Molecular Discrimination of Dinoflagellates Cochlodinium Polykrikoides Margalef, Gyrodinium Impudicum Fraga et Bravo and Gymnodinium Catenatum Graham using RAPD-PCR Method (RAPD-PCR 방법을 이용한 Cochlodinium polykrikoides Gyrodinium impudicum, Gymnodinium catenatum의 분자생물학적 진단)

  • Randomly amplified polymorphic DNA (RAPD) analysis was used to study genetic relationships among C. polykrikoides, G. impudicum and G. catenatum, which possess similar morphological features. Four of 12 primers were selected and 59 amplification products ranged from 0.2 kb to 3.0 kb. The number of polymorphic products in C. polykrikoides, G. impudicum and G. catenatum was 16 (27.1%), 8 (13.5%), and 16 (27.1%), respectively, while 17 were monomorphic. Number of species-specific bounds was 26 (44.0%), 34 (57.6%), 26 (44.0%) in C. polykrikoides, G. impudicum and G. catenatum, respectively. The genetic similarity between C. polykrikoides and G. impudicum/G. catenatum was 0.83, whereas G. impudicum and G. catenatum was 0.78. Our results suggest that C. polykrikoides, G. impudicum and G. catenatum are extremely different on the basis of RAPD analysis, despite similarity based on their morphology. The RAPD technique appears to be efficient in detecting genetic variation in these dinoflagellates.

  • The genus Tilia is characterized by linear form bracts of which the lower part is attached to the peduncle of a cyme. This character is distinguished from the others genus of Malvaceae. The purpose of this study is verifying the phylogenetic relationship of genus Tilia. Phylogenetic analyses were conducted to evaluate relationships of 10 taxa of Tilia in Korea and Japan including one outgroup (Gossypium hirsutum). The molecular phylogenetic analyses were conducted with sequences based on ITS, trnL-F and rpl32-trnL region. The combined data result of ITS, trnL-F and rpl32-trnL was formed by 6 clades. T. kiusiana situated as the most basal clade. T. amurensis, T. taquetii and T. rufa are composed a clade. T. koreana, T. insularis and T. japonica was formed independent clade. T. insularis has the closest relationship with T. japonica. T. miqueliana, T. mandshurica, and T. megaphylla are composed a clade and showed a sister relationship than other species.

  • In fast grown softwood, there are very large changes in material properties going outward from the pith to bark such as anatomical, physical and mechanical characteristics. Some of variations in anatomical properties with annual ring were then examined from Pinus koraiensis, Larix leptolepis, and Chamaecyparis obtusa, which are major softwoods of plantation in Korea. The large variations of annual ring width during young age of tree tended to stabilize after 25year through the transitional period in 17~23year. The ring density was 1.5~2.4 in 1~10year period, and 3.5~6.3 in 30~35year period, in which juvenile and mature wood were certainly assumed to be formed, respectively. Variations of tracheid length showed functional relationships with annual rings as logarithm. Demarcation between juvenile wood and mature wood could be 16~19year, which was determined from increase rate of tracheid length of 0.2%. Cell wall thickness increased with increase of annual ring even though large variations were observed as well. Variations of cell wall thickness within species were pronounced in latewood than earlywood. The increase of cell wall thickness from juvenile wood to mature wood was predominant in Larix leptolepis as 2.0times, and least in Chamaecyparis obtusa as 1.1 times. Cell diameters showed trends of increase during young age of 1~15year, and consistent afterward. The variations of cell diameter between radial and tangential direction were greater in latewood, and most pronounced in Chamaecyparis obtusa.

  • Most intracellular reactive oxygen species (ROS), especially superoxide anion ($O_2^{{\bullet}_-}$) that is converted from oxygen, are overproduced by excessive electron leakage from the mitochondrial respiratory chain. Intracellular oxidative stress that damages cellular components can contribute to lifestyle-related diseases such as diabetes and arteriosclerosis, and age-related diseases such as cancer and neuronal degenerative diseases. We have previously demonstrated that the excessive mitochondrial $O_2^{{\bullet}_-}$ production caused by SDHC mutations (G71E in C. elegans, I71E in Drosophila and V69E in mouse) results in premature death in C. elegans and Drosophila, cancer in mouse embryonic fibroblast cells and infertility in transgenic mice. SDHC is a subunit of mitochondrial complex II. In humans, it has been reported that mutations in SDHB, SDHC or SDHD often result in inherited head and neck paragangliomas (PGLs). Recently, we established Tet-mev-1 conditional transgenic mice using our uniquely developed Tet-On/Off system, which equilibrates transgene expression to endogenous levels. These mice experienced mitochondrial respiratory chain dysfunction that resulted in $O_2^{{\bullet}_-}$ overproduction. The mitochondrial oxidative stress caused excessive apoptosis leading to low birth weight and growth retardation in the neonatal developmental phase in Tet-mev-1 mice. Here, we briefly describe the relationships between mitochondrial $O_2^{{\bullet}_-}$ and aging phenomena in mev-1 animal models

Development of Bovine Specific Leptin Radioimmunoassay and Relationship of Plasma Leptin with Vitamin A and Age of Wagyu

  • Leptin is produced by adipocytes and its role in the regulation of lipid metabolism, feed intake, productive and reproductive performance of domestic animal species has been greatly stressed and extensively investigated in recent years. This study was conducted to develop a radioimmunoassay (RIA) for the estimation of plasma bovine leptin and to determine plasma leptin concentration in fattening Japanese Black cattle (Wagyu) and its crossbreds at commercial farms. Relationships of plasma leptin with plasma vitamin A and age of crossbred cattle were also determined. Recombinant bovine leptin (rbleptin) was produced by the E. coli overexpressed leptin as a GST (glutathione S-transferase)-fusion protein. Then antiserum against bovine leptin was obtained by its immunization in rabbits. Using this antiserum, a bovine specific RIA was developed and plasma leptin level was determined in 120 crossbred fattening cattle (WagyuHolstein, 50:50) at commercial farms. The plasma leptin level increased with the age of cattle and its level was greater in the crossbred heifers than in the steers. Plasma vitamin A level was negatively correlated with plasma leptin level in crossbred heifers and steers. This relationship was stronger in heifers than in steers. Plasma leptin was gradually increased with advancing age in fattening Wagyu cattle. In conclusion, development of a bovine specific RIA to estimate plasma leptin will contribute to better understanding of the role of leptin in cattle.

Comparison of In situ Dry Matter Degradation with In vitro Gas Production of Oak Leaves Supplemented with or without Polyethylene Glycol (PEG)

  • Dry matter (DM) degradation of leaves from Quercus cercis, Quercus libari, Quercus branti, and Quercus coccifera was determined using two different techniques: (i) in vitro gas production and (ii) the nylon bag degradability technique. In vitro gas production in the presence or absence of PEG and in situ DM disappearance were measured at 3, 6, 12, 24, 48, 72 and 96 h. In situ and in vitro DM degradation kinetics were described using the equation y = a+b ($1-e^{-ct}$). At all incubation times leaves from Quercus branti incubated with or without PEG gave significantly higher gas production than the other oak leaves except for 3 and 6 h incubation when leaves from Quercus branti without PEG supplementation only gave higher gas production than Quercus cercis and Quercus coccifera. At all incubation times except at 3, 6 and 12 h the DM disappearance from Quercus branti was significantly higher than the other species. Generally, PEG supplementation considerably increased the gas production at all incubation times and estimated parameters such as gas production rate ($c_{gas}$), gas production (ml) from the quickly soluble fraction ($a_{gas}$), gas production (b) from the insoluble fraction, potential gas production (a+b). However, all oak leaves did not give the same response to the PEG supplementation. Although the increase in gas production at 96 h incubation time was 8.9 ml for Quercus libari the increase was 5.5 ml for Quercus coccifera. It was concluded that except at early incubation times the relationships between the two methodologies seem to be sufficiently strong to predict degradability parameters from gas production parameters obtained in the presence or absence of PEG.

Artificial Inoculation of Inonotus obliquus on Betula platyphylla var. japonica (자작나무를 이용한 차가버섯균의 인공접종)

  • Inonotus obliquus could be isolated from Betula platyphylla var. japonica with diameter in the range of 6$\sim$13 cm that artificially inoculated by the fungus. The diameter and/or inoculation point of tree did not show any significant relationships with the infection rate of the fungus. Inonotus obliquus showed rapid growth on vertical direction of the infected tree while the growth was quite low on radial direction. The isolated fungus from the infected tree did not show vegetative incompatibility with the original fungus used for inoculation. We could isolate 8 contaminants from the inoculated area; Trichoderma reesei, T. atroviride, Cryptococcus neoformans, Botrytis cinerea, Fusarium sp. and 3 unknown species.

  • This study was carried out to clarifyed a taxonomical relationships of Rutaceae on the subfamily, genus and species level in Korea using venation pattern and stomata types. The results are summarized as follows : The leaf venation of the Rutaceae has revealed three major venation types. Semicraspedodromous, Brochidodromous and Cladodromous. The leaf ufmations type of the genus Evodia and Phellodendron were similar to Cladodromous type. They showed gradual evolution from brochidodromous type of Aurantioideae to cladodromous type of Toddaloideae. The stomata ypes were reconized Anomocytic and Actinocytic type, and epidermal cells were Ansio. Polygonal and Medium type. In the correlation analysis, there were highly significant correlation between frequency and length (r=-0.822), frequency and width (r=-0.780), and length and width(r=0.909) at 1% level.

Randomly Amplified Polymorphic DNA Analysis of Listeria Species Isolated from Foods in Korea (국내 식품으로부터 분리한 Listeria Species의 RAPD 분석)

  • This study was carried out for comparing Listeria strains developing genetic markers for Listeroa strains using Listeria sp. genetic markers using Randomly Amlymorphic DNA (RAPD) analysis method. Five of RAPD promers (OPA-01, OP-26-01, OP-26-02, OPB-01, OP-26-10) showed the distinctive polymorphism among Kisteria sp. isolated from domestic foods. RAPD-PCR with five arbitrary primers produced 76 DNA polymorphism. Among them, OPA-01 and OP-26-01 primers produced about 1.5kb and 0.7 kb amplified DNA fragments for all the Listeric relationships of Listeria sp. using NTSYS program were grouped into 7 clusters and showed 0.54 to 0.93 similarity among strains. Especially, No. 3 and No. 20 isolates showed the genetically most similar relationship by 0.94, and No. 7 and No. 24, or No. 7 and N0. 45 isolates showed the least similarty by 0.54 From these results, RAPD analysis method deemed to be successfully applied the classification and genetic analysis for Listeria sp. isolates.

Effects of Predation on Macrobenthic Communities in Seonjae-do Tidal Flat (포식이 선재도 갯벌 대형저서동물 군집에 미치는 영향)

  • We examined the effect of predation by observing the changes in macrobenthic communities in a tidal flat at Seonjae, Korea, following the exclusion of large predators such as birds, fishes and crustaceans using protective screens. We conducted two interference experiments in the field from April to November 2004: (1) Experiment 1 was conducted at a mid-tidal flat in the western part of Seonjae-do, Incheon, Korea, (2) Experiment 2 was set up at a low tidal flat in the eastern part of Seonjae-do. predator exclusion showed different effects in the two experiments. Both the number of species and density were reduced by 20% in Experiment 1. Whereas in Experiment 2, they increased by up to 13% and 69%, respectively. In Experiment 1, a high density of brachyuran crabs observed in the treatment may have caused the difference in community composition between the treatment and the control, and this probably resulted from active predation of macrofauna (e.g., small crabs) under protection from megafaunal predators such as birds and fishes. However, in Experiment 2, as typically observed in other predator exclusion experiments, a lower density of benthic predators and the subsequent reduction of over predation probably resulted in a more diverse and abundant benthic community in the treatment. We confirmed that predation was a contributing factor in the formation of the macrobenthic community. We also demonstrated relationships that exist among different trophic groups (large predators, macrobenthic predators and prey) and what effects and responses occur in each component of the tidal flats.
