대두의 핵산, 단백질 및 물질생산에 미치는 인산비료의 효과 (DNA, RNA, Protein and Yield of the Soybean Plant, Glycine max Merr., as Affected by Phosphorus Nutrition)

  • The effect of phosphorus nutrition on the content of deoxyribonucleic acid(DNA), ribonucleic acid(RNA), crude protein and plant growth of soybean plant(Glycine max, Merr.) was studied. Yields of the above-and under-ground parts of the soybean plant in terms of dry weight, the amounts of crude protein, RNA and DNA continued to increase with increasing phosphorus supply. The amounts of RNA and crude protein were highest in the leaf tissues where most intensive growth was taking place. The relationships among DNA, RNA, crude protein and plant growth appeared to consist of the central dogma which has immortalized, while DNA in plant tissue was subject to charges cuased by external environmental facters such as phosphorus nutrition.

단백질 급원이 흰쥐 혈청 콜레스테롤 농도에 미치는 영향 (Effects of Dietary Proteins on Serum Cholesterol Concentration in Rats)

  • 상이한 단백질원이 흰쥐의 혈청콜레스테롤 농도에 미치는 영향을 조사하고자 명태로부터 조제한 어육단백질과 동물성의 카제인 및 식물성의 대두단백질을 비교 검토하였다. 실험 식이의 단백질 수준은 20%가 되도록 저지방 식이에 준비된 단백질 시료를 혼합하여 S.D. 계통 male rat에 2주간 급여한 후 혈청지질 농도를 분석하였다. 그 결과 혈청중의 총 콜레스테롤, LDL-콜레스테롤 및 총 콜레스테롤/HDL-콜레스테롤 농도는 카제인군에서 가장 높았고 어육단백질군, 대두단백질군 순으로 낮았으며 카제인군과 다른 두 단백질군 간에는 유의성이 있었다(p<0.05). HDL-콜레스테롤 농도는 어육단백질군이 다른 두 단백질군보다 높았지만 유의성은 없었다. 트리글리세라이드 농도의 경우 세 단백질군간 유의성이 있었는데 어육단백질군은 카제인군보다 약 19mg/dl 낮았지만 대두단백질군보다는 약 6mg/dl 높았다. 어육단백질의 Lysine/Arginine비, Glycine/Methionine + Cystine비는 대두단백질의 그것들에 가까운 비율이었다.

유기 반추동물 전용 수입산 유기사료 및 국내산 유기 부산물의 사료적 가치 평가 (Nutritional Evaluation of Imported Organic Feeds and Locally Produced Agricultural By-products for Organic Ruminant Farming)

  • This study was conducted to evaluate the nutritional value of locally produced organic agricultural by-products to substitute imported organic feeds for organic ruminant farming. Imported organic feeds (corn grain, soybean meal, soybean seed, oat grain, barley grain, wheat grain, buckwheat, sunflower seed meal) and byproducts (rice bran, grape seed meal, rice straw, soybean hull, soybean curd, rice hull, green kernel rice, and crushed rice grain) were analyzed for chemical composition and NDF, ADF, mineral, and amino acid contents and anti-nutritional factors. Dry matter, NDF and ADF contents in organic feeds were higher than those in conventional feeds. Especially, the 9.65% fat content of organic soybean meal was 6 times higher than the 1.95% fat content of conventional soybean meal. Fat contents of rice bran, grape seed meal, green kernel rice, and crushed rice grain were 25.66, 6.09, 3.57 and 1.59%, respectively. Protein contents of soybean hull and soybean curd were 14.68 and 19.87%, respectively, which are highest among organic by-products. Levels of aflatoxin in all feeds were below the safety level. Therefore, organic rice bran, green kernel rice and crushed rice as energy source, and soybean hull and soybean curd as protein source could partial replace imported feeds for organic ruminant farming.

Application of the Combination of Soybean Lecithin and Whey Protein Concentrate 80 to Improve the Bile Salt and Acid Tolerance of Probiotics

  • To improve the bile salt and acid tolerance of probiotics against gastrointestinal stresses, we investigated the effects of soybean lecithin and whey protein concentrate (WPC) 80 on the bile salt tolerance of Lacticaseibacillus paracasei L9 using a single-factor methodology, which was optimized using response surface methodology (RSM). The survival rate of L. paracasei L9 treated with 0.3% (w/v) bile salt for 2.5 h, and combined with soybean lecithin or WPC 80, was lower than 1%. After optimization, the survival rate of L. paracasei L9 incubated in 0.3% bile salt for 2.5 h reached 52.5% at a ratio of 0.74% soybean lecithin and 2.54% WPC 80. Moreover, this optimized method improved the survival rate of L. paracasei L9 in low pH condition and can be applied to other lactic acid bacteria (LAB) strains. Conclusively, the combination of soybean lecithin and WPC 80 significantly improved the bile salt and acid tolerance of LAB. Our study provides a novel approach for enhancing the gastrointestinal tolerance of LAB by combining food-derived components that have different properties.

  • Microorganisms, including the strain IJ-3 isolated from soil, were found to secrete an extracellular soybean protein coagulating enzyme and the strain, IJ-3 was identified as Genus Bacillus according to the Bergey's manual . The enzyme coagulated protein in soymilk , thus forming a curd at pHs 5.8-6.4 and at 55-75℃. The optimum temperature for soybean protein coagulating activity was 65-75℃ and the enzyme was stable at temperature below 50℃ and was found to be stable with about 60-100% of the original activity at a with pH ranges(pH6-7). The molecular weight of enzyme was estimated to be 28,000 by SDS-PAGE. The curd formed with the enzyme from Bacilus sp. IJ-3 has a smooth texture, and a mild taste without any bitterness or a beany flavor.

옥수수와 사료용 콩 혼작에 의한 조사료 수량 및 품질 (Yield and Quality of Forage Produced by Mixed Planting of Soybean and Corn)

  • The soybean [Glycine max (L.) Merr.], an edible legume, has a high protein content in both its hay and grain, so it is often used as a supplement for other forages that have a deficient protein concentration. Therefore, this study investigated the forage quality and yield in the case of mixed planting of soybean and corn. The forage yield and quality were assessed for three cropping patterns: soybean mono planting, corn mono planting, and mixed planting of soybean and corn. For planting, this study used a forage corn cultivar, Kwangpyeongok, and three recombinant inbreed lines, W2, W4, and W11, selected from Glycine soja (PI483463)${\times}$G. max (Hutcheson). The mixed planting of soybean and corn produced a higher forage yield than the corn mono cropping. The crude protein and crude fat content were also increased with the mixed planting of soybean and corn when compared with the corn mono cropping. Some decrease of ADF and NDF, and increase for RFV in mixed planting of soybean and corn than corn mono cropping. Therefore, the results show that mixed planting of soybean and corn is an effective cropping system to improve the forage quality.

역삼투법으로 분리, 농축한 두부순물의 이화학적 특성 (Physico-chemical Properties of Soybean Curd Whey Concentrated by Reverse Osmosis)

  • 역삼투 방법을 이용한 두부순물의 정화시 역삼투 잔사의 활용 가능성을 검토하기 위해 두부순물 농축물의 성분 특성 및 분리 단백질의 기능성을 조사하였다. 두부순물의 역삼투 잔사 농축물에는 자당, 라피노오스 및 스타치오스가 건물 중으로 각각 32.59%, 4.76% 및 9.99% 함유되어 있었고, 무기물중 칼륨이 5.23%로 회분의 42.7%를 점하였다. 또한 조단백질 함량은 18.69%이었으며 아미노산 조성 및 전기영동상의 sub-unit 양상은 분리 콩단백질과 다소 차이를 보였다. 한편 두부순물로부터 분리한 단백질의 기능성중 pH에 따른 용해도는 분리 콩단백질에 비해 전반적으로 다소 높은 값을, 유화능과 유화안정성 및 점도는 분리 콩단백질보다 비교적 낮은 값을 나타냈다.

  • 1) 2성분계 도지두부의 유효열전도도($\lambda_e$)를 $0\sim20^{\circ}C$$-5\sim-20^{\circ}C$의 범위내에서 측정한 결과, 수분함양의 증감에 따라 $\lambda_e$값도 증감했으며, 동결점이상의 온도대에서는 온도의 상승과 함께 $\lambda_e$값도 커지고, 동결점 이하에서는 온도의 하강과 함께 $\lambda_e$값이 커졌다. 2) 3성분계 두부의 유효열전도는 수분함양이 증가함에 따라 $\lambda_e$값은 커져가나 지질의 함양이 증가함에 따라 $\lambda_e$값은 작아졌다. $\lambda_e$값의 온도의존성은 2성분계도지두부의 경우와 유사했다. 3)대대단백질의 고유열전도도는 미동결상태에서 $\lambda_p=0.300[W/m{\bullet}k](0\sim-20^{\circ}C)$이고, 동결상태에서 $\lambda_p=0.488[W/m{\bullet}k](-5\sim-20^{\circ}C)$였으며 이들은 실험온도 범위내에서 온도의존성을 나타내지 않았다. 4) 대두단상질의 고유열전도도의 추정치 $\lambda_p=0.300$$\lambda_p=0.488$의 타당성을 지질의 함양이 상이한 2종류의 3성분계 두부에 적용한 결과, 실측치와 계산치는 잘 일치했다.

Independent Inheritance between df2 gene and ti gene in Soybean

  • Dwarfuess and Kunitz trypsin inhibitor (KTI) protein in soybean is useful traits for basic studies. df2 and ti gene control dwarfness and the expression of Kunitz trypsin inhibitor (KTI) protein in soybean, respectively. The objective of this research was to verify genetic linkage or independent inheritance of df2 and ti loci in soybean. The $F_2$ population was made by cross combination between "Gaechuck#2" (Df2Df2titi genotype, KTI protein absence and a normal growth type) and T210 (df2df2TiTi genotype, a dwarf growth type and KTI protein present). A total of 258 $F_2$ seeds were analyzed for the segregation of KTI protein using SDS-PAGE. And so, 198 $F_2$ plants were recorded for the segregation of dwarfness. The segregation ratio of 3 : 1 for Ti locus (201 Ti_ : 57 titi) and Df2 locus (143 Df2_ : 55 df2df2) was observed. Segregation ratio of 9 : 3 : 3 : 1 (116 Ti_Df2_: 44 Ti_df2df2: 27 titiDf2_: 11 titidf2df2) between df2 gene and ti gene was observed ($x^2$=3.53, P = 0.223). These results showed that df2 gene was inherited independently with the ti gene in soybean.

The development of new soybean strain with ti and cgy1 recessive allele

  • Soybean [Glycine max (L.) Merr.] seed is an important dietary source of protein, oil, carbohydrate, isoflavone and other various nutrients for humans and animals. However, there are anti-nutritional factors in the raw mature soybeans. Kunitz trypsin inhibitor (KTI) protein and stachyose are the main anti-nutritional factors in soybean seed. The ${\alpha}^{\prime}$-subunit of ${\beta}$-conglycinin protein exhibit poor nutritional and food processing properties. The genetic removal of the KTI and ${\alpha}^{\prime}$-subunit proteins will improve the nutritional value of the soybean seed. The objective of this research was to develop a new soybean strain with KTI and ${\alpha}^{\prime}$-subunit protein free ($titicgy_1cgy_1$ genotype) and proper agronomic traits. A breeding population was developed from the cross of the Bl-1 and 15G1 parents. A total of 168 $F_2$ seeds from the cross of the BL-1 and 15G1 parents were obtained. The segregation ratios of 9: 3: 3: 1 ($104Ti\_Cgy_{1\_}:\;30Ti\_cgy_1cgy_1:\;21cgy_1cgy_1Ti\_:\;13titicgy_1cgy_1$) between the Ti and $Cgy_1$ genes in the $F_2$ seeds were observed (${\chi}^2=5.12$, P=0.5-0.10). Two $F_4$ plant strains with proper agronomical traits and $titicgy_1cgy_1$ genotype (free of both KTI and ${\alpha}^{\prime}$-subunit protein) were selected and harvested. 2 strains (S1 and S2) had yellow seed coats and hilum. The plant height of the S1 strain was 65 centimeters. The 100-seed weight was 29.2 g. The plant height of the S2 strain was 66 centimeters and 100-seed weight was 26.2 g. The two strains selected in this research will be used to improve the new cultivar that will be free of the KTI and ${\alpha}^{\prime}$-subunit proteins.
