Dudleya brittonii extract promotes survival rate and M2-like metabolic change in porcine 3D4/31 alveolar macrophages

  • Objective: Although alveolar macrophages play a key role in the respiratory immunity of livestock, studies on the mechanism of differentiation and survival of alveolar macrophages are lacking. Therefore, we undertook to investigate changes in the lipid metabolism and survival rate, using 3D4/31 macrophages and Dudleya brittonii which has been used as a traditional asthma treatment. Methods: 3D4/31 macrophages were used as the in vitro porcine alveolar macrophages model. The cells were activated by exposure to phorbol 12-myristate 13-acetate (PMA). Dudleya brittonii extraction was performed with distilled water. For evaluating the cell survival rate, we performed the water-soluble tetrazolium salt cell viability assay and growth curve analysis. To confirm cell death, cell cycle and intracellular reactive oxygen species (ROS) levels were measured using flow cytometric analysis by applying fluorescence dye dichlorofluorescein diacetate and propidium iodide. Furthermore, we also evaluated cellular lipid accumulation with oil red O staining, and fatty acid synthesis related genes expression levels using quantitative polymerase chain reaction (qPCR) with SYBR green dye. Glycolysis, fatty acid oxidation, and tricarboxylic acid (TCA) cycle related gene expression levels were measured using qPCR after exposure to Dudleya brittonii extract (DB) for 12 h. Results: The ROS production and cell death were induced by PMA treatment, and exposure to DB reduced the PMA induced downregulation of cell survival. The PMA and DB treatments upregulated the lipid accumulation, with corresponding increase in the acetyl-CoA carboxylase alpha, fatty acid synthase mRNA expressions. DB-PMA co-treatment reduced the glycolysis genes expression, but increased the expressions of fatty acid oxidation and TCA cycle genes. Conclusion: This study provides new insights and directions for further research relating to the immunity of porcine respiratory system, by employing a model based on alveolar macrophages and natural materials.

  • 극단의 土壤水分( 4 %, 24% )과 溫度(33$^{\circ}C$, -2$^{\circ}C$)에서 뿌리혹線筮의 生存率을 조사한 결과 土壤水分 함량에 따른 차이는 인정되지 않았고, 溫度 處理의 경우 33$^{\circ}C$에서는 Meloidogyne incognita, -2$^{\circ}C$에서는 M. hapla의 생존율이 높았으며, M. arenaria는 兩 處理에서 양자의 중간 정도의 반응을 보였다. 低溫처리에서 tolerance의 증가는 모든 線筮에서 인정되었으며 특히 M. hapla의 tolerance가 가장 높았다.

구리, 카드뮴, 펜벤다졸, 설파티아졸이 국내산 풍년새우 생존에 미치는 영향 (Effects of Cu, Cd, Fenbendazole and Sulfathiazole on the Survival of the Korean Fairy Shrimp Branchinella kugenumaensis)

  • The short term (24-hr) and long term (21 days) effects of copper, cadmium, fenbendazole and sulfathiazole on the survival of the Korean fairy shrimp Branchinella kugenumaensis were evaluated. The 24-hr median lethal concentrations ($LC_{50}$) of copper, cadmium, fenbendazole, and sulfathiazole were 39, 512, 182, and 31,818 ${\mu}g/L$, respectively. The toxicity of copper is highest among 4 chemicals used in this study, while sulfathazole the lowest. After the long term (21 days) exposure experiment, the $LC_{50}$ copper, cadmium, fenbendazole, and sulfathiazole were 1.12, 2.1, 0.1, 6.6 ${\mu}g/L$, respectively. The long term effects of antibiotics were highly enhanced while the short-term effects were not strong. The sensitivities of B. kugenumaensis to copper and cadmium were higher than or comparable to those of other freshwater branchiopods (Streptocephalus spp., Thamnocephalus sp.), and far higher than the marine species (Artemia sp.). There were significant effects on the survival of B. kugenumaensis after long term exposure to relatively lower concentrations of copper, cadmium, fenbendazole and sulfathiazole. Therefore, B. kugenumaensis seems quite a good candidate species for the ecotoxicological assessments of freshwater environments.

바위털갯지렁이(Marphysa sanguinea) 유생의 착저와 초기 성장에 미치는 기질 크기 및 유기물 함량 (Effects of Substrate Sizes and Organic Contents on Larval Settlement and Growth in the Early Stage of the Polychaete Marphysa sanguinea)

  • Marphysa sanguinea is the most well-known polychaete species with a high economic value. However, this species has a high mortality in the early rearing stage of aquaculture. This study was conducted to find out the optimal substrate size and organic contents for the growth and survival rate of M. sanguinea larvae and juveniles. It was observed that the smaller grain size (<Ø 0.063 mm) and high organic contents (5-10%) induced settlement and reduced settlement time of larvae. Moreover, the growth and survival rate of larvae reached high levels at Ø 0.004-0.016 mm of grain sizes and 5-7.5% of organic contents as advantageous substrates for settlement. The survival rate of juveniles reached over 90% in less than Ø 0.016 mm substrate on 15-day experiment for different grain sizes of substrates. These results indicated that substrate compositions of less than Ø 0.016 mm of sand size and 5-7.5% of organic contents in mud will enhance the productivity of M. sanguinea at the early stage.

Potential for artificial symbiosis between marine microalgae and invertebrates: I. survival of marine microalgae injected into the medusa of the moon jellyfish Aurelia aurita

  • Some marine microalgae and cyanobacteria form mutualistic symbioses with diverse invertebrates, particularly cnidarians. Among microalgae, dinoflagellates in the family Symbiodiniaceae are the most well-known symbiotic partners of jellyfish and corals. However, the symbioses involving other dinoflagellate families, nano- and micro-flagellates, diatoms, and cyanobacteria with cnidarians are not well understood. As an initial step, it is essential to explore the survival of these microorganisms inside cnidarians. We monitored the survival of eight microalgal species (nine strains) and one cyanobacterium species every day for seven days after injecting each into the medusa of the moon jellyfish Aurelia aurita. The dinoflagellates Effrenium voratum (free-living [FL] and living-in-coral strains), Cladocopium infistulum, Prorocentrum cordatum, Prorocentrum koreanum, Symbiodinium microadriaticum, the prasinophyte Tetraselmis suecica, the chlorophyte Dunaliella salina, and the raphidophyte Heterosigma akashiwo survived inside the medusa, while the cyanobacterium Synechococcus sp. was not detected. Additionally, E. voratum (FL) survived within the medusa for 60 days and gradually spread to adjacent areas, indicating potential for artificially established symbiosis. The results of this study provide a basis for artificial symbiosis between microalgae and invertebrates.

국내산저서단각류 Mandibulophoxus mai와 Monocorophium acherusicum의 생존, 성장 및 민감도에 대한 온도의 영향 (Influence of Temperature on the Survival, Growth and Sensitivity of Benthic Amphipods, Mandibulophoxus mai and Monocorophium acherusicum)

  • 본 연구는 최근 한국에서 퇴적물 독성 실험생물로 개발되고 있는 저서 단각류 Mandibulophoxus mai와 Monocorophium acherusicum의 생존과 성장 및 민감도(sensitivity)에 대한 온도의 영향를 평가하기 위한 일련의 실험으로 수행되었다. 본 연구에 사용된 각 실험 단각류의 생물학적 영향으로 각기 다른 수온 조건에서의 생존과 성장률을 통해 결정하였다. 표준 독성물질인 카트뮴(Cd)을 평가하는데 있어서 독성 민감도에 대한 온도의 영향은 각기 다른 온도에서 카드뮴이 첨가되지 않은 해수와 카드뮴이 첨가된 해수에 노출된 단각류의 생존률을 비교하여 결정하였다. 연구결과, 온도는 두 실험 단각규 모두의 생존, 성장 및 카드뮴 민감도에 영향을 미치는 것으로 평가되었다(p<0.05). 두 실험종 모두 80%이상의 생존율을 보인 온도범위는 13-22℃인 것으로 확인되었다. 두 실험종의 일일 성장률은 M. mai의 경우 20℃, M. acherusicum의 경우 25℃에서 다른 온도 조건에 비해 상대적으로 높게 나타났다. 반면, Cd 민감노에 대한 영향은 M. mai는 20℃, M. acherusicum은 15℃가 다른 온도 조건에 비해 상대적으로 큰 것으로 평가되었다. 이상의 연구 결과에 근거할 때, 온도는 단각규를 이용한 치사 및 반치사 생물 검정 연구에 있어 매우 중요한 변수인 것으로 판단되었다. 따라서, 독성평가에 활용되는 생물종에 대한 적절한 실험 온도 범위를 결정하는 것은 표준 독성시험법을 개발하는데 우선적으로 고려되어야 학 사항인 것으로 사료된다.

트롤 탈출어류의 생존율 연구 (Study on the survival rate of fishes escaped from trawl net)

  • The aim of this study was to develope a fishing technology for fisheries management for improving the survival rate of young fishes escaped from trawl net. Sea experiments were carried out to investigate the survival rate of the fishes in off Geomun island and Korean southern sea. Cover-net fish cage was designed and manufactured to collect fishes escaped from COD(codend) and TED(trawl escapement device). Fish cage was installed in a shallow site where scuba diver can observe the surviving fishes after separating the cage from trawl net. Most of fishes except horse mackerel(Trachurus japonicus) and mackerel(Scomber japonicus) could escaped through TED(bar space 35mm) easier than COD(mesh size 54mm). Especially, escapement rate of redlip croaker(Pseudosciaena polyactis) escaped from TED was the highest as 73.9%. Survival rate of horse mackerel(Trachurus japonicus) escaped from TED was 85.5% at the point of 96 hours over, while the survival rate for COD showed a rapid decrease according to the time elapse as 85.0% at the point of 24 hours, 76.4% for 48 hours, 62.7% for 72 hours and 56.1% for 96 hours. Over 90% of red seabream(Pagrus major) and rock bream(Oplegnathus fasciatus) escaped from TED were survived 96 hours over and the survival rate of those fishes were excellent compared with the other fish species. All korean pomfret(Pampus echinogaster) escaped from both COD and TED were died within 48 hours. Most of redlip croaker(Pseudosciaena polyactis) were also died within 12 hours because of the weak physiological characteristics by water pressure change. Survival rate of fishes like as horse mackerel(Trachurus japonicus) having hard skin was high while it was low for the fishes like as squid(Todarodes pacificus) and hair-tail(Trichiurus lepturus) having soft skin. We could know that there were big differences in the survival rate offish species escaped from trawl net.

재강슬래그 골재의 TTP 적용성 검토를 위한 연구 (Evaluation of TTP Applicability of Steel Slag Aggregate)

  • In order to evaluate the applicability of steel slag aggregates for tetrapod concrete, the properties of concrete as structural material were investigated. The biochemical research of marine concrete using steel slag aggregates was also carried out. The tested concrete properties are slump, air content, compressive strength, splitting tensile strength, elastic modulus, carbonation, hydration heat, freezing and thawing, sulfate attack, drying shrinkage, etc. The biochemical experiments are carried to research the propagation and reproduction of seaweeds and survival of bottom dwelling species. According to the experimental results, the steel slag aggregate content did not have a significant effect on compressive strength, splitting tensile strength and elastic modulus. The durability of concrete was not influenced by the steel slag aggregate content. From the biochemical research, steel slag aggregate can be evaluated as the material that is ideally suited for promoting propagation and reproduction of seaweeds and survival of bottom dwelling species.

Genistein이 햄스터 난소세포의 항산화효소활성과 발현에 미치는 영향 (Effect of Genistein on Activity and Expression of Antioxidant Enzyme in Hamster ovary cells)

  • Reactive oxygen species (ROS) are produced in the metabolic process of oxygen in cells. The superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GPx) in cells systemize the antioxidant enzymes to control the oxidative stress. Genistein is one of the isoflavonoids, and its role in controlling cellular oxidative stress is presently the active issue at question. In this study; we analyzed genistein-induced survival rates of the CHO-K1 cells, activities of antioxidant enzymes, ROS levels, and expression levels of antioxidant enzyme genes in order to investigate the effect of genistein on cellular ROS production and antioxidative systems in CHO-K1 cells. As results, the survival rate of cells was decreased as the dose of genistein increases (12.5${\sim}$200 ${\mu}$M). Genistein increased cellular ROS levels, while it reduced total SOD activities and the expression of CuZnSOD. In conclusion, we suggest that genistein may induce oxidative stress via down-regulation of SOD.

김치 주.부재료의 활성산소에 대한 피부 세포독성 완화효과 (Effect of Kimchi Ingredients to Reactive Oxygen Species in Skin Cell Cytotoxicity)

  • Kimchi showed protective effect from oxidative damage generated by hydrogen peroxide and paraquat. To investigate the major components of kimchi which reduce the cytotoxicity against reactive oxygen species, keratinocyte(A431, epidermoid carcinoma, human) and fibroblast(CCD-986SK, normal control, human) were cultured under oxidative stress condition provoked by paraquat, a superoxide anion generator, and hydrogen peroxide in the absence or presence of kimchi ingredients. Most keratinocyte and fibroblast cells were killed by hydrogen peroxide and paraquat over 1mM concentration, but kimchi ingredients showed protective effects from oxidative damage generated by hydrogen peroxide and onion, among those, garlic showed the most remarkable preventive effect. Most of kimchi ingredients showed protective effect against paraquat, especially leek notably increased cell survival. For fibroblast cells, ginger had the preventive effect against paraquat, especially leek notably increased cell survival. For fibroblast cells, ginger had the preventive effect from cell killing by high dose of hydrogen peroxide, but most ingredients were not effective against paraquat.
