Characteristics and breeding of a new variety Pleurotus eryngii, Saegonji (큰느타리버섯 신품종 '새곤지' 육성 및 특성)

  • We bred a new strain of Pleurotus eryngii having few number of fruit body per bottle. It's name is 'Saegonji' and it was bred by mating monokaryotic strain isolated from E12-176 and monokaryotic strain 'aerini No.3' in Mushroom Research Institute, Gyonggi province A.R.E.S. The characteristics of a new strain 'Saegonji' are as follows. The optimum temperature for mycelium growth was from 23 to $26^{\circ}C$ on PDA medium and those for the primodium formation and the growth of fruit body were about $15^{\circ}C$. The period from spawn innoculation to harvesting required around 51 days at $20^{\circ}C$. The number of fruit body per bottle was 12.8 and it was 9.3 less than 'Keunneutari No.2'. The length was 135.8 mm. It was longer than 'Keunneutari No.2', The color of cap was white grey, while that of 'Keunneutari No.2' was grey. The yield was about 159 g per bottle(1100cc) and it was same as Keunneutari No.2.

Species Composition and Biology of Major Species of Gobiid Fish in Cheonsu Bay of the Yellow Sea, Korea (한수만(漢水灣) 망둑어과(Family Gobiidae) 어류(魚類)의 계절(季節)에 따른 종조성(種組成) 변화(變化)와 우점종(優點種)의 생태(生態))

  • Gobiid fish were collected monthly in Cheonsu Bay by a beach seine from the shallow water and by an otter trawl from the deep water from September, 1984 to August, 1986. Seasonal variation in species composition and biology of the major species were examined with the data of length and age composition. Fourteen speices of gobies were identified. Gobiid fish were more abundant in the shallow water than in the deep water. In the shallow water, they constituted 66% and 39% of the fish collected in the mud flat and the sand beach, respectively. The suction discs of the gobies explains that they inhabit successfully in the surf zone where the wave action is strong. Of the major species, Chaenogobius mororanus predominated in the shallow mud flat. A large number of adults of this species were obtained in Spring and their juveniles were collected from Summer through Autumn. Favonigobius gymnauchen was abundant in the shallow water, especially in the sand beach. This fish seemed to spawn from May through June, about two months later than Chaenogobius mororanus. Acanthogobius hasta juveniles lived in the shallow water and moved into the deep water as the fish grew. Chaturichthys stigmatias was collected mainly in the deep zone.

Spawning Performance, Embryonic Development and Early Viability under Different Salinity Conditions in a Euryhaline Medaka Species, Oryzias dancena (서로 다른 염분도 조건하에서 광염성 송사리 Oryzias dancena의 산란, 발생 및 초기 생존)

  • Effects of different salinity levels on spawning performance, embryonic development and early viability of a euryhaline medaka species, Oryzias dancena, were examined. O. dancena were able to spawn eggs in a wide range of salinity from 0 to 70$^{\circ}/_{\circ\circ}$, however, the spawning frequency was lowered in complete freshwater (0$^{\circ}/_{\circ\circ}$) and in highly salted water (70$^{\circ}/_{\circ\circ}$). Fertilization success was negatively affected when the environmental salinity was higher than the salt concentration found in normal seawater. Embryonic viability and hatching success were also inversely related with the salinity levels. Typical abnormality was observed in developing embryos incubated at high salinities (30, 45 and 60$^{\circ}/_{\circ\circ}$). In addition, the time to hatch was significantly delayed with increasing salinities: peak hatching occurred at 12~14 days post fertilization (dpf) in freshwater and at least at 17 to 18 dpf in 60$^{\circ}/_{\circ\circ}$. Mean survival rates of the hatched larvae up to 7 days post hatching (dph) were at least 97% in salinity levels ranging from 0 to 30$^{\circ}/_{\circ\circ}$. However, larvae reared in 45 and 60$^{\circ}/_{\circ\circ}$ experienced significant mortality, especially in the early phase, resulting in only 75% and 64% survival rates up to 7 dph, respectively.

  • These studies were conducted to evaluate the bulk pasteurization system in comparison with the conventional mushroom growing technique of and to establish the phase II fermentation method for the system. The results obtained were as follows. 1. From the mushroom compost peak-heated in the bulk pasteurization system higher mushroom yield was obtained than that of the conventional method. 2. The compost fermented in the bulk pasteurization system showed poor mycelial growth and low crop. It was caused by the imperfect blower and its operation, not by ununiform moisture content of the compost and ununiform filling. 3. A bulk pasteurization system which modified the mushroom house was not proper for the fermentation of the mushroom compost and the sporophore yields were lower than the standard pasteurization system. 4. In the bulk pasteurization system, peak-heating time, phase II period and quality of the compost were influenced by the air temperature but its effects were not more significant than those of the conventional method. 5. During phase II in the bulk pasteurization system moisture content of the compost at filling did not affect the fermentation of the compost.

Embryogenesis in the Octopus minor (낙지(Octopus minor)의 배 발생)

  • The small octopus(Octopus minor), a species in the cephalopod class spawns the fewer eggs compared with its relatives such as Octopus vulgaris and Octopus ocellatus and exerts special efforts in nursing eggs hiding in the deep cave beneath the mud flat. Correspondingly, its embryogenic process had been hardly observed. In this study, we induced the small octopus to spawn its eggs in the artificial spawning tube and to nurse the developing embryos under the periodical observation. The early embryogenic change in the fertilized eggs was appearance of blastodisc on the animal pole at 4 days after spawning. A mass of rudiments of several organs developed in the middle of the yolk sac at 28-th day and it moved gradually toward the animal pole. At 38 days after spawning, two eyes appeared on the head and the trunk with heart developed on the apical part of the yolk sac. At 45-th day of embryonic development, the trunk and head occupied a half of the egg capsule and consequently the yolk sac was reduced to a half of its original size. Two eyes became prominent at the bottom of the trunk with several spots at day 60. Embryonic development was completed at about 80 days after spawning and the arms stuffed the lower half of egg capsule. The young fry squeezed out of the egg capsules with their long and slender arms first and mantles later. Soon after hatching, it swam actively and squirted ink.

Sexual Maturation of the Spotted Flounder Verasper variegatus (범가자미 Verasper variegatus의 성성숙)

  • The ovary of the spotted flounder Verasper variegatus is a conical bag in shape and is bilateral structure develops lengthily from the posterior of the abdomen to the end of the anal fin. The testis also is bilateral in structure, usually located in small size in the abdomen. In females, the gonadosomatic index (GSI) showed very low from March to July, and then began to increase from August, thereafter reached the maximum in January through out the year. In males, the GSI reached the maximum (1.7) in January through out the year, as seen in females. Compared with the male GSI in other fishes, the maximum GSI of this species were very lower than those of other fishes. According to the distributions of egg diameter in the spawning season, it is assumed that the spotted flounder spawn four times or more in the spawning season. The total length of female and male reached 50% first sexual maturity were 42.0~44.0 and 28.0~30.0cm, respectively and total length of female and male reached 100% maturity were 44.0 and 32.0cm, respectively. The reproductive cycle could be classified into four successive developmental stages: growing stage (August~October), mature stage (November~December), ripe and spent stage (December~February), degenerative and resting stage (March~July).

Spawning Inducement of Flounder, Paralichthys olivaceus by the Control of Water Temperature and Photoperiod (수온과 광주기 조절에 의한 넙치(Paralichthys olivaceus)의 산란유도)

  • Spawning inducement of flounder, Paralichthys olivaceus, was attemped by the control of water temperature and photoperiod from June 1, 1989 through January 5, 1990 (189 days) Water temperature was gradually decreased from $21.7^{\circ}C$ to $10.6^{\circ}C$ which was the minim biological temperature for spawning of flounder. Water temperature was increased asain to $15^{\circ}C$ gradually, and then maintained at this level through the end of spawning. Photoperiod was also changed gradually from 10L/14D in June 1, 1989 to 14L/10D in July 25, 1989. Spawning of the fish occurred from October 4, 1989 through January 5, 1990 (93 days). The average number of eggs spawned during the spawning season per female were 2.67 billion. The first spawning occurred on the 60th day after the time of minimun water temperature $10.6^{\circ}C$. The water temperature and photoperiod at the first spawning were $13^{\circ}C$ and 14L/10D respectively It took 71 days to spawn since the photoperiod had changed from 10L/14D to 14L/10D. Spawning period can be devided into three terms. The first term was continued for 30 days from the beginning of the spawning. The second term for 41 days was the major spawning period followed by the third term, the final period of the spawning, for 22 days. The percentage of average fertilization rate of the eggs in the first, second and third spawning terms were $37.4{\%},\;54.1\%\;and\;19.6\%$, respectively. Feeding rate was increased in the maturing period but, decreased in the spawing period. During the final spawning period, the abrubt decreasing of the feeding occurred again.

Identification of pathogen and actual culture state of king oyster mushroom (Pleurotus eryngii) (큰느타리버섯 주요재배시 실태조사 및 병원균 분리동정)

  • We have investigated cultural circumstance and given condition of king oyster mushroom(Pleurotus eryngii) growing farmer. We collected many pathogens from King oyster mushroom growing farmer and identified with chemicobiological test and microscope. Most of investigated farmers neglected their's growing room cleaning and washing, after harvesting At pin-heading induction time, humidity degree in growing room was kept of high level and Air ventilation volume was so little that fruit-body formation ratio was low. The collected pathogens were twenty eight strains and identified with Pseudomonas sp., Trichoderma sp. mostly. During the spawn running time and pin-heading induction time, contamination by Trichoderma sp. occurred mostly, but during the fruit-body growing time, contamination by Pseudomonas sp., Erwinia sp. etc, occurred.

Characteristics of 'Baekseung', a new cultivar Flammulina velutipes (팽이버섯 신품종 '백승'의 특성)

  • 'Baekseung', a new variety of Flammulina velutipes, was bred by mating two monokaryotic strains isolated from KMCC 4210 and KMCC 4216 in the Mushroom Research Division, Baekseung ARES in 2016. The Baekseung and Uri1ho strains showed fast mycelial growth and mycelial density on malt extract agar media after 7 days of incubation. The spawn running period on the sawdust substrate required a cultivation period and temperature of 30 days and $25^{\circ}C$, respectively, for primordia formation where in fruiting body development occurred from $11{\pm}1days$ at $14^{\circ}C$ and $14{\pm}1days$ at $7^{\circ}C$. The length of the pilei and stipes of the Baekseung harvested in optimal stag were $11.3{\pm}0.4$ and $89.2{\pm}7.1mm$, respectively, whereas the values for Uri1ho were $10.7{\pm}1.0$ and $91.3{\pm}20.8mm$, respectively. The yield of the Baekseung and Uri1ho strain grown on the sawdust substrate was $153.7{\pm}12.5$ and $139.8{\pm}17.8g$, respectively, per 850 ml in bottle cultivation. The inferred tree exhibited a phylogenetic relationship between the Korean white fruiting body strains of Baekseung, Uri1ho and Fv-14-a-38, Fv-14-a-51, and the Japanese white fruiting body-forming strains of KMCC 4226, and these were confirmed to be genetically related.

Distribution of spontaneously growing mushrooms in the Wolchulsan National Park (월출산국립공원의 자생버섯 분포상)

  • Mushrooms growing in the Wolchulsan National Park were surveyed from April to October 2018. As a result of this survey, 2 divisions, 9 classes, 24 orders, 71 families, 177 genera, and 407 species were found. Among them, 3 classes, 16 orders, 55 families, 150 genera, and 370 species belonged to Basidiomycota, while 6 classes, 8 orders, 16 families, 27 genera, and 37 species belonged to Ascomycota. The species richness was the highest in September at the altitudes between ~100 and 200 m. Among the investigated sites, the most diverse species occurred in the Gyeongpodae area. In this survey we found, Macrolepiota procera, Suillus bovinus, and Amanita pantherina, all of which have been known as climate-sensitive biological indicator species (CBIS). Based on the DNA sequence analyses, Craterellus parvogriseus was confirmed as a new record for Korea.
