Oryeongsan (ORS), a formula composed of five herbal medicines, has long been used to treat impairments of the regulation of body fluid homeostasis. The purpose of this study was to determine the antihypertensive and renal protective effects of ORS in rats with hypertension. Spontaneously hypertensive rats (SHR) were divided into two groups with similar mean baseline systolic blood pressure (SBP). Then, 1 mL/kg of vehicle (distilled water) or 1.5, 3 g/kg of ORS extract were administered orally once a day for 4 weeks. SBP and diastolic blood pressure (DBP) were measured at weeks 1, 2, 3 and 4. At the end of the experiment, blood was collected, and heart were removed for histology. By the 2 weeks after initiation of treatment, the ORS treated group had significantly lower SBP than SHR rats. The ORS treatment significantly improved blood pressure and echocardiogram parameters compared to hypertensive rats. Additionally, the left ventricular (LV) remodeling and LV dysfunction were significantly improved in ORS treated group hypertensive rats. Furthermore, an increase in fibrotic area has been observed in SHR rats compared with Wistar-Kyoto rats (WKY). Furthermore, administration of ORS significantly attenuated cardiac fibrosis in hypertensive rats. Therefore, these findings suggest that ORS has a protective effect on heart failure by alleviating hypertensive heart disease and cardiovascular dysfunction in SHR.
The phospholipase C (PLC)-mediated intracellular signal transduction pathway is considered to be involved in the regulation of blood pressure. However, little information is available concerning the distributional and functional significance of PLC in the genetic hypertensive rats. As the first step of knowing the role of PLC on hypertension, we investigated the distribution of 6 PLC isozymes $(PLC-{\beta}1,\;-{\beta}3,\;-{\beta}4,\;-{\gamma}1,\;-{\gamma}2\;and\;-{\delta}1)$ in the heart and brain, which are concerned with hypertension, in the normotensive Wistar-Kyoto rat (WKY) and spontaneously hypertensive rat (SHR) using the western blotting and immunocytochemistry. The immunoreactivities of PLC isozymes in brain were detected, but there were no distributional and quantitative differences between the WKY and SHR. In the heart, but the immunoreactivities to $PLC-{\beta}1$ and $-{\gamma}2$ in the SHR were higher than those in WKY. In immunocytochemistry to confirm these western blotting data, $PLC-{\beta}1$ and $-{\gamma}2$ were localized in cardiac myocytes and the intensities of immunoreactivity in SHR were stronger than that in WKY. These results suggest that $PLC-{\beta}1$ and $-{\gamma}2$ would have possibility to concern with the establishment of spontaneous hypertension.
Objective : This study was examined to investigate the effect of Gamitongseong-san(GTS) extract on spontaneous hypertension. Methods : After administering GTS extract to SHR for 4 weeks, we measured anti-oxygen effects, weight of body, heart and kidney, blood pressure, heart rate, aldosterone, catecholamine, electrolyte, uric acid, and BUN. Results : 1. GTS increased 2,2-diphenyl-1-picrylhydrazyl(DPPH) scavenging activity and superoxide dismutase(SOD) similar activity depending on the concentration. 2. GTS significantly decreased heart weight in spontaneously hypertensive rat. 3. GTS significantly decreased blood pressure and heart rate in SHR. 4. GTS significantly decreased aldosterone. 5. GTS significantly decreased norepinephrine and epinephrine. 6. GTS significantly decreased $K^+$. 7. GTS significantly decreased BUN. Conclusions : These results suggest that GTS may be usefully applied for the treatment of hypertension.

The present study investigated whether electroacupuncture (EA) can alleviate hypertension and concomitant disorders such as decreased kidney weight and elevation of serum creatinine in spontaneous hypertensive rats. EA (2 Hz, 3 mA, 10 min) was applied to Joksamni (ST36) once daily for 7 days. Body weight, blood pressure and heart rate were measured on Day 0, 2, 6, 8, 10 and 12, and kidney weight and serum creatinine levels were examined after sacrifice (on Day 12 after last examination). In the ST36 group, the blood pressure were significantly decreased from 6thdays and its effects lasted up to Day12 (up to 5days after cessation of acupuncture), compared to control. And, significant decreases of the heart rates after EA at ST36 were also observed on Day 2, 6, and 8. However, there were no significant differences in daily body weight, kidney weight and serum creatinine between acupuncture and control group. These results showed that EA at ST36 caused anti-hypertension by decreasing blood pressure and heart rates in spontaneous hypertenstive rats, although it failed to alleviate concomitant disorders such as the decreased kidney weight and elevation of serum creatinine shown in hypertensive rats.
The present study was carried out to investigate the effect of water extract and 70% ethanol extract from Gastrodiae Rhizoma on cardiovascular activities and plasma levels of catecholamines in unanaesthetized spontaneously hypertensive rats. The depressor response in SHR was observed during three to six hour period after an oral administration of water extract from Gastrodiae Rhizoma(GR). There was a statistically significant correlation between the magnitude of the depressor response induced by an oral administration of water extract from GR and the initial control blood pressure level. The increase in blood pressure induced by norepinephrine was less in Wistar rat treated with GR water extract than those without GR extract. No significant change in heart rate was observed in SHR receiving either water extract or ethanol extract from GR. Associated with the depressor response, there was a concomitant reduction in plasma levels of norepinephrine in SHR at 4 hour after an oral administration of water extract from GR. Plasma levels of norepinephrine and epinephrine were decreased slightly at 2 hour after an oral administration of ethanol extract from GR. These results suggest that the depressor effect of water extract from GR may be due, in part, to a decreased sympathoadrenal activity.

The antihypertensive effects of the Wild Mountain Ginseng Cultured Root Extract (WMGCE) were investigated in spontaneously hypertensive rats (SHR). Daily oral administration of the WMGCE (100 and 200 mg/kg) exhibited a significant decrease in blood pressure in SHR rats during for 8 weeks. The systolic blood pressure was dose- and time-dependently decreased significantly from the second weeks (p < 0.05) to the end of WMGCE treatment in SHR. The WMGCE decreased the plasma levels of sodium, potassium, chlorides, urea and osmolarity in SHR rats but no statistically significant change was observed. Furthermore, no significant changes were noted on heart weight, heart rate and diameter of aorta after WMGCE treatment in SHR. Our results suggest that daily oral administration of WMGCE at the dose of 100 and 200 mg/kg for 8 weeks exhibited antihypertensive activities.
This study explored the effect of dietary levels of Na and Ca on spontaneously hypertensive rats (SHR). SHR were randomly divided into 5 groups and fed a high fat/cholesterol diet containing three levels of Na (0.05, 0.1, 1.5%) and Ca (0.1, 0.5, 1.5%) for 9 weeks. Body weight gain was not influenced by dietary intake but water intake significantly increased in high Na supplementation. Systolic blood pressure was not influenced by dietary Na and Ca levels but was decreased by dietary low Na/high Ca levels at 9 weeks. Angiotensin-II level was affected by dietary Na level but not by Ca levels. Plasma Ca, Mg, K and Na levels were in the normal range regardless of dietary Na and Ca levels. Weight, and K and Na contents of the heart and kidney were not significantly different among those with different dietary Na and Ca levels. Ca and Mg contents of the heart and kidney were significantly higher in the normal Na/normal Ca group. Ca and Mg in the feces were higher in those with high Ca intake. Na in the feces was higher in those with high Na intake. Therefore, Na and Ca had different mechanisms in the hypertension/hyperlipidemia models, respectively. And we suggested that Mg must be supplemented when Ca intake was high because Mg excretion was increased by Ca supplementation.
Effect of BR-900317 on the angiotensin I-induced pressor response in pithed rats and the effects of its single oral administration on plasma angiotensin converting enzyme (ACE) activities in normotensive rats and on the cardiovascular system in hypertensive model rats (SHR, RHR), were compared with those of captopril. BR-900317 attenuated the angiotensin I-induced pressor effects in pithed rats. In a single oral dose administration study, BR-900317 inhibited the plasma ACE activities in a dose-dependent fashion. Duration of the action of BR-900317 was similar to that of captopril. BR-900317 produced antihypertensive effect in spontaneously hypertensive rats and dose-dependent antihypertensive effect in 2-kidney Goldblatt hypertensive rats without affecting heart rate. These results suggest that the main mechanism of the antihypertensive effect of BR-900317 is the suppression of angiotensin II production due to the inhibition of the ACE.
The effects of cardiac ischemia-reperfusion and $\beta$-adrenergic stimulation on metabolic function and energetics were investigated in Lan gendorff-perfused spontaneously hypertensive rat (SHR) hearts. Sarcoplasmic reticulum {TEX}$Ca^{2+}${/TEX}-dependent ATPase and cardiac lactate dehydrogenase (LDH) are additionally studied. The perfusion medium (1.0 mM {TEX}$Ca^{2+}${/TEX}) contained 5 mM glucose(+5 U/L insulin) and 2 mM pyruvate as substrates. Global ischemia was induced by reducing perfusion pressure of 100 to 40 cm {TEX}$H_{2}${/TEX}O, followed by 20 min reperfusin. Isoproterenol (ISO, 1$\mu$M) was infused for 10 min. Coronary vascular resistance and myocardial oxygen consumption ({TEX}$MVO_{2}${/TEX}) of SHR were increased in parallel with enhanced venous lactate during ischemia and reperfusion compared to those of Sprague Dawley (SD) hearts. Although ischemia-induced increase in venous lactate and combined adenosine plus inosine was abolished, coronary vasodilation produced in SD during reperfusion. In SHR, depressed reactive hyperemia was associated with a fall in cardiac ATP and CrP/Pi ratio and a rise in intracellular lactate/Pyruvate ratio. On the other hand, ISO produced coronary functional hyperemia and an increase in {TEX}$MVO_{2}${/TEX}. However, these responses were less than those in SHR hearts. The ATPase activity of SHR was attenuated in free {TEX}$Ca^{2+}${/TEX} concentrations used under basal condition and with ISO compared to that of SD. Venous lactate output and cardiac LDH activity were augmented in SHR as influenced by ISO. These results demonstrate that coronary reactive and functional hyperemia was dpressed in SHR, which cold be explained by alterations in the cytosolic phosphorylation potential and the cytosolic redox state manipulated by LDH, and by abnormal free calcium handling.
In order to investigate the pharmacological properties of ι-muscone, effects of ι-muscone and musk were studied on the cardiovascular system with various experimental models. In isolated rat aorta, ι-muscone and musk made the relaxation of blood vessels in maximum contractile response to phenylephrine (10$^{-6}$ M) in endothelium-containing rings of the rat aorta, but not in endothelium-denuded rings. However, ι-muscone and musk in the presence of the inhibitor of NO synthase and guanylate cyclase did not make the relaxation of blood vessels. In spontaneously hypertensive rats (SHRs), ι-muscone and musk slightly reduced blood pressure but significantly decreased heart rate. In the isolated perfused rat hearts, ι-muscone and musk did not affect significantly on LVDP, contractile force, coronary flow and (-dp/dt)/(+dp/dt). These results suggest that ι-muscone and musk have weak cardiovascular effects with relaxation of blood vessel and decrease of heart rate, but without significant cardiac functions.
