Effects of the Mixture of Vitamin C, Vitamin E, Pycnogenol and Evening Primrose Oil on the UV-Induced Pigmentation and Wrinkle Reductions in Human Skin (비타민과 불포화지방산, 그리고 피크노제놀을 함유한 복합제제의 주름 완화 및 자외선에 의한 색소침착 개선 효과)

  • In this study, the effects of a mixture consisting of vitamin E, vitamin C, pycnogenol and evening primrose oil (mixture LGNC-5) on ultraviolet light (UV) induced pigmentation and wrinkle reductions of normal healthy volunteers were studied. In a double-blind placebo-controlled study, each of 54 subjects took daily either 4 capsules of the mixture LGNC-5 (Group ABC; 282.5 mg/capsule) or placebo (Group Ganada). We irradiated 2.5 MED UV on the upper arms and measured the whitening effect by colorimeter-based L value. The level of wrinkle reduction was determined by image analysis using skin replica around the crow' feet, and the level of serum vitamin E was determined at baseline and 12 weeks. After 12-week oral administration, the treated group showed a significant reduction in skin pigmentation and wrinkles compared with the placebo group (p = 0.011 and p = 0.000005, respectively). Also, the level of serum vitamin E was significantly increased in the treated group after 12-week oral adminstration of the mixture compared with that in the placebo group (p = 0.0001). In conclusion, 12-week oral administration of LGNC-5 as a dietary supplement could be effective to reduce both UV induced pigmentation and skin wrinkle without side effects.

Verification of wrinkle improvement effect by animal experiment of suture for skin wrinkle improvement by applying CO2 gas and RF radio frequency (CO2 gas와 RF 고주파를 적용한 피부 주름 개선용 봉합사 동물 실험에 따른 주름 개선 효과 검증)

  • As the average life expectancy of human beings is extended in addition to the entry of the aging society, there is a tendency for the interest in the appearance of men and women in modern society to increase. The most external judgment of human aging is the wrinkles on the facial skin. People are undergoing various procedures to have clean, wrinkled, and resilient healthy skin. Many thread lifting procedures are being implemented because they tend to want simple and effective procedures during the procedure. In this study, in order to improve lifting effect in thread lifting, animal experiments were conducted to confirm the improvement of wrinkles by injecting RF high frequency and CO2 gas into existing PDO suture procedures. The experimental groups consisted of natural aging groups, PDO treatment groups, groups with RF high frequency in PDO procedures, groups with CO2 gas injected into PDO procedures, and groups with CO2 gas and RF injected simultaneously into PDO procedures. The individuals in the natural aging group had an average wrinkle depth of 0.408mm before the procedure, and the average wrinkle depth of the 10th week was 0.68mm. The depth of wrinkles in the PDO treatment group averaged 0.384mm before the procedure, and 0.348mm on the 10th week after the procedure. The average crease depth of pre-procedure objects injected with RF high frequency in PDO was 0.42mm, and the average crease depth for 10 weeks was 0.378mm. The average crease depth of the CO2 gas injected into the PDO was 0.4mm before the procedure, and the average crease depth was reduced to 0.332mm in the 10th week after the procedure. On average, the number of objects injected with CO2 gas and RF high frequency in the PDO procedure decreased to 0.412mm before and 0.338mm in the 10th week after the procedure. The procedure of injecting CO2 gas and RF into the PDO suture showed the highest reduction rate of 17.96%.

Effects of Nipa fruticans Wurmb Extract on Inhibition of UVB-Induced DNA Damage and MMP Expression (해죽순(Nipa fruticans Wurmb) 추출물의 UVB 유도 DNA 손상 및 MMP 발현 억제 효과)

  • The human skin is an organ that protects the body from physical and chemical factors. The skin is the largest and most massive of the body's organs and is composed of the epidermis, dermis, and subcutaneous tissue. Constant UV exposure to the skin can cause DNA damage, oxidation of proteins, and contribute to adult diseases. Nypa fruticans Wurmb (NF), rich in phytochemicals (polyphenols and flavonoids), has been traditionally used for treating respiratory and other diseases. This study investigated the effects of NF ethyl acetate fraction (ENF) on DNA damage healing and inhibition of wrinkle-related factors in UVB-stimulated Hs68 cells. Westernblotting was used to assess the expression of DNA damage-related proteins and wrinkle-related protein factors. In addition, the wound recovery capability of ENF was confirmed through wound-healing experiments. ENF significantly suppressed the expression of DNA damage-related proteins Phosphorylated H2AX (γ-H2AX), checkpoint kinase 2 (Chk2), protein53 (p53), and Phosphorylated protein53 (p-p53). Furthermore, ENF inhibited the expression of wrinkle-related proteins matrix metalloproteinase-1 (MMP-1), matrix metalloproteinase-3 (MMP-3), and matrix metalloproteinase-9 (MMP-9). High concentrations of ENF also enhanced wound healing in Hs68 cells. ENF is thought to have the potential to heal DNA damage by significantly suppressing the expression of γ-H2AX, Chk2, p53, and p-p53, as well as to inhibit wrinkle formation by suppressing the expression of MMP-1, MMP-3, and MMP-9. These results suggest that ENF can be used as a natural resource to suppress skin damage caused by UVB by regulating the γ-H2AX, Chk2, p53, and MMP pathways in Hs68 cells induced by UVB.

Protective effects of tyndallized Lactobacillus acidophilus IDCC 3302 against UVB-induced photodamage to epidermal keratinocytes cells

  • Photoaging is a consequence of chronic exposure to ultraviolet (UV) radiation and results in skin damage. In this study, whether tyndallizate of the probiotic bacterium Lactobacillus acidophilus IDCC 3302 (ACT 3302) can protect against UVB-induced photodamage to the skin was investigated. For this, HaCaT keratinocytes were used as a model for skin photoaging. HaCaT cells were treated with ACT 3302 prior to UVB exposure and skin hydration factors and matrix metalloproteinase (MMP)-1, MMP-2, and MMP-9 levels in the culture supernatant were evaluated by ELISA. The protective effects of ACT 3302 against UVB-induced oxidative stress in HaCaT cells was also assessed by measuring superoxide dismutase and catalase activity and detecting the expression of pro-inflammatory cytokine-encoding genes and mitogen-activated protein kinase (MAPK) signaling components by reverse transcription-quantitative polymerase chain reaction and western blotting, respectively. UVB exposure increased MMP expression and MAPK activation; these changes were attenuated by pretreatment with ACT 3302. Treatment with ACT 3302 prior to UVB exposure also attenuated inflammation. These results demonstrate that tyndallized ACT 3302 can mitigate photodamage to the skin induced by UVB radiation through the suppression of MMPs and could therefore be used clinically to prevent wrinkle formation.

Anti-wrinkle and Whitening Effects of Essential Oil from Abies koreana (Abies koreana 유래 정유의 항주름 및 미백 효과)

  • The essential oil from Abies koreana E.H. Wilson had been developed, however, its efficacy has not yet been studied especially in terms of skin care research. The aim of this study is to investigate the effects of Abies koreana extracts (AKE) on melanogenesis and wrinkle formation in B16F10 melanoma cells (B16F10) and human dermal fibroblast cell line (HDF). The essential oil was extracted by hydrodistillation method and purified by anhydrous sodium sulfate. At a concentration of $10^{-5}$-fold, viability in these cells had been defined by cytotoxicity assays. Anti-melanogenic effects on B16F10 were evaluated using tyrosinase inhibition assay, and real-time PCR for verifying gene expression of tyrosinase, tyrosinase related protein-1 and -2 (TRP-1 and -2). AKEs reduced about 5-fold of tyrosinase inhibitory activity compared to ${\alpha}$-melanocyte-stimulating hormone (${\alpha}$-MSH)-induced group and about 30% reduction compared to Arbutin induced group. The mRNA levels of three melanin-related factors were increased, separately. To investigate the effects of anti-wrinkle, procollagen type I c peptide synthesis assay (PIP) and Western blot were performed. At AKE-treated group, PIP was up-regulated and the expression of collagen type 1 and matrix metalloproteinase (MMP)-1 were improved. Furthermore, AKE presented anti-wrinkle effects by increasing UVB-inhibited collagen type 1 expression, and reducing UVB-induced MMP-1 production at $60mJ/cm^2$ of UVB radiation. Therefore, Abies koreana extracts has potentials as a safe and an effective skin ingredient for whitening and anti-wrinkle.

  • Phytoestrogens derived from plants and foods, which are diphenolic compounds with structural similarities to natural and synthetic estrogens, have been shown to estrogenic and antiestrogenic actions. Particularly, recent study revealed that phenolic compounds in safflower seed, such as serotonin derivatives, lignans and flavonoids, could be acted as phytoestrogens. Safflower (Carthamus tinctorius L.) seed extract (SID C.SE), therefore, are receiving a renewed interest as potential therapeutic source against skin wrinkles induced by estrogen deficiency. This study was conducted to investigate the anti-wrinkle effect of SID C.SE on normal human fibroblasts through the expression of type I procollagen and UVA-induced MMP-1 in vitro. The SID C.SE increased the type I procollagen expression, comparable to trans-retinol and reduced UVA-induced MMP-1 expression in a dose-dependent manner. The clinical study indicated that cream group treated with $0.1\%$ SID C.SE significantly reduced a skin wrinkles, as compared with a control (non-treated cream group) (p<0.05). These results suggest that the safflower seed extract may be useful as potential source of anti-wrinkle cosmetics.

The Evaluation of Anti-wrinkle Effects in Oriental Herb Extract (한방 원료 추출물의 주름개선 효과를 통한 화장품 원료로서의 가치 평가)

  • The human skin is constantly exposed to environmental irritants such as ultraviolet, smoke and chemicals. Free radicals and reactive oxygen species caused by them play critical roles in cellular damage. They not only injure the skin structure but also participate in the immensely complex inflammatory reaction. Anti-wrinkle effects of the Oriental herb extracts(OHE) were evaluated by the determination of anti-oxidation, collagenase inhibition and collagen synthesis in normal human fibroblast. OHE showed antioxidative activity as high as vitamin C, trolox and DL-penicillamine. Also OHE showed promotive effect on collagen synthesis and inhibitory effect on collagenase activity. These results demonstrated that OHE could be useful as an anti-wrinkle cosmetic ingredient.

Evaluation of Age-dependent Crow′s Feet in Korean Women (한국여성의 연령별 눈가 주름의 정량 분석)

  • In this study, we evaluated the crow's feet of 152 Korean women by using visual assessment established by our research group and mechanical assessment, Skin Visiometer SV 600. The volunteers were divided into 3 groups according to age; the group I of 30's was 43, the group II of 40's was 75, the group III of 50's was 34. The wrinkle score by visual assessment showed tendency to increase according to age and was statistically significant different among those three age groups (P 〈 0.05). But there was no significant difference between the left and the right of crow's feet. The 5 wrinkle parameters (RI, R2, R3, R4, R5) of mechanical assessment varied as age and the Rl, R4, and R5 showed statistically significant difference among three age groups (P 〈 0.05). However there was no significant difference between the left and the right of crow's feet. The volunteers were divided into 3 groups according to wrinkle score; (0-3 grade for group A, 4-6 grade for group B, 7-9 grade for group C). Only R4 and R5 showed significant difference between group A and group C (P 〈 0.05). It was suggested that visual scoring criteria applied in this study was related to R4 and R5 and was useful in investigating in fine wrinkles.

Antioxidation and Anti-photoaging Effects of White Taraxacum Coreanum Extract by Lactobacillus plantarum (Lactobacillus plantarum에 의한 흰 민들레 발효물의 항산화 및 광노화 억제 효과)

  • Lactobacillus plantarum SM4, a strain producing β-glucosidase, was isolated from kimchi and fermented with white Taraxacum coreanum to enhance the production of bioactive compounds. The total polyphenol content(TPC), total flavonoid content(TFC), radical scavenging activity(RSA), tyrosinase inhibitory activities(TIA), and collagenase inhibitory activities(CIA) were measured to evaluate the skin whitening and anti-wrinkle effects of the fermented product. The TPC of fermented white T. coreanum was 41.8±0.26 mg GAE/g DW, which was approximately two times higher than the hot-water extraction of 21.4±0.67 mg GAE/g DW. RSA, an indicator of antioxidant activity, was 65.6±4.7% in fermentation, which is four times higher than that of the hot-water extract. TAI and CAI, which are indicators of the whitening and anti-wrinkle effects, were 87.9±4.73% and 66.7±3.48%, respectively, which were 2.4 and 1.5 times higher than those of hot-water extraction. When comparing the UVA(320 nm) protection effects of fermented and hot-water extraction, the fermented white T. coreanum showed higher protection with an absorption rate of 64.7% and 15.2%, respectively. The white T. coreanum fermented product showed higher bioactive properties and improved skin whitening, anti-wrinkle, and UV protection effects through the production of β-glucosidase from L. plantarum SM4.

Research on the Possibility of Using Wrinkle-improving Functional Materials from Corn By-products (옥수수 부산물을 이용한 주름 개선 기능성 소재 활용 가능성 연구)

  • In this study, to explore the possibility of using discarded by-products of corn as a wrinkle-improving functional material, the active ingredient contained in each part including kernel, cob, husk, and silk of 10 varieties of chodang and waxy corn was identified and the content was analyzed to suggest the possibility of corn by-products as upcycling materials. As a result of evaluating the collagen synthesis efficacy of extracts by part, the cob in chodang and waxy corns showed significant activity at 100 ㎍/mL concentration of 176.5 and 140.0%, respectively. The LC-MS/MS qualitative analysis resulted in the identification of 18 components in the cob, of which N, N'-diferuloylputrescine (DFP) was selected as an active ingredient, and the quantitative analysis was conducted by variety and part. As a result, DFP was confirmed only in the cob and kernel, and it was most frequently detected in the cob of the Goldsugar of chodang corn (39.6 ㎍/g) and Daehakchal of waxy corn (38.4 ㎍/g). The cob is a by-product left over from grain removal, and is expected to create new added value as a material that promotes collagen synthesis. Through this study, we propose the possibility of using discarded cob as a wrinkle-improving functional material.
