In this study, we investigated anticarcinogenic effects of extracts from Gloiopeltis tenax (GT). GT was extracted with methanol (GTM), which was then further fractionated into four fractions by using solvent fractionation method, affording methanol (GTMM), hexane (GTMH), butanol (GTMB) and aqueous (GTMA) soluble fractions. We determined the cytotoxic effects of these fractions on cancer cells by MTT assay. Among various fractions of GT, the GTMM showed the strongest cytotoxic effect at concentration of $150{\mu}g/mL$, displaying 95.97% on HepG2 cell lines and 93.64% on HT-29 cell lines, respectively. And, the anti-proliferative effect of GT was accompanied by a marked in increase of levels of Bad, Bax, Bok and Bak protein and activation of caspase-3, caspase-7 and PARP protein. Also, we observed quinone reductase (QR) induced effects in all fraction layers of GT on HepG2 cells. The QR induced effects of the GTMM and GTMB on HepG2 cells at concentration of $60{\mu}g/mL$ showing inductive indexes of 2.86 and 2.04 compared to the control value of 1.0.
For the effective utilization of diverse and abundant resource of seaweeds in Korea as a food protein supplment, extraction conditions of water, salt, and alkali soluble proteins were investigated in previous work(Ryu and Lee, 1977: Lee et al., 1977: Lee et al., 1978). The present study as a part of the serial work was thus aimed to find the conditions of isolation and purification of extracted proteins, and to evaluate the nutritional quality of the isolated seaweed proteins in terms of amino acid composition, chemical score, protein score, modified essential amino acid index(MEAAI), and in vitro digestibility presented as pepsin-pancreatin digest residue index (PPDRI). As for the isolation of extracted proteins, TCA treatment was more effective for the proteins from rhodophyceae and Chlorophyceae while the precipitation at isoelectric point was more desirable for Phaeophyceae proteins. In amino acid composition, water soluble protein fraction was superior to the other fractions in Porphyra suborbiculata whereas both water and alkali soluble fractions seemed to bo more benefitial for Enteromorpha linza and Ulva pertusa; the extraction with alcohol-alkali mixed solvent for Undaria pinnatifida and Sargassum fulvellum. Glutamic acid and aspartic acid content was particularly high in all protein fractions examined. The total amino acid content of Porphyra suborbiculata and Enteromorpha linza was almost equivalent to that of dried whole egg although the essential amino acid content was lower. A comparative analysis was made on the inedexes between raw seaweed powder and isolated protein. Chemical score of Porphyra suborbiculata and Ulva pertusa was approximately 35 and 56 in cafes of raw powder and isolated protein respectively while only 10 to 16 for raw powder of Undaria pinnatifida and Sargassum fulvellum and 30 to 35 for their isolated proteins. Protein score of all isolated proteins was in the range of 63 to 73 which indicates that isolated protein would be mere valuable than the fern of raw seaweed powder. Digestibility by means of PPDRI was found to be extremely low in case of raw powder but it could be doubled in case of isolated protein yielding 67 to 70 for Porphyra suborbiculata and Ulva pertusa.
In order to elucidate the behavior of bensulfuron-methyl, a sulfonylurea herbicide, in a soil/plant microecosystem, rice plants (Oryza sativa L.) were grown for 12 weeks in the specially made stainless steel pots (17cm I.D. $\times$ 10cm H.) containing two different paddy soils treated with fresh and 13-week-aged residues of [phenyl-$^{14}C$]bensulfuron-methyl, respectively. During the aging period, the mineralization to $^{14}CO_2$ from soil A (OM, 3.59%; CEC, 7.65 $cmol^+\;kg^{-1}$; texture, sandy clay loam) and B (OM, 1.62%; CEC, 4.51 $cmol^+\;kg^{-1}$; texture, sandy loam) amounted to 6.79 and 10.15% of the originally applied $[^{14}C]$bensulfuron-methyl, respectively. The amounts of $^{14}CO_2$ evolved from the soils with fresh residues were higher than those from the soils with aged residues. At harvest after 12-week growing, $^{14}C$-radioactivity absorbed and translocated into rice plants from soils A and B containing fresh residues of bensulfuron-methyl was 1.53 and 4.40%, while 4.04 and 6.37% in the two soils containing aged residues, respectively. Irrespective of aging and soil type, the $^{14}C$-radioactivity remaining in soil ranged from 80.41 to 98.87% of the originally applied $[^{14}C]$bensulfuron-methyl. The solvent extractability of tile soils was $39.25\sim70.39%$, showing the big differences among the treatments. Most of the nonextractable soil-bound residues of $[^{14}C]$bensulfuron-methyl were incorporated into the fulvic acid fraction$(61.32\sim76.45%)$. Comparing the microbial activity of the soils with rice plants grown with that of the soils without them, the former was $1.6\sim3.0$ times higher than the latter. However, it did not correlate with the $^{14}CO_2$ evolution.
Antimicrobial activity of 18 different traditional medicinal herbs extracts against Staphylococcus aureus was determined by a paper disc method. The Prunella vulgaris, Caesalpinia sappan and Rhus javanica extracts in 5 mg/ml, Poncirus trifoliata, Lonicera japonica and Seutellaria baicalensis extracts in 10 mg/ml and Schizandra chinensis, Alpinia katsumadai, Siegesbeckia orientalis extracts in 30 mg/ml showed a significant antimicrobial activity against Staphylococcus aureus. Minimum inhibitory concentrations of medicinal herbs extracts were in the range of $1{\sim}34\;mg/ml$ and $1{\sim}46\;mg/ml$, in the case of MeOH extracts and EtOH extracts, respectively. In addition, the antimicrobial activity of each solvent fraction was most significant with EtOAc layer. Optical density at 620nm after 24 hours incubation of Staphylococcus aureus in the presence of 100, 300 or 500 ppm of Caesalpinia sappan extract ranged from 0.02 to 0.03 compared to 0.4 in the absence of Caesalpinia sappan extract, indicating that growth of Staphylococcus aureus was significantly inhibited within 24 hours by the addition of at least 100 ppm of Caesalpinia sappan extract. Optical density at 620 nm after 24 hours incubation of Staphylococcus aureus in the presence of 300 ppm of Rhus javanica extract ranged from 0.02 to 0.03 compared to 0.4 in the absence of Rhus javanica extract, indicating that growth of Staphylococcus aureus was also significantly inhibited within 24 hours by the addition of at least 300 ppm of Rhus javanica extract. Optical density at 620 nm after 24 hours incubation of Staphylococcus aureus in the presence of 300 ppm of Seutellaria baicalensis extract ranged from 0.02 to 0.07 compared to 0.4 in the absence of Seutellaria baicalensis extract, indicating that growth of Staphylococcus aureus was also significantly inhibited within 24 hours by the addition of at least 300 ppm of Seutellaria baicalensis extract. In conclusion, these findings suggest that extracts from medicinal herbs may play important roles for antimicrobial activities against Staphylococcus aureus.
Entomopathogenic fungus Cordyceps militaris is famous for its medicinal efficacies. It has been reported to have various pharmacological activities such as anti-tumour, insecticidal, antibacterial, immunomodulatory and antioxidant. In this study, we investigated the effect of the extract of C. militaris (MPUN8501), which was identified by the analysis of the nucleotide sequences of 5.8S ribosomal RNA, on the function of liver. C. militaris powder was extracted using hot water extracts method as time, volume and temperature and using method as differential polarity of organic solvent. Each fraction was tested for the improvement of hepatic enzyme alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase (ALDH) activity. The BuOH extracts (CME) had highest activity which was used for the test of toxicity and efficacy of C. militaris. The enhancing effect of CME on the activity of ADH and ALDH was much more than medicine, drink, natural tea etc. Thus CME promoted the resolution of alcohol and acetaldehyde in rats, inducing recovery to normal condition rapidly. Furthermore, oral administration of CME effectively protected the carbon tetrachloride-induced acute hepatic injury as revealed by the hematological parameters (levels of sGOT and sGPT) and histological observation. CME was ascertained to be safe by regulatory toxicity studies of single dose toxicity and genotoxicity. These results suggest that CME would be useful for the maintaining normal hepatic activity as a functional health food.
This study was performed to investigate the effect of ethanol extract of Pimpinella brachycarpa(PBE) on chronically ethanol-induced hepatotoxicity in rat liver. Sprague-Dawley rats weighing 90-130 g were divided into 5 groups; normal group(NOR), ethanol(35%, 10 ml/kg) treated group(CON), PBE 200 mg/kg treated group(P1), PBE 200 mg/kg and ethanol treated group(P2), and PBE 400 mg/kg and ethanol treated group(P3). PBE was also fractionated by the following solvent: n-hexane, chloroform, ethylacetate and n-butanol. The antioxidative capacity of the n-hexane fraction was the highest among fractions and was similar to that of butylated hydroxytoluene(BHT). The body weight gain and feed intake of the rats were decreased by ethanol administration, but were gradually increased to the similar levels of the NOR group by administering PBE. The AST activity in serum elevated by ethanol was significantly decreased by administering the high dosage of PBE, but exerted no significant change on serum ALT activity. It was also observed that the hepatic activities of xanthine oxide(XO), catalase and glutathione peroxidase(GSH-Px) increased by ethanol were markedly decreased in the combined ethanol and PBE administered groups(P2 and P3), but not in the activity of superoxide dismutase(SOD) as compared with the CON group. The glutathione(GSH) contents were decreased by ethanol adminstration, however, increased after administering PBE. These results suggest that ethanol extract of Pimpinella brachycarpa has a possible positive effect on the liver function in hepatotoxicity-induced rats by ethanol administration.
In this study, a preliminary evaluation of the antioxidant and anti-inflammatory activity of the Ficus erecta var. sieboldii (Miq.) King (FES) leaf extract has been performed to assess its potential as a natural resource for food and medicinal materials. FES was extracted using 70% EtOH and then fractionated sequentially using n-hexane, $CH_2Cl_2$, EtOAc, and n-BuOH. To screen for antioxidant and anti-inflammatory agents effectively, the inhibitory effect of the FES extracts on the production of oxidant stresses (DPPH, xanthine oxidase, and superoxide) and pro-inflammatory factors (NO, iNOS, COX-2, $PGE_2$, IL-6, and $IL-1{\beta}$) in the murine macrophage cell line RAW 264.7 activated with lipopolysaccharide (LPS) was examined. Among the sequential solvent fractions of FES, the $CH_2Cl_2$ and EtOAc fractions showed decreased production of oxidant stresses (DPPH, xanthine oxidase and superoxide), and the hexane and $CH_2Cl_2$ fractions of FES inhibited the production of pro-inflammatory factors (NO, iNOS, COX-2, and $PGE_2$). The $CH_2Cl_2$ fraction also inhibited the production of pro-inflammatory cytokines ($TNF-{\alpha}$, IL-6, and $IL-1{\beta}$). These results suggest that FES has a significant effects on the production of oxidant stresses and pro-inflammatory factors and may be used a natural resource for antioxidant and anti-inflammatory agents.
