Association of the ubiquitin specific peptidase 9X -linked and Afadin expression patterns with sexual maturation in boar testis

  • Closely correlated expression patterns between ubiquitin specific peptidase 9X-linked (USP9X) and adherens junction formation factor (Afadin) in mouse testis development suggests that Usp9x regulates the deubiquitination of Af-6 (also known as Afadin, AFDN), and subsequently, the cell adhesion dynamics during gametogenesis. However, this relationship has not yet been tested in other domestic animals. The study was examined the temporal and spatial expression patterns of porcine USP9X and AFDN from the pre-pubertal to adult stages using real time-PCR and immunohistochemistry. Furthermore, we detected the transcripts of USP9X and AFDN in the testis of 1-, 6- and 12-months old boar, respectively. USP9X and AFDN were found to have similar expressions patterns, with basal expression after 1 month followed by a significant up-regulation from 6 months (puberty) onwards. In addition, neither the AFDN or USP9X proteins were detected in spermatogenic cells but they were expressed in the leydig cells and sertoli cells. USP9X was detected around the basal lamina during pre-puberty, and predominantly expressed in the leydig cells at puberty. Finally, in adult testis, USP9X was increased at the sertoli cell-cell interface and the sertoli cell-spermatid interface. In summary, closely correlated expression patterns between USP9X and AFDN in boar testis supports the previous findings in mice. Furthermore, the junction connections between the sertoli cells may be regulated by the ubiquitination process mediated via USP9X.

  • To assess the testicular toxicity induced by DA-125, a new anthracycline anticancer agent, the test substance was intraveneously administered to male beagle dogs at dose levels of 0, 0.0023, 0.0375, 0.15, and 0.6 mg/kg/day, 6 days a week for 26 weeks. At 0.6 mg/kg/day, 1 out of 3 dogs had died on day 42 of treatment and the other dogs were sacrificed on days 46 and 122 of treatment due to the increasingly severe clinical condition. Clinical signs considered to be related to treatment were included anorexia, vomiting, salivation, decreased activity, mucous and/or dark faeces, diarrhea, and swelling, abscess and/or ulceration of injection sites. Suppression in body weight gain, reduction in food intake, decreases in testicular weight and size, and hemorrhage of epididymis were also observed in male dogs. Microscopically, severe degenerative changes such as atrophy of seminiferous tubules, loss of germ cells, degeneration of germ cells, vacuolization of Sertoli cells, and hyperplasia of Leydig cells were observed in all dogs. Azoospermia in epididymal tubules, atrophy of epithelia in the cauda epididymis, and prostate atrophy were also found. At 0.15 mg/kg/day, anorexia, vomiting, salivation, diarrhea, and swelling of injection sites were observed. In addition, suppression in body weight gain and decreases in testicular weight and size were found in male dogs. Atrophy of seminiferous tubules, decrease of germ cells, degeneration, exfoliation and retention of germ cells, vacuolization of Sertoli cells, and hyperplasia of Leydig cells were observed by histopathological examination. Azoospermia in epididymal tubules and prostate atrophy were also found. At 0.0375 mg/kg/day, there were no clinical signs considered to be indicative of a reaction to treatment, but testicular size was significantly reduced. Microscopically, decreases in the number of spermatogonia and epidydimal speramtozoa were found. There were no evidences of general or testicular toxicity at 0.0023 mg/kg/day. These results indicate that DA-125 produces significant and persistent damage to the spermatogenic compartments of the testes in male beagle dogs.

Gonadal Development and Reproductive Cycle of the Top Shell, Omphalius rusticus(Gastropoda: Trochidae)

  • Gonadal development, reproductive cycle, gonad index, meat weight rate, and first sexual maturity of the top shell, Omphalius rusticus were Investigated monthly by histological observations. Specimens were collected from the west coast of Korea during the period from January to December in 1999. O. rusticus is dioecious and oviparous. The gonad is widely situated on the surface of the digestive g1and located in the posterior spiral meat part in the shell. The ovary and the testis were composed of a number of oogenic follicles and several spermatogenic follicles, respectively. Ripe oocytes were approximately 120-130 $\mu$m in diameter. The meat weight rate peaked in June (27.7%), and then rapidly decreased in September (19.5%). Monthly changes in the gonad index in both sexes reached the maximum in June, and then sharply decreased in September. Percentages of first sexual maturity of female and male snails ranging from 9.0 to 9.9 mm in shell heights were 58.3% and 54.5%, respectively, and 100% in those over 11.0 mm in both sexes participated in reproduction. Reproductive cycle of this species can be categorized into five successive stages: in females, early active (October to April), late active (December to June), ripe (April to September), spawning (July to September) and recovery (September to January): in males, early active (November to March), late active (December to June), ripe (April to September), spawning (July to September) and recovery (September to December). Gonadal development, gametogenesis, reproductive cycle, and spawning were closely related to the seawater temperature.

Gametogenesis and Reproductive Cycle of the Rock Shell, Reishia (Thais) clavigera (Neogastropoda: Muricidae), on the West Coast of Korea

  • Gonadal development, gametogenesis, reproductive cycle, and first sexual maturity of Reishia clavigera were investigated monthly from July 1998 to June 1999 through cytological and histological observations. R. clavigera had separate sexes, and was an internal fertilizer. The ma1e penis was located near the two tentacles. The ovary and testis were composed of a great number of oogenic lobules and spermatogenic tubules, respectively. The size of ripe oocyte ranged from 130 to 140 ${\mu}$m in diameter. The peripheral cytoplasm of the germinal vesicle of the ripe oocyte in many cases were surrounded by smaller yolk granules, while the eccentric cytoplasm was occupied with larger ones. The reproductive cycle of R. clavigera could be classified into five successive stages: early active, late active, ripe, spawning, and recovery. Spawning of females occurred from early July to August when the seawater reached above 24.8$^{\circ}C$. Spawning of males occurred from early June to August in the water above 22.8$^{\circ}C$. Minimum size for sexual maturity of both sexes was above 10.0 mm in shell height. Each egg capsule was a cylinder or spindle in shape, 4-6 mm in length and 1-2 mm in width. Colors of newly spawned egg capsules showed yellowish white or pale yellow, while those with veliger larvae showed pale black, and released larvae or dead egg capsules showed black violet. The fecundity in an egg capsule ranged from 70 to 91 eggs (mean=80.28 eggs).

극지역 물개류의 번식생물학: 정자형성 (Reproductive Biology of the Seals in Polar Region: Spermatogenesis)

  • Abstract - Global decline in wildlife mammals has been accelerated during past decades. Especially the conservation the wild life mammals in polar areas, is urgent. In an effort to understand the reproduction of the seals dwelling in the polar area, spermatogenesis in the seals was reviewed. Seals breed seasonally and in most of the seal species, delayed implantation is frequently observed. To date, histological and endocrinological evaluation revealed highly cyclic nature in supermatogenesis and steroidogenesis in testis. Seasonal changes in blood testosterone level together with melatonin is closely related with changes in light cycle between summer and winter. In adult testis at breeding seasons, spermatogenesis is manifested by consecutive 18 stages of germ cell development. Three kinds of Leydig cells different in steroidogenic activity as well as cellular morphology appear during the testis development. During non-breeding season, spermatogenic arrest and Leydig cell hypoplasia are frequently found. Interestingly, blood circulation through the anastomoses of pelvic veins cooled the testes and thus guarantees spermatogenesis within the body trunk. Endocrine disruptors and heavy metals have been found in the body tissues of several seals species and alter steroidogenesis in seals, suggesting environmental pollutants together with decrease in habitats are potentially threatening the reproductive success in seal species.

Spermatogenic index and hormonal profile in the rats received chromatographic fractions of ethanol extract of Crotalaria juncea L. seeds

  • The ethanol extract of the Crotalaria juncea seeds, which showed promising antispermatogenic and antiandrogenic activities in albino mice, was taken up further for the isolation of the active fractions present in it. Two fractions that were obtained from thin layer chromatography were subjected for testing to know their antispermatogenic and antiandrogenic activities. After preliminary trials the fraction I showed maximum antifertility activity at the dose level of 200 mg/kg body weight when administered orally to the rats for 50 days. The fraction I was found to affect spermatogenesis as well as the endocrine functions of the testis as indicated by gravimetric, histopathological and biochemical changes. Further this fraction has caused degenerative changes in the seminiferous tubules and Leydig cells of the testis. The accessory reproductive organs like epididymis, seminal vesicles, vas deferens, prostrate, Cowper's gland and Levator Ani muscle showed significant malfunction. Cauda epididymal sperm count and sperm motility were reduced significantly. The treatment has also resulted in increase in the cholesterol level and alkaline phosphatase activity, and decrease in protein, glycogen, sialic acid contents and acid phosphatase activity in testis. It is noteworthy that RIA studies have shown significant reduction in serum FSH, LH and testosterone. Scanning electron microscopic observations revealed abnormalities in sperm structure.

Effects of a mild heat treatment on mouse testicular gene expression and sperm quality

  • The decrease in sperm quality under heat stress causes a great loss in animal husbandry production. In order to reveal the mechanism underlying the sperm quality decrease caused by heat stress, we first established a mild heat-treated mouse model. Then, the sperm quality was identified. Further, the testicular proteome profile was mapped and compared with the control using 2D electrophoresis and mass spectrometry. Finally, the differential expressed proteins involved in the heat stress response were identified by real-time PCR and Western blotting. The results showed that heat stress caused a significant reduction in mouse sperm quality (P<0.05). Further, 52 protein spots on the 2D gel were found to differ between the heat-shocked tissues and the control. Of these spots, some repair proteins which might provide some explanation for the influence on sperm quality were found. We then focused on Bag-1, Hsp40, Hsp60 and Hsp70, which were found to be differently expressed after heat shock (P<0.05). Further analysis in this heat-shocked model suggests numerous potential mechanisms for heat shock-induced spermatogenic disorders.

  • Spermatogenesis and sperm ultrastructure of the marsh clam (Corbicula japonica) were investigated by electron microscopic observations. Testis of the marsh clam consists of numerous spermatogenic follicle containing germ cells in the different developmental stages. Spermatogonia are located nearest the outer wall of the follicle, while spermtocytes and spermatids are positioned nearer to the lumen. Spematogonia are oval-shaped and about $5{\mu}m$ In diameter. Spermatogonia develop into spermatocyte, spermatid and spermatozoon. In the spermatid to about $2{\mu}m$ in diameter, cytoplasm decreases and mitochondria move to the base of the nucleus and fuse into several spheres, the centrioles become orthogonally oriented, a flagellum appears, and an acrosomal vesicle forms. The mature sperm has primitive type, consisting of a head, a midpiece and a tail. The sperm was arrow-shaped, and its head is about $8{\mu}m$ long and comprised of a long nucleus and an acrosome. The four mitochondria encircled the centrosome in midpiece. The flagellum had the classical 9+2 axoneme structure, and axonemal lateral fins in the tail were observed.

  • The aims of the study were to establish a short-term screening test for detecting testicular toxicity of chemicals in rats and to determine whether a 2-week administration period is sufficient to detect testicular toxicity of 2-bromopropane (2-BP) as an example. Male Sprague-Dawley rats were subcutaneously administered with 1000 mg/kg/day of 2-BP or its vehicle for 2 weeks. Ten male rats each were sacrificed on days 3, 7 and 14 after the initiation of treatment. Parameters of testicular toxicity included genital organ weights, testicular sperm head counts, epididymal sperm counts, motility and morphology, and qualitative and quantitative histopathologic examinations. The early histopathological changes observed on day 3 of treatment included degeneration of spermatogonia and spermatocytes, multinuclear giant cells, mature spermatid retention, vacuolization of Sertoli cells, and decreased number of spermatogonia in stages II and V. On day 7 of treatment, atrophy of seminiferous tubules, exfoliation of germ cells, degeneration of spermatogonia and spermatocytes, multinuclear giant cells, mature spermatid retention, vacuolization of Sertoli cells, decreased number of spermatogonia in stages II and V, and decreased number of spermatocytes in stages VII and XII. On day 14 after treatment, a significant decrease in the weights of testes and seminal vesicles was found. Atrophy of seminiferous tubules, exfoliation of germ cells, degeneration of spermatogonia and spermatocytes, mature spermatid retention, vacuolization of Sertoli cells, decreased number of spermatogonia in stages II and V, and decreased number of spermatocytes in all spermatogenic stages were also observed. In addition, a slight non-significant decrease in testicular sperm head counts, daily sperm production rate and epididymal sperm counts was found. The results showed that 2 weeks of treatment is sufficient to detect the adverse effects of 2-BP on male reproductive organs. It is considered that the short-term testicular toxicity study established in this study can be a useful tool for screening the testicular toxic potential of new drug candidates in rats.
