Research Trends in the Development of Cosmetic Ingredients for Skin Barrier Improvement

  • In 2022, the domestic production performance of functional cosmetics in South Korea reached 4.6 trillion won, accounting for 33.85% of the total cosmetics production. The number of functional cosmetics reviewed increased by about 7.5% from the previous year, totaling 974 items. Especially with the increasing importance of the skin barrier function due to skin sensitivity caused by various environmental pollutants, domestic cosmetic companies are showing interest in the development of new ingredients and products related to this area. This study aims to analyze academic research trends related to in vitro experiments for the development of cosmetics improving the skin barrier, to provide practical information for the cosmetic industry. The findings are as follows: Academic research mainly focused on the efficacy of natural ingredients in improving the skin barrier, but there is a significant lack of quantitative accumulation of research. For the development of skin barrier-improving cosmetic ingredients, efficacy evaluation indicators were set, including hyaluronic acid production, expression of filaggrin gene, loricrin, formation of cornified envelope (CE), and expression of ceramide synthesis enzyme genes. Moreover, effective cosmetic ingredients for improving the skin barrier included lemongrass and perilla leaf extracts, flavonoids, Lactococcus lactis subsp. lactis, Exosomelike Nanovesicles derived from apple callus, Eleutherococcus sessiliflorus, Acanthopanax sessiliflorus, Eleutherococcus gracilistylus, Acer okamotoanum extracts, Aloe vera adventitious root extract, ethanol extract of Aruncus dioicus, and organic solvent fraction of Dracocephalum argunense.

황금추출물의 지방장벽 생성을 통한 염증완화 효과 (Effect of Anti-Inflammation through Creation of Skin Fat Barrier on Scutellaria baicalensis extract)

  • Objectives The purpose of this study is to confirm the effect of alleviating inflammation through creating skin fat barrier from Scutellaria baicalensis extract. Methods Four-week-old Balb/C mice were divided into four groups: control group (Ctrl), lipid barrier eliminated group (LBEG), dexamethasone (DM) treated group after lipid barrier elimination (DMTG), and Scutellaria baicalensis (SB) treated group after lipid barrier elimination (SBTG). Scutellaria baicalensis extract were administered for 5 days after removal of the fat barrier. Changes in skin condition, improvement of the fat barrier, and relief of inflammation were observed in each group. Results Compared to LBEG and DMTG, pathological skin damage and tissue changes were less in SBTG, and transepidermal water loss (TEWL) and pH were also significantly reduced. Filaggrin was also significantly increased in SBTG. KLK7, PAR-2, and TSLP in SBTG also showed significant reduction compared to the LBEG and DMTG. Conclusions Scutellaria baicalensis extract restores skin barrier and relieves inflammation through the creation of skin fat barrier. This means that the Scutellaria baicalensis extract can regulate Th2 differentiation through the creation of the epithelial fat barrier.

황련감초 추출물의 상피지방장벽 생성을 통한 Th2 분화 조절 (Effect of Th2 Differentiation Control through Formation of Skin Fat Barrier on Coptidis Rhizoma & Glycyrrhiza Uralensis extract)

  • Objectives This study is conducted to evaluate Th2 skewed condition control through skin fat barrier formation from the treatment using Coptidis Rhizoma and Glycyrrhiza uralensis extract. Methods The 6-week-old NC/Nga mice were divided into 4 groups: Control group (Ctrl), lipid barrier eliminate treatment group (LBET), Coptidis Rhizoma and Glycyrrhiza uralensis feeding treatment after lipid barrier elimination group (CGFT), dexamethasone feeding treatment after lipid barrier elimination group (DxFT). After 3 days, differences in skin condition, improvement of skin fat barrier, and control of Th2 skewed condition of each group were observed. Results Pathologic skin damage and tissue changes were less in the CGFT group than in the LBET and DxFT groups, and Transepidermal water loss (TEWL) and pH were also significantly decreased (p < 0.05). The filaggrin intensity and positive response also increased significantly in the CGFT group (p < 0.05). Kallikrein-related peptidase (KLK) 7, Protease activated receptor (PAR)-2, Thymic stromal lymphopoietin (TSLP), Interleukin (IL)-4, and the products of the Th2 differentiation process also showed a significant decrease compared to the LBET and DxFT groups (all p < 0.05). Conclusions The Coptidis Rhizoma and Glycyrrhiza uralensis extract causes skin barrier recovery and function recovery through the formation of skin fat barrier. This leads to the conclusion that Coptidis Rhizoma and Glycyrrhiza uralensis extract can control Th2 differentiation through the formation of skin fat barrier.

Effects of Petrolatum and Glycerin on Acetone Damaged Canine Skin Barrier

  • The purpose of this study is to investigate whether the effects of topically applied petrolatum and glycerin on the barrier repair of acetone-induced skin damage in 6 beagle dogs. To confirm the effects of petrolatum and glycerin on acetone disrupted skin models, we performed to evaluate the characteristics of transepidermal water loss and SC hydration and scanning electron microscopic observations. TEWL and SC hydration measurements were carried out 3, 6, 12, 24, 48h after applying petrolatum and glycerin during recovery from acute disruption. Our results showed that there were some different effects between petrolatum and glycerin on the acetone damaged skin such as barrier function repair process and SC hydration status. The results indicate that the significant improvement could be observed in glycerin apply more than petrolatum after acetone damages, and further study will be required.

Improvement of skin barrier function using lipid mixture

  • Dry skin is caused mainly by the perturbation of stratum corneum lipids which affected by ageing, change of season, excess use of surfactant and the effect of disease like atopic dermatitis and psoriasis. Intercellular lipid structures in stratum corneum are responsible for the barrier function of mammalian skin. The major lipd classes that can be extracted from stratum corneum are ceramides, cholesterol and fatty acid, which make up approximately 50, 25, 10 percent of the stratum corneum lipid mass, respectively. Small amount of cholesterol sulfate, phospholipids, glycosylceramide and cholesterol esters are also present. Recent studies have shown that application of one or two these lipids to the perturbed skin delays barrier recovery; only equimolar mixtures allow normal recovery. We observed that barrier recovery rate was improved in hairless mouse by topical application of single neutral lipids (ceramide, free fatty acid, cholesterol) and lipid mixtures. Whereas the application of single lipid didn’t allows a significant enhancement comparing with normal barrier repair, the equimolar mixtures of 3 components(including synthetic pseudoceramide PC104) improved barrier repair, as assessed by the transepidermal water loss. At clinical study to the volunteers aged over sixty, skin dryness recuperated by the increase of moisture(capacitance) and the reduction of scaling. Utilization of physiologic lipid mixture containing natural ceramides or synthetic pseudoceramide could lead to new forms of topical therapy for the dryness and dermatoses(e.g., psoriasis, atopic dermatitis and irritant dermatitis).

루꼴라(Eruca sativa) 추출물의 항균활성과 피부장벽 개선 효과 (Antimicrobial activities and skin barrier improvement effect of Eruca sativa extract)

  • 본 연구에서 루꼴라(Eruca sativa)에 대한 다양한 생리활성을 조사하여 기능성소재 응용가능성을 검토하였다. 루꼴라 추출물은 B. subtilis, E. coli, C. albicans에 대한 항균활성이 매우 우수하게 나타났으며 특히 그람양성세균 B. subtilis에 대한 항균활성이 높았다. 또한, 인볼루크린 발현에 대한 면역조직화학 분석을 인공피부를 이용하여 분석한 결과 루꼴라 추출물을 처리(50 mg/L)한 경우에 0.1% DMSO로 처리한 대조군에 비하여 인볼루크린 발현이 뚜렷하게 증가하였으며 동일농도로 처리된 합성물질 WY14643과 비교하였을 때에도 비슷한 발현양상을 보여 주었다. 이와 같은 결과로 미루어 볼 때 루꼴라 추출물은 피부염 개선 및 피부장벽 기능을 향상할 수 있는 천연물 기반의 소재로 활용하기 위한 매우 효과적인 재료가 될 수 있다고 사료된다.

In Vitro Anti-Inflammatory and Skin Protective Effects of Codium fragile Extract on Macrophages and Human Keratinocytes in Atopic Dermatitis

  • Codium fragile has been traditionally used in oriental medicine to treat enterobiasis, dropsy, and dysuria, and it has been shown to possess many biological properties. Atopic dermatitis (AD) is one of the types of skin inflammation and barrier disruption, which leads to chronic inflammatory skin diseases. In the current investigation, the protective effects of C. fragile extract (CFE) on anti-inflammation and skin barrier improvement were investigated. In LPS-stimulated RAW 264.7 cells, nitric oxide generation and the expression levels of interleukin (IL)-1β, IL-4, IL-6, iNOS, COX-2, and tumor necrosis factor-alpha (TNF)-α were reduced by CFE. CFE also inhibited the phosphorylation of NF-κB-p65, ERK, p-38, and JNK. Additionally, CFE showed inhibitory activity on TSLP and IL-4 expression in HaCaT cells stimulated with TNF-α/interferon- gamma (IFN-γ). Enhanced expression of factors related to skin barrier function, FLG, IVL, and LOR, was confirmed. These findings implied that CFE may be used as a therapeutic agent against AD due to its skin barrier-strengthening and anti-inflammatory activities, which are derived from natural marine products.

Experimental Studies on the Skin Barrier Improvement and Anti-inflammatory Activity based on a Bibliometric Network Map

  • Atopic dermatitis is a chronic inflammatory skin diseases caused by skin barrier dysfunction. Allium victoralis var. Platyphyllum (AVP) is a perennial plant used as vegetable and herbal medicine. The purpose of this study was to suggest that AVP is a new cosmetic material by examining the effects of AVP on the skin barrier and inflammatory response. A bibliometric network analysis was performed through keyword co-occurrence analysis by extracting author keyword from 69 articles retrieved from SCOPUS. We noted the anti-inflammatory activity shown by the results of clustering and mapping from network visualization analysis using VOSviewer software tool. HPLC-UV analysis showed that AVP contains 0.12 ± 0.02 mg/g of chlorogenic acid and 0.10 ± 0.01 mg/g of gallic acid. AVP at 100 ㎍/mL was shown to increase the mRNA levels of filaggrin and involucrin related to skin barrier function by 1.50-fold and 1.43-fold, respectively. In the scratch assay, AVP at concentrations of 100 ㎍/mL and 200 ㎍/mL significantly increased the cell migration rate and narrowed the scratch area. In addition, AVP suppressed the increase of inflammation-related factors COX-2 and NO and decreased the release of β-hexosaminidase. This study suggests that AVP can be developed as a functional cosmetic material for atopy management through skin barrier protection effects, anti-inflammatory and anti-itch effects.

Ginsenoside Rg3 에 의한 3D 피부 모델의 장벽 기능 강화 및 지질 패킹 구조 개선에 관한 연구 (A Study on the Enhancement of Barrier Function and Improvement of Lipid Packing Structure in a 3D Skin Model by Ginsenoside Rg3)

  • 피부의 장벽 구조는 표피의 각질 형성세포의 분화과정에 의해서 생성된다. 이 구조는 케라틴 단백질로 구성되는 각질세포와 그 사이를 채우고 있는 세포간 지질로 구성된다. 이때 표피의 기저층의 세포의 막을 이루던 인지질 등의 성분은 분해되어 없어지고, 세라마이드 등이 성분이 신규로 합성되어 각질층의 세포간 지질을 구성한다. 본 연구에서는 피부 장벽의 세포간 지질 구조의 패킹과 장벽기능에 진세노사이드 Rg3성분이 미치는 영향을 확인하였다. 이를 위해 3D피부 세포의 분화과정에 Rg3성분을 처리하였다. 3D피부를 대상으로 FT-IR 및 TEWL를 분석한 결과, 각질 세포간 지질의 orthorhombic패킹이 강화되고 장벽기능이 강화되는 것을 확인하였다. 또한 HaCaT세포에 Rg3를 처리한 경우, 긴 체인 길이의 지질을 합성하는 EVOL1 및 EVOL4의 발현 증가와 짧은 길이의 세라마이드의 합성을 당하는 CERS6의 감소 그리고 피토스핑고신을 사용하는 세라마이드를 분해시키는 ACER6의 증가를 검출하였다. 이를 통해 Rg3가 표피 분화 과정 중 지질의 합성에 영향을 주어 장벽 기능 변화를 가져올 가능성을 제시하였다.

용아초(龍芽草) 유기용매 분획물의 항산화 및 피부 장벽 개선 효과 (Anti-oxidant Activity and Skin Barrier Function Measured in Different Solvent Fractions Obtained from Agrimoniae Herba)

  • Objectives : Agrimoniae Herba is a herbal medicine widely distributed in Asia and contains flavonoids including catechin, quercitrin, rutin, hyperoside, and quercetin. This study aimed to investigate the anti-oxidant activity and skin barrier function of different solvent fractions (Hexane; methylene chloride, MC; ethyl acetate, EA; n-butanol, Bu; Water) obtained from Agrimoniae Herba. Methods : Anti-oxidant activity of different solvent fractions obtained from Agrimoniae Herba was investigated through total polyphenol contents, total flavonoid contents, 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging activity, and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging activity measurements. Then, filament aggregating protein (Filaggrin), Type I collagen, ceramide synthase (CERS) 3, and CERS4 were analyzed to evaluate the skin barrier strengthening effect of different solvent fractions obtained from Agrimoniae Herba on UVB-stimulated HaCaT cells. Results : As a result of measuring total polyphenol contents, total flavonoid contents, DPPH free radical scavenging activity, and ABTS radical scavenging activity, antioxidant activity was found to be excellent in the order of EA > Bu > MC > Hexane > Water. As a result of measuring mRNA gene expression of Type I collagen, Filaggrin, CERS3, and CERS4 after UVB-stimulated was applied to HaCaT cells treated with different solvent fractions obtained from Agrimoniae Herba, it was found to increase significantly in the Bu-treated group. Conclusion : Our findings show that the Bu sample obtained from Agrimoniae Herba has excellent anti-oxidant ability, which increases Type I collagen, Filaggrin, and ceramide synthetase in UVB-stimulated HaCaT cells to control the skin barrier improvement effect.
