Nicotiana tabacum과 N. rustica 체세포 잡종식물의 육성 (Somatic hybridization between Nicotiana rustica and N. tabacum through protoplast fusion)

  • Mesophyll protoplasts derived from young leaves of Nicotiana rustica and N. tabacum cv Burley 21 were fused with the aid of polyethylene glycol(PEG). Cytological examination of protoplasts after PEG treatment revealed 12.8 % heterokaryocytes. After 7 weeks culture, the hybrid calli showing greenish white with a compact appearance were selected in contrast to parental type calli tinged with white or green color. The somatic hybrid plants were verified by morphological, biochemical and cyclological analysis. A heterosis effect for plant vigor and height was observed but the shape of leaves and flower characteristics were intermediate between N. tabacum and N. rutstica. The isozyme banding patterns for peroxidase of somatic hybrid lines were compared with the parent species. A number of isozyme bands derived from both parental species were found in the hybrids. Somatic hybrid plants have been successfully backcrossed to the parental N. tabacum particularly with somatic hybrid plants as female parents. These hybrid plants yielded small seeds, only few which were germinable.

  • Petunia hybrida와 Nicotiana sanderae의 원형질체를 융합하여 체세포잡종식물을 얻고자 융합산물의 배양에 의해 얻은 개체들에 대해 유전분석을 하였다. 재분화 개체중 융합된 것으로 인정되는 개체의 peroxidase pattern은 두 모본의 band를 모두 가진 개체(A-D patterns)에서 나타난 band중 p. hybrida에서 나타난 band 이외에 최상부에 1개의 band가 추가되었는데 이는 융합을 판별할 수 이는 중요한 marker 로서 가능성이 있으며, aspartate aminotransferase isoenzyme pattern은 두 모본의 양상이 모두 나타나거나 N. sanderae의 상부 band가 소실된 개체도 있었다. 염색체 검경에서 P.hybrida는 2n=14, M sanderae는 2n=18개로 나타났으며, 동위효소검정에서 체세포 잡종일 것으로 추정되는 개체들은 2n=32-36으로 나타났다. 표현형 관찰에서 체세포잡종으로 인정되는 개체의 형태적 특징은 두 모본의 중간형태를 나타내었다.

Production and Characterizations of Somatic Hybrids between Brassica campestris L. ssp pekinensis and Brassica of oleracea L. var capitata

  • Protoplasts isolated from inbred lines of Brassica oleracea L. var capitata (cabbage) and Brassica campestris L. ssp. pekinensis (Chinese cabbage) were fused by PEG-mediated method, and somatic hybrid cells were differentiated into plants. for the identification of somatic hybrid plants, ploidy level, plant morphology, and cytological analysis were performed. All of the regenerated plants derived from fused protoplasts were shown to be 2X-4X, or higher ploidy level, presumably due to somatic hybridization or chromosome doubling. The morphology of leaves, petioles, and flowers showed an intermediate phenotype between Chinese cabbage and cabbage. Chromosome numbers in these somatic hybrids ranged mostly from 33 to 38. According to Genomic in situ hybridization (GISH) pattern, signals from both fusion parents of B.campestris or B.oleracea were detected in different colors when chromosomes of putative somatic hybrids were observed.

Effect of IOA and Media on Production of Somatic Hybrid by the Asymmetric Protoplast Fusion Between Three Solanum Species

  • In this study, we surveyed the influence of IOA (iodoacetamide) and media upon protoplast fusion for the efficient production of the somatic hybrid among S. sisymbriifolium and other Solanum species (S. intergrifolium and S. toxicarium). Regardless of a breed, as the IOA concentration increases, the cell division tends to decrease. As the influence of the media on the colony formation, we could get 5 colonies from the fusion of S. sisymbriifolium and IOA-treated S. integrifolium protoplast, but none was observed in other fusions. As a result of analyzing their IDH isozyme, we found a somatic hybrid in 2 objects.

Current Status of Comparative Mapping in Livestock

  • Comparative maps, representing chromosomal locations of homologous genes in different species, are useful sources of information for identifying candidate disease genes and genes determining complex traits. They facilitate gene mapping and linkage prediction in other species, and provide information on genome organization and evolution. Here, the current gene mapping and comparative mapping status of the major livestock species are presented. Two techniques were widely used in comparative mapping: FISH (Fluorescence In Situ Hybridization) and PCR-based mapping using somatic cell hybrid (SCH) or radiation hybrid (RH) panels. New techniques, using, for example, ESTs (Expressed Sequence Tags) or CASTS (Comparatively Anchored Sequence Tagged Sites), also have been developed as useful tools for analyzing comparative genome organization in livestock species, further enabling accurate transfer of valuable information from one species to another.

High frequency somatic embryogenesis and plant regeneration of interspecific ginseng hybrid between Panax ginseng and Panax quinquefolius

  • Background: Interspecific ginseng hybrid, Panax ginseng ${\times}$ Panax quenquifolius (Pgq) has vigorous growth and produces larger roots than its parents. However, F1 progenies are complete male sterile. Plant tissue culture technology can circumvent the issue and propagate the hybrid. Methods: Murashige and Skoog (MS) medium with different concentrations (0, 2, 4, and 6 mg/L) of 2,4-dichlorophenoxyacetic acid (2,4-D) was used for callus induction and somatic embryogenesis (SE). The embryos, after culturing on $GA_3$ supplemented medium, were transferred to hormone free 1/2 Schenk and Hildebrandt (SH) medium. The developed taproots with dormant buds were treated with $GA_3$ to break the bud dormancy, and transferred to soil. Hybrid Pgq plants were verified by random amplified polymorphic DNA (RAPD) and inter simple sequence repeat (ISSR) analyses and by LC-IT-TOF-MS. Results: We conducted a comparative study of somatic embryogenesis (SE) in Pgq and its parents, and attempted to establish the soil transfer of in vitro propagated Pgq tap roots. The Pgq explants showed higher rate of embryogenesis (~56% at 2 mg/L 2,4-D concentration) as well as higher number of embryos per explants (~7 at the same 2,4-D concentration) compared to its either parents. The germinated embryos, after culturing on $GA_3$ supplemented medium, were transferred to hormone free 1/2 SH medium to support the continued growth and kept until nutrient depletion induced senescence (NuDIS) of leaf defoliation occurred (4 months). By that time, thickened tap roots with well-developed lateral roots and dormant buds were obtained. All Pgq tap roots pretreated with 20 mg/L $GA_3$ for at least a week produced new shoots after soil transfer. We selected the discriminatory RAPD and ISSR markers to find the interspecific ginseng hybrid among its parents. The $F_1$ hybrid (Pgq) contained species specific 2 ginsenosides (ginsenoside Rf in P. ginseng and pseudoginsenosides $F_{11}$ in P. quinquefolius), and higher amount of other ginsenosides than its parents. Conclusion: Micropropagation of interspecific hybrid ginseng can give an opportunity for continuous production of plants.

Production of Triploid Somatic Hybrids Between Mandarin and Grapefruit through Electrofusion

  • Somatic hybrids were produced by electrofusion between embryogenic callus protoplasts of 'Syougun' mandarin and leaf protoplasts of grapefruit. Hybridity of the two plants was confirmed by leaf morphological characteristics and random amplified polymorphic DNA (RAPD) analysis. The cpDNA analysis using PCR-RFLP could not distinguish those of both parents. These plants showed normal growth and had chromosome number of 27. These unexpected triploid somatic hybrids might be derived from fused cells between diaploid protoplast of embryogenic calli and diploid protoplast of leaf, because polysomaty, a mixture of haploid cells and diploid cells was observed in the lactose medium-pretreated embryogenic calli of 'Syougun' by flow cytomehy analysis.

Doritaenopsis 체세포배의 내배수성 특성과 절편체의 내배수성 세포에 기인한 체세포변이의 발생 (Endoreduplication Pattern of Somatic Embryos and Variants Occurrence Affected by Pre-existed Endoreduplicated Cells in Doritaenopsis)

  • 일반적으로 체세포배를 경유한 대량증식은 증식효율이 높다. 그러나 종종 배수성 변이와 같은 체세포 변이를 야기하기도 한다. 본 연구에서는 Doritaenopsis hybrid의 체세포배 증식시 발생하는 배수성 변이체와 체세포배의 세포에 내재되어 있는 내배수화된 세포와의 관계를 구명하여 배수성 변이체 발생 원인을 밝히고자 실시되었다. 내배수성 분석결과 체세포배의 상부는 $2C{\sim}16C$의 DNA를 함유한 세포들로 구성되어 있는 반면, 중부와 하부 조직에는 $2C{\sim}32C$의 내배수성이 높은 세포들이 내재되어 있었다. 또한 체세포배 연령별로는 2주된 체세포배가 $2C{\sim}16C$ 정도의 세포로 구성되어 있는 반면 10주령의 체세포배는 $2C{\sim}64C$까지 고도로 내배수화가 진행된 세포들이 존재하였다. 이는 한 식물체에서도 부위, 연령에 따라 내배수성이 다를 수 있음을 보여주는 결과이다. 체세포배 부위중 내배수성이 낮은 상부와 높은 하부로 절단하여 각각의 절편체를 배양한 결과 배수성 변이체의 발생은 내배수성이 높은 하부를 배양하였을 때 3배 이상 높았다. 본 연구결과는 변수성 변이체의 발생원인이 내배수화된 세포에 있음을 보여주며 이를 기초로 Dtps. hybrid의 안정적인 증식체계를 구축하는데 기초가 되리라 생각된다.

Nicotiana tabacum과 Petunia inflata의 전기적 원형질체융합에 의한 속간 체세포 잡종의 생성 (Intergeneric Somatic Hybrids by Electrofusion of Protoplasts Between Nicotiana tabacum and Petunia inflata)

  • Leaf mesophyll protoplasts of Nicotiana tabacum (nitrate reductase deficient mutant) were fused with cell suspension protoplasts of albino Petunia inflata in an electric field. Hybrid cell colonies were selected for nitrate reductase proficiency and chlorophyll synthesis. Five hybrid plant lines, regenerated from the selected calli lines, were analysed by electrophoresis, number of chromosomes and morphological characters. Somtic hybrid plants showed both parent patterns in the isozymesof isoleucine aminopeptidase and esterase. The hybrids had the expected chromosome number of 62 and exhibited an intermediate floral morphology when compared with the parents, but plant height and leaf arrangement were similar to N. tabacum.

체세포 염색체 배가법을 이용한 백합 OA 종간 잡종의 불임극복과 후대획득 (Overcoming F1 Sterility of Intersectional OA Lily Hybrids by Somatic Chromosome Doubling and Production of Subsequent Progeny)

  • 본 연구는 나리의 종간잡종 육성체계를 확립하기 위한 기초자료를 얻고자 Oriental과 Asiatic 잡종 (OA-hybrid) 의 후대 획득에 필요한 실험을 수행하여 얻은 결과이다. 체세포 배수화를 통한 이질 4배체 OA-hybrid 3계통으로 화분발아 실험을 수행한 결과, 0-80% 범위로 계통간 차이가 현저하였다. 이질 4배체 OA-hybrid를 자방친 또는 화분친으로 사용하고 Asiatic 또는 Oriental hybrid와 교배시 후대 획득수에 차이가 있었는데 4배체 OA-hybrid를 화분친으로 이용하였을 때, 그리고 상대친을 Asiatic hybrid로 교배하였을 때 후대획득이 현저히 용이하였다. 그러나, 이질 4배체 F$_1$ OA-hybrid를 자방친으로 사용하거나 Oriental hybrid와 교배하면 후대 획득률이 현저히 저하되거나 또는 전혀 후대생산이 이루어지지 않았다. 이질 4배체 F$_1$ OA-hybrid와 교배시 상대친의 ploidy level은 4배체 보다는 2배체가 유리하였다.
