The capability of higher ascomyceteous fungi to cause typical soft-rot decay for wood under laboratory conditions is reviewed and discussed. Fungi tested were extremely active in the decomposition of timbers. Scanning electron micrographs illustrated typical soft-rot decay pattern of higher wood decay ascomycetes, with the exception of H. trugodes that caused white-rot decay. Most of the fungi tested could be grouped as soft-rot fungi that showed typical soft-rot type II. Hypha confined primarily to the resin canals in softwoods or vessel elements in hardwoods and spread tracheid to tracheid via pits of cell wall to cell wall with mechanical force.

The heartwood of cengal (Neobalanocarpus heimii) is known to have a high degree of decay resistance by virtue of its high extractive content. After 30 years in ground contact an utility pole of this tropical hardwood was found to be degraded only in the surface layers by cavity-forming soft rot fungi. The present work was undertaken 1) to characterize the degradation of cengal heartwood from the aspect of ultrastructure and chemistry and 2) to investigate the correlation between soft rot decay and its extractive microdistribution in wood tissues. The chemical analysis of cengal heartwood revealed the presence of a high amount of extractives as well as lignin. The wood contained a relatively high amount of condensed lignin and the guaiacyl units. Microscopic observations revealed that vessels, fibers and parenchyma cells (both ray and axial parenchyma) all contained extractives in their lumina, but in variable amounts. The lumina of fibers and most axial parenchyma were completely or almost completely filled with the extractives. TEM micrographs showed that cell walls were also impregnated with extractives and that pit membranes connecting parenchyma cells were well coated and impregnated with extractives. However, fungal hyphae were present in the extractive masses localized in cell lumina, and indications were that the extractives did not completely inhibit fungal growth. The extent of cell wall degradation varied with tissue types. The fibers appeared to be more susceptible to decay than vessels and parenchyma. Middle lamella was the only cell wall region which remained intact in all cell types which were severely degraded. The microscopic observations suggested a close correlation between extractive microdistribution and the pattern and extent of cell wall degradation. In addition to the toxicity to fungi, the physical constraint of the extractive material present in cengal heartwood cells is likely to have a profound effect on the growth and path of invasion of colonizing fungi, thus conferring protection to wood by restricting fungal entry into cell walls. The presence of relatively high amount of condensed lignin is also likely to be a factor in the resistance of cengal heartwood to soft rot decay.

A soft stem rot disease was observed on Chamaecereus silvestrii (Korean name: Sanchui), a scion of graft-cactus, in major growing areas of Suwon (National Horticulture Research Institute), Anseong, Eumseong, Cheonan, Daegu, and Goyang, Korea during 2000 and 2001. Typical symptoms were soft rots characterized by moist and watery decay of the whole cactus stem, which initiated as small water-soaked lesions and enlarged rapidly to the entire stem. The causal organism isolated from the infected stems was identified as Pectobacterium carotovorum subsp. carotovorum (Erwinia carotovora subsp. carotovora) based on its physiological and biochemical characteristics and confirmed by the cellular fatty acid composition and Biolog analyses. Artificial inoculation of the bacterium produced the same soft rot symptoms on the cactus stems, from which the same bacterium was isolated and identified. This is the first report of the P. carotovorum subsp. carotovorum in the graft-cactus C. silvestrii in Korea.
경기도 평택 희곡리 토탄층(약 5,700~5,900년전(前))에서 수침상태로 출토된 물푸레나무속 1종과 오리나무속 1종을 대상으로 전자현미경 관찰을 통하여 수종에 따른 분해정도와 형태를 조사하였다. 관찰결과 두 수종 모두 연부후와 박테리아에 의한 세포벽 공격형태와 분해형태는 거의 유사하였다. $S_2$층부터 복합세포간층 방향으로 점진적으로 불규칙하게 분해하는 침식형 박테리아와 목재세포벽을 분해하여 $S_2$층에 터널을 만드는 터널형 박테리아가 관찰되었다. 침식형 박테리아는 종종 연부후균에 의한 공동과 유사하지만 크기가 작은 공동을 형성하였다. 도관요소를 제외한 다른 세포들의 세포벽은 극심히 분해되었지만 세포벽의 외형은 수분으로 인해 그 고유한 형태를 유지하고 있었다. 해양천공충의 식흔 흔적이 발견되지 않고 연부후균과 박테리아에 의한 분해 형태만 발견되어 평택희곡리 토탄지역 형성 당시인 5,700~5,900여년전이 온난화에 의한 홀로세 기후최적기 기간중이기 때문에 서해안 해수면 상승에 의해 해수에 침수되었을 것이라는 가설은 사실이 아닌 것으로 해석되었다.
감자 괴경에 타박상을 입혔거나 세균현탁액을 주입했을 때 낮은 water potential(-6.46 bar) 보다 높은 water potential (-6.06 bar)에서 더 쉽게 부패를 초래하였으며, 높은 water potential을 가진 괴경과 낮은 water potential을 가진 괴경 사이에 있어서의 부패의 진전에는 큰 차이를 발견할 수 있었으며, 현탁액의 농도의 종류에 따른 괴경의 연부병의 이병정도는 높은 water potential에서 높았으며, 세균 현탁액 농도의 $ED_{50}$은 높은 water potential에서 $10^{8.5}\;cells/ml$이며 낮은 water potential에 서는 $100^{9.8}\;cells/ml$이었으며, 높은 water potential과 낮은 water potential $ED_{50}$에서는 작은 차이를 인정할 수 있었다. 이 시험에서 감자는 상처나 타박상이 나지 않도록 다루어야 하며 수확후 충분히 건조하여야 오랜 운반과 저장중에 연부병 발생을 크게 줄일 수 있다는 것을 나타낸다.
화재를 예방하고자 방염 처리 한 일부 고건축재에서 발견된 미생물에 의한 고목재의 분해를 전자현미경을 사용하여 관찰하였다. 방염 처리한 고건축 부재는 2차벽 중층에 마이크로배열 방향과 동일한 동공을 가지고 있는 전형적인 연부후를 보였다. 또한 주사형 및 투과형 전자현미경을 사용하여 관찰한 결과 일부 세포벽은 세균에 의해 분해된 것으로 나타났다. 방염 처리한 고건축재에서 나타난 이같은 부후형태는 수분으로 포화된 수침 고목재에서 나타나는 전형적인 부후 형태이다. 부후 형태로부터 본 연구에 사용된 고건축 부재는 방염 처리 후 흡수성이 급격히 증가하였으며, 이같은 함수율의 증가는 사용된 방염제 때문인 것으로 생각된다. 본 연구 결과 사찰 및 궁질 등 고건축물에 사용하고 있는 방염제에 대한 보다 더 체계적이고 심도있는 연구가 필요함을 보여주었으며 아울러 방염제가 목재의 흡수성과 강도에 미치는 영향에 대한 연구가 뒤따라야함을 제시하였다. 본 논문은 또한 기상 열화에 의한 일부 목재의 표층부에 나타난 중간층의 분해에 대해서도 논의하였다.
This study was conducted to evaluate the resistance of heat-treated wood using the catalyst to decay caused by fungi for sapwood and heartwood of two tree species, Korean red pine (Pinus densiflora) and Japanese larch (Larix kaempferi), respectively. Wood samples were immersed for 10 min in sulfuric acid (7.5%) and then heat-treated at $130^{\circ}C$ for 90 min. Fomitopsis palustris, a brown-rot fungus, was used to examine the decay resistance of Korean red pine and Japanese larch wood. Weight and density of wood from the all conditions increased after heat treatment using the catalyst. Weight loss after decay resistance test was also dropped with a heat treatment. The lowest weight loss indicated at heat-treated heartwood of Japanese larch. Heat treatment using the catalyst effectively increased the resistance of wood to decay caused by fungi.
Antifungal activities of natural substrances from Eucalyptus darlympleana, E. globules, E. gunnii and E. unigera were evaluated against postharvest pathogens of kiwifruits, Botrytis cinerea, Botryosphaeria dothidea, and Diaporthe actinidiae, to screen effective natural substances as an alternative to chemical fungicides. Methanol extract of the Eucalyptus trees showed strong antagonistic activity against the pathogenic fungi. Among them, E. unigera and E. darlympleana effectively inhibited mycelial growth of the pathogens. For chemical identification of the antifungal substances, the methanol extract of E. darlympleana leaves was successively partitioned with $CH_2Cl_2$, EtOAc, n-BuOH and $H_2O$. Among the fractions, $CH_2Cl_2$ and n-BuOH showed strong inhibitory activity of mycelial growth of the fungi. Five compounds were isolated from EtOAc and n-BuOH fractions subjected to $SiO_2$ column chromatography. Two phenolic compounds(gallic acid and 3,4-dihydroxybenzoic acid) and three flavonoid compounds(quercetin, quercetin-3-O-$\alpha$-L-rhamnoside, quercetin-3-O-$\beta$-glucoside) were identified by $^1H$-NMR and $^{13}C$-NMR spectroscopy. Among them, only gallic acid was found to be effective in mycelial growth and spore germination of B. cinerea at relatively high concentrations. The results suggest that gallic acid can be a safer and more acceptable alternative to current synthetic fungicides controlling soft rot decay of kiwifruit during postharvest storage.
고구마 저장중의 성분변화(成分變化), 연부병(軟腐病)에 대한 감수성(感受性) 및 연부병균포자(軟腐病菌胞子)의 치사(致死)에 미치는 방사선(放射線)($^{60}Co-{\gamma}$)의 영향을 조사하여 다음과 같은 결과를 얻었다. 1) $^{60}Co-{\gamma}$선조사(線照射)에 의하여 고구마의 수용성당(水溶性糖)은 증가하고 환원형(還元型) ascorbic acid는 현저히 감소하였다. 2) 조사선량(照射線量)이 높을수록 直後)의 호흡율(呼吸率)은 증가하나 시간이 경과됨에 따라 고선량(高線量)의 조사구(照射區)에서는 조직(組織)의 갈변(褐變)과 아울려 호흡율(呼吸率)이 감소하였다. 3) 고선량(高線量)($2.0{\times}10^5$ rad 이상)의 조사(照射)는 고구마 저장(貯藏)중 냉해(冷害)의 영향과 같이 연부병균(軟腐病菌)의 감염율(感染率)을 증가시켰다. 4) 고구마연부병균(軟腐病菌)인 Rhizopus nigricans 포자(胞子)의 평균치사선량(平均致死線量)은 $3{\times}10^5$ rad이며 $1.25{\times}10^5$ rad에서 완전사멸(完全死滅)되었다.
2011년 8월 경상북도 봉화군 재산면 농가포장 5년생 인삼포장에서 잎이 푸른색을 띈 채 급격하게 마르는 증상이 발견되었다. 잎이 시든 직후 인삼의 뿌리를 채취하였으나, 뿌리나 줄기에서 특별한 병징은 없었으며, 뿌리의 수분이 약간 빠져나간 상태였다. 이러한 증상이 발생된 지 10일이 경과된 뿌리에서는 뿌리가 물러지면서 썩는 증상이 나타났다. 이러한 병징과 건전부의 경계부위에서 세균이 분리되었으며, 이 세균에 의한 뿌리썩음병이 의심되었다. 세균을 배양한 후 건전 인삼에 접종하자, 동일한 썩음 증상이 발현되었으며, 병반으로부터 동일한 세균이 재분리되었다. 이 세균을 BioLog system에 의한 탄소원 이용여부, Vitek 2 system을 이용한 생화학적 반응, GC-MIDI Sherlock system을 이용한 지방산 조성, 16S rRNA 염기서열을 분석한 결과, 모두 Serratia plymuthica로 동정되었다. 이에 따라 인삼 뿌리에 발생하는 이 병을 Serratia plymuthica에 의한 인삼 세균뿌리썩음병으로 명명하여 보고하고자 한다.
