyloliquefaciens JCK-12 and chemical fungicides in combination may result from the cell wall damage and altered cell membrane permeability in the phytopathogenic fungi caused by the CLP mixtures and subsequent increased sensitivity of F. graminearum to fungicides. In addition, B. amyloliquefaciens JCK-12 showed the potential to reduce trichothecenes mycotoxin production. The results of this study indicate that B. amyloliquefaciens JCK-12 could be used as an available biocontrol agent or as a chemosensitizer to chemical fungicides for controlling FHB disease and as a strategy for preventing the contamination of harvested crops with mycotoxins.

  • Entomogenous fungi which attack living insects are powerful means for microbiological insecticide. The purpose of this study is to establish the culture conditions and media for mass production of Beauveria sp. C208 which has a broad host range as a potential microbiological pesticide. The temperature and pH range for optimal cultivation of this strain were 28$circ$C and pH 5.0-7.0. For Beauveria sp. C 208, 2% rice straw and 0.6% tryptone were found as the proper carbon and nitrogen sources, considering cell mass, enzyme activities such as chitinase, protease and lipase, and spore concentration.
