Cultural Characteristics of Cauliflower Mushroom, Sparassis crispa (꽃송이버섯(Sparassis crispa)의 배양적 특성)

  • This study was carried out to obtain the basic data for the mycelial growth conditions of cauliflower mushroom, Sparassis crispa. Twenty-one isolates were collected from domestic and abroad. The optimal temperature and pH for the mycelial growth of these isolates were $23^{\circ}C$ and pH 6.0, respectively. The mycelial growth was the best in the HBA medium, but very poor in the Lilly medium. However no mycelial growth in the CzapekDox medium. The utilization of carbon source was the best with fructose, and that of nitrogen source was the best with glutamine when compared to other tested sources. The selected isolate ASI150010 produced the highest mycelial weight in liquid culture containing soybean mill ($15\;g/{\ell}$) and potato ($200\;g/{\ell}$) extract. And uncleaned rice, wheat and barley were found to be good substrates for the mycelial growth S. crispa.

Protein and Phosphorus Availabilities of Five Different Dietary Protein Sources in Juvenile Olive flounder (Paralichthys olivaceus) as Determined by Growth Performance and Phosphorus Retention

  • The present study aims to evaluate protein and phosphorus availabilities of five different dietary protein sources during the 6-week feeding trial in juvenile olive flounder, Paralichthys olivaceus as determined by growth performance and phosphorus retention. Five diets containing blood meal (BM), poultry by-product (PBP), squid liver powder (SLP), feather meal (FM) and soybean meal (SM) were prepared by mixing a basal diet (BD) with one of five test ingredients at the ratio of 7 to 3. As a reference diet, BD contains three different protein sources such as white fish meal, casein and gelatin. After 2 weeks of the conditioning period, fish initially averaging 2.7$\pm$0.02g (mean$\pm$SD) were randomly distributed into each aquarium as a group of 30 fish reared in the recirculating system. Fish of triplicate groups were fed one of six experimental diets (BD+five test diet). After 6-week feeding trial, pro- tein efficency ratio (PER) of fish fed BM diet was the lowest in experiment groups. While fish fed PBP diet showed a significant higher PER as compared to the FM diet, and fish fed SLP diet and BD were a higher PER than did fish fed PBP diet. However, there was no significant difference in PER among fish fed SLP diet, BD and SM diet, and between SM diet and PBP diet. Phosphorus retention efficiency of bone (PR $E_{b}$) of fish fed BM diet was the lowest in all the diets, and fish fed FM diet showed a higher PE $R_{b}$ than fish fed BD and SM diet. However, there was no significant difference in PER among fish fed FM diet, SLP diet and PBP diet, and among SLP diet, PBP diet, SM diet and BD. These results indicate that SLP could be a suitable protein source for low pollution diets of olive flounder in the future fish feeds market. Furthermore, PBP and SM are available protein source to reduce P waste in the oliver flounder aquaculture with the use of proper mixture of other protein sources and more processing to improve protein availibility of these.ese.

Effects of different energy and rumen undegradable protein levels on dairy cow's production performance at mid-lactation period (에너지 및 반추위 미분해단백질 수준을 달리한 사료급여가 비유중기 유우에 미치는 영향)

  • Sources of energy and rumen undegradable protein (RUP) have been used to meet nutrient requirements for high producing dairy cows. However studies for manipulation the levels of energy and RUP in diets have been mainly achieved using dairy cows at early-lactation period. The objective of this study thus, was to investigate the effects of different energy and rumen undegradable protein (RUP) levels on dry matter intake and milk yield in Holstein cows at mid-lactation period. Basal diet was prepared as TMR to meet nutrient requirements for dairy cows at mid-lactation according to NRC recommendation. Cows of control group (Con) were fed only basal diets while ground corn (0.5 kg/d), heat-treated soybean meal (0.5 kg/d), and their mixture (0.25 kg of each supplements/d) were added to diets for cows of treatment groups (T1, T2, and T3 respectively) to modulate the level of energy and RUP contents in diets. Addition of energy or RUP source in basal TMR did not affect in total DMI while TMR intake tended to be higher in Con compared to T3. Cows fed T3 diets tended to show increased milk yield and MUN content than those of Con. Cows for T2 as well as T3 had lower ADG (P<0.05) compared with those of Con. We concluded that the addition of RUP source in diets for dairy cows on mid-lactation period might cause the decrement of DMI and ADG.

Role of Mesophyll Morphology in Determination of Leaf Photosynthesis in Field Grown Soybeans (포장생육대두의 엽광합성과정에서 엽육세포 형태의 역할)

  • Photosynthetic variation in field grown soybean [Glycine max (L.) Merr. cv Hodgson78] was studied in relation to leaf anatomical variation. Variations in mesophyll morphology were accentuated by manipulating source and sink size. At R3 stage, two treatments were started: one was thinning and continu-ous debranching(6. 5 plants rather than 26 plants per m of row and remaining plants were debranched weekly), and the other was continuous partial depodding (allowing only one pod to develop at each mainstem node). Gas exchange characteristics, mesophyll cell volume and surface area per unit leaf surface, and microclimatic parameters were measured on the intact terminal leaflet at the 10th node. Observations were made 5 times with 3 to 4 day intervals starting R4 stage. Two models were used to compute leaf photosynthetic rates: one considered no effect of mesophyll morphology on photosynthesis, and the other considered potential effects of variations in mesophyll cell volume and surface area on diffusion and biochemical processes. Seventy nine percent of total photosynthetic variations observed in the experiment was explained by the latter, while 69% of the same variations was explained by the former model. By incorporating the mesophyll morphology concept, the predictability was improved by 14.6% in the field condition. Additional Index Words: photosynthesis model, leaf anatomy, Glycine max (L.) Influences of Culture Medium Components on the Production Poly (γ-Glutamic Acid) by Bacillus subtilis GS-2 Isolated Chungkookjang (청국장에서 분리한 Bacillus subtilis GS-2에 의한 Poly(γ-Glutamic Acid) 생산의 최적 배양조건)

  • A bacterium strain GS-2 isolated from the Korean traditional seasoning food, Chungkookjang and was determined to produce large amounts of ${\gamma}$-PGA with high productivity when provided with simple nutrients (L-glutamic acid 2.0%, glucose 1.0%, $NH_4Cl$ 0.5%, $KH_2PO_4$ 0.05%, $MgSO_4{\cdot}7H_2O$ 0.01%, pH 7.0). In this study, the culture medium for this strain was optimized for the production of ${\gamma}$-PGA. The Bacillus subtilis GS-2 required supplementation with L-glutamic acid and other nutrients for maximal production of ${\gamma}$-PGA. The optimal culture conditions for ${\gamma}$-PGA production were a 48 hr culture time, a temperature of $33^{\circ}C$ and initial pH of 6.5 by rotary shaking (220 rpm). A maximum ${\gamma}$-PGA production of 31.0 $g/{\ell}$ was obtained with L-glutamic acid (30 $g/{\ell}$), sucrose (the main carbon source, 30 $g/{\ell}$), $NH_4Cl$ (the main nitrogen source, 2.5 $g/{\ell}$), $KH_2PO_4$ (1.5 $g/{\ell}$) and $MgSO_4{\cdot}7H_2O$ (0.15 $g/{\ell}$) in the culture medium.

Reproductive Performance, Milk Composition, Blood Metabolites and Hormone Profiles of Lactating Sows Fed Diets with Different Cereal and Fat Sources

  • Different dietary cereal sources and fat types in the lactation diet were evaluated to investigate their effects on reproductive performance, milk composition, blood metabolites and hormones in multiparous sows. Twenty-four sows were randomly assigned to one of four treatments according to a 2${\times}$2 factorial arrangement of treatments. Each treatment had 6 replicates comprising 1 sow. Two cereal (corn or wheat) and two fat (tallow or soybean oil) sources were used to prepare iso-caloric and iso-nitrogenous diets. Sows fed corn-based diets lost less body weight (p = 0.003) and backfat thickness (p = 0.034), consumed more feed (p = 0.032) and had shorter wean-to-estrus interval (p = 0.016) than sows fed wheat-based diets. Fewer piglets and lower body weight of piglets (p<0.05) at weaning were noted in sows fed wheat-based diets than in sows fed corn-based diets. However, no significant effects (p>0.05) of dietary fat source and its interaction with dietary cereal source on sow body condition and reproductive performance were observed during lactation. Feeding of a corn-based diet improved (p<0.05) sow milk total solid, protein and fat, increased blood urea nitrogen (p = 0.032) and triglyceride (p = 0.018), and decreased blood creatinine (p = 0.011) concentration at weaning when compared with sows fed wheatbased diets. Sows fed corn-based diets had higher concentration of insulin (p = 0.048) and LH (p<0.05) at weaning than sows fed wheatbased diets. The results indicate that feeding corn-based diets to lactating sows improved sow body condition and reproductive performance compared with wheat-based diets regardless of fat sources.

Effects of Salmonella typhymurium Lipopolysaccharide Challenge on the Performance, Immune Responses and Zinc Metabolism of Laying Hens Supplemented with Two Zinc Sources

  • The study was conducted to determine the effect of Salmonella typhymurium lipopolysaccharide (LPS) challenge on egg-laying performance, inflammatory response, zinc metabolism in layer fed diets supplemented with organic or inorganic zinc since 3-wk-old. The three dietary treatments were corn-soybean meal basal diet without supplemental zinc or with supplemental zinc at 60 mg/kg zinc from $ZnSO_4$ or zinc amino acid complex (ZnAA). At the age of 58 wk-old, twelve hens from each dietary treatment were allotted into two sub-groups. On day 1, 3, 5, 7 of the $58^{th}$ week of age, six birds of one sub-group were injected intraperitoneally (i.p.) with 2 ml LPS (1.0 $\ell$/ml) or sterile saline. Neither zinc source ${\times}$ immune challenge interaction nor zinc source effect on egg production performance was observed (p>0.05), LPS-challenge decreased egg production (p<0.04) and increased percentage of cracked eggs (p <0.01). With LPS challenged, the fever response of hens fed ZnAA peaked and subsided earlier than in hens fed $ZnSO_4$ or basal diet. Serum IL-1$\beta$ at 3 h was higher (p<0.01), but lower (p<0.001) at 12 h post-challenge with LPS in hens fed ZnAA than $ZnSO_4$. In salinetreated groups, serum IL 1$\beta$ was higher in hens fed ZnAA than the basal diet at 3 h post-injection (p<0.01). LPS-challenged birds had lower serum zinc and higher zinc sequestered in liver and spleen (p<0.001). In saline-treated birds, there was no difference in zinc concentration of serum, liver and spleen among different dietary treatments (p>0.05). Supplementation of 60 mg/kg zinc from either ZnAA or $ZnSO_4$ significantly (p<0.05) elevated metallothionein (MT) concentration in liver and spleen. MT concentration in liver of birds fed ZnAA diet was higher than in those fed $ZnSO_4$ diet (p<0.05). The magnitude of increase of hepatic and splenic MT due to LPS challenge was higher by supplementation of ZnAA than $ZnSO_4$. The results suggest that zinc amino acid complex enhanceed MT synthesis and zinc sequestered in liver and spleen and increased the sensitivity to immune response due to LPS challenge.

Production and Characterization of Ethanol- and Protease-Tolerant and Xylooligosaccharides-Producing Endoxylanase from Humicola sp. Ly01

  • This paper reports the production and characterization of crude xylanase from the newly isolated Humicola sp. Ly01. The highest (41.8 U/ml) production of the crude xylanase was obtained under the optimized conditions (w/v): 0.5% wheat bran, 0.2% $KH_2PO_4$, and 0.5% peptone; initial pH 7.0; incubation time 72 h; $30^{\circ}C$; and 150 rpm. A considerable amount of the crude xylanase was induced using hulless barley bran or soybean meal as the carbon source, but a small amount of the enzyme was produced when supplementary urea was used as the nitrogen source to wheat bran. The crude xylanase showed apparent optimal cellulase-free xylanase activity at $60^{\circ}C$ and pH 6.0, more than 71.8% of the maximum xylanase activity in 3.0-30.0% (v/v) ethanol and more than 82.3% of the initial xylanase activity after incubation in 3.0-30.0% (v/v) ethanol at $30^{\circ}C$ for 2 h. The crude xylanase was moderately resistant to both acid and neutral protease digestion, and released 7.9 and 10.9 ${\mu}mol/ml$ reducing sugar from xylan in the simulated gastric and intestinal fluids, respectively. The xylooligosaccharides were the main products of the hydrolysis of xylan by the crude xylanase. These properties suggested the potential of the crude enzyme for being applied in the animal feed industry, xylooligosaccharides production, and high-alcohol conditions such as ethanol production and brewing.

Effects of Eco-friendly Squid Todarodes pacificus Liver and Patagonian Toothfish Dissostichus eleginoides Muscle Oils on the Serum Lipids and Adipose Tissues of Rats (살오징어(Todarodes pacificus) 간유 및 비막치어(Dissostichus eleginoides) 근육지질이 흰쥐의 혈청지질 및 지방조직에 미치는 영향)

  • This study investigated the effects of squid Todarodes pacificus liver oil on the serum lipids and adipose tissue weight of rats. The rats in the soybean oil (SO) group received a basic diet prepared according to the recommendations of the American Institute of Nutrition (AIN-93G), with 7% SO as lipid source. The rats in the Patagonian toothfish Dissostichus eleginoides muscle oil (PTFO) group received 5% PTFO and 2% SO as the lipid source, while the rats in the eco-friendly squid liver oil (EFSO) group received 5% EFSO and 2% SO. EFSO was prepared according to the method outlined in the author's patent. PTFO was extracted from muscle using a physical method in the author's laboratory. The prominent polyunsaturated fatty acids in the dietary oils were 18:2n-6 (54.3%) and 18:3n-3 (5.64%) in SO, DHA (6.77%) and EPA (3.61%) in PTFO, and DHA (26.2%) and EPA (9.94%) in EFSO. After feeding for 4 weeks, the serum lipid levels (e.g., triglyceride, total cholesterol, and low-density lipoprotein cholesterol) of the rats in the EFSO group were significantly reduced as compared with those in the SO group (P<0.05); those of the rats in the PTFO group were also reduced, albeit not significantly, compared with the SO group. On the other hand, the total white adipose tissue (visceral, epididymal, and perirenal) weight was significantly reduced in the EFSO and PTFO groups compared to the SO group. These results demonstrate that EFSO is an effective lipid modifying agent in rat blood, and that EFSO and PTFO have greater anti-obesity effects than SO.

Production and properties of exoinulase from Streptomyces sp. S34 (Streptomyces sp. S34의 exoinulase 생산 및 성질)

  • An exoinulase-producing bacterium was isolated from soil, and identified as Streptomyces sp. The maximum inulase production was achieved when inulin as carbon source and soybean meal as organic nitrogen source were included in the culture. The exoinulase was considered to be a constitutive enzyme produced not only by inulin but also by soluble starch or glucose. The purified enzyme on DEAE-cellulose and Sephadex G-200 column showed the maximal activity at $pH\;5.5{\sim}6.0$ and $50^{\circ}C$, but lost 65% inulase activity at $50^{\circ}C$ after 1 hour incubation. This exoinulase was activated by $Mn^{+2}$, wherease more that 80% inactivation was observed with $Ag^+$, $Hg^{+2}$ and $Fe^{+3}$. The enzyme was possibly a metalloenzyme in that EDTA and 8-hydroxyquinoline inhibited the enzyme. Km values for inulin (16.51 mM) and sucrose (14.62 mM) were in very close range suggesting mostly equal affinity toward the subatrates. However, the maximum velocity for inulin was 10 times greater than for sucrose.
