Taxonomic and Functional Changes of Bacterial Communities in the Rhizosphere of Kimchi Cabbage After Seed Bacterization with Proteus vulgaris JBLS202

  • Maintenance of a beneficial microbial community, especially in the rhizosphere, is indispensable for plant growth and agricultural sustainability. In this sense, plant growth-promoting rhizobacteria (PGPR) have been extensively studied for their role in plant growth promotion and disease resistance. However, the impact of introducing PGPR strains into rhizosphere microbial communities is still underexplored. We previously found that the Proteus vulgaris JBLS202 strain (JBLS202) promoted growth of Kimchi cabbage and altered the relative abundance of total bacteria and Pseudomonas spp. in the treated rhizosphere. To extend these findings, we used pyrosequencing to analyze the changes in bacterial communities in the rhizosphere of Kimchi cabbage after introduction of JBLS202. The alterations were also evaluated by taxon-specific realtime PCR (qPCR). The pyrosequencing data revealed an increase in total bacteria abundance, including specific groups such as Proteobacteria, Acidobacteria, and Actinobacteria, in the treated rhizosphere. Time-course qPCR analysis confirmed the increase in the abundance of Acidobacteria, Actinobacteria, Alphaproteobacteria, and Betaproteobacteria. Furthermore, genes involved in nitrogen cycling were upregulated by JBLS202 treatment indicating changes in ecological function of the rhizosphere soil. Overall, these results indicate that introduction of JBLS202 alters both the composition and function of the rhizosphere bacterial community, which can have direct and indirect effects on plant growth. Therefore, we propose that long-term changes in bacterial composition and community-level function need to be considered for practical use of PGPRs.

옥수수의 벼검은줄오갈병 (Occurrence of Rice black-streaked dwarf fijivirus in Maize)

  • 2005년 7월 전북고창 지역에서 옥수수에 줄무늬 위축증상을 나타내는 이병주를 채집하여 게놈 dsRNA 분리 및 RT-PCR 반응을 실시하였다. 게놈 dsRNA 분리는 이병엽 100mg에서 직접 dsRNA를 추출하여 agarose gel에서 전기영동 하였으며, 10개의 분절을 확인하였다. 추출한 dsRNA를 주형으로 하여 S7, S8, S10 full-length 특이적인 primer를 제작하여 RT-PCR을 실시하였다. 그 결과 각각의 분절에서 예상되는 크기의 밴드를 확인하였다. 고창지역의 사료용 옥수수 재배지에서 RBSDV의 발생면적은 약 22ha에 달하였으며 발생이 심한 곳은 약 80%의 이병율을 나타내기도 하였다.

경주국립공원 서악 지구의 관속식물상 (Vascular Plants of Seoak District in Gyeongju National Park)

  • The purpose of this study is to offer the raw data for establishing the management and restoration plan by objective surveying and analysing the flora distributed in Seoak District, Gyeongju National Park, Korea. The flora summarized as 411 taxa including 92 families, 285 genera, 363 species, 2 subspecies, 41 varieties and 5 forms. The rare plants designated by Korea Forest Service were 2 taxa including Exochorda serratifolia and Potentilla discolor. The Korean endemic plants were 3 taxa including Philadelphus schrenkii, Lespedeza maritima and Weigela subsessilis. The specific plants by floristic region were 16 taxa including Asplenium sarelii, Pyrrosia petiolosa, Vitex negundo var. incisa and so forth. The target plants adaptable to climate change were 2 taxa including Lespedeza maritima and Carpesium macrocephalum, and the plants with approval for delivering oversea were 4 taxa including Exochorda serratifolia, Glycine soja, Lespedeza maritima and Weigela subsessilis. The naturalized plants were 51 taxa including Phytolacca americana, Viola papilionacea, Lamium purpureum and so forth. The invasive alien plants were 3 taxa including Ambrosia artemisiifolia, Aster pilosus and Lactuca scariola. NI(Naturalized Index) was 12.4% of all 411 taxa of surveyed flora in this study and UI(Urbanized Index) was 15.9% of all 321 taxa of naturalized plants in Korea. Potentilla discolor in rare plant, Philadelphus schrenkii in endemic plant and Dictamnus dasycarpus in specific plant were established the conservation plan. Whereas, invasive alien plants such as Ambrosia artemisiifolia, Aster pilosus and Lactuca scariola should be removed as soon as possible. The damaged sites in Seoak District were divided between wildfire area and farmland. To restore a forest fire site, we will have to apply a natural renewal and community planting. In case of farmland, we will have to do ecological planting using native species and constrcut a forest wetland.

현사시나무(Populus alba × P. glandulosa)에서 분리한 non-specific Lipid Transfer Protein (ns-LTP) 프로모터의 특성 분석 (Characterization of a non-specific Lipid Transfer Protein (ns-LTP) promoter from poplar (Populus alba × P. glandulosa))

  • 나무의 유전 공학적 연구를 위해서는 목본 고유의 유전자 및 프로모터 연구가 필수적이다. 우리는 포플러(P. alba ${\times}$ P. glandulosa)의 Pagns-LTP 유전자의 867 bp 프로모터를 분리하였고, ${\beta}$-glucuronidase (GUS) reporter 유전자를 이용한 프로모터의 형질전환 포플러를 제작하여 특성 분석하였다. Pagns-LTP 유전자는 어린뿌리에서 강하게 발현되었고 어린잎에서는 약하게 발현되었으며, 그밖에 다른 조직에서는 발현되지 않았다. 또한, 프로모터의 활성은 뿌리와 어린잎에서 한정되었으며 어린뿌리의 세포 전체에서 강한 활성을 나타내었다. 이에 포플러 ns-LTP 프로모터 내의 cis-element를 조사하고 현사시나무에서 Pagns-LTP 프로모터를 분리한 후 활성을 분석하였다. 프로모터 내의 cis-element를 분석한 결과, 조직 특이적 발현과 호르몬 및 스트레스에 반응하는 다양한 cis-element가 존재함을 확인하였다. 이를 통해 포플러의 ns-LTP는 생장뿐만 아니라, 스트레스에도 관여할 것이라고 추측할 수 있었다. 본 연구는 목본의 유전자 기능 분석 및 다양한 응용 연구를 위해 유용하게 이용될 수 있는 도구로서의 가능성을 제시하였다.

철강 산업의 산업공정부문 CO2 실측 배출계수 산정에 관한 연구 (An Estimation of Plant Specific Emission Factors for CO2 in Iron and Steel Industry)

  • The development of domestic plant specific emission factors is very important to estimate reliable national emissions management. This study, for the reason, was carried out to obtain advances emission factor for Carbon Dioxide ($CO_2$) by source-specific emission tests from the iron and steel industry sector which is well known as one of the major sources of greenhouse gases ($CO_2$). Emission factors estimated in this study were compared with those of IPCC for evaluation and they were found to be of similar level in the case of $CO_2$. There was no good information available on $CO_2$ plant specific emission factors from the iron and steel industry in Korea so far. The major emission sources of $CO_2$ examined from the iron and steel manufacturing precesses were a hot blast stove, coke oven, sintering furnace, electric arc furnace, heating furnace, and so on. In this study, the concentration of $CO_2$ from the hot blast stove process was the highest among all processes. The $CO_2$ emission factors for a ton of Steel and Iron products (using B-C oil) were estimated to be 0.315 $CO_2$ tonne (by Tier 3 method) and 4.89 $CO_2$ tonne. In addition, emission factor of $CO_2$ for heating furnace process was the highest among all process. Emission factors estimated in this study were compared with those of IPCC for evaluation and they were found to be of similar level in the case of $CO_2$.

Managing Within-Field Spatial Yield Variation of Rice by Site-Specific Prescription of Panicle Nitrogen Fertilizer

  • Rice yield and protein content have been shown to be highly variable across paddy fields. In order to characterize this spatial variability of rice within a field, two-year experiments were conducted in 2002 and 2003 in a large-scale rice field of $6,600m^2$ In year 2004, an experiment was conducted to know if variable rate treatment (VRT) of N fertilizer, that was prescribed for site-specific management at panicle initiation stage, could reduce spatial variation in yield and protein content of rice while increasing yield compared to conventional uniform N topdressing (UN, 33kg N/ha at PIS) method. VRT nitrogen prescription for each grid was calculated based on the nitrogen (N) uptake (from panicle initiation to harvest) required for target rice protein content of $6.8\%$, natural soil N supply, and recovery of top-dressed N fertilizer. The required N uptake for target rice protein content was calculated from the equations to predict rice yield and protein content from plant growth parameters at panicle initiation stage (PIS) and N uptake from PIS to harvest. This model· equations were developed from the data obtained from the previous two-year experiments. The plant growth parameters for the calculation of the required N were predicted non-destructively by canopy reflectance measurement. Soil N supply for each grid was obtained from the experiment of year 2003, and N recovery was assumed to be $60\%$ according to the previous reports. The prescribed VRT N ranged from 0 to 110kg N/ha with an average of 57kg/ha that was higher than 33 kg/ha of UN. The results showed that VRT application successfully worked not only to reduce spatial variability of rice yield and protein content but also to increase rough rice yield by 960kg/ha. The coefficient of variation (CV) for rice yield and protein content was reduced significantly to $8.1\%$ and $7.1\%$ in VRT from $14.6\%$ and $13.0\%$ in UN, respectively. And also the average protein content of milled rice in VRT showed very similar value of target protein content of $6.8\%$. In conclusion the procedure used in this paper was believed to be reliable and promising method for reducing within-field spatial variability of rice yield and protein content. However, inexpensive, reliable, and fast estimation methods of natural N supply and plant growth and nutrition status should be prepared before this method could be practically used for site-specific crop management in large-scale rice field.

고추와 토마토 종자에서 종자전염 세균 및 바이러스의 동시 검출을 위한 One-step Multiplex RT-PCR 방법 (One-step Multiplex RT-PCR Method for Simultaneous Detection of Seed Transmissible Bacteria and Viruses in Pepper and Tomato Seeds)

  • 우리나라에서 주로 재배되는 가지과작물의 종자 전염 병원균 중에서 세균성 병원균 Xanthomonns campestris pv. vesicatoria(Xcv), Clavibacter michiganensis subsp. michiganensis (Cmm), Erwinia carotovora subsp. carotovora (Ecc)와 바이러스 병원균 Pepper mild mottle virus (PMMoV) and Tobacco mild green mosaic virus (TMGMV)를 종자로부터 동시검출하기 위한 One-step Multiplex RT-PCR을 개발하였다. 각각의 병원균을 특이적으로 증폭시키는 병원균 검출용 프라이머 15세트를 primer-blast 프로그램을 이용하여 제작하였고 각각의 병원균을 특이적으로 검출할 수 있는 5세트의 프라이머 세트(Cmm-F/R, Ecc-F/R, Xcv-F/R, PMMoV-F/R, TMGMVF/R)를 선발하였다. 이들 프라이머 세트는 프라이머간의 또는 병원균 cDNA간의 간섭없이 타겟 병원균만을 검출하였다. 가지과 작물 중에서 유통 중인 고추종자 20품종과 토마토종자 10품종에 대한 종자 감염 병원균 검출을 위한 PCR을 수행한 결과, 2개 품종의 토마토종자에서 Ecc가 검출되었고 4개 품종의 고추종자에서도 Ecc가 검출되었다. 특히 고추 종자에선 Ecc가 검출된 4개 품종 외에 1개 품종에서 PMMoV, TMGMV가 동시 검출되었다. 이로 인해 개발된 One-step multiplex RT-PCR은 서로 간섭 현상 없이 병원균을 효율적으로 검출할 수 있음을 보여 주었다.

식물 특정효소저해제의 생물활성 조사에 의한 신규제초제 작용점 탐색 (Searching of Possible Target Enzymes for Herbicide Development using Commercial Plant-Specific Inhibitors)

  • 본 연구는 새로운 제초제 후보물질을 탐색하기 위하여 식물특이적 효소 저해제로 알려진 107개 기존 화합물에 대하여 생물활성을 조사하였다. Germination test, seedling assay, wheat leaf disc assay, cyanobacteria assay, whole plant assay를 통하여 15종의 저해제를 선발하였고 이들은 34종 효소를 저해하는 것으로 확인되었다. 이들 화합물 중에서 phenylhydrazine, purine, o-phenanthroline, oleylamine, 7,8-benzoquinoline, aminooxyacetic acid, dicyclohexylcarbodiimide 등은 성체를 이용한 온실 실험에서 높은 제초활성을 나타내었다. 7,8-benzoquinone, 8-hydroxyquinoline, 2,2'-dipyridyl 및 o-phenanthroline 등은 피, 벼, 토마토의 발아를 $1.25{\sim}5{\mu}M$의 농도에서도 억제하였다. 7,8-benzoquinoline, cyanuric fluoride, 4-methylpyrazole, tranylcypromine, oleylamine과 trifluoperazine 등은 $30{\sim}100{\mu}M$ 농도에서 cyanobacteria의 생육을 저해하였다. Dicyclohexyl carbodiimide와 chlorpromazine은 $100{\mu}M$ 농도에서 wheat leaf disc의 백화현상을 유기시켰다. 이상과 같이 생물학적 활성을 갖는 식물 특이적 효소저해제들은 신규제초제 후보물질을 선발하기 위한 새로운 대상효소로 이용될 수 있을 것으로 생각된다.

Comparison of ecophysiological and leaf anatomical traits of native and invasive plant species

  • Background: To address the lack of evidence supporting invasion by three invasive plant species (Imperata cylindrica, Lantana camara, and Chromolaena odorata) in tropical ecosystems, we compared the ecophysiological and leaf anatomical traits of these three invasive alien species with those of species native to Sempu Island, Indonesia. Data on four plant traits were obtained from the TRY Plant Trait Database, and leaf anatomical traits were measured using transverse leaf sections. Results: Two ecophysiological traits including specific leaf area (SLA) and seed dry weight showed significant association with plant invasion in the Sempu Island Nature Reserve. Invasive species showed higher SLA and lower seed dry weight than non-invasive species. Moreover, invasive species showed superior leaf anatomical traits including sclerenchymatous tissue thickness, vascular bundle area, chlorophyll content, and bundle sheath area. Principal component analysis (PCA) showed that leaf anatomical traits strongly influenced with cumulative variances (100% in grass and 88.92% in shrubs), where I. cylindrica and C. odorata outperformed non-invasive species in these traits. Conclusions: These data suggest that the traits studied are important for plant invasiveness since ecophysiological traits influence of light capture, plant growth, and reproduction while leaf anatomical traits affect herbivory, photosynthetic assimilate transport, and photosynthetic activity.

Gui Valley: A High Yielding Potential and Good Processing Potato Cultivar

  • The main objective of this breeding program is to develop high yielding, disease resistance and good processing potato cultivar. 'Gui Valley' is a clonal selection resulting from a cross between 'ND2471-8' and 'Cona'. It has medium plant height and light green foliage. 'Gui Valley' has medium flowering habit and light pink flowers. 'Gui Valley' is a medium maturing potato cultivar and tubers are smooth, yellow skin, light yellow flesh, long-oval tuber shape, distinct red eyes with medium depth and medium dormancy. It has high level of tuber uniformity and good keeping quality. 'Gui Valley' demonstrates resistance to potato virus Y (PVY), soft rot, but moderately susceptible to late blight and common scab. It is also resistant to most of the internal and external physiological disorders particularly dehiscence, hollow heart and internal brown spot. The specific gravity of 'Gui Valley' is significantly higher (1.097) than that of 'Shepody' (1.078). 'Gui Valley' has suitable for processing mainly French fries and chips. This cultivar has high level of tuber uniformity and capable of yielding 37.6 $t{\cdot}ha^{-1}$, which is 18.2% higher than the control potato cultivar 'Shepody' under optimum agronomical practices.
