Characteristics of Rhizome Rot incidence of Platycodon grandiflorus by Ridge width and Depth and Cultivation Period in the Seeding Place (이랑폭과 고랑깊이 및 재배년수에 따른 도라지뿌리썩음병 발생양상)

  • This study was carried out to investigate the characteristics of Rhizome rot incidence of Platycodon grandiflorus by cultivation period and ridge width and furrow depth. The three types of ridge width 0.8, 1.0, and 1.2 m and several levels of furrow depth and 6 levels cultivation period. This experiment was done in farmer's farm and the treatments were also classified in the same spot. In this investigation, ridge width, soil water content, soil hardness, and cultivation period were positively related with Rhizome rot incidence, however, furrow depth was negatively related with that. So this experiment could draw a conclusion : excess water damage and soil hardness could directly or indirectly effect on the Rhizome rot incidence, so cultivation method should be developed such as making underground ditch or cultivation in well draining soil for escaping excess waster damage.

Desalinization Effect of Off-season Crop Cultivation in Long-term Oriental Melon Cultivated Plastic Film House Soils (휴경기 후작물 재배에 의한 참외 장기연작 비닐하우스 토양의 제염 효과)

  • BACKGROUND: During the off-season, the cultivation of Chinese cabbage and water dropwort is often used to desalinize plastic film house soils. The objective of this study was to verify the effect of double-cropping systems on the salt removal in oriental melon cultivated plastic film house soils.METHODS AND RESULTS: Electrical conductivity (EC) and soluble salt contents were measured in soils collected from plastic film houses of oriental melon cultivation before and after the off-season crop cultivation. Also the same measurements were performed in the next oriental melon season to estimate the desalinization effect of double-cropping systems. During the cultivation of Chinese cabbage under open-field condition, ECeof surface soil was reduced from 6.0 to 0.8 dS/m. Double-cropping of water dropwort in flooded soil was also efficient in removing the salts accumulated during oriental melon cultivation. In the house soils where salts were removed during the off-season crop cultivation, soil ECewas maintained below 3 dS/m during the next oriental melon cultivation season.CONCLUSION: The off-season cropping under open-field or flooded condition was effective in desalinization of plastic film house soils. Since the salt removal effect is not expected to last for several years, the double-cropping system should be introduced every season to maintain soil EC below the critical level.

Growth, Vitamin C, and Mineral Contents of Sedum sarmentosum in Soil and Hydroponic Cultivation (토경 및 수경재배에서 돌나물의 생육, 비타민 C와 무기성분 함량 변이)

  • The growth, yield and quality of Sedum sarmentosum between soil and hydroponic cultivation were compared to produce high-quality shoots in summer season. The plants were collected from four places in Korea and cultivated in a plastic film house with 50% shading. A 1:1:1 mixtures of sand, upland soil and substrate (N-P-K: 330-220-400 $mg{\cdot}L^{-1}$) was used for soil cultivation, and a nutrient film technique (NFT) with Yamazaki-lettuce nutrient solution used for hydroponics. After 30 days of cultivation, plant height ranged from 18.4 to 21.8 cm and showed no significant difference between soil and hydroponic cultivation. Number of shoot, leaf length, leaf width, number of leaf, fresh weight, and dry weight significantly increased in hydroponics compared to those in soil cultivation. Fresh and dry weights in hydroponics were greater in 'Gunsan' and 'Wando' collections than those in other collections. Stem diameter and compression-force significantly increased in soil cultivation rather than hydroponics with significant differences among collections. However there were no significant differences in shoot bitterness and vitamin C content between cultivation methods or among the collections. Most mineral contents except Fe in hydroponics were higher than those in soil cultivation. K, Na, Zn contents in hydroponics were over 2 times higher than those in soil cultivation.

Effect of Plant Growth and Environmental Enhancement of Soils through Nanoparticle Application

  • Silver nanoparticles (AgNPs) have been manufactured in recent years and widely used in various fields. Reactive oxygen species (ROS), which occur in AgNPs, destroy cell membranes. It is widely accepted that ROS generated in this manner inhibit microorganisms growth and causes toxic effects, However, it does not affect cell membranes directly but positively affects growth in plants with cell walls. The nanoball used in this experiment is a new material that generates ROS stably and is used in aqueous solution. Results of this study indicate a 30% increase in yield of Ginseng mixed with culture soil. The analysis of soil condition after cultivation showed that the possibility of repetitive cultivation in soil mixed with Nanoball was high. This suggests that Nanoball is an antimicrobial active material due to the microbial / extermination effect of pathogenic microorganisms. Therefore, there may be potential applications in agricultural cultivation sites as a repetitive cultivation technology that reuses soil.

Effect of Soil Fumigation and Maize Cultivation on Reduction of Replant Failure in Ginseng (토양 훈증 및 녹비작물 재배가 인삼의 연작장해 경감에 미치는 영향)

  • Background: Dazomet are widely used as soil fumigant to solve soilborne problems, and the degradation intermediates are toxic to nematodes, fungi, bacteria, insects and weeds. Methods and Results: The effects of cultivation of green manure crop, maize before and after soil fumigation on the control of ginseng root rot disease were compared using soil where 6-years-old ginseng was harvested. Fumigant (dazomet) were used for soil fumigation in May and September, respectively. Maize was grown for soil management before and after soil fumigation. After May fumigation, the sowing date of maize was delayed by 15 days and thus its dry weight was decreased significantly. Maize cultivation after May fumigation increased pH but decreased EC, $NO_3$, $P_2O_5$, and K significantly. Maize cultivation after May fumigation decreased fungi population and the ratio of fungi and bacteria. Growth of 2-years-old ginseng was improved and the incidence of ginseng root rot was significantly decreased by maize cultivation after May fumigation. After harvesting 2-years-old ginseng, the population of Cylindrocarpon destructans was not different between treatment of May and September, but Fusarium solani showed a significant increase in September fumigation after maize cultivation. Conclusions: Maize cultivation after soil fumigation was effective in inhibiting ginseng root rot by the amendment of mineral composition and microorganism in fumigated soil.

Biochemical Characterization of a Psychrophilic Phytase from an Artificially Cultivable Morel Morchella importuna

  • Psychrophilic phytases suitable for aquaculture are rare. In this study, a phytase of the histidine acid phosphatase (HAP) family was identified in Morchella importuna, a psychrophilic mushroom. The phytase showed 38% identity with Aspergillus niger PhyB, which was the closest hit. The M. importuna phytase was overexpressed in Pichia pastoris, purified, and characterized. The phytase had an optimum temperature at $25^{\circ}C$, which is the lowest among all the known phytases to our best knowledge. The optimum pH (6.5) is higher than most of the known HAP phytases, which is fit for the weak acidic condition in fish gut. At the optimum pH and temperature, MiPhyA showed the maximum activity level ($2,384.6{\pm}90.4{\mu}mol{\cdot}min^{-1}{\cdot}mg^{-1}$, suggesting that the enzyme possesses a higher activity level over many known phytases at low temperatures. The phytate-degrading efficacy was tested on three common feed materials (soybean meal/rapeseed meal/corn meal) and was compared with the well-known phytases of Escherichia coli and A. niger. When using the same amount of activity units, MiPhyA could yield at least $3{\times}$ more inorganic phosphate than the two reference phytases. When using the same weight of protein, MiPhyA could yield at least $5{\times}$ more inorganic phosphate than the other two. Since it could degrade phytate in feed materials efficiently under low temperature and weak acidic conditions, which are common for aquacultural application, MiPhyA might be a promising candidate as a feed additive enzyme.

Effect of Nitrogen Application Levels on Nitrate Concentration in Soil Solution under Plastic Film House

  • This study was conducted to investigate investigated the effect of nitrogen fertilizer on nitrate concentration in soil solution and to determine the relationship between yield and nitrate concentration in soil solution for cucumber cultivation under plastic film house. Nitrogen as urea was applied at rates of 0, 120, 240, 360, and $480kg\;N\;ha^{-1}$ as an additional fertilizer by trickle irrigation during cucumber cultivation. Monitoring of nitrate concentration in soil solution was investigated using porous cups at 25 cm depth under soil surface. Nitrate concentration in soil solution increased with increasing the rate of additional nitrogen. Correlation coefficient between EC value and nitrate concentration was positive in soil and soil solution (p<0.05). An additional nitrogen of about $300kg\;ha^{-1}$ was shown the highest yield of cucumber, and improved yield by 5% compared to N recommendation of $240kg\;N\;ha^{-1}$. The highest yield was determined at nitrate concentration of $82mg\;L^{-1}$ in soil solution by regression equation ($Y=74.2+0.73X+0.000504X^2$, $R^2=0.629^*$). These results means indicate that nitrate concentration in soil solution would be useful method to rapid determination for additional nitrogen during cucumber cultivation under plastic film house.

Effect of Rye Cultivation for Reduction of Phytophthora Blight in Red Pepper Field (노지고추에서 고추역병 경감을 위한 녹비작물 호밀의 재배효과)

  • This study was carried out to evaluate the effect of rye as green manure crop on the improvement of soil environment and reduction of Phytophthora blight in red pepper of open field where Phytophthora blight occurred frequently. Soil physical properties such as bulk density and porosity were increased in rye cultivation. In addition, gaseous was increased but liquid was decreased compared with conventional cultivation. The analysis of phospholipid fatty acids extracted from soil showed that rye cultivation significantly increased relative abundance of microbial community and ratio of aerobic to anaerobic bacteria. Furthermore, ratio of saturated to unsaturated fatty acids and cyclo-fatty acids to precursor. the indicators of increasing in environmental stresses, were reduced in rye cultivated field. Occurrence of Phytophthora blight in rye cultivation was reduced 30.7% compared with conventional cultivation. These results suggest that rye cultivation in red pepper of open field where Phytophthora blight occurred can improve soil environment and reduce damage of Phytophthora blight.

Current Status of Ginseng Cultivation and Soil Characteristics of Northeastern Three Provinces in China

  • This study was conducted to improve the continuous techniques for international competitiveness of ginseng industry to Korea-China FTA negotiation and conclusion, and provide the basic information for ginseng industry development of Korea. It was carried out the visiting of the northeastern three provinces (Jilin, Liaoling and Heilongjang) in China for 3-year from 2014 to 2016 and observed the farmers' fields of ginseng cultivation with soil environmental status. The types of ginseng cultivation could be observed in small scales of 0.5~3.0 ha, in middle scales of 4.0~10.0 ha and in large scales of 30~700 ha with the kinds of imhasam, Chinese ginseng, Korean ginseng and western ginseng. Also ginseng was cultivated in newly reclaimed land of forest in two types of direct seeding and transplanting of ginseng seedlings. The field beds of ginseng growing were covered with vinyl films in arch design of 100~130 cm height and vinyl was painted in spraying with blue, green and yellow colours for shading. It was investigated in status of the physico-chemical properties of soils. The physical information on the field soils were silt loam, loam and sandy loam in soil textures, and some plain in low slope, some alluvial fan or local valley in forest of land topography. Soil pH ranged within 5.0~5.2, soil EC was $0.93{\sim}3.78dS\;m^{-1}$, organic matter was $37{\sim}35g\;kg^{-1}$, nitrate nitrogen $63{\sim}490mg\;kg^{-1}$, available $P_2O_5$ $55{\sim}163mg\;kg^{-1}$, and in exchangeable cations, K was 0.30~0.98, Ca was 6.5~14.0, Mg was $1.1{\sim}5.3cmol_c\;kg^{-1}$ in ranges. Farmers used the fertilizer for ginseng cultivation in 10~11 t of compost, $200{\sim}400kg\;ha^{-1}$ of complex fertilizer and $750kg\;ha^{-1}$ of oil cakes. The northeastern three provinces of China can use the newly lands with large areas of ginseng cultivation in soil sickness by continuous cropping. and the soil basic fertility is batter than that of Korean in standard guide of ginseng cultivation soil.

Comparison of Soil Physico-chemical and Microbial Characteristics in Soil of 'Niitaka' Pear Orchards between Organic and Conventional Cultivations (유기농법과 관행농법에 의해 재배한 '신고'배 과원 토양의 물리화학적 및 미생물학적 특성 비교)

  • Consumers' interest and government's support for the fruits rapidly increased organic fruit productions. This study was examined to compare the soil physicochemical and microbial properties of orchards soil in conventionally and organically management systems. Organic cultivation had lower soil bulk density, solid phase, and penetration resistance than the conventional cultivation. Soil pH and organic matter contents increased from March to August, and the values were greater in the organic cultivation than the conventional cultivation. Total nitrogen (N) and phosphorous concentrations decreased from March to August, and the organic soils had greater N but lower phosphorous concentrations than the conventional soils. Soil microbial carbon biomass increased 36% and 15% for organic and conventional cultivations, respectively, from March to August. Soil microbial N biomass was greater in June than March or August, and the organic cultivation had a greater biomass N compared to the conventional cultivation. Soil dehydrogenase and chitinase activities were greater in June than in March or August. ${\beta}$-glucosidase activity declined in both cultivations, while the phosphatase activity increased. Organic cultivation had greater enzyme activities in March, June, and August, except for the acid phosphatase activity in June.
