Antimutagenic and Antioxidative Effects of Water Dropwort and Small Water Dropwort (미나리와 돌미나리의 돌연변이 유발 억제작용과 항산화 효과)

  • The purpose of this study is to research the antimutagenic and antioxidative effects of water dropwort and small water dropwort by Ames test and by measuring malondialdehyde(MDA) production. Water dropwort and small water dropwort were extracted with methanol and then further fractionated to hexane, chloroform, ethyl acetate, butanol and water, stepwise. The methanol extracts from both samples reduced the mutagenicities by aflatoxin $B_1(AFB_1)$ in Salmonella typhimurium TA 98. The production of MDA also decreased when the methanol extracts were added to the system. The hexane, chloroform and ethyl acetate fractions revealed higher antimutagenic activities against $AFB_1$ than the butanol and water fractions. Among the five fractions, the ethyl acetate fraction showed the highest level of antioxidant activity. From the results of the experiment, water dropwort and small water dropwort seem to be good antimutagenic and antioxidative sources of food.

Effects of Phytol and Small Water Dropwort Extract on the T Subset in the Sarcoma 180-Transplanted Mice (Phytol과 들미나리추출물이 Sarcoma 180마우스의 T Subset에 미치는 효과)

  • Several studies have shown that phytol isolated from perilla leaf and small water dropwort (Oenanthe javanica (BL) D.C., wild type) extract reveal antirumor activities. In the present study we investigated the effect of phytol and the small water dropwort extract on the T subset in the sarcoma 180-transplanted mice in order to elucidate the immunological mechanism of antiturmor activity of these substances. The results obtained from the experiment were as follows : When phytol was injected into the sarcoma-180 transplanted mice (control), the levels of T cell and T subset by inoculation of the tumor cells were enhanced, but that of T cells in case of small water dropwort extract was similar to the control. Asialo GM1$^{+}$ cells were increased when phytol or small water dropwort extract with injected into tumor-transplanted mice. In normal mice the number of asialo GM1$^{+}$ cells increased with phytol injection and decreased with small water dropwort extract injection. L3T4$^{+}$/Lyt-2$^{+}$cell ratios were decreased when phytol was injected into tumor-transplanted mire, but increased in case of small water dropwort extract injection. In normal mice the ratios showed large decreases with phytol or small water dropwort extract injection. These results indicate that phytol or small water dropwort extract ran activate the proliferation of natural killer cells that are effector cells in tumor-bearing mice.

The Antimutagenic Activity and the Growth Inhibition Effect of Cancer Cells on Methanol Extracts from Small Water Dropwort (돌미나리 메탄올 추출물의 항돌연변이 작용과 암세포증식 억제효과)

  • The study was carried out to evaluate the antimutagenic and anticancer effects of small water dropwort. The methanol extracts from small water dropwort significantly reduced the mutagenicity induced by aflatoxin $B_1\;(AFB_1)$ and N-methyl-N'-nitro-N-nitrosoguanidine(MNNG) in Salmonella typhimutium TA 100. Also, the methanol extracts inhibited the growth of AZ-521 human gastric cancer cells and HT-29 colon cancer cells. The chloroform fraction from methanol extracts of small water dropwort inhibited $40\;to\;80\%$ of the mutagenicity by $AFB_1$ in Sal. typhimurium TA 100 by the addition of 2.5 to $10\%$. To separate active compounds, the chloroform fraction was subjected to column chromatography on a silica gel and separated into five fractions. Among the five fractions, fraction 4 showed the highest antimutagenic effect against $AFB_1$ and an anticancer effect in the HT-29 colon cancer cell. As the result of the analysis in GC-MS, 1-napthalene carbonitrile, 5,6,7,8-tetrahydrol and benzene, 1,1'-(1,4-pentadiene-1,5-diyl) bis-,(E,E) were identified potentially from fraction 4.

  • This study was conducted to examine the stages showing the antimutagenic effects on the microbial mutation by addition of the juice extracted from small water dropwort. It was not able to find out the signal showing the genic derepression or change of gene repair system by addition of the juice. And it was hardly possible to expect the conversion of 2-AF to inactive form by the juice. however the longer 2-AF and S-9 mix were contacted before addition of the juice, the stronger the microbial mutagenisity of 2-AF was, and after addition of the juice, the mutagenicity was decreased rapidly. It seems that some components in the juice act as inhibitor of a enzyme in S-9 mix, and block the conversion of 2-AF to the ultimate mutagen.

Inhibitory Effect of Green-Yellow Vegetables on the Mutagenicity in Salmonella Assay System and on the Growth of AZ-521 Human Gastric Cancer Cells (녹황색 채소류의 돌연변이유발 억제 및 AZ-521 위암세포의 성장 저해효과)

  • The antimutagenic effect of green-ye1low vegetables on the mutagenicities induced by N-methyl-N'-nitro-N-nitrosoguanidine(MNNG) and N-nitrosodimethylamine (NDMA) in Salmonella assay system and also their inhibitory effect on AZ-521 human gastric cancer cells were studied. Twenty-four items from twenty six kinds of vegetables(92%) revealed antimutagenic activity toward MNNG (p< 0.0l, 0.05). Perilla leaf, Korean cabbage, cauliflower, lettuce, mustard leaf, water dropwort, small water dropwort, carrot and burdock inhibited the mutagenicity more than 80%. The methanol extracts of the vegetables also showed the antimutagenic activity toward NDMA (p< 0.01, 0.05). Especially, perilla leaf, kale, soybean sprout and onion inhibited more than 80% of the NDMA induced mutagenicity in S. typhimurium TA100. Small water dropwort and perilla leaf exhibited the strong inhibitory effect (97~100%) on the growth of the AZ-521 human gastric cancer cells. Soybean sprout, water dropwort, broccoli, crown daisy, green red pepper, red pepper leaves, spinach, cabbage and sweet potato also inhibited growth of the cancer cells (p < 0.001~0.05).

Protective Effect of Yellow-Green Vegetable Juices on DNA Damage in Chinese Hamster Lung Cell Using Comet Assay (Comet Assay를 이용한 케일, 명일엽, 당근, 돌미나리 녹즙의 Chinese Hamster Lung 세포 DNA 손상 보호 효과)

  • The present study was attempted to investigate the antioxidant capacity of popular yellow-green vegetable juices (kale, Angelica keishei, carrot, small water dropwort) and to investigate the effect of vegetable juices on protecting oxidative damage to DNA in cultured Chinese hamster lung (CHL) cells. Antioxidant capacity was analyzed by TRAP assay (Total radical-trapping antioxidant potential). Cellular DNA dmamage was measured by SCGE (single-cell gel electrophoresis, also known as comet assay. Cells incubated in medium with PBS (negative control) or with various concentration of the freeze dried green juices (25, 50, 100, 250 $\mu\textrm{g}$/$m\ell$) resuspended in PBS were treated with $H_2O_2$ (200 ${\mu}{\textrm}{m}$) as an oxidative stimulus for 5 min at 4$^{\circ}C$. The physiological function of each vegetable juice on oxidative DNA damage was analyzed and expressed as tail moment (tail length X percentage migrated DNA in tail) . Kale juice had the highest TRAP value suggesting that kale has the highest antioxidant capacity followed by Angelica keishei, small water dropwort and carrot. Cells treated with $H_2O_2$ had extensive DNA damage compared with cells treated with PBS or pre-treated with vegetable juice extracts. All green juices inhibited $H_2O_2$-induced DNA damage with kale being the most effective juice among the tested juices. These results indicate that green juice supplementation to CHL cells followed by oxidative stimulus inhibited damage to cellular DNA, supporting a protective effect against oxidative damage induced by reactive oxygen species. Effect of Powder of Small Water Dropwort (Oenanthe javanica DC) and Brewer's Yeast(Saccharomyces cerevisiae) on the Liver Function and Serum Lipid Metabolism in Alcohol-Consumed Rats (돌미나리 가루와 맥주 효모가 알코올을 섭취한 흰쥐의 간 기능 및 혈청 지질대사에 미치는 영향)

  • This study investigated the effect of the powder of the small water dropwort(PSWD) and brewer's yeast(BY) on the liver function and serum lipid metabolism in alcohol-consuming rats. Male Sprague-Dawley rats were fed an AIN-93 diet(control), ethanol plus control(A), ethanol plus PSWD(AS), ethanol plus BY(AB), ethanol plus PSWD and BY(ABS) diet for seven weeks. The feed intake, body weight gain and feed efficiency ratio were the lowest and in the alcohol-consuming groups. The weight of the liver, kidney, spleen and epididymis of the anatomized rats showed positive correlation with the body weight gain. Compared with group A, the content of the lipid in the serum was significantly low in the AS and AB groups. In particular, it was the most effective in the AB group. The GOT, GPT and ALP level in the serum showed a significant difference in the alcohol-consuming groups. Compared with the A group, they showed a significantly low difference in the AS and AB groups. The total cholesterol and triglyceride levels in the liver were similar in all groups. The weight of the feces was significantly different but there was no significant difference in the content of total cholesterol and triglyceride levels in the AS and AB groups. These results suggest that BY and PSWD improve the liver function, and had an effect on reducing the lipid content of the serum and feces of alcohol-consumed rats. In particular, the effect of BY which contained protein, dietary fiber and vitamin B was higher than that of PSWD.

  • The antimutagenic effects of green-yellow vegetables toward aflatoxin B$_1$(AFB$_1$) and 4-nitroquinoline-1-ox-ide (4-NQO) using the Ames assay system with Salmonella typhimurium TA98 and TA100 were studied. Forty six to fifty percent of the methanol extracts of the vegetable samples inhibited the mutagenicity induced by AFB$_1$in TA98 and TA100. Perilla leaf, lettuce, broccoli, crown daisy, water dropwort, small water dropwort, red pepper, red pepper leaves, amaranth, spinach and radish root were significantly reduced the mutagenicity of AFB$_1$(p< 0.01). Whereas 25 out of 27 samples (93%) exhibited antimutagenicity toward a direct mutagen of 4-NQO (p< 0.01. 0.05). The samples which showed the strong antimutagenicity (>60%) were cabbage, kale, lettuce, broccoli, mustard leaf, green red pepper, green sweet pepper, spinach, amaranth, soybean sprout and immature pumpkin. The juices from the several samples also showed antimu-tagenic activity toward AFB$_1$. Cabbage, perilla leaf, small water dropwort and spinach reduced TAT100 revertants dose dependently in the range of 50-500$m\ell$/plate, however, cucumber and carrot showed little effect.

Antimutagenic Effects of the Edible Mountain Herb Juices Against Trp-P-1 and 2-AF (Trp-P-1과 2-AF에 대한 산채류 생즙의 항돌연변이 효과)

  • On the mutagenicity induced by 3-amino-l,4-dimethyl-5-H-pyrido[4,3-b]indol (Trp-P-1) and 2-aminotluorene (2-AF), the antimutagenic effects of edible muntain herb juices were examined by the Ames assay using Salmonella typhimurium TA98 and TA100. Juices prepared from aralia bud, small water dropwort, mugwort, roots of belltlower and sedum didn't have mutagenicity. Most of sample juices showed the antimutagenicity. Especially, juices prepared from aralia bud, small water dropwort and mugwort were found to possess strong antimutagenic effects. Sedum was moderatly effective and root of belltlower had little effect on mutagenicity caused by Trp-P1 and 2-AF. The experimental results with TA98 were similar to those with TA100 in the antimutagenicity test of edible mountain herb juices. In this study, antimutagenicity on Trp-P-1 was more effective than that on 2-AF.

Effects of Edible Herbs on the Growth of In Vitro Intestinal Microorganisms (산채류가 장내세균의 In Vitro 생육에 미치는 영향)

  • This study was aimed to screen edible herbs which control the composition of intestinal microflora. With in vitro experiments, we screened the water or ethanol extracts of about 60 edible herbs and wild plants in terms of the inhibition activity on the growth of the harmful Clostridium perfringens and growth promoting activity for the beneficial Bifidobacteria. The water extracts of mugwort and small water dropwort inhibited the growth of Cl.perfringens both in agar diffusion method and broth culture. On the other hand, the water extracts of petasites, mugwort, yellow day-lily and bitter cress have shown the promotion effect on the growth of Bifidobacterium longum. In the culture test using human feces as starter, the extracts of the above selected herbs increased the population of Bifidobacteria and Lactobacillus while they reduced the numbers of Cl.perfringens and E.coli.
