SUMO Proteins are not Involved in TGF-${\beta}1$-induced, Smad3/4-mediated Germline ${\alpha}$ Transcription, but PIASy Suppresses it in CH12F3-2A B Cells

  • TGF-${\beta}$ induces IgA class switching by B cells. We previously reported that Smad3 and Smad4, pivotal TGF-${\beta}$ signal-transducing transcription factors, mediate germline (GL) ${\alpha}$ transcription induced by TGF-${\beta}1$, resulting in IgA switching by mouse B cells. Post-translational sumoylation of Smad3 and Smad4 regulates TGF-${\beta}$-induced transcriptional activation in certain cell types. In the present study, we investigated the effect of sumoylation on TGF-${\beta}1$-induced, Smad3/4-mediated $GL{\alpha}$ transcription and IgA switching by mouse B cell line, CH12F3-2A. Overexpression of small ubiquitin-like modifier (SUMO)-1, SUMO-2 or SUMO-3 did not affect TGF-${\beta}1$-induced, Smad3/4-mediated $GL{\alpha}$ promoter activity, expression of endogenous $GL{\alpha}$ transcripts, surface IgA expression, and IgA production. Next, we tested the effect of the E3 ligase PIASy on TGF-${\beta}1$-induced, Smad3/4-mediated $GL{\alpha}$ promoter activity. We found that PIASy overexpression suppresses the $GL{\alpha}$ promoter activity in cooperation with histone deacetylase 1. Taken together, these results suggest that SUMO itself does not affect regulation of $GL{\alpha}$ transcription and IgA switching induced by TGF-${\beta}1$/Smad3/4, while PIASy acts as a repressor.

A New Dual Connective Network Resource Allocation Scheme Using Two Bargaining Solution (이중 협상 해법을 이용한 새로운 다중 접속 네트워크에서 자원 할당 기법)

  • In order to alleviate the limited resource problem and interference problem in cellular networks, the dual connectivity technology has been introduced with the cooperation of small cell base stations. In this paper, we design a new efficient and fair resource allocation scheme for the dual connectivity technology. Based on two different bargaining solutions - Generalizing Tempered Aspiration bargaining solution and Gupta and Livne bargaining solution, we develop a two-stage radio resource allocation method. At the first stage, radio resource is divided into two groups, such as real-time and non-real-time data services, by using the Generalizing Tempered Aspiration bargaining solution. At the second stage, the minimum request processing speeds for users in both groups are guaranteed by using the Gupta and Livne bargaining solution. These two-step approach can allocate the 5G radio resource sequentially while maximizing the network system performance. Finally, the performance evaluation confirms that the proposed scheme can get a better performance than other existing protocols in terms of overall system throughput, fairness, and communication failure rate according to an increase in service requests.

Heterologous Expression of Yeast Prepro-$\alpha$-factor in Rat $GH_3$ Cells

  • Yeast pheromone a-factor is a 13-amino acid peptide hormone that is synthesized as a part of a larger precursor, prepro-$\alpha$-factor, consisting of a signal peptide and a proregion of 64 amino acids. The carboxy-terminal half of the precursor contains four tandem copies of mature $\alpha$-factor. To investigate the molecular basis of intracellular sorting, proteolytic processing, and storage of the peptide hormone, yeast prepro-$\alpha$-factor precursors were heterologously expressed in rat pituitary $GH_3 cells. When cells harboring the precursor were metabolically labeled, a species of approximately 27 kD appeared inside the cells. Digestion with peptide: N-glycosidase F (PNG-F) shifted the molecular mass to a 19 kD, suggesting that the 27 kD protein was the glycosylated form as in yeast cells. The nascent polypeptide is efficiently targeted to the ER in the $GH_3 cells, where it undergoes cleavage of its signal peptide and core glycosylation to generate glycosylated pro-a-factor. To look at the post ER intracellular processing, the pulse-labelled cells were chased up to 2 hrs. The nascent propeptides disappeared from the cells at a half life of 30 min and only 10-25% of the newly synthesized, unprocessed precursors were stored intracellularly after the 2 h chase. However, about 20% of the pulse-labeled pro-$\alpha$-factor precursors were secreted into the medium in the pro-hormone form. With increasing chase time, the intracellular level of propeptide decreased, but the amount of secreted propeptide could not account for the disappearance of intracellular propeptide completely. This disappearance was insensitive to lysosomotropic agents, but was inhibited at $16^{circ}C or 20^{\circ}C$, suggesting that the turnover of the precursors was not occurring in the secretory pathway to trans Golgi network (TGN) or dependent on acidic compartments. From these results, it is concluded that a pan of these heterologous precursors may be processed at its paired dibasic sites by prohormone processing enzymes located in TGN/secretpry vesicles producing small peptides, and that the residual unprocessed precursors may be secreted into the medium rather than degraded intracellularly.

  • Nitric oxide(NO) has recently been shown to play an important role on central nervous system(CNS) function in mammals. It is synthesized from L-arginine by the enzyme NO synthase. In this study, we examined this enzyme's existence in CNS of rainbow trout(Oncorhynchus mykiss) and described the forms of microglia. astrocytes. and oligodendrocytes. Two forms of microglia are distributed in CNS. one resembling their mammalian counterpart(large microglia : LM). and the other comprising small microglia(SM) with very little cytoplasm. CNS contained astrocytes of a distinct type which form reticular network, but lack connections to capillaries. The oligodendrocyte was generally a much denser cell than the astrocyte. We have detected NOS($1.04{\pm}0.12\;pg/min/mg$) from rainbow trout CNS. It could be inhibited reversibly or irreversibly by $N^{G}MMA$ and EGTA. These result suggest that the formation of NO from L-arginine in CNS is calcium-dependent and a pathway of early evolutionary orgin.

  • Up to this day, mobile communications have evolved rapidly over the decades, mainly focusing on speed-up to meet the growing data demands of 2G to 5G. And with the start of the 5G era, efforts are being made to provide such various services to customers, as IoT, V2X, robots, artificial intelligence, augmented virtual reality, and smart cities, which are expected to change the environment of our lives and industries as a whole. In a bid to provide those services, on top of high speed data, reduced latency and reliability are critical for real-time services. Thus, 5G has paved the way for service delivery through maximum speed of 20Gbps, a delay of 1ms, and a connecting device of 106/㎢ In particular, in intelligent traffic control systems and services using various vehicle-based Vehicle to X (V2X), such as traffic control, in addition to high-speed data speed, reduction of delay and reliability for real-time services are very important. 5G communication uses high frequencies of 3.5Ghz and 28Ghz. These high-frequency waves can go with high-speed thanks to their straightness while their short wavelength and small diffraction angle limit their reach to distance and prevent them from penetrating walls, causing restrictions on their use indoors. Therefore, under existing networks it's difficult to overcome these constraints. The underlying centralized SDN also has a limited capability in offering delay-sensitive services because communication with many nodes creates overload in its processing. Basically, SDN, which means a structure that separates signals from the control plane from packets in the data plane, requires control of the delay-related tree structure available in the event of an emergency during autonomous driving. In these scenarios, the network architecture that handles in-vehicle information is a major variable of delay. Since SDNs in general centralized structures are difficult to meet the desired delay level, studies on the optimal size of SDNs for information processing should be conducted. Thus, SDNs need to be separated on a certain scale and construct a new type of network, which can efficiently respond to dynamically changing traffic and provide high-quality, flexible services. Moreover, the structure of these networks is closely related to ultra-low latency, high confidence, and hyper-connectivity and should be based on a new form of split SDN rather than an existing centralized SDN structure, even in the case of the worst condition. And in these SDN structural networks, where automobiles pass through small 5G cells very quickly, the information change cycle, round trip delay (RTD), and the data processing time of SDN are highly correlated with the delay. Of these, RDT is not a significant factor because it has sufficient speed and less than 1 ms of delay, but the information change cycle and data processing time of SDN are factors that greatly affect the delay. Especially, in an emergency of self-driving environment linked to an ITS(Intelligent Traffic System) that requires low latency and high reliability, information should be transmitted and processed very quickly. That is a case in point where delay plays a very sensitive role. In this paper, we study the SDN architecture in emergencies during autonomous driving and conduct analysis through simulation of the correlation with the cell layer in which the vehicle should request relevant information according to the information flow. For simulation: As the Data Rate of 5G is high enough, we can assume the information for neighbor vehicle support to the car without errors. Furthermore, we assumed 5G small cells within 50 ~ 250 m in cell radius, and the maximum speed of the vehicle was considered as a 30km ~ 200 km/hour in order to examine the network architecture to minimize the delay.

  • The objective of this study is to conduct a thermal-hydraulic analysis on the spent fuel pool and to evaluate a parametric effect for the thermal-hydraulic analysis of spent fuel pool. The selected parameters are the Reynolds Number and the gap flow through the oater gap between fuel cell and fuel bundle. The simplified flow network for a path of fuel cells is used to analyze the natural circulation phenomenon. In the flow network analysis, the pressure drop for each assembly from the entrance of the fuel rack to the exit of the fuel assembly is balanced by the driving head due to the density difference between the pool fluid and the average fluid in each spent fuel assembly. The governing equations ore developed using this relation. But, since the parameters(flow rate, pressure loss coefficient, decay heat, density)are coupled each other, iteration method is used to obtain the solution. For the analysis of the YGN 3&4 spent fuel rack, 12 channels are considered and the inputs such as decay heat and pressure loss coefficient are determined conservatively. The results show the thermal-hydraulic characteristics(void fraction, density, boiling height)of the YGN 3&4 spent fuel rack. There occurs small amount of boiling in the cells. Fuel cladding temperature is lower than 343.3$^{\circ}C$. The evaluation of parametric effect indicates that flow resistances by geometric effect are very sensitive to Reynolds number in the transition region and the gap flow is negligible because of the larger flow resistance in the gap flow path than in the fuel bundle.

Development of Fender Segmentation System for Port Structures using Vision Sensor and Deep Learning (비전센서 및 딥러닝을 이용한 항만구조물 방충설비 세분화 시스템 개발)

  • As port structures are exposed to various extreme external loads such as wind (typhoons), sea waves, or collision with ships; it is important to evaluate the structural safety periodically. To monitor the port structure, especially the rubber fender, a fender segmentation system using a vision sensor and deep learning method has been proposed in this study. For fender segmentation, a new deep learning network that improves the encoder-decoder framework with the receptive field block convolution module inspired by the eccentric function of the human visual system into the DenseNet format has been proposed. In order to train the network, various fender images such as BP, V, cell, cylindrical, and tire-types have been collected, and the images are augmented by applying four augmentation methods such as elastic distortion, horizontal flip, color jitter, and affine transforms. The proposed algorithm has been trained and verified with the collected various types of fender images, and the performance results showed that the system precisely segmented in real time with high IoU rate (84%) and F1 score (90%) in comparison with the conventional segmentation model, VGG16 with U-net. The trained network has been applied to the real images taken at one port in Republic of Korea, and found that the fenders are segmented with high accuracy even with a small dataset.

The Effects of the Tumor Mass Size Inoculated in Immunologically Competent Balb/c Mice on Delayed-type Hypersensitivity Response (정상 면역 생쥐에 접종된 암세포주의 종괴 형성이 숙주의 지연성과민반응에 미치는 영향)

  • Background: Based on outstanding progresses in animal experiments, vaccines for some human tumors have been developed. However, clinical effects of these vaccines have been far below than expected. This discrepancy might come from differences between animal models and human patients with respect to immunocompetency. The immune status of mice after tumor inoculation has not been well studied, which make us cautious in interpreting and applying the results from mice to human. We evaluated cell-mediated immune responses in mice after tumor cell inoculation. Methods: Mice were inoculated with TA3Ha, CT26, or 4T1. Delayed-type hypersensitivity (DTH) responses were induced 2-4 weeks after inoculation using 2,4-dinitro-1-fluorobenzene as an antigen. The relationships between the severity of DTH responses and the duration of tumor inoculation or the size of tumor mass were analyzed. Results: In T A3Ha groups, DTH response was elevated 2 weeks after inoculation, but depressed after 4 weeks, compared to the control group. When analyzed based on the sizes of tumor masses elicited, DTH responses were inversely related to the mass size, especially in those greater than 10 mm in diameter. In CT26 groups, while the duration after inoculation did not affect the severity of DTH responses, those with large mass showed depressed responses regardless the duration of inoculation. 4T1 cells grew so slowly that the size of tumor mass was small even 4 weeks after inoculation, and this group showed much higher DTH responses compared to that of tumor-free group. Conclusion: At least in an experimental setting where tumor model was induced by inoculating tumor cell lines into immunologically competent mice, the host immune response was elevated in early stage, and then depressed in late stage when the mass grew over a critical size.

Potentiality of Anti-idiotypic Antibodies Mimicking GD2 to Induce Cellular Immunity (GD2 유사 항이디오타입 항체의 세포면역 유발 잠재성)

  • Background: Disialoganglioside GD2 is a tumor-associated antigen that is overexpressed on tumor cells of neuroectodermal origin, such as melanoma, small cell lung carcinoma and neuroblastoma. Immunity against GD2 has anti-tumor activities, but GD2 is poorly immunogenic. Anti-idiotypic antibodies that mimic GD2 may induce more effective immune responses than GD2 antigen itself, because they are protein antigens and are known to be able to break immune tolerance. In our previous study, we produced anti-idiotypic antibodies mimicking GD2 (3A4 and 3H9), which induced humoral immunity. However, cellular immunity is essential to eradicate tumor cells in vivo as well as humoral immunity. In the present study, we investigated whether these anti-idiotypic antibodies 3A4 and 3H9 could induce cellular immunes responses. Methods: BALB/C mice were immunized with anti-idiotypic antibody 3A4 or 3H9, or normal mouse IgG as a negative control. Lymphoproliferative responses, cytokine production responses, and delayed-type hypersensitivity reactions were measured in mice immunized with the anti-idiotypic antibodies. Results: Both the anti-idiotypic antibody 3A4 and 3H9 induced GD2-specific lymphoproliferative responses and $IFN-{\gamma}$ production of lymph node lymphocytes in BALB/C mice. Only anti-idiotypic antibody 3H9 induced significant GD2-specific delayed-type hypersensitivity in the mice. Conclusion: These results show that anti-idiotypic antibodies 3A4 and 3H9 have the potentiality of inducing GD2-specific cellular immune responses that cannot be induced by the native antigen GD2 itself.

Development of High-Efficient Small Euel Cells : I. Synthesis of Organic-Inorganic Nanocomposite Electrolyte Membranes (고효율 소형 연료전지의 개발 : I.유기-무기 나노복합 전해질막의 합성)

  • New fast proton-conducting organic-inorganic nanocomposite membranes were successfully fabricated using polymer matrix obtained through proper oxidation of thiol ligands in (3-Mercaptopropyl) trimethoxysilane (MPTS) and hydrolysis/condensation reaction of (3-glycidoxypropyl) trimethoxysilane (GPTS). The obtained nanocomposite membranes showed relatively hirh proton-conductivity over $10^{-2}S/cm$ at $ 25^{circ}C$. The proton conductivities of the fabricated composite membranes increased up to $3.6{\times}10^{-1}$ S/cm cm by increasing temperature and relative humidity to $70^{circ}C$ and 100 $100RH\%$. The high proton conductivity of the composites Is due to the proton conducting path through the GPTS-derived 'pseudo-polyethylene oxide 'network in which sulfonic acid ligands work as a proton donor.
