A rare case of bovine tuberculosis caused by Mycobacterium bovis in a domestic rabbit

  • A 12-month-old domestic rabbit died suddenly without specific clinical signs. Gross examination revealed irregular yellowish hepatic nodules with pus in the submandibular muscles, lungs, uterus, and small intestines. Histopathological examination of the liver showed granulomatous inflammation with acid-fast-positive bacteria. Mycobacterium bovis SB1040 was identified by polymerase chain reaction and spoligotyping, and Pasteurella multocida was isolated from the multiple lesions. This report demonstrates the pathological features of rare bovine tuberculosis (bTB) in a domestic rabbit, the first case in the Republic of Korea. To ensure public safety, we recommend routine monitoring of rabbits to control the incidence of bTB.

  • This paper describes the evaluation of noise influence for residental and boundary areas in 75MW thermal power plant. It includes the measurements of noise level around the boundary area of the plant, identification of noise propagating path, and discussion on the measures. Noise assessments are carried out based on the ISO 3744, ISO 9613-1 and ISO 9613-2 to predict the noise distribution of specific locations from the noise sources such as power transformers, flash vent-pipe, I.D.fan, and stack. It is identified the vent-pipe of flash tank in thermal power plant as the root cause of the environmental noise during the plant commissioning process.

Variability in the coat protein genes of two orchid viruses from Phlaenopsis orchids in Korea

  • This study was conducted to designing conserved regions of molecules for virus-derived resistance to transgenic Phlaenopsis orchids to protect against two major orchid viruses, Cymbidum mosaic virus (CymMV) and Odontoglossum ringspot virus (ORSV). Infected leaf samples of Phalaenopsis were randomly screened by the RT-PCR with specific primers to both of viruses. RT-PCR products of the viruses were cloned and their nucleotide sequences were determined. Multiple alignments of coat protein (CP) genes of the viruses revealed that over the 88 % and 94 % identities with CymMV and ORSV, respectively, were observed. These data can be useful for selection of highly conserved regions of CP gene of the viruses for transgenic orchid experiments.

배추 뿌리혹병균 Plasmodiophora brassicae의 종 특이적 프라이머 개발 (Development of Species-Specific Primers for Plasmodiophora brassicae, Clubroot Pathogen of Kimchi Cabbage)

  • Plasmodiophora brassicae는 십자화과 작물에 뿌리혹병을 일으키는 주요 병원균이다. 본 연구에서는 뿌리혹병균의 신속 정확한 검출을 위해서 뿌리혹병균에 대한 새로운 종 특이적 프라이머를 개발하고자 하였다. 새롭게 개발된 프라이머들은 10종의 주요 토양병원균을 비롯하여 기주인 배추 DNA와는 반응하지 않고 P. brassicae와만 반응하는 특이성을 갖고 있었다. 그 가운데 Primer ITS1-1/1-2는 민감도 검정 결과, 10 spores/ml의 DNA까지 검출이 가능함으로써, first round PCR용임에도 불구하고 이전의 검출법 보다 감도가 높고 정확한 결과를 얻었다. Quantitative real-time PCR로 분석할 경우에는 더 적은 수의 포자까지 안정적으로 검출해 낼 수 있어 새로운 P. brassicae 종 특이적 프라이머로서의 유용성을 확인할 수 있었다.

Development of New Molecular Markers for the Identification of Male Sterile Cytoplasm in Peppers (Capsicum annuum L.)

  • Cytoplasmic male sterility (CMS) induced by mutant mitochondria genome, has been used for commercial seed production of $F_1$ hybrid cultivars in diverse crops. In pepper (Capsicum annuum L.), two sterile cytoplasm specific gene organization, atp6-2 and coxII were identified. An open reading frame, orf456 nearby coxII gene has been speculated to induce male sterility (MS) by mutagenic analysis. Moreover, molecular markers for atp6-2 and coxII of mitochondrial genotype (mitotype) were developed. However, the Cytoplasmic MS specific markers, atp6SCAR and coxIISCAR markers appeared in both N and S cytoplasms when polymerase chain reaction (PCR) cycles prolonged more than 40 cycles. Since the reported molecular markers were dominant markers, the presence of the faint sterile-specific band in normal cytoplasm may lead to the mis-classification of pepper breeding lines. To solve this problem, one common forward primer and two different reverse primers specific to normal coxII and sterile orf456 genes were designed after analyzing their gene organizations. By using these three primers, N and S coxII specific bands were co-amplified in male-sterile lines, but only normal coxII specific band was amplified in maintainer lines. Since the reverse primer for sterile coxII was specifically designed 275 bp downstream of orf456, relatively stable PCR amplification patterns were observed regardless of the number of PCR cycles. These primer sets easily identified different mitotypes among the divergent breeding lines, commercial cultivars and diverse germplasms.

Antioxidant Systems of Plant Pathogenic Fungi: Functions in Oxidative Stress Response and Their Regulatory Mechanisms

  • During the infection process, plant pathogenic fungi encounter plant-derived oxidative stress, and an appropriate response to this stress is crucial to their survival and establishment of the disease. Plant pathogenic fungi have evolved several mechanisms to eliminate oxidants from the external environment and maintain cellular redox homeostasis. When oxidative stress is perceived, various signaling transduction pathways are triggered and activate the downstream genes responsible for the oxidative stress response. Despite extensive research on antioxidant systems and their regulatory mechanisms in plant pathogenic fungi, the specific functions of individual antioxidants and their impacts on pathogenicity have not recently been systematically summarized. Therefore, our objective is to consolidate previous research on the antioxidant systems of plant pathogenic fungi. In this review, we explore the plant immune responses during fungal infection, with a focus on the generation and function of reactive oxygen species. Furthermore, we delve into the three antioxidant systems, summarizing their functions and regulatory mechanisms involved in oxidative stress response. This comprehensive review provides an integrated overview of the antioxidant mechanisms within plant pathogenic fungi, revealing how the oxidative stress response contributes to their pathogenicity.

백련초 분말을 첨가해 제조한 반죽형 케이크와 쿠키의 품질 특성 (Effect of Angelica plant powder on the Quality Characteristics of Batter Cakes and Cookies)

  • 백련초 분말의 기능성을 지닌 케이크와 쿠키의 제조를 위해서 밀가루의 1%, 3%, 5%를 백련초 분말로 대체한 후에 케이크와 쿠키의 물리적, 관능적 특성을 평가하였다. 백련초 분말의 첨가로 케이크의 비체적은 유의적으로 증가하였고, 경도는 감소하였으나 유의적 차이는 없었다. 쿠키의 비체적은 대조군과 첨가군 간에 차이가 없었고 반죽내 수분함량에 의해 결정되는 퍼짐성은 첨가량이 많을수록 낮은 값을 보였다. 케이크의 껍질과 케이크 내부 색도 측정 결과, 첨가량이 증가할수록 L값은 낮아졌고, a값과 b값은 증가하였다. 쿠키의 색도 변화 역시 케이크와 동일한 양상이었는데 특히, a값은 시료간 큰 유의성을 보였다. 관능검사 결과, 대조군에 비해 백련초 분말을 첨가한 케이크의 풍미, 색깔, 조직감 등에 대한 평가가 높았고 전체적인 기호도에 대한 평가는 1% 첨가군이 가장 높은 값을 나타내었다. 쿠키의 관능검사에서는 대조군에 비해 맛, 풍미, 색깔, 조직감 등에서 백련초 분말의 첨가량이 증가할수록 높은 값을 나타내었고, 전체적인 기호도 평가 결과는 3% 첨가군이 가장 높은 선호도를 보였다.

형질전환된 식물세포에서 고정화 방법을 통한 hCM-CSF의 생산성 증대 연구 (Enhanced Production of hGM-CSF by Immobilized Transgenic Plant Cell Cultures)

  • Plant cell immobilization can protect plant cells from shear forces and increase the stability of gene. An additional advantage of immobilization is the easiness for performing continuous culture with cell recycling. Therefore plant cell immobilization can overcome the limitations of plant cell applications. In addition, target protein should be selected from pharmaceutical proteins to get rid of low expression level problem. The enhanced production of human granulocyte-macrophage colony-stimulating factor (hGM-CSF) was investigated in immobilized Nicotiana tabacum suspension cell cultures. When the cells were immobilized in polyurethane foam, specific production of hGM-CSF was higher than that in alginate bead immobilization. Optimum continuous culture condition was the addition of 60 g/L sucrose in growth media with exchanging media every 6 day. Under the same condition, specific hGM-CSF production was 7 times higher in a 500-mL spinner flask than that in 100-mL Erlenmeyer flasks. Therefore, development of an effective immobilization process would be possible when the advantage of easy cell recycling was used. Consequently, enhanced production of target proteins could be possible in immobilized continuous cultures when the advantages of immobilization were applied.

Identification of Subspecies-specific STS Markers and Their Association with Segregation Distortion in Rice(Oryza sativa L.)

  • Two subspecies, japonica and indica, have been reported in rice, which differ in several ecotypic traits. However, reproductive barriers in hybrid progenies between subspecies have been major obstacles in breeding programs using inter-subspecific hybridization. As the first step to elucidate the reproductive barriers, we developed subspecies-specific(SS) STS markers in this study. A total of 765 STS primers were designed through comparing DNA sequences at every $2{\sim}3$cM interval between japonica and indica rices, which are available at Web DBs such as IRGSP, NCBI, TIGR, and GRAMENE, and tested for subspecies-specificity using 15 indica and 15 japonica varieties of diverse origin. Of them, 67 STS markers were identified as SS STS markers and their subspecies-specificity scores were estimated. The SS markers were dispersed throughout the genome along chromosomes. Of them, 64 SS markers were mapped on an RIL population derived from a Dasanbyeo(indica)/TR22183(japonica) cross. Genomic inclination of RILs was evaluated based on the genotyping with different types of markers. Association test between markers and segregation distortion revealed that segregation distortion might not be the cause of generating SS markers. The SS markers will be applicable to estimate the genomic inclination of varieties or lines and to study the differentiation of indica and japonica, and ultimately to breed true hybrid rice varieties in which desirable characters from both subspecies are recombined.
