Fungi, 이스트, 그리고 streptomyces에서 L-phenylalanine ammonia-lyase의 활성도 비교 (L-phenylalanine ammonia-lyase activity of fungi, yeasts and streptomyces)

  • Microorganisms isolated from soil (150 strains), fungi (39 strains), yeasts (9 strains) and Streptomyces species (39 strains) were assayed for L-phenylalanine ammonia-lyase(PAL) activity. 17 strains of fungi and 46 strains of soil isolates were proved to produce PAL, Aspergillus panamensis, Penicillium varioti and 11 soil isolates showed comparatively large PAL activity. When PAL activity was assayed with cell-free extracts of these 13 strains and 7 strains of Rhodotorula and Rhodosporidium geni, Rhodosporidium toruloides (IFO 0559) showed the highest PAL activity with 0.333 units per g of the wet cell weight.

Taxonomical Studies on Red Yeasts in El-Minia City, Egypt

  • 227 red yeast strains were isolated from air (60 isolates), plant flowers (45 isolates), soil (40 isolates), water (37 isolates) and dairy products (45 isolates). On the basis of 33 different physiological and morphological properties, the isolated strains were assigned to 6 species belonging to 4 genera. Rhodotorula mucilaginosa and Cryptococcus albidus were the most dominant species among red yeasts of the air, plant flowers, water and dairy products, whereas Cryptococcus albidus and Rhodotorula glutinis were prevailed in soil. Cryptococcus laurentii was represented by considerable number of strains, whereas the other spesies were of low occurrence. Noteworthy was the isolation of 2 different groups of isolates belonging to Rhodotorula glutinis. These groups were differentiated from each other on the basis of rhamnose, cellobiose and arabinitol assimilation and growth at $37^{\circ}C$. Systematic position of Rhodotorula glutinis was discussed.

Description of unrecorded wild yeasts from soil in Republic of Korea under cold conditions

  • The purpose of this study was to isolate and identify wild yeasts from soil collected in Daegu City and Cheongyang County, Republic of Korea. Among 11 strains isolated in this study, nine strains were previously reported and two strains were unreported in Republic of Korea. To identify wild yeast strains, pairwise sequence comparisons of the D1/D2 region of the 26S rRNA gene sequence were done using Basic Local Alignment Search Tool (BLAST). The cell morphologies were observed by phase contrast microscope and assimilation test are done using API 20C AUX kit. All strains were assigned to the phylum Basidiomycota. Of the two unrecorded yeast strains, CY-9-10C belongs to the genus Mrakia (family Mrakiaceae, order Cystofilobasidiales, class Tremellomycetes) and PG3-4-10C belongs to the genus Slooffia (family Chrysozymaceae, order Microbotryomycetes incertae sedis, class Microbotryomycetes). Both strains had oval-shaped and polar budding cells. This research described the morphological and biochemical properties of the two unreported yeast species that had not officially reported in Korea.

Diversity of Yeasts Associated with Natural Environments in Korea

  • Biodiversity af yeasts in various natural environments including soils, swamps and plants was investigated. By molecular identification methods based on the partial sequences of 265 rDNA, 69 isolates were assigned to 44 taxa including 27 known species. The remaining 17 taxa could potentially form new species. All of them were classified into Ascomycota, Hymenomycetes, Urediniomycetes and Ustilaginomycetes. Ascomycetous and ustilaginomycetous yeasts were generally isolated from flower samples, and hymenomycetous and urediniomycetous yeasts were generally isolated from soil samples. Distribution of yeast groups exhibited geographical variation. Yeast biodiversity of root sail also varied according to the associated plant species.

Isolation and characterization of unrecorded yeasts species in the family Metschnikowiaceae and Bulleribasidiaceae in Korea

  • The goal of this study was to isolate and identify wild yeasts from soil samples. The 15 wild yeast strains were isolated from the soil samples collected in Pocheon city, Gyeonggi Province, Korea. Among them, four yeast stains were unrecorded, and 11 yeast stains were previously recorded in Korea. To identify wild yeasts, microbiological characteristics were observed by API 20C AUX kit. Pairwise sequence comparisons of the D1/D2 domain of the 26S rRNA were performed using Basic Local Alignment Search Tool(BLAST). Cell morphology of yeast strains was examined by phase contrast microscope. All strains were oval-shaped and polar budding and positive for assimilation of glucose, 2-keto-ᴅ-gluconate, N-acetyl-ᴅ-glucosamine, ᴅ-maltose and ᴅ-saccharose (sucrose). There is no official report that describes these four yeast species: one strain of the genus Kodamaea in the family Metschnikowiaceae and three strains of the Hannaella in the family Bulleribasidiaceae. Kodamaea ohmeri YI7, Hannaella kunmingensis YP355, Hannaella luteola YP230 and Hannaella oryzae YP366 were recorded in Korea, for the first time.

Occurrence of Yeasts in Cultivated Soils in El-Minia City, Egypt

  • Two-hundred two yeast strains were isolated from rhizosphere(87 strains) and nonrhizosphere(115 strains) areas of potato, maize, vegetable marrow, and cabbage plants. On the basis of 26 morphological and physiological properties, the isolated yeast strains were assigned to 9 genera and 15 species. Trichosporon beigelii, Kluyveromyces marxianus and Torulaspora delbrueckii were the dominant species. Cryptococcus humicolus and Candida tropicalis were represented by considerable numbers of strains. Of low occurrence were Saccharomyces cerevisiae and Candida blankii. Other yeast species were represented by single or two strains. Total counts of yeast cells per gram dry soil ranged from $1.1{\times}10^3$ to $6.6{\times}10^3$ in soil samples of rhizosphere areas and from $6.5{\times}10^2$ to $5.6{\times}10^3$ in soil samples of nonrhizosphere areas. Types of the tested plants affected not only the total counts of yeast cells but also spectra of yeast species. Relationships of age of potato plant, moisture contests of soil samples, and its pH values and total counts of yeast cells were discussed.

  • 대전광역시 유성구 복용 자연공원 부엽토와 주변 토양 85점을 채취하여 야생효모들을 분리, 동정한 결과, 야생효모 49종 91균주들이 분리, 동정되었다. 가장 많이 분리된 야생효모는 Cryptococcus속 균이었고 Cryptococcus aureus이 11균주로 가장 많았다. 또한 분리균 중 Kwoniella mangroviensis JSS0500, Candida corydali JSS0501, Candida bombi JSS0503, Candida multigemmis JSS0504, Cryptococcus dimennae JSS0506, Cryptococcus saitoi JSS0507, Cryptococcus victoriae JSS0508, Metschnikowia pulcherrima JSS0502, Papiliotrema aurea JSS0505, Debaryomyces vanrijia JSS0509, Occultifur externus JSS0510, Filobasidium floriforme JSS0511과 Yamadazyma scolyti JSS0512 등 13균주들이 국내 미기록 효모 균주들로 선별되었고 이들 야생효모들은 구형으로 출아법으로 영양증식하였다. Candida corydali JSS0501 등 7균주들이 vitamin을 함유하지 않은 YPD 배지에서도 잘 생육하였고 Cryptococcus victoriae JSS0508 등은 10% NaCl를 함유한 YPD 배지에서 생육하는 호염성 효모들이었다.

향신료 재배 토양과 주변 산림 토양으로부터 야생효모의 분리 및 국내 미기록 효모들의 특성 (Isolation and Characterization of Unrecorded Wild Yeasts Obtained from Soils of Spice Fields and Mountains)

  • 향신료 재배 토양 내에서 분리한 효모 중 이들의 분포와 국내 미기록 야생효모들의 특성을 조사한 결과, 충청남도 금산군 진산면의 향신료 재배토양에서 35종 58균주의 야생효모들을 분리, 동정하였고 Trichosporon moniliiforme균이 우점균으로 확인되었다. 또한, 대둔산 일대 산림토양에서 46균주의 야생효모들을 분리하였고 이들 향신료 토양들과 산림 토양 등에서 분리한 야생효모들 중 아직까지 국내에 보고되지 않은 Debaryomyces vindobonensis GHY31-3, Pseudozyma alboarmeniaca CD23-5, Filobasidium stepposum SFG1-4, Aureobasidium caulivorum HD17-5, Metschnikowia noctiluminum HD29-2, Papiliotrema anemochoreius NK2-12, Hannaella surugaensis DCH25-5, Microbotryozyma collariae DCH1-4, Rhodosporidiobolus lusitaniae DCH30-2, Wickerhamiella azyma DCH46-3등 10균주의 국내 미기록 효모들을 선별하였다. 이들 미기록 균주들은 구형-타원형으로 출아, 분열법으로 영양 증식하였다. Filobasidium stepposum SFG1-4균주를 포함하는 5균주들은 포자를 형성하였고 Pseudozyma alboarmeniaca CD 23-5등 3 균주들은 비타민을 첨가하지 않은 YPD 배지에서도 모두 생육하였다. Wickerhamiella azyma DCH 46-3는 20% 포도당을함유한 YPD 배지에서, Debaryomyces vindobonensis GHY31-3 균주는 5% NaCl을함유한 YPD 배지에서 잘 자라는 특성을 보였다. 또한, 이들의 무세포추출물들의 항산화 활성은 거의 없거나 10% 미만이었지만, 항통풍성 xanthine oxidase 저해활성은 Filobasidium magnum SFG1-3가 28.6%로 높게 나타났으며, 미백성 tyrosinase 저해활성은 Rhodotorula mucilaginosa GHP40-2 균주의 무세포추출물이 26%로 가장 높게 나타났다.

인삼과 당귀 재배 토양으로부터 야생효모들의 분리 및 미백성 Tyrosinase 저해활성 (Isolation and Tyrosinase Inhibitory Activity of Wild Yeasts Obtained from Soil in the Fields of Medicinal Plants, Ginseng and Korean Angelica)

  • 약용식물 재배지 토양의 야생효모 분포특성을 조사하고 이들 야생효모들의 산업적 응용성을 위한 자료를 얻고자 먼저 약용식물 집단 재배지인 충남 금산의 인삼과 당귀 재배지의 토양들을 2018년 3월에 50점을 채취하여 야생효모들을 분리, 동정하였다. 이들 시료들로부터 21종 43균주의 야생효모들이 분리되었고 인삼재배 논 토양에서는 Rhodotorula glutinis 균주가 6주, 당귀 재배 밭 토양에서는 Cyberlindnera saturnus 균주가 역시 6균주로 가장 많이 분리되었다. 이들 야생효모들을 이용하여 미백성 기능성 화장품 소재를 개발하고자 이들의 무세포추출물을 제조하여 tyrosinase 저해활성을 측정한 결과 저온성 야생효모인 Naganishia globosa G1-7가 28%의 가장 높은 저해활성을 보여 우수효모로 선발하였다.

충남 서천군 갈대밭 주변 토양에서 야생효모의 분리 및 미기록종 특성 (Isolation of Wild Yeasts from Soils of Reed Fields in Seocheon-gun County, Chungcheongnam-do, South Korea, and Characterization of Unrecorded Yeasts)

  • 충남 서천군 한산면 신성리 금강하구 갈대밭 토양의 야생효모 분포특성을 알아보기 위하여 갈대밭 토양 20점을 채취하여 야생효모들을 분리하여 총 11종 20균주의 야생효모를 동정하였다. Candida속의 효모가 10균주로 가장 많이 분리되었고, 그 중 Candida subhashii 균이 6균주로 가장 많았다. 분리된 균주 중에서 Candida tropicalis (Y5-1,Y5-2, Y6-1), Candida subhashii (J7-1, J7-3, J8-1, Y9-1, Y9-2, Y9-3), Lachancea thermotolerans (MJS 007, MJS 019) 균주 등 12균주들이 알코올 발효능을 보였다. Bullera japonica YJ10-1, C. subhashii J7-1, Kluyveromyces yarrowii YJ11-1, Ustilago shanxiensis Y10-1등 4균주들이 국내 미기록 효모 균주들로 최종 선별되었다. 이들 국내 미기록 균주들은 구형으로 Candida subhashii J7-1만이 자낭포자와 의균사를 형성하였다. Kluyveromyces yarrowii YJ11-1은 비타민을 함유하지 않은 배지에서도 생육하였고 Candida subhashii J7-1는 YM과 YPD 배지 및 5% NaCl을 함유한 YPD 배지에서 생장하지 않았다.
