Sorption kinetics on the removal of zinc ion using scoria

  • Recently, considerable researches have been focused to find out inexpensive sorbents. for removal of heavy metals in aquatic environments. In particular, various natural materials including geologic media have been attractive. In order to evaluate the applicability of the scoria taken from the Jeju island, Korea, to remove Zn(II) from aqueous solutions, the kinetic sorption experiments were performed in this study. The batch-type kinetic sorption tests were carried out under different conditions, such as different initial Zn(II) concentration, particle size of the scoria, and sorbate/sorbent ratio. The results indicated that the removal of Zn(II) by scoria increased with decreases in initial Zn(II) concentration, particle size of the scoria, and sorbate/sorbent ratio. However, the sorption capacity of the scoria decreased with increasing amount of the scoria. The sorption behavior of Zn(II) onto scoria seemed to be mainly controlled by cation exchange.

  • The Budget method used currently in Codex for estimation of use levels of food additives was investigated and the specific approach that can be applicable for Koreans has been proposed. Theoretical maximum daily intakes(TMDIs) of benzoate, $\rho$-hydroxybenzoic acid ester, butylated hydroxyanisole(BHA), butylated hydroxytoluene (BHT), t-butylhydroquinone(TBHQ), nitrite, nitrate, sorbate, and sulfite were estimated and compared with corresponding acceptable daily intakes(ADIs). Additives requiring further precise evaluation were screened. TMDIs of benzoate, BHA, BHT, TBHQ, nitrite, and sulfite exceeded ADIs. On the other hand, it is appeared that TMDIs of $\rho$-hydroxybenzoic acid ester, nitrate, and sorbate were below ADIs. It is expected that the Budget method may be used as one approach for the estimation of use level and risk assessment.

Shelf-life and Quality Characteristics of Potassium Sorbate-free Meat Products (솔빈산 칼륨이 첨가되지 않은 육제품의 저장 수명과 품질 특성)

  • This study aimed for the examination of quality characteristics and safety of potassium sorbate-free meat products. Therefore, experiments were carried out on the frankfurter sausage and pressed ham, which were stored at 4 and 30$^{\circ}C$ for up to 40 days. The potassium sorbate concentrations of the frankfurter sausage and pressed ham obtained from local market ranged from 1.087 to 1.449g / kg, which were below the permitted value as prescribed in the Korean Hygienic Regulation. At the 0 day the total aerobic bacterial counts of frankfurter sausage and pressed ham were in the level of around 103.0 and 103.4 CFU / g, respectively. However, they were prominently increased after 20 days at 4$^{\circ}C$ and 10 days at 30$^{\circ}C$ to higher than 105 CFU / g. After 30 days the counts were increased to 106.5 and 107.2 CFU / g, respectively. The growth curve of lactic acid bacteria was shown to be similar with that of total bacteria. the counts of lactic acid bacteria of the products stored at 4$^{\circ}C$ were 101∼102 CFU / g lower than those stored at 30$^{\circ}C$. Coliform bacteria was not detected in both of the products stored at 4$^{\circ}C$ even after 40 days storage, but after 10 days at the 30$^{\circ}C$. No significant differences in the microbial counts examined in this study were observed between frankfurter sausage and pressed ham. The biochemical tests on the isolated colonies from Clostridein agar showed no presence of Clostridium botulinum and Clostridium perfringens in the meat products examined. The pH of frankfurter sausage and pressed ham at the beginning was about 6.6, which level was maintained relatively constant during the storage at 4$^{\circ}C$, but it was increased after decrease to about 5.5 during the storage at 30$^{\circ}C$. TBA value was increased slightly till 30 days, but after that time increased sharply. VBN value was increased slowly during the whole storage, but it was more than 30 mg% for the samples stored at 30$^{\circ}C$.

Effect of Some Materials on the Content of Nitrate, Nitrite and Vitamin C in Kimchi during Fermentation (배추김치 숙성(熟成) 중(中) 일부(一部) 첨가재료(添加材料)가 질산염(窒酸鹽), 아질산염(亞窒酸鹽) 및 Vitamin C 함량(含量)에 미치는 영향(影響))

  • This study was intended to observe the changes of the nitrate, nitrite and vitamin C content during the fermentation of Kimchies by some added materials. Eight different types of Kimchi, were prepared with chinese cabbage and seasonings, to which added respectively materials such as soused anchovy, soused shrimp, garlic, mustard leaf, K-sorbate, ascorbic acid, radish. After they were prefermented at $18^{\circ}C$ for 24 hours, stored 35 days at $4^{\circ}C$. Generally optimal maturity of Kimchi showed pH 4.4 to 4.6, lactic acid content 0.3 to 0.4% at salt content belows 2.5%. The content of total vitamin C in Kimchies was approximately 19.8-24.7 mg/100g at the initial stage of fermentation and then slightly decreased. When the process of the fermentation was active, the content of total vitamin C increased up to the same level or higher than that of the initial stage and then gradually decreased. In the case of Kimchi which added garlic, the content of vitamin C was relatively higher then the other samples. In the initial stage of fermentation, the nitrate and nitrite content in the Kimchi which added garlic and raddish were relatively higher than other samples. Nitrate content reached its minimum by the 21st day, at that time content was 290-342 ppm. At this time, the nitrite was not detected and total vitamin C content in all samples decreased.

Biochemical Characteristics of Micrococcus varians, Staphylococcus carnosus and Staphylococcus xylosus and Their Growth on Chinese-Style Beaker Sausage

  • This study was conducted to investigate protein and carbohydrate utilization of Micrococcus varians, Staphylococcus carnosus and Staphylococcus xylosus. Sensitivity to pH, sodium chloride, potassium sorbate and sodium nitrite of these strains was also determined. In Chinese-style beaker sausage manufacturing, the growth rate of these strains during the curing period ($20^{\circ}C$ and $30^{\circ}C$) was evaluated. The results indicated that no strains could hydrolyze azo-casein and sarcoplasmic protein and only S. xylosus could hydrolyze gelatin at $30^{\circ}C$. All of these strains could oxidize and ferment fructose and mannitol. S. carnosus and S. xylosus could slightly oxidize lactose and utilize citrate. Arabinose was oxidized by S. xylosus and sorbitol was oxidized by S. carnosus. Growth of M. varians was restricted at pH 5.0 and S. carnosus and S. xylosus were restricted at pH 4.5. S. xylosus and S. carnosus were able to grow with 0.1~0.5% potassium sorbate, 50~200 ppm sodium nitrite or 1~15% sodium chloride. S. xylosus had a higher growth rate than the other strains. Staphylococcus species grew well during curing period of Chinese-style beaker sausage then followed by Micrococcaceae.

Inhibitory Effect of Pine Needle(Pinus densiflora S.) Extract on Potato Polyphenol Oxidase (감자 polyphenol oxidase에 대한 솔잎 추출물의 저해효과)

  • The inhibitory effect of pine needle (Pinus densiflora S.) on potato polyphenol oxidase (PPO) was investigated. The addition of the pine needle extract exhibited a higher inhibitory effect on the potato polyphenol oxidase activity than that of the citric acid or potassium sorbate. The enzyme activity was strongly inhibited in a pH range of 7.0-8.0. When the incubation time of reaction mixture was increased, the potato polyphenol oxidase activity was markedly inhibited. The pine needle extract inhibited the potato polyphenol oxidase non-competitively. And also the pine needle extract subjected to a heat treatment at $100^{\circ}C$ for 10 min or to an acid treatment at pH 2.0, 3.0, and 4.0 for 3 hours still retained inhibitory effect on potato polyphenol oxidase.

Synergistic Effect of Combined Treatment of Bacteriocin Produced by Enterococcus faecalis MJ-231 and Potassium Sorbate on Growth of Food-Borne Pathogenic Bacteria (식중독균 생육에 대한 Enterococcus faecalis MJ-231의 박테리오신과 소르빈산칼륨의 혼합처리 효과)

  • The alone and combined effects of bacteriocin produced from Enterococcus faecalis MJ-213 and potassium sorbate against the food-borne pathogenic bacteria were studied. Bacteriocin minimal inhibitory concentration (MIC) values for Staphylococcus aureus ATCC 6538 and Salmonella enteritidis ATCC 13076 were 50 and 100 ${\mu}g$/ml, respectively. Bacteriocin (100 ${\mu}g$/ml) alone was active against S. aureus and S. enteritidis, but it was lower in antimicrobial effectiveness than the combination of bacteriocin (100 ${\mu}g$/ml) with potassium sorbate (100 ${\mu}g$/ml), which reduced initial counts (6 log cycle) of S. aureus and S. enteritidis by 1 and 3 log cycle, respectively. The bactericidal activity of bacteriocin of E. faecalis MJ-213 heated at $100^{\circ}C$ for 30 min or $121^{\circ}C$ for 15 min was markedly decreased as compared with the control. Moreover, the activity of bacteriocin was completely abolished by pepsin or protease II, but not affected by ${\alpha}$-amylase or lipase. The activity of bacteriocin adjusted to pH 6.0-8.0 showed almost the same inhibition ratio compared with the bacteriocin unadjusted pH, and though the inhibition ratio against pathogenic bacteria was reduced than the control, the bacteriocin was stable at pH 4.0 or 10.0, relatively. Furthermore, the combined treatment of bacteriocin and potassium sorbate than the alone treatment of bacteriocin significantly decreased (p<0.05) the viable cell counts of S. aureus or S. enteritidis inoculated on grind beef during storage at $4^{\circ}C$.

  • Potassium Sorbate (PS) is a potassium salt version of sorbic acid, which has antimicrobial and fungistatic features in foods. Therefore, PS is used as a food preservative against fungi and mold. PS has been found to be non-toxic even when taken in large quantities given its trait to be broken down in the body into water and carbon dioxide. Gap Junctional Intercellular Communication (GJIC) is essential in the maintenance of tissue homeostasis during development and differentiation. This study was made of the effects of PS on GJIC in WB-F344 rat liver epithelial (WB) cells. We found dramatic decrease of cell viability in time- and dose-dependent manners when WB cells were treated with PS. The effect of PS on GJIC is strong inhibition, and it took place in parallel with a hyperphosphorylation of connexin 43 expression. The finding that PS interferes with gap junction functionality should be considered with respect to the mechanism of PS-induced hepatotoxicity.

Keeping Quality of Yogurt Beverage Prepared from Soy Protein Concentrate (농축대두단백(濃縮大豆蛋白)으로 제조된 유산균음료의 저장성)

  • Soy yogurt beverage(SYB) was prepared from soy protein concentrate and the effect of stabilizers on the sedimentation of SYB curd and the microbiological quality of SYB during storage were investigated. The curd in SYB precipitated significantly during the first one hour of storage and the sedimentation of curd was reduced by the addition of CMC of over 0.6%, PGA of over 0.2% or Na-alginate of over 0.05%. SYB could be kept for 66 days at $5^{\circ}C$ without significant change in viable cell count of lactic acid bacteria. pH, titratable, acidity and viable cell count of SYB were significantly changed from 42 days of storage at $25^{\circ}C$. Some of SYB samples were contaminated by fungi and the growth of fungi was greatly inhibited by the addition of K-sorbate of 0.005%.

The Effect of Various Humectants on Equilibrium Moisture Content and Storage Stability of Seasoned Squid (여러가지 보습제가 조미오징어 평형수분함량에 미치는 영향)

  • This study was, firstly, to investigate water holding capacity in terms of variation of moisture sorption isotherms of seasoned squid treated with sodium lactate, glycerol, propylene glycol, sorbitol, mannitol, sodium benzoate, potassium sorbate and calcium propionate, and secondly, the effect of humectant treatments on storage stability was studied. The criteria for storage stability was based on three quality factors, namely, lipid oxiations, color development by non-enzymatic browning reactions and lipid oxidation, and mold growth. The effect of humectants on equilibrium moisture content was in the following increasing order; mannitol < sorbitol < sodium lactate < propylene glycol < glycerol. The experimental data indicated that sodium lactate has, in practice, potentially positive effect on processing of seasoned squid. During the storage period of 60 days, TBA values increased in all samples tested as humectants concentrations increased up to 10%. However, in the range of 1-7% sodium lactate treatment, the degree of lipid oxidation, browning reactions and mild growth were not high enough to affect the quality of seasoned squid, when compared with conventionally manufactured ones.
