Fucoidan Enhances the Survival and Sustains the Number of Splenic Dendritic Cells in Mouse Endotoxemia

  • Fucoidan is a sulfated polysaccharide derived from brown algae that has been reported to perform multiple biological activities, including immunostimulation. In this study, we investigated whether fucoidan has beneficial effects on endotoxemia induced by LPS, a septic model in mice. The focus of this study was on survival rates and spleen function of the mice upon treatment. We found that fucoidan had prophylactic effects on the survival rate of mice with endotoxemia. Flow cytometric analysis using antibodies for subset-specific markers revealed that fucoidan profoundly reversed the depleted population of dendritic cells in mice with endotoxemia. According to Western blot analysis, the spleen cells of LPS/fucoidan-treated mice showed a higher expression of anti-apoptotic molecules compared to those of LPS-treated mice. Also, fucoidan-treated spleen cells were more responsive to mitogens. Taken together, these results demonstrate that fucoidan pre-treatment has beneficial effects on the survival rate and function of the spleen in mice with endotoxemia. This study may broaden the use of fucoidan in clinical fields, especially endotoxemia.

면역시킨 마우스의 비장세포, 혈청 또는 모유를 통해 얻을 수 있는 Naegleria fowleri 감염에 대한 방어 능력 (Resistance to Naegleria fowleri infection passively acquired from immunized splenocyte, serum or milk)

  • Naegleria fowleri로 면역시킨 마우스의 혈청이나 비장세포를 다른 마우스에 주입시켜 N. fowleri 감염에 대한 면역효과가 나타나는지 살펴보았다. 또 어미 마우스를 N. fowleri로 면역시 킨 다음 면역시키지 않은 어미에서 태어난 새끼 마우스에게 면역시킨 마우스의 모유를 먹게한 뒤 모유를 통한 면역 효과의 전달 여부를 관찰하였다. 면역시킨 마우스의 비장세포나 정상 마우스의 혈청을 다른 마우스에 주입시킨 경우 감염 후 사망률이 감소하였으나 혈중 IgG는 증가되지 않았다. 한편, 면역시킨 마우스의 혈청이나 정상 마우스의 비장세포를 준 경우 혈중 보G가 증가되고 감염 후 생존 기간은 연장되었으나 결국은 모든 마우스가 사망하였다. 즉 면역시킨 마우스 또는 정상 마우스의 혈청이나 비장세포를 다른 마우스에 주입시키면 사망률이 감소되거나 생존 기간이 연장됨을 알 수 있었다. 또 면역시킨 어미 마우스의 혈중, 모유 내, 또는 그 모유를 먹은 새끼 마우스의 혈 중에서, N. fowleri에 대한 IgG는 증가되었으나 IgA는 증가하지 않았으며 증가된 항체도 감염 후 의 사망률이나 생존 기간에는 큰 영향을 주지 않았다.

  • 1) 자라의 비장을 싸고있는 皮膜은 筋內細胞가 없으며 哺乳類의 그것과 마찬가지로 망상섬유와 탄력섬유 및 교원섬유를 함유하고 있다. 2) 자라의 비장의 脾株는 개구리 비장보다 발달되고 짧은 脾株가 잘 발달되어 있다. 3) 자라 비장의 血管系는 哺乳類와 거의 비슷하나 특히 中心動脈이 잘 발달되고 脾洞의 발달은 포유류의 脾洞構造나 數量面에서 下等性을 보인다. 4) 자라 비장의 造血細胞의 출현빈도가 포유류와 양서류등 다른 脊椎動物보다 훨씬 많이 出現된다.

우(牛), 마(馬), 돈(豚) 및 견(犬)의 실질장기내(實質臟器內) Mast Cell 분포(分布)와 Rompun을 투여(投與)한 견(犬)의 Mast Cell 탈과립소견(脫顆粒所見) (Distribution of the Mast Cells in the Parenchymal Organs of the Cattle, Horses, Pigs, Dogs, and Rompun-induced Mast Cell Degranulation in the Dog)

  • This paper dealt with the distribution of normal mast cells in the spleen, liver and lung on cattle, horses, pigs and dogs, and also degranulation of mast cells in the dogs infected with Rompun (2% Xylazine HCl). The results observed are summarized as follows. Normal mast cells were distributed in spleen, liver and lung on cattle, horse, pig and dog. Mast cells were observed in both red pulp and surroundings of white pulp of the spleen in horse, in the white pulp of the spleen in cattle, in the trabeculae of the spleen in pigs, and in white pulp and red pulp of the spleen in dogs, respectively. Mast cells were observed in the portal triad of the liver in cattle and horses, in both portal triad and interlobular connective tissues of the liver in pigs, and not only the portal triad but also walls of the sinusoids and the central veins in dogs. A large number of mast cells were observed in the interlobular septa and peribronchioles of lung on all the species in this experiment. The mast cells are more numerous in the lungs than other organs. Author considers that numbers of normal mast cells distributed in the tissue is related to the dosage of Rompun in animal. The degranulation of mast cells were observed in the subcutaneous tissues of dog intramuscularly injected with Rompun(0.5ml/times) for 4 or 5 times and subcutaneously injected with Rompun(0.3ml/times) for 4 times. In dog intradermally injected with 0.1ml of Rompun, mast cells were decreased in number at 30 minutes and markedly decreased in number at 2 hours, but more or less increased in number at 3 hours after injection. In addition, the granules of the mast cells were decreased in number at 30 minutes and marked degranulation of the mast cells were recognized at 2 hours after injections, but normal mast cells begun to appear in subcutaneous tissue with the lapse of time from 3 hours after injection. There was also observed local infiltration of neutrophils in subcutaneous tissues of dogs intradermally injected with 0.1ml of Rompun at 30 minutes. At 2 hours after injection, numerous neutrophils and a small number of eosinophils were observed in the site of injection. Conclusionally, Rompun was regarded as a factor which causes the degranulateon of mast cell and the authors considered that histamine released from the mast cells by Rompun might cause relaxation of skeletal muscle.

한우 황체세포의 Progesterone 및 IGF-I 분비에 대한 비장세포의 역할 (Roles of Spleen Cells in the Regulation of Progesterone and IGF -I Secretion in the Hanwoo Luteal Cells)

  • 본 연구는 한우 난소의 황체세포를 분리ㆍ체외배양하여 progesterone 과 IGF-I 분비기능에 대한 비장세포의 첨가효과를 검토하여 난소기능에 대한 기초정보를 제공하는데 있다. 도축장에서 도축되는 한우 난소로부터 황체를 분리ㆍ효소처리하여 LLC 와 SLC (1$\times$$10^{6}$ cells/$m\ell$)를 회수하였으며 10% FCS와 antibiotic가 첨가된 D-MEM 배양액에 24 시간 체외배양하였다. 비장세포는 성숙한 거세한우의 비장에서 회수하여 5%, 10% 및 20%를 황체세포에 각각 첨가하여 공배양하였다. 황체일령별 조직내 progesterone 농도는 발정주기 중 중기황체 (CL-3)가 유의적으로 높았다. 비장세포를 5%, 10% 및 20%를 각각 황체세포에 첨가하여 배양한 결과, 배양액 중의 progesterone 농도는 대조구에 비해 유의적인 차이가 발견되지 않았으나 LH(100ng/$m\ell$) 첨가구와 비장세포 5%, 10%, 20% 첨가와 함께 LH 를 각각 공배양구에서 대조구(LH+BP)에 비해 유의적 (p<0.05)으로 높은 progesterone 분비를 나타내었다. 한편, 황체세포의 체외배양에 있어서 IGF-I은 일정하게 분비하였으나 비장세포와 LH+비장세포 5%, 10% 및 20%와의 공배양은 대조구에 비해 큰 차이가 없었으나 LH 단독처리구만이 대조구에 비해 유의적으로 (p<0.05) 높은 수준을 보였다. 이상의 결과로, 비장세포는 황체세포에 작용하여 LH 의 progesterone 분비기능을 촉진시킴으로서 황체세포의 progesterone 분비를 촉진하는 기능이 있으나 IGF-I의 분비기능은 없는 것으로 사료된다.

Chlorogenic acid 및 인동등 ethyl acetate 분획의 비장 및 흉선 세포에서의 유전자 발현 분석을 통한 면역조절효과 (Immunomodulatory effects of chlorogenic acid and ethyl acetate fraction from Lonicera japonica on cytokine gene expression profiles in spleen and thymus)

  • Objective : Lonicera japonica contains anti complementary polysaccharides and polyphenolic compound. Among these polyphenolic substances, chlorogenic acid is the major active component of this plant. However, the immunological mechanisms for these activities, have not been elucidated, nor the active components. To clarify immunomodulatory effects of those we examined the relationship between the activity of CD8+ T cell-mediated lysis and the frequency of cytokine profiles in spleen, thymus (especially IFN-${\gamma}$, IL-4, GM-CSF etc.) expressing CD8+ T cells activated by IL-2. Methods : To study immunomodulatory effects ethyl acetate fraction from Lonicera japonica, chlorogenic acid on cytokine gene expression from spleen, thymus cells, RT-PCR was performed after quantitative normalization for each gene by a densitometry using ${\beta}$-actin gene expression. A modified standard $^{51}Cr$-release assay was used to measure cytotoxic activities of cytotoxic T cells. Spleen, thymus cells from NOD mice were stained with CD3, CD4, CD44, CD69 in staining buffer and analyzed by two color flow cytometry. Results : We showed that ethyl acetate fraction from Lonicera japonica in combination with IL-2 resulted in a significant enhancement of PCR products for IFN-${\gamma}$, IL-4, IL-10, GM-CSF, IL-6 and cytotoxtic CD8+ T cell proportion in spleen and thymus T cells in NOD mice. This suggests that IFN-${\gamma}$, IL-6 like IL-4 may be acting as a regulatory rather than proinflammatory cytokine. Conclusions : In conclusion, based on the results of the present study which showed that ethyl acetate fraction from Lonicera japonica and chlorogenic acid upregulating cytokine gene expression in spleen and thymus, we are tempted to speculate that some of the therapeutic efficacies such as anti-diabetic activity of Lonicera japonica are due to the immunomodulatory its ethylacetate fraction and chlorogenic acid.

향사양위탕(香砂養胃湯) 전탕액이 노화주의 비장, 췌장, 위장 세포의 항산화능에 미치는 영향 (Anti-oxidant Effect of Hyangsayangyi-tang Decoction in Stomach, Spleen and Pancreas Cell of SD Rats)

  • 1. Objectives The purpose of this study was to observe the anti-oxidant effects of Hyangsayangyi-tang(HY) in SD rats. 2. Methods This experiment was used the tissue of stomach, spleen and pancreas cells of 6, 52 and 68 weeks old SD rats. Each age group was again divided into three groups. One group, as normal group, was not-treated cells, another group, as control group, was saline-treated cells, and the last group, as experimental group, was HY-treated cells. After culture for 48 hours, each groups measured the level of SOD, GSH, MDA and NO in the tissue of stomach, spleen and pancreas cells. 3. Results and Conclusions The activity of SOD were significantly increased in spleen cell of 52, 68 w-HY group, pancreas cell of 68 w-HY group and in stomach cell of 52, 68 w-HY group compared with those of the normal and the control groups. The level of GSH were significantly increased in spleen cell of 52, 68w-HY group and in pancreas cell of 68w-HY group compared with those of the normal and the control groups. The level of MDA were significantly decreased in pancreas cell of 68 w-HY group compared with those of the normal and the control groups. The level of NO were significantly decreased in spleen cell of 68 w-HY group, pancreas cell of 52, 68w-HGD groups compared with those of the control groups. According to results, HY showed anti-oxidant effect. Investigation into the clinical use of the HY is suggested for future research.

마우스 내장 림프조직에서 우세하게 발현되는 IgA Isotype Switching 관련 전사체의 분석 (Preferential Expression of IgA Isotype Switching-associated Transcripts in Mouse Intestinal Lymphoid Tissues)

  • Background: Transforming growth factor-${\beta}$ (TGF-${\beta}1$) directs class switch recombination (CSR) to IgA isotype, which is a predominant antibody in mucosal surfaces. Although IgA is preferentially committed in mucosal lymphoid tissues, it is not definitely established whether hallmarks of IgA CSR such as IgA germ-line transcripts (GLT ${\alpha}$), post-switch transcripts (PST ${\alpha}$) and circle transcripts (CT ${\alpha}$) are readily expressed in such tissues. Therefore, we compared the expression of these transcripts among mouse Peyer's patches (PP), mesenteric lymph nodes (MLN), and spleen. Methods: Levels of GLTs, PSTs and CTs were measured by RT-PCR in isolated PPs, MLNs and spleen cells. Results: GLT ${\alpha}$ and PST ${\alpha}$ were well expressed in PP and MLN cells but in spleen cells. Similar patterns were observed in the expression of GL ${\gamma}$2b and PST ${\gamma}$2b. On the other hand, these transcripts were only inducible in spleen cells upon stimulated with LPS and TGF-${\beta}1$. In addition, CT${\alpha}$ and CT${\gamma}$2b were detected in PP cells. Conclusion: PP B cells readily express IgA GLT, PST, and CT. Overall expression patterns of these transcripts were similar in MLN cells. Thus, these results suggest that microenvironment of PP and MLN influences spontaneous IgA CSR, which lacks in systemic lymphoid tissues such as spleen.

어성초(漁腥草) 약침(藥鍼)이 B16흑색종(黑色腫) 암(癌)모델에 대한 항종양효과(抗腫瘍效果) 및 면역반응(免疫反應)에 미치는 영향(影響) (Experimental Study on the Effect of Houttuyniae Herbal Acupuncture on the Growth of Melanoma 816 in Mice)

  • Purpose ; In order to study the effect of Houttuyniae herbal acupuncture on growth of melanoma B 16 and immunity in mice, the Control group with normal saline acupunchure after subcutaneous inoculation of B16BL6 cells, the Sample I with Houttuyniae herbal acupuncture manufactured by distilled water extraction method after subcutaneous inoculation of B16BL6 cells and the Sample II with Houttuyniae herbal acupuncture manufactured by alcohol extraction method after subcutaneous inoculation of B16BL6 cells were divided. Methods ; To evauluate the effect of Houttuynias herbal acupuncture on growth of melanoma B16 in mice, the weight of mouse melanoma, body weight, spleen index( $\sqrt(Weight of spleen/Body weight){\times}100$), lymphocytes numbers in mouse peripheral blood and spleen tissue, the percentage of CD4+ T cells, CD8+ and the ratio of CD4+/CD8+ in mouse peripheral blood and spleen tissue are measured. Results ; In study of the weight of mouse melanoma, Houttuyniae herbal acupuncture groups(Sample I, Sample II) showed statistically significant inhibitory effect. And in study of effect of reduction of change in body weight of mouse, Sample I showed statistically significant inhibitory effect, too. In study of reduction of spleen index, lymphocytes numbers in mouse spleen tissue, and the percentage of CD4+ and the ratio of CD4+/CD8+ T cells in mouse peripheral blood, Houttuyniae herbal acupuncture groups(Sample I, Sample II ) showed with statistically significant inhibitory effect. Also, Sample I showed inhibitory effect of reduction of the percentage of CD8+ T cells in mouse peripheral blood and spleen tissue and Sample II showed inhibitory effect of reduction of the percentage of CD4+ and the ratio of CD4+/CD8+ T cells in mouse spleen tissue with statistical significance. Conclusions ; The inhibitory effect of reduction of lymphocytes numbers in mouse peripheral blood and IL-2 productivity, Houttuyniae herbal acupuncture groups(Sample I, Sample II) didn't show significant effect.

귀전우 Petroleum Ether 추출물의 세포독성 (Cytotoxicity of Petroleum Ether Extract of Euonymus alatus)

  • The purpose of this research was to investigate the effects of petroleum ether extract of Euonymus alatus (EAP) on the proliferation of human tumor cells. EAP inhibited the proliferation of HeLa, Hep G2, KHOS/NP and A431 cells. The cytotoxicity of doxorubicin on human tumor cells and Balb/c 3T3 cells were increased by the combination of EAP. EAP did not affect the proliferation of Balb/c 3T3 cells, mouse spleen cells and human lymphocytes. These results suggest that EAP has the cytotoxicity on human tumor cells without cytotoxicity on Balb/c 3T3 cells, mouse spleen cells and human lymphocytes, and increase the cytotoxicity of doxorubicin.
